Imunolocalização do fator de crescimento semelhante à insulina (IGF-I) e do fator de crescimento epidermal (EGF) em ovários caninos e seus efeitos sobre a maturação in vitro de oócitos : avaliação nuclear /

Foram selecionados 787 complexos cumulus-oocitos (COCs), provenientes de ovários de 34 cadelas, em diferentes fases do ciclo estral, submetidas a ovário-histerectomias eletivas. Os COCs foram dividos em três grupos para diferentes tratamentos, A (TCM 199, n=199), B (TCM 199 + 100ng/mL de EGF, n=208) e C (TCM 199 + 100 ng/mL IGF-1, n=204). Após 96 horas de incubação a 38 °C em atmosfera de 5% de CO2, 50% dos oócitos, desprovidos das células do cumulus, de cada grupo foram corados com Bisbenzimida H33342 (Hoechst) e 50% com 4',6-Diamidino-2-phenylindole (Dapi). A adição de EGF ou de IGF-1 ao meio TCM 199 não foi benéfica à maturação in vitro (MIV) de oócitos de cadelas, e a coloração Hoechst foi mais eficaz para a avaliação dos estágios de maturação nuclear de oócitos de cadelas. Os tratos reprodutivos das 34 cadelas foram coletados, fixados, processados e incluídos em parafina. Os fragmentos de ovários foram cortados, montados em lâmina, desparafinIzados e reidratados. A técnica de Imuno-histoquímica foi realizada utilizando os seguintes anticorpos: Anti-EGF-pc (Ab-3, Calbiochem), Anti- IGF-1-pc (Ab-2, Calbiochem). As amostras foram incubadas overnight. O anticorpo primário foi detectado utilizando IgG secundária biotinilada de cabra anti-coelho ou de coelho anti-cabra, com complexo avidina-biotina-peroxidase. A detecção da reação foi feita com o AEC (Vector). A imuno-histoquímica revelou a presença de EGF e do IGF-1 no ovário da cadela. O EGF foi imunolocalizado no citoplasma das células foliculares (CF) e nos oócitos de folículos secundários e terciários e também na parede vascular, nos cordões... (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: The aim of the present study was to assess the effect of the addition of EGF or IGF-1 to the culture medium TCM 199 on nuclear maturation of canine oocytes and to determine the distribution of these growth factors in the ovary of the bitch. 787 comulus-oocyte complexes (COCs) were sampled during different phases of the oestral cycle from 34 bitches submitted to elective ovariohysterectomy. The COCs were distributed into three groups: A (TCM 199, n= 199); B (TCM 199 + 100ng/ml of EGF, n=208) and; C (TCM 199 + 100ng/ml of IGF-1, n=204). Following 96 hours of incubation at 38 ºC, in sterile atmosphere with 5% CO2, the nuclear maturation stage was evaluated following bisbenzimide 33342 (50%) or 4',6-Diamidino-2-phenylindole staining (50%). The addition of EGF or IGF-1 to the TCM 199 did not favour the IVM of canine oocytes. The bisbenzimide 33342 staining was better than 4',6-Diamidino-2- phenylindole staining in the assessment of nuclear maturation stage of canine oocytes. . Reproductive tracts from 34 bitches at the different phases of the estrous cycle were collected, fixed and processed with paraffin embedding. Sections were cut, mounted, dried, deparaffinized and rehydrated. Immunocytochemistry was performed using a dilution of the following antisera: anti-EGF-pc (Ab-3, Calbiochem) and Anti-IGF-1-pc (Ab- 2, Calbiochem ). Samples were incubated overnight. Primary antibody binding was detected using a biotynilated secondary goat-anti-rabbit or rabbit anti-goat IgG with avidin-biotin-peroxidase complexing. The detection reagent was AEC (Vector). Comparação da resposta imune humoral, da capacidade de neutralização dos soros anticrotálicos produzidos em ovinos jovens, avaliação clínica e ponderal entre animais inoculados com veneno de serpente Crotalus durissus terrificus nativo ou irradiado com Cobalto-60 /

Resumo: A técnica de ELISA foi utilizada para avaliar e comparar a resposta imune humoral de ovinos jovens para a produção de soro anticrotálico. Durante o processo de soroprodução, foi realizada a avaliação clínica e ponderal dos animais. Os parâmetros utilizados foram hemograma completo, dosagem de uréia, creatinina, aspartato aminotransferase, proteínas totais, albumina, globulina. O peso dos animais foi aferido quinzenalmente durante o experimento. A capacidade de neutralização do soro produzido a partir de veneno de serpente Crotalus durissus terrificus, nativo (VN) e irradiado (VIr) com Cobalto-60 foi verificada por meio de desafios in vitro. Um grupo de seis animais recebeu veneno nativo, o segundo grupo recebeu veneno irradiado e o terceiro grupo foi o controle. Os animais receberam seis imunizações durante 84 dias com intervalo de 14 dias. Houve diferença significativa (p<5%) no teste de ELISA do perfil de anticorpos produzidos pelos grupos experimentais (VN<VIr). Não houve diferença significativa (p<5%) para os testes bioquímicos, hemograma e peso dos animais entre os três grupos testados. O grupo imunizado com veneno irradiado apresentou perfil de anticorpos maior que o grupo imunizado com veneno nativo. A capacidade de neutralização do soro produzido a partir do VIr foi cinco vezes maior quando comparado ao soro produzido com VN. A avaliação clínica e ponderal, mostrou que ovinos em fase pós-desmame, não tiveram alterado seus perfis fisiológicos e