Dessa forma, a utilização de fungos nematófagos associados, no controle biológico, pode ser uma alternativa eficaz e vantajosa para melhorar os resultados de tratamentos com fungos isoladamente e potencializar o controle dos nematoides que parasitam bovinos. O objetivo do presente trabalho foi avaliar a compatibilidade entre os fungos nematófagos do grupo dos predadores Arthrobotrys robusta (isolado I31) e Duddingtonia flagrans (isolado AC001) em laboratório e a ação conjunta destes na redução ambiental de formas infectantes de nematoides parasitos gastrintestinais de bovinos jovens Holandês-Zebu. Primeiramente, a presença de compatibilidade entre os fungos A. robusta e D. flagrans in vitro foi verificada por testes de confronto direto, de antibiose e de compostos voláteis. Esses testes permitem identificar a competição por espaço entre os organismos e a produção de antibióticos e de compostos voláteis por algum desses fungos. No teste de confrontação direta, A. robusta colonizou aproximadamente 2/3 da placa, o que sugere competição por espaço e, portanto, antagonismo. No teste de antibiose, nenhum fungo produziu antibióticos sobre o meio de cultura, o que resultou na ausência de halo de inibição da cultura subsequentemente semeada na mesma placa. No teste de compostos voláteis, o isolado de A. robusta reduziu (p<0,05) o crescimento de D. flagrans. No ensaio in vivo, cada animal do grupo tratado (A) recebeu 2g/10 kg de peso vivo (PV) de péletes de alginato de sódio com os isolados fúngicos AC001 (0,2 g de micélio/10 kg de PV) e I31 (0,2 g de micélio/10 kg de PV) com ração comercial duas vezes por semana durante 26 semanas, enquanto os animais do grupo controle (B) receberam somente ração comercial. Ao final do experimento, os animais do grupo A apresentaram em relação aos do grupo B os resultados: redução de 72,87% no número de ovos por grama de fezes (OPG) e de 6,07% no número de larvas recuperadas das coproculturas vii respectivamente. Porém, o número total de larvas (L3) recuperadas por quilograma de matéria seca de pastagem foi semelhante (p>0,05) entre os grupos tratado e controle ao final do experimento. / Worldwide, gastrointestinal parasites in ruminants caused by nematodes produce economic losses often not visible or quantified for livestock production. Thus, the use of associated nematophagous fungi, as a form of biological control, can be an effective and advantageous alternative to improve the results of treatments using only fungi and to enhance the control of nematodes that parasitize cattle. The purpose of this study was to evaluate the compatibility between nematophagous fungi from the predator group Arthrobotrys robusta (isolate I31) and Duddingtonia flagrans (isolate AC001) under laboratory conditions and the joint action of these fungi in the environmental reduction of infective forms of gastrointestinal nematodes on young Holstein-Zebu cattle. Firstly, we detected the in vitro compatibility between A. robusta and D. flagrans through direct confrontation, antibiosis and volatile compounds testing. These tests allow the identification of the existence of competition for space between the organisms and the production of volatile compouds and antibiotics for some of these fungi. In direct comparison test, the A. robusta colonized approximately 2/3 of the plate, which suggests that there is competition for space between them and thus a form of antagonism. In antibiosis test, the production of antibiotics on culture medium by any fungus was not observed, which resulted in the absence of inhibition zone of fungus subsequently sown in the same plate. In the volatile compounds test, A. robusta reduced (p <0.05) the growth of D. flagrans. On the in vivo test each animal in the treated group (A) received 2 g/ 10 kg body weight (BW) of sodium alginate pellets containing the fungal isolates AC001 (0.2 g mycelium / 10 kg of BW) and I31 (0.2 g mycelium / 10 kg of BW) with commercial ration twice a week for 26 weeks, whereas animals in the control group (B) received only commercial feed. At the end of the experiment, the animals of group A compared to group B showed the following results: 72.87% reduction in the average number of eggs per gram of feces (EPG) and 6.07% of the ix average number of larvae recovered of coproculture, respectively. However, there was no difference (p> 0.05) of the total number of larvae (L3) recovered per kilogram of dry grassland between the treated and control groups at the end of the experiment.

