Os herpesvirus possuem genoma DNA e são envelopados. Aproximadamente 70% dos adultos já tiveram contato com Herpes Simplex tipo 1 (HSV-1). O HSV-1 causa úlceras ou até mesmo lesões mais graves como encefalite, e após a infecção, o vírus comumente fica latente e pode ser ou não reativado. Células do sistema immune inato, através dos “Receptores do Tipo Toll” (TLRs), reconhecem “padrões moleculares associados a patógenos” (PAMPs) orquestrando a resposta imune contra o vírus. Quando ativados, TLRs iniciam uma cascata de sinalização que culmina na ativação de genes relacionados à defesa imune inata (óxido nítrico-NO, citocinas e quimiocinas). Outros estudos do grupo mostraram que animais nocautes (KO) para TLR9 ou TLR2/9 apresentam alta mortalidade na infecção por HSV-1, ao contrário dos C57BL/6 selvagens (WT) e TLR2KO, que são capazes de controlar o vírus. Objetivamos pesquisar a interrelação na expressão dos TLRs 1, 2, 3, 6, 7 e 9 em animais TLR2 KO, TLR9 KO e TLR2/9 KO infectados por HSV-1 e principais citocinas, células e mecanismos imune celulares contra infecção por HSV-1. No presente trabalho camundongos WT, TLR2KO, TLR9KO e TLR2/9KO foram infectados via intranasal com 106 u.f.p. de HSV-1, e seus controles receberam PBS. A eutanásia dos animais foi realizada no 5º dia após infecção (dpi), que previamente já foi demonstrado ser o pico da infecção neste modelo, e então gânglios trigêmeos (TG) e cérebros foram coletados. A expressão de TLRs 1, 2, 3, 6, 7 e 9 foi avaliada por PCR em Tempo Real. Camundongos WT infectados apresentaram diferença significativa na expressão destes TLRs comparados com seus respectivos controles no TG, mas não no cérebro. Nos KOs, houve aumento da expressão dos TLRs tanto nos TGs (mas com níveis mais moderados comparado aos WT) quanto nos cérebros. Também foi avaliada a expressão de gp91phox, p22phox e iNOS. A expressão de iNOS em C57BL/6 WT infectados foi maior que nos outros grupos, mas o mesmo não ocorreu para gp91phox, p22phox. Adicionalmente, foi mensurada a produção de NO por macrófagos intraperitoneais, através da reação de Griess, que demonstrou maior produção por macrófagos WT, comparados aos grupos TLR KOs. Ensaios de histopatologia e imunofluorescência demonstraram maior celularidade nos TGs infectados, além de confirmar a presença de CD8, macrófagos, e aumento da produção de iNOS nos C57BL/6 WT infectados, o que não ocorreu com os camundongos TLRs KO. Em ensaio de sobrevivência com C57BL/6 WT, CCL3 KO, CD8 KO, RAG KO e iNOS KO a importância das células T e do iNOS foi confirmada, uma vez que CD8 KO, RAG KO e iNOS KO tiveram 0% de sobrevida. Camundongos C57BL/6 WT, RAG KO, CCL3 KO e iNOS KO tiveram seus TGs analisados por PCR em tempo real para verificar a expressão do gene viral VP-16 e MCP-1, iNOS, IP-10, Rantes e TNF-_ no 5º dpi. Estes dados indicaram a relevância de iNOS e das célúlas T na resposta inata contra o HSV-1, uma vez que os RAG KO foram irresponsivos à expressão das citocinas, enquanto os iNOS KO apresentaram uma resposta exacerbada das mesmas. Ensaio de citometria indicou que IFN_ produzido pelas células T CD8, nos TGs, e produzido pelas células T CD4 e Natural Killer (NK), nos linfonodos regionais, são um possível mecanismo de controle contra a infecção de HSV-1 dos animais WT infectados. Hipotetizamos que a expressão coordenada de TLRs reflete na orquestração eficiente do sistema imune inato, através da produção equilibrada de quimiocinas e citocinas e da atividade imune celular adequada, com produção de NO por macrófagos e ação de células T CD8, T CD4 e NK controlando a infecção por HSV-1. / This work was carried out in the Immunopathology laboratory of the Centro de Pesquisa René Rachou (CPqRR) and Virus laboratory (Labvirus) of the Universidade Federal de Minas Gerais (UFMG). Herpes viruses have DNA genome and are enveloped. Approximately 70% of adult people have had contact with the Herpes Simplex Virus type 1 (HSV-1). After infection, HSV-1 cause cold sores, or more serious injuries such as encephalitis, and after infection, the virus usually become latent and may be reactivated or not. Cells of innate immune system recognize the “pathogen associated molecular patterns” (PAMPs) through “Toll Like Receptors” (TLRs), orchestrating the immune response against the virus. When activated, TLRs start a signalization cascade that culminates in the activation of genes related to the innate immune defense (NO, cytokines and chemokines). Previous research from our group demonstrated that knockout (KO) mice to TLR9 and TLR2/9 showed high mortality after HSV-1 infection, unlike C57BL/6 wild type (WT) and TLR2KO, which were able to control the virus. Research aimed in interrelation of the expression of TLRs 1, 2, 3, 6, 7 and 9 in TLR2 KO, TLR9 KO and TLR2 / 9 KO mice infected with HSV-1 and the main cytokines, cells and cellular immune mechanisms against infection by HSV-1. In this work, TLR2KO, TLR9KO, TLR2/9KO and C57BL/6 WT mice were intranasally infected with 106 p.f.u. of HSV-1, and their respective controls received PBS. Mice euthanasia was at the 5th day post infection (dpi), previously showed to be the viral multiplication peak in our model, and then trigeminal ganglia (TG) and brain were collected. The expression of TLRs 1, 2, 3, 6, 7 and 9 were measured by Real Time PCR. Infected WT mice showed