apresentaram um excelente ganho de peso durante o período experimental. Estes resultados demonstram uma nova perspectiva para a utilização de ovinos visando a produção comercial de soro anticrotálico, que poderá ser aplicado no tratamento do envenenamento de seres humanos e animais. O custo para sua produção poderá ser reduzido pela posterior utilização dos ovinos hiperimunizados na alimentação humana. / Abstract: The ELISA technique was used to evaluate and compare the humoral immune response of young ovines to anticrotalic serum production. During serum production, the clinical and weight evaluation of the animals was performed. The parameters utilized were complete blood count, and dosage of urea, creatinine, aspartate aminotransferase, total proteins, albumin and globulin. The animals weight was verified fortnightly during the experiment. The neutralization capacity of the serum produced from the snake Crotalus durissus terrificus natural (NV) and Cobalt-60-irradiated venom (IrV) was evaluated by in vitro challenges. One group of six animals received natural venom, the second group received irradiated venom, and the third was the control group. The animals received six immunizations during 84 days with an interval of 14 days. There was a significant difference (p<5%) in the ELISA test for the profile of the antibodies produced by the experimental groups (NV<IrV). There was no significant difference (p<5%) for biochemical tests, complete blood count, and animals weight between the three groups tested. The group immunized with irradiated venom showed antibodies profile higher than the group immunized with natural venom. The neutralization capacity of the serum produced from the IrV was fivefold higher when compared to the serum produced with NV. The clinical and weight evaluation showed that the ovines in post-weaning phase did not have their physiological profiles altered, and showed an excellent increase in weight during the experimental period. These results indicate a new perspective for the utilization of ovines, aiming the commercial production of anticrotalic serum, which may be applied in the treatment of human and animal envenomation. Carlos / Banca: Sueli Calvi / Não disponível / Abstract: Silibinin is the major active component of silymarin (Silybum marianum), a polyphenolic plant flavonoid that has anti-inflammatory, cytoprotective and anticarcinogenic effects. The modulatory effect of silibinin on monocyte function against Paracoccidioides brasiliensis (Pb18) has not yet been demonstrated. The present study investigated whether the effect of silibinin on NF-kB pathways may affects the production of tumor necrosis factor-alpha (TNF-α), interleukin-10 (IL-10), Transforming growth factor beta (TGF-β1), prostaglandin E2 (PGE2) and nitric oxide (NO), as well as the fungicidal activity of human monocytes challenged in vitro with Pb18. Peripheral blood monocytes from healthy individuals were treated with or without 5 uM and 50 uM silibinin and challenged with Pb18 or were stimulated with lipopolissaccharide (LPS). After 18 h of culture TNF-α, IL-10, TGF-β1 and PGE2 were determined by ELISA, while NO release was quantified by the accumulation of nitrite in the supernatants using the standard Griess assay. Fungicidal activity of monocytes against Pb18 was assessed after cell incubation with interferon-gamma, culture with or without silibinin for 18 h and challenge with Pb18 for 4h. NF-kB activation in the cultures was evaluated by flow cytometry and ELISA after monocyte stimulation with Pb18 or LPS for 30 min. Silibinin at 50uM concentration partially inhibited p65NF-kB activation as observed by reduction in the number of cells expressing this factor, that was confirmed by low concentration detection of p65NF-kB in the nucleus. This silibinin effects resulted in suppression of TNF-α, IL-10, TGF-β1 and PGE2 and NO production, but did not affect fungicidal activity of monocytes against Pb18. The modulatory effect of silibinin on the monocytes inflammatory response against P. brasiliensis might be important to establish an efficient and beneficial immune response of the host against the fungus / Doutor

Algumas substâncias podem influenciar as respostas do sistema imune de peixes, como o levamisol, através da modulação de parâmetros imunes, aumentando a resistência contra diversos agentes. Foram avaliados os efeitos do levamisol na dieta em dois experimentos. O primeiro experimento avaliou a suplementação com 0, 125, 250 e 500 mg kg-1 do imunoestimulante administrado por sete e 15 dias, no qual foram avaliados os parâmetros da imunidade inata, hematológicos e bioquímicos. A administração do levamisol por sete e 15 dias promoveu alterações em parâmetros imunológicos e hematológicos. A administração do levamisol pode promover imunomodulação, entretanto a determinação do efeito não ficou claro devido às respostas contraditórias em cada parâmetro avaliado. O segundo experimento consistiu em suplementação com 0, 125, 250 e 500 mg kg-1 do imunoestimulante administrada por sete dias conjuntamente com a imunização com bactérias Aeromonas hydrophila. A imunização e a administração de levamisol promoveram aumento do título