As doenças parasitárias, principalmente as causadas por endoparasitas ocupam lugar de destaque dentre os fatores que interferem no desenvolvimento pleno da produção animal. Dentre as alternativas promissoras para o combate não químico de parasitos, pode-se destacar a utilização do controle biológico feito através de fungos nematófagos. O potencial antagônico do fungo Arthrobotrys cladodes var. macroides sobre larvas infectantes de nematoides parasitas gastrintestinais de bovinos foi avaliado por meio de microscopia eletrônica de varredura e foi realizado teste in vivo da resistência e viabilidade do fungo em uma formulação a base de alginato de sódio a passagem por via oral de bovinos. O fungo estudado apresentou alta produção de conídios e clamidósporos. Em teste de interação do fungo com os nematoides foi observada uma redução de 68,65% das larvas infectantes de nematóides nas placas de Petri do grupo tratado, em comparação com o controle. Sendo que a interação entre o fungo e os nematoides foi confirmada pela microscopia eletrônica de varredura. A observação das placas contendo as amostras de fezes coletadas após uma administração única por via oral de 100 gramas de péletes contendo o fungo A. cladodes mostrou que o mesmo foi capaz de sobreviver à passagem pelo trato gastrintestinal de ruminantes, crescer sobre o ágar, formar armadilhas e predar as larvas de terceiro estádio de parasitas gastrointestinais. Com o objetivo de avaliar a eficácia do fungo nematofago Arthrobotrys cladodes var. macroides no controle biológico de nematoides gastrintestinais de bovinos, doze bovinos mestiços holandês x zebu com sete a nove meses de idade, foram divididos aleatoriamente em dois grupos (grupo tratado e grupo controle) e distribuídos em piquetes de Brachiaria decumbens, naturalmente infestados com larvas de helmintos. Os animais do grupo tratado receberam 1g de pélete em matriz de alginato de sódio para cada 10 kg de peso vivo animal, contendo o fungo Arthrobotrys cladodes var macroides administrados duas vezes por semana conjuntamente com ração comercial. Já no grupo controle cada animal recebeu 1 g de pélete para cada 10 kg de peso vivo animal, sem micélio fúngico adicionado a ração. Amostras de fezes e pastagens foram coletadas quinzenalmente durante 12 meses. Os gêneros de nematoides mais prevalentes nas coproculturas foram Haemonchus sp., Cooperia sp. e Oesophagostomum sp. refletindo os resultados encontrados na forragem. A pastagem que continha os animais que receberam tratamento com o fungo apresentou redução de 59% e 52% de larvas recuperadas nas distâncias de 20cm e 40cm do bolo fecal, respectivamente . As médias mensais de ovos por grama de fezes e peso não diferiram significativamente (p>0,05) entre o grupo tratado e controle. Amostras de fezes e solo de ambos os grupos foram colonizadas pelo fungo A. cladodes e por outros fungos. A administração do micélio de Arthrobotrys cladodes var macroides por meio de matriz de alginato de sódio, duas vezes na semana, diminuiu a infestação de larvas na pastagem, indicando que esse fungo pode ser um controlador biológico promissor no combate das formas infectantes de nematoides presentes no meio ambiente. / Every year, Brazilian participation in international trade has been increasing, with emphasis on beef production. The parasitic diseases, especially those caused by endoparasites occupy a prominent place among the factors that interfere in the full development of animal production. Among the promising alternatives for the non- chemical control of parasites, one can highlight the use of biological control made by nematophagous fungi. The antagonistic potential of the fungus Arthrobotrys cladodes var. macroides on infective larvae of bovine gastrointestinal parasitic nematodes was evaluated by scanning electron microscopy and an in vivo test of the resistance and viability of the fungus in a formulation based on sodium alginate was carried out orally by bovines. The fungus studied showed high production of conidia and chlamydospores. In a test of interaction of the fungus with the nematodes, a 68.65% reduction of the nematode infecting larvae in the Petri dishes of the treated group was observed, in comparison with the control. The interaction between the fungus and the nematodes was confirmed by scanning electron microscopy. Observation of plates containing feces collected after oral administration of 100 grams of pellets containing the A. cladodes fungus showed that it was able to survive passage through the gastrointestinal tract of ruminants, grow on the agar, form traps and predatory larvae of the third stage of gastrointestinal parasites. In order to evaluate the efficacy of the nematophagous fungus A. cladodes var. macroides of the bovine gastrointestinal nematodes, twelve crossbred Dutch - zebu cattle with seven to nine months of age were randomly divided into two groups (treated group and control group) and distributed in Brachiaria decumbens pickets, naturally infested with larvae of helminths. The animals in the treated group received 1g of sodium alginate matrix pellet for every 10 kg of animal live weight containing the fungus A. cladodes var macroides administered twice a week in conjunction with commercial feed. In the control group, each animal received 1 g of pellet for every 10 kg of animal live weight, without fungal mycelium added to the feed. Samples of feces and pastures were collected biweekly for 12 months. The most prevalent nematode genera in the coprocultures were Haemonchus sp., Cooperia sp. and Oesophagostomum sp. reflecting the results found in forage. The pasture that contained the animals that received treatment with the fungus presented reduction of 59% and 52% of larvae recovered at distances of 20cm and 40cm of the fecal mass, respectively. The mean monthly egg per gram stool and weight did not differ significantly (p> 0.05) between the treated and control groups. Feces and soil samples from both groups were colonized by the fungus A. cladodes and by other fungi. Administration of the mycelium of Arthrobotrys cladodes var macroides by means of a sodium alginate matrix twice weekly reduced the infestation of larvae in the pasture, indicating that this fungus may be a promising biological controller in the control of the nematode infecting forms present in the environment.

