Modelling nematode infections in sheep and parasite control strategies

Gastrointestinal parasitism in grazing lambs adversely affects animal performance and welfare, causing significant production losses for the sheep industry. Control of gastrointestinal parasitism using chemotherapeutic treatment is under threat due to the emergence of anthelmintic resistance, thus stimulating research into alternative control strategies. Whilst investigating control strategies experimentally can be costly and time consuming, using a mathematical modelling approach can reduce such constraints. A previously developed model which describes the impact of host nutrition, genotype and gastrointestinal parasitism in a growing lamb, provided an appropriate starting point to explore control strategies and their impact on host-parasite interactions. Two contrasting mechanisms have previously been proposed to account for the occurrence of anorexia during parasitism. These were reductions in either intrinsic growth rate or relative food intake. Thus, the existing individual lamb model was modified to evaluate these mechanisms by exploring the relationship between anorexia and food composition (Chapter 2). For foods that did not constrain food intake, published data was found to be consistent with the predictions that arose from anorexia being modelled as a reduction in relative food intake. Reported genetic parameter estimates for resistance and performance traits appear to vary under differing production environments. In order to explore the impact of epidemiological effects and anthelmintic input on genetic parameter estimates the model was extended to simulate a population of lambs in a grazing scenario (Chapter 3). Whilst estimates of heritabilities and genetic correlations for drenched lambs remained constant, for lambs given no anthelmintic treatment, the heritability of empty body weight (EBW) reduced and the genetic correlation between faecal egg count (FEC) and EBW became increasingly negative with increasing exposure to infective larvae. Thus differences in anthelmintic input and pasture larval contamination (PC) may provide plausible causes for the variation in genetic parameter estimates previously reported. To investigate the interactions between host resistance and epidemiology (Chapter 4) a population of 10,000 lambs were simulated and FEC predictions used to assign the 1,000 lambs with the highest and lowest predicted FEC to ‘susceptible’ (S) and ‘resistant’ (R) groups, respectively. R and S groups were then simulated to graze separate pastures over 3 grazing seasons. The average FEC and PC predictions of these groups diverged during the first 2 grazing seasons and stabilised during the third, such that the difference in FEC predictions between R and S groups were double those predicted when grazed with the population. This was found to be consistent with experimental data. Further, anthelmintic treatment and grazing strategies were predicted to have no impact on the EBW of resistant lambs, suggesting that control strategies should be targeted towards susceptible animals. Targeted selective anthelmintic treatment (TST) has been proposed to reduce risks of anthelmintic resistance with minimal impacts on performance. To describe the short- and long-term impacts of TST and drenching frequency on sheep production and the emergence of anthelmintic resistance, the model was extended to include a description of anthelmintic resistance genotypes within the nematode population (Chapter 5). Reducing the proportion of treated animals was predicted to increase the duration of anthelmintic efficacy, whilst reducing the drenching frequency increased the long-term benefits of anthelmintic on sheep production. Various determinant criteria for use in TST regimes were compared (Chapter 5) including performance traits such as live weight and growth rate, and parasitological traits such as FEC. Using FEC as the TST criterion was predicted to allow the greatest reduction in the number of anthelmintic treatments administered whilst maintaining the highest average EBW, whilst live weight and growth rate were predicted to give little to no improvement in comparison to selecting animals at random for TST. Using estimated breeding values (EBVs) for FEC as the determinant criterion for TST regimes was compared to using measured FEC (Chapter 6). The EBV for true FEC across the entire growth period, akin to perfect genomic selection, was predicted to be a better criterion than measured time-specific FEC (including a sampling error) for a TST regime. EBVs calculated using measured time-specific FEC showed little benefit compared to measured FEC. The information gained from these simulation studies increases our understanding of control strategies and their impact on host-parasite interactions under various scenarios that may not have been possible using experimental methods. It is important to remember that the aim of alternative or complimentary control strategies is to maintain the sustainability of sheep production systems, and as such the production gain of any control strategy needs to be weighed against the financial, labour and time costs involved in implementation.