de anticorpos, atividade bactericida do soro, hematócrito, número de eritrócitos, leucócitos totais e trombócitos nos pacus. A administração de levamisol por sete dias e a imunização de pacus promoveu melhora de alguns parâmetros da imunidade adquirida e inata de defesa. Entretanto outros protocolos devem ser estudados para avaliar o efeito do levamisol sobre o sistema imune de pacu / Abstract: The emergence of diseases in fish farming is a problem faced in Brazil and worldwide. Some substances can influence the immune system responses of fish, like levamisole, increasing resistance against various etiological agents. Were evaluated the effects of levamisole in diet in two experiments. The first experiment evaluated supplementation with 0, 125, 250 and 500 mg kg-1 imunoestimulante administered by seven and 15 days in which have been assessed the parameters of innate immunity, haematological and biochemists. The administration of levamisole by seven and 15 days promoted changes in haematological and immunological parameters. The administration of levamisole can promote immunodulation, however the determination of the effect not clear due to contradictory answers evaluated in each parameter. The second experiment consisted of supplementation with 0, 125, 250 and 500 mg kg-1 imunoestimulante administered by seven days together with immunization with inactivated Aeromonas hydrophila bacteria with formaldehyde. Immunization and administration of levamisole promoted increase antibody titre, serum bactericidal activity, hematocrit, number of erythrocytes, leucocytes and thrombocytes in pacus totals. Administration of levamisole for seven days and immunization pacus promoted improves some parameters of innate and acquired immunity defense. Parâmetros produtivos, fisiológicos e imunológicos de juvenis de pacu (Piaractus mesopotamicus) submetidos à restrição alimentar e estresse de manejo /

Além disso, a redução na oferta de alimento é um manejo utilizado para evitar o desperdício de ração, redução da mão-de-obra, e principalmente manutenção da qualidade de água. Entretanto o efeito desse tipo de manejo deve ser mais conhecido para ser melhor explorado. Estre estudo avaliou a influência da restrição alimentar controlada no desempenho zootécnico, homeostase metabólica, fisiológica e imunológica em pacu (Piaractus mesopotamicus). Juvenis de pacu foram submetidos a quatro estratégias de alimentação (controle; duas semanas; três semanas e quatro semanas de restrição alimentar). Após o período de restrição, todos os tratamentos foram realimentados por duas semanas seguidas. Cada tratamento teve três repetições (viveiros com cerca de 40 m²), sendo utilizado um total de 12 tanques. Os peixes foram amostrados ao final de cada período de restrição (n=12 para cada tratamento) e ao final do período de realimentação de cada tratamento (n=12 para cada tratamento). A segunda fase experimental teve início ao final da amostragem da primeira fase (após a restrição de alimento), para avaliar a higidez dos peixes. Os peixes restantes foram capturados e submetidos ao manejo de transporte, após o qual foram novamente amostrados. em seguida à amostragem pós-transporte, os peixes foram distribuídos em caixas de 100 L, com aeração e renovação constante de água. Peixes de cada tratamento da primeira fase foram distribuídos em 12 caixas (seis peixes/caixa) onde permaneceram por 72 horas, de modo a simular um período de recuperação. (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: A study of strategies to improve feeding management in fish is the use of restricted feeding for a while to promote compensatory growth during refeeding. Food restriction is a management used to avoid feed wastage, reduction of manpower, especially maintenance of water quality. However no one knows what the effects of this type of management on the animal performance, metabolic, physiological and immunological homeostasis of pacu. Juvenile pacu (Piaractus mesopotamicus) underwent four feeding strategies (control; two weeks; three weeks; and four weeks of food restriction). After the restriction period, all treatments were fed back for two weeks straight. Each treatment had three replicates (tanks with about 40 m²), used a total of 12 tanks. Fish were sampled at the end of each restriction period (n= 12 for each treatment) and the end of the refeeding from each treatment (n= 12 for each treatment). The second phase began at the end of sampling after refeeding, to assess fish health, captured animals were submitted to the management of transport, where they were sampled again. After the post-transport sampling, fish were distributed in polyethylene aquarium of 100 L with aeration and constant water flow. Fish per treatment of first stage were distributed in 12 boxes (six fish/aquarium) where they remained for 72 hours in order to simulate a recovery period. There were significant differences (p<0.05) in grow parameters, metabolic, hematologic and stress parameters. The feed restriction followed by refeeding resulted in total compensatory growth of juvenile pacu, and has proved an effective management, which should not affect the state of health of animals / Mestre
