Efeitos dos processos de preservação sobre os fungos Arthrobotrys robusta e Monacrosporium thaumasium predadores de nematóides / Effects of long-term storage of Arthrobotrys robusta and Monacrosporium thaumasium nematode trapping fungi

Os fungos nematófagos produzem estruturas em forma de armadilha, responsáveis pela captura e morte dos estágios pré-parasitários dos nematóides. A manutenção contínua da atividade predatória dos isolados fúngicos é um dos pré-requisitos básicos para o sucesso desta forma de controle. Neste trabalho foi verificado o comportamento dos isolados I31 de Arthrobotrys robusta e NF34a de Monacrosporium thaumasium, submetidos a manutenção em temperatura de 4 o C, criopreservados com e sem adição de DMSO e glicerol e mantidos em sílica-gel. Os resultados demonstraram que os isolados NF34a e I31 não apresentaram variação no crescimento radial, quando armazenados em temperatura de 4 o C e congelados com ou sem adição de crioprotetores. Não houve diferença na produção massal de micélio seco dos isolados quando armazenados a temperatura de 4 o C e congelados com adição de crioprotetores. O armazenamento em sílica-gel e o congelamento sem crioproteção pareceram interferir negativamente na capacidade dos fungos em produzir massa micelial. O armazenamento de NF34a em temperatura de 4 o C e o congelamento com adição de DMSO foram os tratamentos em que se obteve maior redução das larvas infectantes in vitro. Não foi verificada diferença na capacidade predatória in vitro do isolado I31 quando mantido em temperatura de 4 o C ou quando congelado com adição de DMSO ou glicerol. A manutenção dos isolados em sílica-gel e o congelamento sem crioproteção interferiram negativamente na capacidade predatória, porém estes ainda foram capazes de reduzir a população de larvas infectantes em relação ao grupo controle. No teste de passagem dos isolados pelo trato gastrintestinal de bovinos, a administração de 20 g de micélio aos animais foi suficiente para recuperação de material fúngico nas fezes e pico de redução no número de larvas infectantes nas fezes após 24 horas da administração das amostras. O isolado NF34a foi responsável pelos maiores índices de redução das populações de larvas de helmintos após a passagem pelo trato gastrintestinal dos animais e o armazenamento em 4 o C foi o tratamento em que se observou maior taxa de redução. A criopreservação parece ser um método eficiente para manutenção dos isolados, porém neste trabalho foi evidenciada diminuição da capacidade predatória do isolado NF34a. A manutenção dos isolado em sílica-gel deve ser utilizado para manutenção das amostras por períodos prolongados, quando não houver disponibilidade de nitrogênio líquido. / Biological control of helminths parasites of cattle is an alternative to the use of anthelmintics, and its main goal is to reduce the amount of infective larvae in the pasture. The nematophagous fungi produce trap-shaped structures, which are responsible for capturing and destroying the pre-parasite stages of nematodes. Continuous maintenance of the fungi isolates’ predatory activity is one of the basic prerequisites for the success of this form of control. This research analyzed the behaviour of I31 isolates of Arthrobotrys robusta and NF34a isolates of Monacrosporium thaumasium, submitted to maintenance at a temperature of 4 o C, cryopreserved with and without DMSO and glicerol, and kept in silica gel. Results showed that the NF34a and I31 isolates did not present variation in radial growth when stored at a temperature of 4 o C and frozen with or without adding cryoprotectants. There was no difference in production of dry biomass by the isolates when stored at a temperature of 4 o C and frozen with added cryoprotectants. Silica gel storage and freezing without cryoprotection seemed to interfere negatively with the fungi’s capacity to produce biomass. Storing NF34a at a temperature of 4 o C and freezing it with DMSO were the treatments in which the largest in vitro reduction of infective larvae was obtained. There was no difference in terms of in vitro predatory capacity of the I31 when stored at a temperature of 4 o C or when frozen with DMSO or glicerol. Storage of the isolates in silica gel and freezing without cryoprotection interfered negatively in their predatory capacity, but they were still able to reduce the population of infective larvae in relation to the control. When the passage of isolates through the gastrointestinal tract of bovines was tested, the administration of 20 g of mycelium was enough to recover fungi material in feces and for a peak reduction of the number of infective larvae in feces 24 hours after the administration of the samples. The NF34a isolate was responsible for the largest reduction rates in helminth larvae populations following passage through the gastrointestinal tract of the animals, and storage at 4 o C was the treatment in which the largest reduction rate was observed. Cryopreservation seems to be an effective method for keeping the isolates, although, in this research, a reduction of the NF34a’s predatory capacity became evident. Storage of the isolates in silica gel should be chosen when keeping the samples for long periods of time, whenever liquid nitrogen is not available. / Dissertação importada do Alexandria
