Formulation and evaluation of castro-retentive floating tablet of griseofulvin

Thesis (M.Pharm. (Pharmaceutics) -- University of Limpopo, 2018 / Griseofulvin is an antibiotic fungistatic drug used in the treatment of dermatophyte and ringworm infections. About 50% of a dose of griseofulvin passes the gastro- intestinal tract unabsorbed and is excreted in faeces. Since griseofulvin is highly soluble in acidic pH, a gastro-retentive floating matrix system was developed to control dissolution rate and thereby enhance solubility in an effort to develop an improved and convenient dosage form. Preformulation studies included selection of excipients and evaluation of their compatibility with griseofulvin. Using the chosen excipients, floating tablets of griseofulvin were formulated. Floating tablets containing 100 mg of griseofulvin were prepared by wet granulation technique with varying ratios of Methocel™, Accurel MP and Polyvinylpyrrolidone as determined by Design Expert software. Pre and post-compression studies, buoyancy capability and dissolution studies were carried out to assess the influence of the tablet components. Results obtained revealed that a density of less than 0.00091 g/cm3 was necessary for tablet floatation. Tablets that float immediately upon contact with dissolution medium and continue floating for over 12 hours were achieved with at least 28% Accurel MP by mass of the tablet. Dissolution studies revealed that an increase in tablet hardness reduced the rate of griseofulvin release only up to 120 minutes. From 120 minutes onwards, tablet hardness had no significant influence on griseofulvin release from tablets. Methocel™ had the most significant influence on griseofulvin release. The amount of Methocel™ included in the formulation was indirectly proportional to the rate of griseofulvin release. Using Design Expert software, optimized formulation was achieved with 1% Polyvinylpyrrolidone, 30% Methocel™, 60% Accurel MP and hardness ranging between 8 – 9 N. Pre and post-compression parameters of the optimized tablets were found to be within pharmacopoeial limits and thus compressed tablets were of acceptable quality. Tablets produced floated immediately upon contact with the medium and remained floating for at least 12 hours. Griseofulvin was released from the optimized tablets in a near zero order fashion, with a total of 80.8% griseofulvin released at the end of the 12 hour dissolution test period. Results of accelerated stability studies indicated potential stability of the manufactured tablets months.

CADMIUM AND CALCIUM TRANSPORT ALONG THE GASTRO-INTESTINAL TRACT OF RAINBOW TROUT: MORE THAN “GUT FEELINGS” ON MECHANISMS OF UPTAKE

<p>Teleosts take up metals by two major pathways: gills and/or gut. Past research is heavily focused on branchial uptake despite evidence that the gastro-intestinal tract (GIT) is the dominant route in some natural environments. To address this information gap, my thesis characterizes uptake mechanisms of Cd and Ca along the GIT of <em>Oncorhynchus mykiss</em>. Toxic effects of Cd, protective effects of Ca against Cd uptake, and Cd distribution within fish after ingestion are also explored.</p> <p>Four-week dietary Cd exposure affected growth and Ca regulation, while causing toxicity at the subcellular level. Elevated Ca in diets protected against accumulation and altered subcellular handling of Cd. Pre-exposure to different diets changed unidirectional uptake and binding rates of Cd and Ca, although they remained highly correlated. Binding rates of Cd to mucus can predict absorption rates of Cd<strong>.</strong></p> <p>A variety of <em>in vitro </em>and <em>in vivo </em>experiments on four distinct GIT segments (stomach, anterior-, mid-, and posterior- intestine) were undertaken to determine specific mechanisms of Cd and Ca uptake in freshwater trout. Cd transport was unaffected by solvent drag, but was stretch- and temperature- sensitive. Strong evidence for a common pathway for Ca and Cd transport was obtained. Cd also appeared to be taken up in part by zinc (ZIP-like) transporters and the divalent metal transporter DMT1. Ca uptake along the GIT appeared to be carrier-mediated, time- and concentration- dependent, but was not affected by solvent drag, or by Na concentration. Mucosal Cd did not inhibit Ca uptake. Ca, but not Cd, uptake rates were much lower in sea water-acclimated trout. Seawater and freshwater fish accumulated similar whole body Cd concentrations when fed contaminated diets for three weeks, but the majority of Cd in seawater trout remained in the posterior intestine tissue, while freshwater trout had higher internal burdens.</p> <p>This thesis advances the knowledge of metal uptake along the GIT of trout.</p> / Doctor of Science (PhD)

Polystomes of the world (Polystomatidae: Monogenea) : an appraisal of intestinal morphology and species diversity / Michelle Delport