Além disso, as mudas micropropagadas podem ser tratadas com microrganismos benéficos, que conferem proteção e vigor extras, agregando valor ao produto. O fungo Pochonia chlamydosporia já é conhecido por sua capacidade de controlar nematoides fitoparasitas e por promover o crescimento vegetal. Sendo assim, este trabalho foram avaliados os benefícios que esse fungo endofítico pode agregar ao ser introduzido em mudas micropropagadas de bananeira durante o período de aclimatação, antes de serem expostas ao contato com o nematoide das galhas radiculares, Meloidogyne javanica. Doses do produto Rizotec®, à base do isolado Pc-10 do fungo P. chlamydosporia, a fim de gerarem concentrações de 2.000, 3.000, 4.000 e 5.000 clamidósporos.g -1 de substrato, foram comparadas quanto à capacidade de conferir proteção e promover crescimento às mudas de banana. As diferentes concentrações do fungo resultaram em redução do número de galhas e de ovos de M. javanica por grama de raiz. O número de galhas e de ovos de M. javanica.g -1 de raízes, na concentração de 5.000 clamidósporos.g -1 de substrato, foram reduzidos em até 47,6% e 67%, respectivamente, em relação ao tratamento controle. Para as variáveis de desenvolvimento da planta, a altura da parte aérea, massa da parte aérea seca, diâmetro do pseudocaule e massa fresca das raízes de bananeira houve maior incremento na concentração de 5.000 clamidósporos.g -1 de substrato quando comparado ao controle. O fungo foi detectado colonizando as raízes das plantas e o substrato ao final do experimento, mostrando sua capacidade de se instalar, multiplicar e sobreviver no ambiente da muda micropropagada, e foi eficiente na redução dos danos causados pelo nematoide e na promoção do crescimento das plântulas de bananeira. / Micropropagation of banana seedlings has become increasingly common due to the gain in scale and time in the production, as well as avoiding the spread of pathogens, which occurs frequently in the natural propagation of the crop. In addition, micropropagated seedlings may be treated with beneficial microorganisms, which provide extra protection and vigor, adding value to the product. The fungus Pochonia chlamydosporia is already known for its ability to control plant-parasitic nematodes and to promote plant growth. Thus, this work evaluated the benefits that this endophytic fungus can add if introduced in micropropagated seedlings during the acclimatization period, before they are exposed to contact with the root gall nematode, Meloidogyne javanica. Doses of the Rizotec® product, based on the P. chlamydosporia fungus Pc-10 isolate, to generate 2000, 3000, 4000 and 5000 chlamydospore.g -1 concentrations in the substrate were compared for their ability to confer protection and promote the growth of banana seedlings. The different concentrations of the fungus resulted in a reduction in the number of galls and eggs of M. javanica per gram of root. The number of root galls showed a greater reduction in the concentration of 5,000 chlamydospores.g -1 than in the other treatments, with a reduction of up to 47.6% when compared to the control treatment. The number of eggs of M. javanica.g -1 of roots obtained a reduction of 67% at the concentration of 5,000 chlamydospores.g -1 of substrate, in relation to the control treatment. The dry shoot mass increased by up to 23% to 5,000 chlamydospores.g - soil. The diameter of the pseudostem also increased with the fungus application, showing an increase of 14%. The fresh root mass of the plants was also higher in the treatments with the fungus, with a 11.4% increase in the concentration of 5,000 chlamydospores.g -1 , in relation to the control treatment. The fungus was detected colonizing the roots of the plants and the substrate at the end of the experiment, showing its ability to install, multiply and survive in the micropropagated seedling environment, and was efficient in reducing the nematode damage and in promoting banana seedling growth.