significant increase in the expression of these TLRs compared to respective controls in TG, but not in brain.KOs mice had increased expression of TLRs in TG (but in more moderated levels compared to WT) and in the brains. The expression of gp91phox, p22phox and iNOS were also measured. iNOS expression in infected C57BL/6 WT was higher than in other groups, that not occurs to gp91phox, p22phox. In addition, NO production by intraperitoneal macrophages was measured through Griess reaction, that demonstrated a more pronounced production by WT macrophages, compared to TLR KOs groups. Histopathology and immunofluorescence assays demonstrated higher cellularity in infected TG, and besides confirmed the CD8 and macrophages presence and increase of iNOS production in infected C57BL/6 WT, what did not occurred with TLR KO mice. In survival assay with C57BL/6 WT, CCL3 KO, CD8 KO, RAG KO and iNOS KO, we showed the importance of the T cells and iNOS, because CD8 KO, RAG KO and iNOS KO had 0% of survival. C57BL/6 WT, RAG KO, CCL3 KO and iNOS KO mice had their TG analyzed by Real Time PCR, to verify the expression of viral gene VP-16 and MCP-1, iNOS, IP-10, Rantes and TNF-_ in 5th dpi. These data show us that iNOS and T cells are important in innate responses against HSV-1, once RAG KO were irresponsive to cytokines expression, while iNOS KO showed an excessive response to cytokines expression. The cytometry assay indicated that IFN_ produced by T CD8 cells, in the TG, and by T CD4 and Natural Killer cells (NK), in region lymph nodes, are a possible mechanism to control HSV-1 infection of WT infected mice. We hypothesize that coordinated expression of TLRs leads to an efficient orchestration of the innate immunity system, through cytokines and chemokines stable production and proper cellular immune activity, with NO production by macrophages and T CD8, T CD4 e NK cells action controlling HSV-1 infection.

Seu papel na proteção contra vários patógenos, incluindo protozoários está bem estabelecido. Entretanto, pouco se sabe sobre o papel dos TLRs na malária. No presente estudo, investigamos o papel dos TLRs durante a malária murina e humana. Nossos resultados mostraram que camundongos com deficiência para MyD88, um adaptador essencial para a sinalização dos TLRs, produzem níveis de citocinas pró-inflamatórias significativamente menores e apresentam sintomas mais amenos durante a infecção por Plasmodium chabaudi. Entretanto, estes animais retêm a capacidade de controlar a parasitemia sugerindo que os TLRs possuam um papel na patogênese e não na proteção contra a malária. Posteriormente, mostramos que ambas, a infecção natural humana por P. falciparum e a experimental murina por P. chabaudi, aumentam a expressão e a responsividade dos TLRs nas células do sistema imune inato. O estado hiper-responsivo das células durante a malária é derivado da ativação de TLR9 e a produção de IFN por células T, levando a uma alta susceptibilidade ao choque séptico durante a malária aguda. Finalmente, em colaboração com a EISAI Research Institute, desenvolvemos um antagonista de TLR9 e testamos seu efeito na Malária Cerebral (CM), uma das manifestações clínicas mais graves da malária. O tratamento oral com este composto inibiu os sintomas, tais como extravasamento vascular cerebral, protegendo camundongos da morte por CM. Em conjunto, nossos resultados mostram um importante papel dos TLRs, especialmente TLR9, na patogênese da malária e que a intervenção na função destes receptores é uma potencial quimioterapia anti-inflamatória contra essa doença. / Toll-like receptors (TLRs) have been involved in almost every know disease that afflict human health so far and their role in resistance to several pathogens, including protozoan parasites, has been well established. The role of TLRs in malaria, however, still remains to be elucidated. Here we studied the role of TLRs in experimental and naturally acquired malaria infection. We showed that mice with deficiency to MyD88, an essential adaptor to TLRs signaling, produce significant lower levels of proinflammatory cytokines and commensurate better clinical outcome upon infection with Plasmodium chabaudi. Nevertheless, these mice can still control parasite loads suggesting that TLRs are involved in pathogenesis rather than protection during malaria. We further studied cellular responsiveness of innate immune responses during human and murine malaria. We showed that both natural acquired P. falciparum infection in humans and experimental infection of mice with P. chabaudi increase TLR expression in innate immune cells causing pro-inflammatory priming of TLR responses. The cellular hyper-responsiveness during Malaria is caused by TLR9 activation and IFN production by T cells conferring high susceptibility to septic shock during the acute disease. Finally, in collaboration effort with EISAI Research Institute, we develop and tested an antagonist of TLR9, on Cerebral Malaria (CM), one of the most severe manifestations of malaria. Oral treatment of mice with this compound inhibited CM symptoms, such as vascular leakage, and prevented death from CM. All together our results show an important role of TLRs, especially TLR9, in malaria pathogenesis and that the therapeutic targeting of TLRs is a potential anti-inflammatory chemotherapy against malaria.