The studies were conducted in support of Industry and Government initiatives to reinvigorate Falklands’ agriculture. The current industry is based on broad diameter wools harvested from sheep grazing poor quality native pastures, and sheep suffer low growth rates, poor productivity and high mortality, and farmers rely on small flocks to produce a single commodity for their total income. The dramatic collapse of wool markets in the 1980’s crippled the Falklands’ wool industry and farmers have relied heavily on subsidies for more than 12 years. The studies in this thesis were initiated to support the efforts to reinvigorate Falklands’ agriculture, and they examined basic aspects of the nutrition provided to sheep by native pastures and evaluated practical methods to improve the survival of sheep, their subsequent rate of growth and productive value to the economy. Five studies examined specific hypotheses posed at the outset of the research programme and they were broadly supported by the data presented in the thesis: · A wether trial was used to benchmark the productivity of wool sheep in the Islands, and the trial demonstrated that substantial genetic variation exists within the national flock that may be exploited in the future to improve the productivity of the wool industry, · Microhistological techniques were used in conjunction with n-alkane marker technology to investigate the diets consumed by sheep grazing native pastures. The diet studies concluded that the low quality of native pasture in the Falklands during autumn, winter and spring restricts growth and productivity of young sheep, · Studies with weaner sheep treated with controlled release anthelmintics established that infection with gastrointestinal nematodes exacerbates poor nutrition of young sheep consuming native pastures, · A short-term metabolism study demonstrated that native pasture hay fortified with molasses and urea, or winter forage crops may be used as supplements to enhance sheep productivity, and · Forage crops were grown and evaluated under Falklands’ conditions, and when lambs were fed the resulting highly productive and nutritious forages, the study demonstrated that lamb growth can be increased significantly, and lamb carcasses offer local farmers the potential to diversify farm income by using existing Falklands’-bred lambs to develop an export meat industry The data presented in this thesis demonstrate that the potential to improve the productive and financial position of Falklands’ farmers is substantial. Selon les nouvelles orientations agroécologiques qui s’imposent largement, il convient d’élaborer des systèmes d’élevage adaptés aux contraintes environnementales, efficients et pourvoyeurs de services écosystémiques bénéfiques pour l’environnement et la société. La question se pose singulièrement dans nos territoires antillais et ceci dans le cadre d’une demande pressante en produits carnés de qualité et à forte valeur patrimoniale comme l’est la viande caprine en Guadeloupe. Les petits ruminants, principalement élevés au pâturage, sont exposés aux infestations par les nématodes gastro-intestinaux (NGI) qui constituent une cause majeure de morbidité et de mortalité. Le déploiement de techniques adaptées et applicables sur le terrain s’avère nécessaire. Aussi des méthodes de gestion intégrée du parasitisme sont préconisées parmi lesquelles sont étudiées les stratégies d’alimentation des animaux. Il convient d’améliorer la capacité de l’animal hôte à tolérer les effets négatifs (résilience) ou à contrôler les infestations par les NGI (résistance) par la complémentation alimentaire. L’objectif de cette thèse était de définir des stratégies de complémentations des chevreaux en croissance, stade physiologique le plus touché par les NGI et dont les performances zootechniques conditionnent directement le revenu des producteurs (la production de viande).L’apport de la complémentation a été combinée à une série de recommandations de la gestion intégrée du parasitisme (voie génétique, méthode Famacha ®) et singulièrement à une stratégie de gestion du pâturage mixte avec des génisses. Ont été obtenus, un accroissement des résultats zootechniques (croissance et carcasse), de la note d’état corporel ainsi qu’une amélioration de la réponse physiologique (taux d’hématocrite). Une diminution du nombre de traitements anthelmintiques est observée aussi. Cet essai a abordé l’opérationnalité de la complémentation pour un éleveur. D’une façon générale, ce travail a ouvert aussi des questionnements scientifiques sur l’interaction nutrition*parasitisme avec un focus sur la nutrition azotée à déconnecter de l’alimentation énergétique. / New agroecological guidelines are needed around the world to develop livestock systems that are adapted to the environmental, efficient and environmentally and socially beneficial ecosystem services requirements. The question arises singularly in our Caribbean territories and this in the context of an urgent demand in meat products of quality and ofhigh patrimonial value as is the goat meat in Guadeloupe. Small ruminants, mainly grazed, are exposed to gastrointestinal nematode (NGI) infestations, which are a major cause of morbidity and mortality. The deployment of adapted and applicable techniques in the field is necessary. Therefore, methods of integrated management of parasitism are recommended among which are studied the strategies of feeding of the animals. The ability of the host animal to tolerate adverse effects (resilience) or to control infestations by NGI (resistance) should be enhanced by food supplementation. The objective of this thesis was to define supplementation strategies for growing kidneys, the physiological stage most affected by NGI and whose zootechnical performances directly affect the incarne of producers (meat production).Discussions focus on the development of targeted complementing strategies and attempt to answer questions: how to complement? (ln energy, nitrogen or both); When to complement? (Delay effects or effects during an established infestation). lt was also a question oftesting the life-size recommendations and on several variables (physio­pathological responses and zootechnical responses including carcass characteristics). In this sense this work has presented a certain originality. There was no statistically significant effect on carcass quality, however, in carcass­infected animals the carcasses were more degraded, especially with a low-protein diet. The complementation contribution was combined with a series of recommendations of integrated management of parasitism (genetic pathway, famacha method) and particularly a management strategy of mixed grazing with heifers.