Species interact and exploit one another for a number of reasons, including transportation, shelter or nutrition such as in parasitic relationships. Parasitism is an important aspect in life and is common in all taxonomic groups. Parasites are often host-specific and can be endoparasites or ectoparasites. The phylum Platyhelminthes includes the class Monogenea or monogenetic parasitic flukes. Monogeneans are mainly parasitic in fish but the family Polystomatidae, also commonly referred to as polystomes, are found on the skin and gills of the Australian lungfish, tadpole gills, kidneys and urinary bladders of frogs, gills and skin of salamanders, cloaca and phalodeum of caecileans, on the eye, in the nose, mouth or urinary bladder of freshwater turtles and on the eye of the hippopotamus. Polystomes have a cosmopolitan distribution, and are found on all hospitable continents. Polystome species were first discovered in the 1758. Between 1961 and 1980 French researchers focussed on Central and West Africa and described a large number of parasites. Polystome discovery has steadily decreased in the last 30 years, however despite this, new species are still being discovered annually. The list of currently known polystomes is most likely only a small portion of the species that exists. Wherever scientists searched for polystomes, new species were discovered. The current distribution of polystomes is not at all a true reflection of their global distribution but merely an indication of research effort. Monogenean flatworms exhibit many variations in the morphology of the intestinal tract. These parasites display two distinct diets, where one group mainly feeds on blood while the other mainly feeds on mucus and epithelial tissues. Thus the feeding habits and other factors such as the shape of the caeca, the presence/absence and number of medial and lateral diverticula as well as anastomosis may play a role in the morphology of the intestinal tract, which can be used as a classification tool to classify polystome species into specific genera. This diploma thesis focuses on the description of a chosen gastro-intestinal disease - colorectal carcinome, unspecified chronic intestinal infections, and ulcerous gastroduodenal disease. It pursues the causes, symptoms, diagnosis, treatment, and most attention is given to the prevention of a chosen disease in gastro-intestinal tract. Concerning prevention, it focuses also on nutrition because that is what plays a significant role in keeping human population healthy. The empiric part of the thesis has been processed using a qualitative research survey, using the method of an in-depth semi-standardized interview. The first research group was formed by healthy individuals. The second research group was formed by nurses working with a practical doctor at the internal department and at the gastro-enterological department. The third research group was formed by patients suffering from gastroduoenal disease. The fourth research group was formed by patients sufferring from non-specific chronic intestinal infections. The fifth research group was formed by patients suffering from colorectal carcinome. The obtained results have proven that people know what a healthy lifestyle is, but compliance to it is rather difficult. One of its positive effects is that most people get their medical check-up regularly. The research group of ill individuals proves that the area of terciary or quarterly prevention is not made clear to the probands. The nurses´ research group has proven the fact that the prevention area is not made properly clear to the nurses, thus they do not provide the patients with sufficient information about their illness. This diploma thesis should aim at nurses and make sure they do not ignore the necessity of gastro-intestinal disease preention. T inform patients about options of prevention they have available. Patients themselves also need to realize how important their health is in their lives. Research survey results have been processed to form an information brochure meant for gastro-intestinal disease patients. To deepe knowledge about this issue we suggest offering the obtained results to be published in an expert magazine called Gastroenterology and Haemathology - Czech Gastroenterological Society.

Selection of probiotic lactic acid bacteria for horses based on in vitro and in vivo studies

