An agent-based approach to dynamically represent the pharmacokinetic properties of baicalein / CUHK electronic theses & dissertations collection

Purpose: To develop an agent-based, discrete-event, synthetic model that integrates the existing knowledge about intestinal absorption and disposition of baicalein (Ba) and dynamically represents the pharmacokinetic properties of Ba. To validate the model by matching simulated observations to bi-directional Caco-2 transport profiles of Ba. / Methods: A 3D multi-agent system extending the previous 2D in silico analogue of Caco-2 cell monolayer was used to study the pharmacokinetic properties of Ba. The model specification was based on previous study findings. Our model consisted of three 3D spaces and two 2D membranes: apical space (S1), intracellular space (S2), basolateral space (S3), apical membrane (M1), and basolateral membrane (M2). Validated enzyme components (UGTs and SULTs) and binder components (BINDERs) were placed in S2. Validated efflux transporter components (BCRPs, MRP2s, MRP1s and MRP3s) were placed at M1 and M2 respectively. Initially, Solutes (BA) were loaded to either S1 or S3. From there, they penetrated into S2 according to a validated, passive transport algorithm. Within S2, BA could be transformed into Baicalein-Glucuronide (BG) by UGTs or to Baicalein-O-Sulfate (BS) by SULTs. These metabolites were pumped out to either S1 or S3 by the active transport of BCRPs, MRP2s, MRP1s and MRP3s. Simulated results were then compared to corresponding wet-lab data to assess similarity based on the pre-specified similarity criteria. An iterative refinement protocol combined with Monte-Carlo simulation was employed to parameterize the model. Finally, the in silico parameters had also been mapped to the classical pharmacokinetic parameters. / Results: The simulated results captured the preset qualitative and quantitative features of the wet-lab observations. The feasible parameter set showed that substrate inhibition happened in both conjugation pathway of Ba. The simulation results suggested that sulfation pathway was dominated at low concentrations and that SULT was more inclined to substrate inhibition than UGT. All these findings were consistent with the previous outcomes from a catenary model. In addition, a micro-hypothesis that BS’s apical efflux transporter could be inhibited by Ba at high loading concentration was implemented to address the changing preference of the distribution of BS. / Conclusion: The mechanisms represented by our model are plausible. Our novel modeling approach could dynamically represent the pharmacokinetics of bi-directional transport of Ba in Caco-2 system. A study of infraspecific flavonoid variation of Cannabis sativa L. (Cannabaceae)

The genus Cannabis has been treated taxonomically by several authors, but two main hypotheses predominate. Small and Cronquist (1976) concluded that the genus is monotypic and contains only Cannabis sativa L. Subspecies were named according to the economic purpose of the plants, i.e. fibre or drug, and varieties were assigned according to habitat (wild or cultivated). Schultes (1974) believed the genus is polytypic, with Cannabis sativa L. representing the fibre plants, C. indica Lam, the drug plants, and C. ruderalis Janis. the wild and weedy plants. A study of flavonoid variation was undertaken to determine which of the taxonomic schemes would be supported by this variation. Individual plants were examined by thin-layer chromatography and presence/absence data for nine flavonoids were analyzed by three types of computer programs. The infraspecific variation of flavonoids in the genus Cannabis supports the monotypic theory of Small and Cronquist (1976). No consistent, distinguishable groups were noticeable in the data. Wild taxa were more closely related to their cultivated progenitors than they were to each other. Qualitative Analysis of Sequence Specific Binding of Flavones to DNA Using Restriction Endonuclease Activity Assays

Flavones, found in nature as secondary plant metabolites, have shown efficacy as anti-cancer agents. We have examined the binding of two flavones, 5,7-dihydroxy-3,6,8-trimethoxy-2-phenyl-4H-chromen-4-one (5,7-dihydroxy-3,6,8- trimethoxy flavone; FlavA) and 3,5-dihydroxy-6,7,8-trimethoxy-2-phenyl-4H- chromen-4-one (3,5-dihydroxy-6,7,8-trimethoxy flavone; FlavB), to phiX174 RF DNA using restriction enzyme activity assays employing the restriction enzymes Alw44, AvaII, BssHII, DraI, MluI, NarI, NciI, NruI, PstI, and XhoI. These enzymes possess differing target and flanking sequences allowing for observation of sequence specificity analysis. Using restriction enzymes that cleave once with a mixture of supercoiled and relaxed DNA substrates provides for observation of topological effects on binding. FlavA and FlavB show differing sequence specificities in their respective binding to phiX. For example, with relaxed DNA, FlavA shows inhibition of cleavage with DraI (reaction site 5′TTTAAA) but not BssHII (5′GCGCGC) while FlavB shows the opposite results. Evidence for tolological specificity is also observed, Molecular modeling and conformational analysis of the flavones suggests that the phenyl ring of FlavB is coplanar with the flavonoid ring while the phenyl ring of FlavA is at an angle relative to the flavonoid ring. This may account for aspects of the observed sequence and topological specificities in the effects on restriction enzyme activity.

Ces travaux de thèse concernent l’étude phytochimique de cinq plantes médicinales dont les espèces sélectionnées sont endémiques du Venezuela : Palicourea demissa Standl. et Hydrocotyle umbellata L., mais aussi Bauhinia cumanensis H.B.K, Guazuma ulmifolia Lam. et Urena sinuata L. Vingt-deux composés ont été purifiés et identifiés à partir des différents extraits étudiés ; il s’agit notamment de terpènes, de flavones et de saponines. Six de ces substances naturelles, parmi lesquelles figurent plusieurs saponines de types spirostane et ergostane, sont décrites pour la première fois. / This work is dedicated to the phytochemical study of five medicinal plants whose selected species are endemic of Venezuela: Palicourea demissa Standl., and Hydrocotyle umbellata L., but also Bauhinia cumanensis H.B.K, Guazuma ulmifolia Lam., and Urena sinuata L. Twenty-two compounds were purified and identified from the various studied extracts; they are in particular terpenes, flavones and saponins. Six of these natural products, among which appear several spirostane- and ergostane-types saponins, are described for the first time.
