This thesis deal with looking for optimal conditions for determination of Sudan dyes. High performance liquid chromatography (HPLC) was used for determination. During searching several various columns and mobile phases were tested. For validation of separation conditions Ascentis Express F5 column (100 x 4,6 mm, 2,7 μm) was used. Detection was measured at a wavelength of 500 nm and 426 nm. Separation used gradient elution with mobile phase of acetonitrile - water at a flow rate 1 ml/min. Injection volume was set to 5 μl. During measuring validation parametres was used on-line SPE extraction in HPLC system on precolumn Ascentis Express C18 (5 x 4,6 mm, 2,7 μm) with washing mobile phase 5% solution of acetonitril - water before separation. In recent years, Canadian governments and courts have increasingly responded positively to the demands of lesbian and gay communities for legal rights. As a result, in several instances, such rights have been extended, at both statutory and constitutional levels. In this thesis, I consider the politics of struggles for lesbian and gay legal equality in Canada. Although I explore several developments in this area, I focus my analysis upon two key examples: the struggle, in 1986, to add a "sexual orientation" ground to Ontario's Human Rights Code; and a key legal rights case launched in the late 1980s, and still on-going as of this writing (Mossop). More specifically, I address three key questions: [1] how are lesbian and gay subjects and subjectivities constituted through human rights law and what forces produce these legal constructions? [2] how capable are liberal democracies of accommodating 'sexual pluralism', and what are the implications of this for other areas of social transformation? [3] what is the relationship between the lesbian and gay rights movement, its principal opponents the New Christian Right, and 'the state' - how do the struggles of social movements for interpretive authority shape the law-making process (and vice versa)? In responding to these questions, I draw upon diverse approaches in legal theory, sociology, feminism, and lesbian and gay studies. My analysis centres upon the role of law as a site of struggle. I explore the engagements between the lesbian and gay rights movement, and its key opponent the New Christian Right. I assess the effects of lesbian and gay rights campaigns in both the short and long terms, considering issues to do with social movement mobilisation, effective political communication, and the role of these struggles in shifting dominant frameworks of meaning. I offer a detailed discussion of the role of rights, as goal and rhetoric, within political action. And I consider the relationship between law, and other forms of knowledge. I argue that the effects of legal struggle are complex, contradictory, and unpredictable. Lesbian and gay rights reforms have both entrenched and undermined dominant paradigms of sexuality, and the effects of legal struggle in this and other areas must be assessed in the long-term. This thesis contributes to knowledge in four key areas: critical rights theory; theories of law and social change; the sociology of social movements and religions; and lesbian and gay politics. I use a combination of legal, sociological, feminist, and historical methodologies.

This work deals with suggested possibilities of cooperation between regional institutions and Elementary School Miloše Šolleho Kouřim, Kolín District in utilization of educational potential of local cultural heritage. Theoretical part firstly goes in an overview of regional literature about Kouřim and used pedagogical literature. Then follows brief history of settlement in Kouřim from origins through its prime as royal town up to its decline with emphasis on its dominants, which decorate the town until now. First part of the work is closed by chapter Pedagogical outlets, which presents five major topics for creation of historical walks. Practical part of the master's thesis presents three concrete walks, which took place in the autumn of 2010 in cooperation with Elementary School Miloše Šolleho Kouřim, Kolín District and Regional museum of Kolín, respectively with regional office in Kouřim. Corresponding chapters concern preparation, realization and valuation of assessed walks and their potential for further utilization not only for Elementary School Miloše Šolleho Kouřim, Kolín District, but also by other elementary schools from nearby and faraway as well. .

Le cinéma chinois indépendant se développe dans les années 1980, alors que la Chine vient de commencer une politique d’ouverture et de modernisation. Le pays, dont la croissance est exceptionnelle et continue, devient rapidement une puissance économique émergente. Il connaît ainsi un processus de mutations qui bouleverse ses territoires et les conditions de vie de ses populations. Jia Zhang-ke, qui est l’un des principaux cinéastes indépendants chinois, s’attache tout au long de son œuvre à filmer et à interroger ce processus de mutation. Son projet est de garder une trace de ce que les mutations détruisent, font disparaître, de ce qui le remplace, et également de ce moment même de la mutation. Pour tenter de filmer ce passage, Jia Zhang-ke met en œuvre des dispositifs filmiques d’entre-deux et d’alternance, s’enracinant dans les héritages de l’art pictural chinois et des modernités cinématographiques européennes. Son cinéma, en cela, questionne le processus de mondialisation, ou de globalisation, et participe à l’élaboration de la connaissance, en même temps qu’il peut être considéré comme une forme de résistance. / The independent Chinese cinema develops in the 1980s, while China has just begun a policy of openness and modernization. The country, the growth of which is exceptional and continuous, quickly becomes an emergent economic power. It so knows a process of transformations which upsets its territories and the living conditions of its populations. Jia Zhang-ke, who is one of the main Chinese independent film-makers, works throughout his work to film and to question this process of transformation. His project is to keep a track of what the transformations destroy, remove, of what replaces it, and also of this same moment of the transformation. To try to film this passage, Jia Zhang-ke operates cinematic devices of jump ball and alternation, taking root in the inheritances of the Chinese pictorial art and the European film modernities. His cinema, in that respect, questions the process of globalization, and participates in the elaboration of the knowledge, at the same time as he can be considered as a shape of resistance.