Este tipo de controle se destaca por ser uma alternativa “limpa”, sem o uso de produtos químicos que possam gerar possíveis resíduos. Nesse contexto, fungos nematófagos produzem enzimas extracelulares envolvidas em diversas etapas da infecção, como liberação de nutrientes para o crescimento do microrganismo, penetração da cutícula pela degradação proteica e digestão do tecido hospedeiro. O mecanismo molecular da ação patogênica de fungos contra nematoides não é completamente conhecido. Dessa forma, o presente trabalho teve como objetivo otimizar a produção, purificar e caracterizar enzimas extracelulares produzidas por fungos nematófagos, bem como avaliar sua capacidade nematicida. Micélios obtidos da transferência de discos de cultura de três isolados de fungos nematófagos (Monacrosporium thaumasium (NF34), M. sinense (SF53) e Pochonia chlamydosporia (VC4)) mantidos em corn-meal-ágar 2% (CMA 2%) e transferidos para frascos contendo meio de cultura que teve sua composição otimizada para a produção de protease e quitinase. Em seguida, as enzimas foram purificadas e caracterizadas em relação ao pH e temperatura. Por fim, foi demonstrada a atividade nematicida dos extratos brutos e das enzimas purificadas sobre larvas de nematoides. Duas varíaveis (pH e tempo de incubação), apresentaram um efeito significativo (p<0,05) sobre a produção de protease do fungo nematófago Monacrosporium sinense (SF53). Além disso, este fungo produziu três diferentes proteases (Ms1, Ms2 e Ms3) com atividade nematicida sobre larvas de Panagrellus redivivus, sugerindo que essas proteases sejam produzidas como um complexo enzimático eficaz na destruição de larvas de primeiro estágio de Angiostrongylus vasorum. Em relação a M. thaumasium (NF34), foi observada atividade nematicida (p<0.01) de seus conídios e de seu extrato bruto proteolítico em coproculturas sobre larvas de primeiro estádio de A. vasorum. Os efeitos das variáveis (quitina, quitina coloidal, glicose, nitrato de sódio (NaNO 3 ), umidade) sobre a produção xiide quitinase em fermentação no estado sólido pelos isolados de Monacrosporium NF34 e SF53 foram analisados usando o planejamento estatístico Plackett-Burman. Para o isolado NF34, nos níveis avaliados, a presença de quitina e NaNO 3 nos meios de cultura foram significativas (p<0,05) para a produção de quitinase. Por outro lado, para o isolado SF53, nos níveis avaliados, apenas a presença de glicose foi significativa (p<0,05) para a produção de quitinase. Em relação a influência do pH na atividade enzimática, para M. thaumasium (NF34), o valor de pH com maior atividade obtida foi 5.5. Entretanto, para M. sinense (SF53), a maior atividade foi observada em pH 8. Além disso, observou-se que o isolado NF34 produziu duas quitinases distintas com a atividade nematicida sobre larvas de P. redivivus. Por fim, a atividade ovicida do fungo nematófago Pochonia chlamydosporia (VC4) e de suas proteases e quitinases foi avaliada sobre ovos de Dioctophyma renale. Houve diferença significativa (p <0,01) na destruição de ovos em relação ao grupo de controle, não se verificando diferença estatisticamente significativa (p> 0,01) entre as concentrações de P. chlamydosporia na destruição de ovos. No entanto, houve uma tendência de aumento da mortalidade com o aumento da concentração de P. chlamydosporia. As proteases e quitinases do isolado VC4, causaram uma redução de percentagem significativa (p <0,01) no número de ovos viáveis de D. renale, em relação ao controle, com os seguintes valores de redução: 27,8% (proteases), 29,4% (quitinases) e 43,4% (proteases + quitinases). Mais estudos devem ser conduzidos no sentido de otimizar as condições de produção de enzima e aplicação destas no combate