Thesis (MSc)--Stellenbosch University, 2011. / ENGLISH ABSTRACT: The equine gastro-intestinal tract (GIT) is a relatively unexplored niche concerning the presence of natural microbiota. Studies have shown that disruption of the microbial population naturally present in the GIT leads to the onset of several forms of gastro-intestinal disorders. To maintain a balanced microbiota, probiotic bacteria need to be administered at specific levels. Beneficial microorganisms assist with digestion of the feed, absorption of nutrients from the GIT, strengthens the immune system and improves the animal‟s growth. Various combinations of lactic acid bacteria (LAB) have been administered to horses, but have failed to benefit the host in any of the latter criteria. The screening for alternative strains with probiotic properties is thus necessary. Two strains (Lactobacillus equigenerosi Le1 and Lactobacillus reuteri Lr1) were originally isolated from horse faeces. Lactobacillus plantarum 423 and Enterococcus mundtii ST4SA, both bacteriocin-producing strains, were isolated from sorghum beer and soy beans, respectively. All four strains survived growth at acidic conditions (pH 3) and the presence of 0.5%, 1.0% and 1.5% (w/v) bile salts. L. reuteri Lr1 was the most resistant to these conditions. All strains adhered to buccal (cheek) epithelium cells sampled from horses. L. equigenerosi Le1 and E. mundtii ST4SA, however, invaded the cells, but without visible signs of disrupting the cells. None of the strains contained genes encoding adhesion to collagen (Ace), resistance to vancomycin A, B and C, or, production of aggregation substance (AS), cytolysin (Cyl) and, non-cytolysin (β hemolysin III), suggesting that they are non-virulent. Of all strains, L. equigenerosi Le1 competed the best with Clostridium sp. C6 for adherence to epithelial cells. L. equigenerosi Le1 and L. reuteri Lr1, showed the highest level of co-aggregation with Clostridium sp. C6. When the four strains were administered to horses over a period of 10 days, L. reuteri Lr1 was retained the longest (8 days) in the GIT. The numbers of viable cells of Clostridium spp. and Salmonella spp. remained constant during administration of the four strains. Blood analyses showed no negative effects from administering the strains. Total white blood cell counts remained unchanged. However, a small but tentative increase in neutrophil and eosinophil cell numbers has been recorded, suggesting that the LAB may have elicited a mild, transient, intolerance reaction. The glucose, lactate and urea levels decreased during administration with the four LAB strains. / AFRIKAANSE OPSOMMING: Die spysverteringstelsel (SVS) van die perd is 'n relatief onbekende nis wat die voorkoms van natuurlike mikrobiota betref. Studies het getoon dat versteuring van die natuurlike mikrobiese populasie in die SVS aanleiding kan gee tot die ontwikkeling van menige vorms van gastro-intestinale ongesteldhede. Om 'n gebalanseerde mikrobiota te verseker, moet probiotiese bakterieë teen 'n spesifieke vlak toegedien word. Voordelige mikroorganismes bevorder vertering en absorpsie van nutriënte vanaf die SVS, versterk die immuunsisteem en bevorder die groei van die dier. Verskeie kombinasies van melksuurbakterieë is reeds aan perde toegedien, maar sonder ooglopende voordele vir die dier. Die soeke na alternatiewe stamme met probiotiese eienskappe is dus noodsaaklik. Twee melksuurbakterieë (Lactobacillus equigenerosi Le1 en Lactobacillus reuteri Lr1) is oorspronklik uit perdemis geïsoleer. Lactobacillus plantarum 423 en Enterococcus mundtii ST4SA, beide bakteriosienproduserende stamme, is afsonderlik van sorghumbier en sojabone geïsoleer. Al vier spesies groei by lae pH (pH 3) en in die teenwoordigheid van 0.5%, 1.0% en 1.5% (m/v) galsoute. L. reuteri Lr1 is die mees bestand onder hierdie toestande. Al vier stamme het aan wang epiteelselle van perde geheg. L. equigenerosi Le1 en E. mundtii ST4SA het egter die epiteelselle binnegedring, maar sonder opsigtelike vernietiging van die selle. Nie een van die stamme besit gene wat kodeer vir aanhegting aan kollageen (Ace), bestandheid teen vankomisien A, B en C, of produksie van, sel-aggregasie (AS), sitolisien (Cyl) en nie-sitolisien (β-hemolisien III), wat daarop dui dat hulle nie-virulent is. Van al die stamme het L. equigenerosi Le1 die beste met Clostridium sp. C6 vir aanhegting aan epiteelselle gekompeteer. L. equigenerosi Le1 en L. reuteri Lr1, het die beste vlak van ko-aggregasie met Clostridium sp. C6 getoon. Met die toediening van 'n kombinasie van die vier stamme aan die perde oor 'n periode van 10 dae, het L. reuteri Lr1 die langste retensie (8 dae) in die SVS getoon. Die aantal lewende selle van Clostridium spp. en Salmonella spp. het konstant gebly tydens toediening van die vier stamme. Toediening van die vier stamme het geen negatiewe effek getoon met resultate verkry van bloed analises nie. Die totale witbloed seltellings het onveranderd gebly. 'n Klein, maar tentatiewe, toename in neutrofiel- en eosinofiel selgetalle is waargeneem, wat daarop dui dat die melksuurbakterieë 'n geringe allergiese reaksie teweeggebring het. Die glukose, laktaat en ureum vlakke het gedaal tydens die toediening van die vier melksuurbakterie stamme.

