This thesis analyzes the engineering-geological and geotechnical conditions MSW landfills in Úholičky. It includes the methodology of field mapping and documentation work, evaluation of natural conditions the area of interest, the assessment of geotechnical properties of rocks and soils and geotechnical conditions at the landfill and finally deals with risk analysis MSW landfill in Úholičky. At the same thesis includes a search of domestic and foreign literature. The outcome of their work is the engineering map, evaluation and interpretation of the data from the MSW landfill settlement Úholičky. Development of a simplified thermal analysis procedure for insulating glass units

A percentage of insulating glass (IG) units break each year due to thermally induced perimeter stresses. The glass industry has known about this problem for many years and an ASTM standard has recently been developed for the design of monolithic glass plates for thermal stresses induced by solar irradiance. It is believed that a similar standard can be developed for IG units if a proper understanding of IG thermal stresses can be developed. The objective of this research is to improve understandings of IG thermal stresses and compare the IG thermal stresses with those that develop in monolithic glass plates given similar environmental conditions. The major difference between the analysis of a monolithic glass plate and an IG unit is energy exchange due to conduction, natural convection, and long wave radiation through the gas space cavity. In IG units, conduction, natural convection, and long wave radiation combine in a nonlinear fashion that frequently requires iterative numerical analyses for determining thermal stresses in certain situations. To simplify the gas space energy exchange, a numerical propagation procedure was developed. The numerical propagation procedure combines the nonlinear effects of conduction, natural convection, and long wave radiation into a single value. Use of this single value closely approximates the nonlinear nature of the gas space energy exchange and simplifies the numerical analysis. The numerical propagation procedure was then coupled with finite element analysis to estimate thermal stresses for both monolithic glass plates and IG units. It is shown that the maximum thermal stresses that develop in IG units increase linearly with input solar irradiance during the transient phase. It is shown that an initial preload stress develops under equilibrium conditions due to the thermal bridge effects of the spacer. It is shown that IG units develop larger thermal stresses than monolithic glass plates under similar environmental conditions. Finally, it is shown that the use of low-e coatings increase IG thermal stresses and that the location of low-e coating as well as environmental conditions affect which glass plate develops larger thermal stresses.

The regulation of AID function by transcription factors PU.1 and IRF4 in chicken B cells

B cells are capable of producing antibodies of diverse antigen specificities and effector functions to counter infection by a wide range of pathogens. The diversification of immunoglobulin (Ig) is achieved through a series of programmed DNA recombination and mutagenic events during B cell maturation. A key factor involved in the Ig diversification process is Activation Induced Cytidine Deaminase (AID). AID is a B cell specific enzyme that is critical for three distinct pathways of Ig diversification: class switch recombination, somatic hypermutation and Ig gene conversion. AID functions by deaminating cytosine to uracil in target DNA at the Ig loci. Although essential for effective immunity, the mutagenic activity of AID needs to be confined to the Ig loci in order to protect genomic integrity, but the underlying mechanism is not fully understood. In this study, I show that two lymphoid specific transcription factors, PU.1 and IRF4, play important roles in regulating AID function in chicken B cells. PU.1 and IRF4 have been implicated in many aspects of B cell development and function. The two factors could form a heterodimer and regulate target gene expression cooperatively. However, we found that PU.1 and IRF4 appear to have different impacts on AID function. We show that PU.1 is important for the expression of AID gene in chicken B cells, and the regulation appears to involve direct interaction of PU.1 with the AID gene. By comparison, IRF4 plays a minor role in AID expression. On the other hand, both PU.1 and IRF4 are required for efficient gene conversion that is mediated by AID at the Igλ locus. Moreover, the gene dosage of PU.1 is critical for AID function, since a severe gene conversion defect is observed in PU.1+/- cells. The function of PU.1 and IRF4 in AID-mediated gene conversion involves binding sites for the PU.1/IRF4 complex within a regulatory element at the Igλ locus. Future studies will be directed at understanding how PU.1 and IRF4 regulate AID-mediated gene conversion. / text

The Biophysics of Titin in Cardiac Health and Disease

The giant protein titin is the third myofilament in the cardiac sarcomere. It is responsible for generating passive forces in stretched myocardium and maintaining sarcomere structure. The force generation properties of titin are determined by titin's elastic springlike elements, and this dissertation focuses on the determination of the physical properties of these springlike elements using atomic force microscopy. The primary project of this dissertation investigates the link between a single point mutation in one of titin's subdomains and arrhythmogenic cardiomyopathy.

Intergenerational Appeal in Luxury Product Advertising: Impacts of Brand-Gender Extension and Brand Duration

¡§Intergenerational (IG) appeal¡¨ is an ad focusing on the relationship between parents and children in order to enhance heritage image of a brand and facilitate the likelihood that parents introduce the brand to their sons and daughters. This study examines the application of IG appeal on advertising. An experimental study is conducted to examine the moderating effects of brand duration and (in)congruent brand-gender extension on the effect of IG appeal. The experiment in a 4 advertising appeal (IG appeal: male vs. female, non-IG appeal: male vs. female) x 2 brand duration (long vs. short) x 2 brand-gender extension (congruent vs. incongruent) factorial design is developed. A series of analysis of variance (ANOVA) are used to test the proposed hypotheses. Results of the experiment indicate that brand duration facilitates the effects of IG appeal. In addition, congruent brand extension increases the effects of IG appeal. More importantly, a three-way interaction is observed. In incongruent brand-gender extension, IG appeals used to present brands with short brand duration are more effective than Non-IG appeal. These findings should be incorporated into heritage branding when media campaign is developed to increase branding influences.