de nematoides. / Biological control is one of the possible biotechnological applications of fungal enzymes. This type of control is known for being a "clean" alternative, without the use of chemicals that may generate residues. In this context, nematophagous fungi produce extracellular enzymes that are involved in various stages of infection, such as release of nutrients for microorganism growth, cuticle penetration through protein degradation and digestion of host tissue. However, the detailed molecular mechanism of pathogenic action of fungi against nematodes is not complete known. Thus, the present study aimed to optimize production, purify and characterize extracellular enzymes produced by nematophagous fungi and evaluate their nematicidal ability. Mycelia were collected through the transfer of culture dishes of three isolates (Monacrosporium thaumasium (NF34), M. sinense (SF53) and Pochonia chlamydosporia (VC4)) of nematophagous fungi kept in 2% corn-meal-agar (2% CMA) and transferred to vials containing culture medium that had the composition optimized for the production of protease and chitinase. Then, the enzymes were purified and characterized with respect to pH and temperature. Finally, the nematicidal activity of crude extracts and purified enzymes on nematodes larvae was demonstrated. Two variables (pH and incubation time) showed a significant effect (p <0.05) on the production of protease from nematophagous fungus Monacrosporium sinense (SF53). Moreover, this fungi produced three different proteases (Ms1, Ms2 and MS3) with nematicidal activity against Panagrellus redivivus larvae. It is also suggested that these proteases are produced in an enzymatic complex that was effective in the destruction of first-stage larvae of Angiostrongylus vasorum. Regarding M. thaumasium (NF34), it was observed nematicidal activity (p <0.01) of its conidia and crude extract in proteolytic coprocultures on first stage larvae of A. vasorum. The effect of the variables (chitin, colloidal chitin, glucose, sodium nitrate (NaNO 3 ), moisture) on chitinase production in solid state fermentation by Monacrosporium isolates SF53 and NF34 were analyzed using Plackett- xivBurman statistical design. For the isolate NF34, at the evaluated levels, the presence of chitin and NaNO 3 in culture medium was significant (p <0.05) for the production of chitinase. On the other hand, for isolate SF53, at the evaluated levels, only the presence of glucose was significant (p <0.05) for chitinase production. Regarding the influence of pH on enzyme activity, for M. thaumasium (NF34) the pH value with the highest activity was 5.5. However, for M. sinense (SF53), the highest activity was observed at pH 8. Furthermore, it was observed that the isolate NF34 produced two different chitinases with nematicidal activity against larvae of P. redivivus. Finally, the ovicidal activity of the nematophagous fungus Pochonia chlamydosporia (VC4) and its proteases and chitinases was evaluated on Dioctophyma renale eggs. There was a significant difference (p <0.01) in the destruction of eggs in relation to the control group. Moreover, there was no difference (p> 0.01) between the concentrations of P. chlamydosporia in the destruction of eggs. However, there was a trend for increased mortality with increasing its concentration. Proteases and chitinases of isolate VC4 have caused a significant percentage reduction (p <0.01) in the number of viable eggs of D. renale, compared to control, with the following values of reduction: 27.8% (proteases), 29 4% (chitinase) and 43.4% (proteases + chitinases). More studies should be conducted to optimize enzyme production conditions and the application of these on nematodes.