O trabalho objetivou avaliar o efeito de probióticos e antibióticos utilizados como promotores do crescimento sobre o desempenho de frangos de corte e a capacidade dos agentes de alterar o ecossistema intestinal de aves criadas em baterias e sobre piso. Adicionalmente foi estudado o efeito dos probióticos sobre a presença de oocistos na cama das aves. Os antibióticos tiveram sua eficácia, como promotores de crescimento, comprovada para aves criadas sobre piso, mas não em bateria. Foram observadas alterações na estrutura da comunidade de Bacteria no trato intestinal de frangos criados em baterias e sobre piso e suplementados com antibióticos. Não houve evidência de efeito favorável dos probióticos sobre o desempenho e incidência de oocisto na cama das aves. Os probióticos não tiveram a capacidade de colonizar o epitélio intestinal de frangos de corte. Foram observadas discretas modificações na estrutura da comunidade de Bacteria de frangos criados em bateria e sobre piso e suplementados com dietas contendo probióticos. A estrutura da comunidade de Bacteria do intestino delgado de frangos foi modificada em função do ambiente no qual frangos, suplementados com probióticos e antibióticos, foram criados. Frangos isentos de qualquer tipo de promotor de crescimento apresentaram 15 unidades taxonômicas operacionais distintas na microbiota intestinal aderida ao epitélio, predominantemente Lactobacillus e Pseudomonas. / This study was conducted to evaluated the effects of growth promoter probiotic and antibiotics on the perfomance and organ morphometry of broiler chickens raised in floor pens and in batteries and the ability of the additive to promote changes in the intestinal ecosystem. Additionally, the effect of probiotics on the presence of coccidia oocysts in the litter was evaluated. The efficacy of antibiotics as growth promoters was observed when the chickens were raised in floor pens but not in batteries. Antibiotic supplementation caused changes in the structure of Bacteria community of the intestinal tract of chicken raised in floor pens or in batteries. The probiotic additives tested did not result in improvement in performance in both environmental or in oocyst incidence in the litter. Also, the probiotic did not have the ability to colonize the intestinal epithelium of the birds. Discrete changes in the structure of Bacteria community were observed when probiotics were supplemented to chickens raised in floor pens or in batteries. Bacteria community structure in the small intestine of chicken was modified as a function of the environmental in which the birds were raised. Chicken fed diets devoid of growth promoters had 15 distinct phylogenetic groups in the microbiota adhered to the intestinal epithelium.

Analyse molekularer Mechanismen der ERα- und ERβ-vermittelten Wirkung spezifischer Liganden und des Phytoestrogens Genistein

Die Behandlung menopausaler und postmenopausaler Beschwerden ist mit einem erhöhten Risiko verbunden, an Mamma- und Endometriumskarzinomen zu erkranken. Darüber hinaus zeigen epidemiologische Studien, dass in ostasiatischen Ländern postmenopausale Beschwerden, osteoporotische Frakturen und Herz-Kreislauferkrankungen seltener auftreten als in westlichen Ländern. Vor diesem Hintergrund war es Ziel der Untersuchungen dieser Arbeit, in dem Tiermodell der ovarektomierten Ratte die mögliche Bedeutung von estrogenrezeptorsubtypspezifischen Einflüssen für hormonell bedingte Erkrankungen und Beschwerden zu untersuchen. Hierbei sollten gewebespezifische Wirkungen estrogenrezeptorsubtypspezifischer Liganden untersucht und explizit die Bedeutung der Estrogenrezeptorsubtypen ERα und ERβ bei der Gewebehomöostase in Knochen und Darm analysiert werden. Darüber hinaus sollten vor dem Hintergrund estrogenrezeptorsubtypspezifischer Wirkungsweisen gewebespezifische Einflüsse des Phytoestrogens Genistein (Gen) näher charakterisiert werden. Es konnte gezeigt werden, dass nach einer subkutanen Applikation der knochenprotektive Einfluss von Gen mit dem von Estradiol (E2) vergleichbar ist, durch die Kombination mit Bewegung verstärkt wird und über den ERα vermittelt zu sein scheint. Es zeigte sich außerdem, dass der stimulierende Einfluss von E2 auf den motorischen Antrieb ERα-vermittelt ist und ERβ-spezifische Liganden ebenso wie Gen diesen Effekt antagonisieren. Des Weiteren wurde deutlich, dass E2, nicht aber Gen über den ERα Einfluss auf die Körperfettverteilung nimmt. Mit einer phytoestrogenreichen Diät konnten in adulten Ratten physiologisch relevante Gen/Dai-Plasmaspiegel erreicht werden, allerdings blieben hierbei die nach einer subkutanen Applikation beobachteten knochenprotektiven Effekte dieser Phytoestrogene aus. Bei der näheren Betrachtung der Gewebehomöostase im Dünndarm zeigte sich, dass über den im Darm verstärkt exprimierten ERβ antiproliferative und proapoptotische Effekte vermittelt werden und Gen in diesem Gewebe wie ein ERβ-spezifischer Agonist wirkt. Bezogen auf eine hormonell bedingte Osteoporose, wie sie bei einem Großteil postmenopausaler Frauen auftritt, scheint das Phytoestrogen Genistein eine mögliche Alternative zur Hormonersatztherapie darzustellen. Außerdem zeigt sich, dass Genistein gewebe- und estrogenrezeptorsubtypspezifische antagonistische und agonistische Einflüsse hat und somit die Charakterisierung als „Phyto-SERMs“ (pflanzlicher selektiver Estrogenrezeptormodulator) zutreffend ist. Sollten sich in weiterführenden Studien die beobachteten Effekte im Dünndarm auch für die Gewebehomöostase im Kolon beschreiben lassen, können vor diesem Hintergrund Genistein und ERβ-spezifische Liganden für die Darmkrebsprävention diskutiert werden…