Contudo, foram estudados os fatores determinantes da freqüência de fungos totais e nematófagos, da atividade enzimática e da distribuição de nematóides de solos não rizosféricos e rizosféricos sob alface (Lactuca sativa L.); banana (Musa cavendishii L.); grama batatais (Paspalum notatum) e Impatiens (Impatiens valleriana). A atividade predadora dos fungos nematófagos foi avaliada contra o nematóide Panagrellus redivivus. O número de fungos totais variou de 6,9 x 105 a 31,2 x 105 UFC g-1 solo seco no solo não-rizosférico e de 6,9 x 105 a 25,6 x 105 UFC g-1 solo seco no solo rizosférico. As contagens dos fungos nematófagos corresponderam a 23-41 % e 23-34 % dos fungos totais encontrados no solo rizosférico e não-rizosférico, respectivamente. Enquanto as contagens dos fungos totais do solo rizosférico se distribuíram na seguinte seqüência: alface > banana > grama-batatais > Impatiens, os nematófagos diminuíram na seguinte ordem: alface > Impatiens > banana > grama-batatais. A matéria orgânica e a umidade do solo influenciaram a distribuição tanto dos fungos totais como dos nematófagos do solo não-rizosférico. A distribuição dos nematóides nas amostras de solo e raiz foram Rotylenchulus sp., Helicotylenchus sp. e Tylenchus sp. para alface, Meloidogyne sp., Helicotylenchus sp., Pratylenchus sp. e Rotylenchulus sp. para banana, Meloidogyne sp., Helicotylenchus sp., Pratylenchus sp. e Rotylenchulus sp. para grama-batatais, Meloidogyne sp., Helicotylenchus sp., Pratylenchus sp., Rotylenchulus sp. e Tylenchus sp. para Impatiens. We investigated the factors affecting the frequency of the total and the nematophagous fungi, soil enzymatic activity as well as the distribution of the nematodes in rhizospheric and bulk soil of lettuce (Lactuca sativa L.); Impatiens (Impatiens valleriana); banana (Musa cavendishii L.) and Bahiagrass (Paspalum otatum). The effect of the nematophagous fungi were evaluated against the nematode Panagrellus redivivus. The total fungal number ranged from 6,9 x 105 to 31,2 x 105 UFC g-1 dry soil in the bulk soil and from 6,9 x 105 to 25,6 x 105 UFC g-1 dry soil in the rhizosphere. Nematofagous fungi counts were to 23-41 % and 23-34 % of the total fungi found in the rhizosphere and bulk soil, respectively. Agentes microbianos no controle de nematóides e fungos fitopatogênicos de soja e sua compatibilidade com agroquímicos /

Resumo: Nematóides de galhas e cisto, além de fungos de solo causadores de doenças constituem importante grupo de patógenos da cultura da soja, sendo o manejo integrado uma das principais medidas de controle visando a redução de perdas econômicas. O objetivo deste trabalho foi avaliar a eficácia dos fungos Paecilomyces lilacinus e Pochonia chlamydosporia e de um produto comercial à base de Bacillus sp. e do nematicida químico Aldicarb no controle de Meloidogyne incognita, Heterodera glycines, Fusarium solani e Rhizoctonia solani em soja, variedade M-SOY 6101, para o que foram feitos ensaios in vitro, em casa de vegetação e em campo. Foi também avaliada a produção dos fungos em dois substratos, sua compatibilidade com agroquímicos utilizados em soja e capacidade de colonização de raízes. Os fungos colonizaram satisfatoriamente grãos de arroz e milheto. Também foram compatíveis a inseticidas baseados em Fipronil. P. chlamydosporia colonizou endofiticamente raízes de soja. Nenhum dos fungos, utilizados em tratamento de sementes inibiu a germinação ou causou tombamento em plântulas de soja, porém não foram eficientes em prevenir esta doença quando o substrato estava inoculado com F. solani e R. solani. Nos ensaios em casa de vegetação com infestação de plantas com M. incognita apenas aldicarb reduziu o número de juvenis no solo e ovos nas raízes, porém os tratamentos biológicos reduziram o número de ovos nas raízes, tendo P. lilacinus favorecido a manutenção do peso seco das raízes. The objective of this work was to evaluate the efficacy of the fungi Paecilomyces lilacinus and Pochonia chlamydosporia, a commercial product based on Bacillus sp. and aldicarb on the control of Meloidogyne incognita, Heterodera glycines Ichinohe, Fusarium solani and Rhizoctonia solani on soybean, cultivar M-SOY 6101. Mass production of the fungi on rice and millet was tested, as well as the compatibility of them with some chemical pesticides. Patogenicidade de fungos a mosca-negra-dos-citros e compatibilidade entre agrotóxicos e Purpureocillium lilacinum /

O objetivo deste trabalho foi avaliar a patogenicidade e virulência de isolados de fungos entomopatogênicos sobre a mosca-negra-dos-citros, Aleurocanthus woglumi (Hemiptera: Aleyrodidae) em condições de laboratório, e o efeito de diferentes produtos fitossanitários sobre o desenvolvimento do fungo Purpureocillium lilacinum in vitro e semicampo. No teste de patogenicidade, foram utilizadas dez ninfas de segundo e terceiro ínstares de Aleurocanthus woglumi e os fungos Beauveria bassiana, Metarhizium anisopliae, Lecanicillium muscarium, Fusarium proliferatum e Purpureocillium lilacinum. Para determinação da concentração letal 50 (CL50) e do tempo letal médio 50 (TL50) foi escolhido o fungo P. lilacinum, sendo cada tratamento representado por uma concentração 1 x 106, 5 x 106, 1 x 107, 5 x 107, 1 x 108 e 5 x 108 conídios/mL. Nos testes de compatibilidade de agrotóxicos sobre o fungo P. lilacinum, foram testados os produtos Ampligo® (clorantraniliprole + lambda-cialotrina), Kohinor 200 SC® (imidacloprido), Provado 200 SC® (imidacloprido), Recop® (oxicloreto de cobre), Iharol® (óleo mineral), Talstar 100 EC® (bifentrina), Stimulate® (ácido 4-indol-3-ilbutírico + ácido giberélico + cinetina) e Neenmax® (azadiractina), nas doses recomendadas para campo. No experimento em casa de vegetação, mudas de limão Cravo foram pulverizadas com os produtos e o patógeno P. lilacinum (4,76 x 107 conídios/mL) de três formas diferentes, primeiramente os agrotóxicos e depois o fungo; primeiro o fungo e posteriormente o agrotóxico e os dois simultaneamente. The objective this study was to evaluate the pathogenicity and virulence of entomopathogenic fungi on citrus blackfly, Aleurocanthus woglumi (Hemiptera: Aleyrodidae) under laboratory conditions, and effect of different pesticides on the development of fungus Purpureocillium lilacinum in vitro and greenhouse. Efeitos dos processos de preservação sobre os fungos Arthrobotrys robusta e Monacrosporium thaumasium predadores de nematóides / Effects of long-term storage of Arthrobotrys robusta and Monacrosporium thaumasium nematode trapping fungi

Os fungos nematófagos produzem estruturas em forma de armadilha, responsáveis pela captura e morte dos estágios pré-parasitários dos nematóides. A manutenção contínua da atividade predatória dos isolados fúngicos é um dos pré-requisitos básicos para o sucesso desta forma de controle. Neste trabalho foi verificado o comportamento dos isolados I31 de Arthrobotrys robusta e NF34a de Monacrosporium thaumasium, submetidos a manutenção em temperatura de 4 o C, criopreservados com e sem adição de DMSO e glicerol e mantidos em sílica-gel. Os resultados demonstraram que os isolados NF34a e I31 não apresentaram variação no crescimento radial, quando armazenados em temperatura de 4 o C e congelados com ou sem adição de crioprotetores. Não houve diferença na produção massal de micélio seco dos isolados quando armazenados a temperatura de 4 o C e congelados com adição de crioprotetores. O armazenamento em sílica-gel e o congelamento sem crioproteção pareceram interferir negativamente na capacidade dos fungos em produzir massa micelial. O armazenamento de NF34a em temperatura de 4 o C e o congelamento com adição de DMSO foram os tratamentos em que se obteve maior redução das larvas infectantes in vitro. Não foi verificada diferença na capacidade predatória in vitro do isolado I31 quando mantido em temperatura de 4 o C ou quando congelado com adição de DMSO ou glicerol. A manutenção dos isolados em sílica-gel e o congelamento sem crioproteção interferiram negativamente na capacidade predatória, porém estes ainda foram capazes de reduzir a população de larvas infectantes em relação ao grupo controle. No teste de passagem dos isolados pelo trato gastrintestinal de bovinos, a administração de 20 g de micélio aos animais foi suficiente para recuperação de material fúngico nas fezes e pico de redução no número de larvas infectantes nas fezes após 24 horas da administração das amostras. O isolado NF34a foi responsável pelos maiores índices de redução das populações de larvas de helmintos após a passagem pelo trato gastrintestinal dos animais e o armazenamento em 4 o C foi o tratamento em que se observou maior taxa de redução. A criopreservação parece ser um método eficiente para manutenção dos isolados, porém neste trabalho foi evidenciada diminuição da capacidade predatória do isolado NF34a. A manutenção dos isolado em sílica-gel deve ser utilizado para manutenção das amostras por períodos prolongados, quando não houver disponibilidade de nitrogênio líquido. / Biological control of helminths parasites of cattle is an alternative to the use of anthelmintics, and its main goal is to reduce the amount of infective larvae in the pasture. The nematophagous fungi produce trap-shaped structures, which are responsible for capturing and destroying the pre-parasite stages of nematodes. Continuous maintenance of the fungi isolates’ predatory activity is one of the basic prerequisites for the success of this form of control. This research analyzed the behaviour of I31 isolates of Arthrobotrys robusta and NF34a isolates of Monacrosporium thaumasium, submitted to maintenance at a temperature of 4 o C, cryopreserved with and without DMSO and glicerol, and kept in silica gel. Results showed that the NF34a and I31 isolates did not present variation in radial growth when stored at a temperature of 4 o C and frozen with or without adding cryoprotectants. There was no difference in production of dry biomass by the isolates when stored at a temperature of 4 o C and frozen with added cryoprotectants. Silica gel storage and freezing without cryoprotection seemed to interfere negatively with the fungi’s capacity to produce biomass. Storing NF34a at a temperature of 4 o C and freezing it with DMSO were the treatments in which the largest in vitro reduction of infective larvae was obtained. There was no difference in terms of in vitro predatory capacity of the I31 when stored at a temperature of 4 o C or when frozen with DMSO or glicerol. Storage of the isolates in silica gel and freezing without cryoprotection interfered negatively in their predatory capacity, but they were still able to reduce the population of infective larvae in relation to the control. When the passage of isolates through the gastrointestinal tract of bovines was tested, the administration of 20 g of mycelium was enough to recover fungi material in feces and for a peak reduction of the number of infective larvae in feces 24 hours after the administration of the samples. The NF34a isolate was responsible for the largest reduction rates in helminth larvae populations following passage through the gastrointestinal tract of the animals, and storage at 4 o C was the treatment in which the largest reduction rate was observed. Cryopreservation seems to be an effective method for keeping the isolates, although, in this research, a reduction of the NF34a’s predatory capacity became evident. Storage of the isolates in silica gel should be chosen when keeping the samples for long periods of time, whenever liquid nitrogen is not available. / Dissertação importada do Alexandria

Em 1987, foram estimadas perdas de 14,2%. No Brasil, perdas associadas à Tylenchulus semipenetrans foram estimadas em R$ 319 milhões para a safra de 1999/2000 no Estado de São Paulo. Para Pratylenchus jaehni, observou-se perdas de 30% na produção, de pomares de laranja no município de Itápolis-SP. O manejo dessas espécies de nematoides, em pomares infestados é feito, exclusivamente, com uso de aldicarbe (Temik® 150G), contudo o controle biológico pode tornar-se uma alternativa viável de manejo desses nematoides. Os ensaios foram conduzidos nos Municípios de Taquaritinga, Itápolis e Fernandópolis-SP, no período de setembro de 2008 a fevereiro de 2011. Os objetivos desse trabalho foram: avaliar a sobrevivência dos fungos nematófagos nas parcelas que foram tratadas, a campo, com uma formulação de cinco espécies desses agentes de biocontrole; avaliar a influência da adição de matéria orgânica de diferentes fontes para promover o estabelecimento e potencializar a ação dos fungos; estudar o controle químico com doses reduzidas de aldicarbe, associado aos fungos nematófagos no manejo dos nematoides dos citros; avaliar a distribuição espacial nos sentidos norte, sul, leste e oeste de cinco fungos nematófagos aplicados em pomar comercial de citros. A sobrevivência dos fungos nematófagos concluiu-se que as espécies de Arthrobotrys e Monacrosporium já estavam presentes na área e foram recuperadas até a avaliação de 9 meses após a aplicação. No experimento de distribuição geográfica dos fungos nematófagos concluiu-se não existe um padrão de distribuição sob a copa das plantas. To Pratylenchus jaehni, there was loss of 30% in the production of orange orchards in the county of Itápolis-SP. The management of these species of nematodes in infested orchards is made exclusively with the use of aldicarb (Temik ® 150G), but biological control is emerging as a viable alternative for management of these nematodes. The objectives of this study were to evaluate the survival of nematophagous fungi on plots that were treated, the field, with a formulation of five species of biocontrol agents, to evaluate the influence of the addition of organic matter from different sources to promote the establishment and enhance action of fungi, to study the chemical control with reduced doses of aldicarb associated with nematophagous fungi the management of the citrus nematode; evaluate the spatial distribution in the directions north, south, east and west with applied to five nematophagous fungi in commercial citrus orchards.
