Diagnostic tools were developed and utilised to detect Toxoplasma gondii infection in a range of Australian marsupial species and identify epidemiological trends. An ELISA was developed to detect anti-T. gondii IgG in macropod marsupials. When compared with the commercially available MAT (modified agglutination test), the ELISA was in high agreement and yielded a ê coefficient of 0.96. Of 18 western grey kangaroos (Macropus fuliginosus) tested for the presence of T. gondii DNA by PCR, the 9 ELISA positive kangaroos tested PCR positive and the 9 ELISA negative kangaroos tested PCR negative indicating that the ELISA protocol was both highly specific and sensitive and correlated 100% with the more labour intensive PCR assay.
A T. gondii seroprevalence study was undertaken on free ranging Australian marsupials. There was a T. gondii seroprevalence of 15.5% (95%CI: 10.7-20.3) in western grey kangaroos located in the Perth metropolitan area. The T. gondii seroprevalence in male western grey kangaroos was significantly less than their female counterparts (p=0.038), which may be related to behavioural differences causing differences in exposure to oocysts or recrudescence of T. gondii infection in pregnant females. Marsupial populations located in islands free from felids had a low overall T. gondii seroprevalence. A case control study determined that marsupials located in areas where felids may roam are 14.20 (95%CI: 1.94-103.66) times more likely to be T. gondii seropositive than marsupials located on felid-free islands.
PCR, immunohistochemistry and serological techniques were used to detect T. gondii infection in marsupial dams and their offspring. T. gondii DNA was detected in the pouch young of chronically infected western grey kangaroos and a woylie (Bettongia penicillata). T. gondii DNA was also identified in the mammary gland of the woylie dam suggesting that infection of the woylie pouch young was from suckling milk from the mammary gland. Results of the study demonstrate that vertical transmission of T. gondii occurs in Australian marsupials and may be of importance in the maintenance of T. gondii infection in Australian marsupial populations.
Animal tissue and meat from Australia, predominately from Australian marsupials, were screened for T. gondii DNA using PCR primers for the multi-copy, T. gondii specific B1 gene. Sequencing of the B1 gene revealed atypical genotypes in 7 out of 13 samples from Australia. These 7 isolates contained single nucleotide polymorphisms (SNPs) in the B1 gene that could not be matched with known sequences from strains I, II, III and X. Six unique genotypes were identified out of the 7 atypical isolates; two out of the 7 isolates had the same unique sequence at the B1 gene whereas the other 5 isolates each had different combinations of SNPs at the B1 gene. A majority of T. gondii isolates sampled from native Australian marsupials were of an atypical genotype. The discovery of atypical strains of T. gondii in Australia leads to further questions regarding the origin and transmission of these atypical strains. Additional studies linking atypical strains with their clinical manifestation are also warranted.

Jaw disease in kangaroos and wallabies, while uncommon in animals living in the free state, is a major cause of death in captive macropods. There are two major schools of thought regarding its aetiology. The first accepts the findings of Fox (1923) and considers the disease to be actinomycosis while the second believes jaw disease to be one manifestation of necrobacillosis. Regardless of the belief of the authors the clinical and pathological descriptions indicate the condition attacking macropods throughout the world is a single disease. / Over one hundred affected macropods were examined during the course of this study and few species were found to be spared from jaw disease. Cases of lumpy jaw most commonly occurred in the winter months. All ages of macropods were affected although a higher prevalence was recorded in younger animals; however this may have been a reflection of the age of macropods in the zoo under study. The most common site of infection was the premolar and anterior molar region of the oral cavity, regardless of the age of the animal. The gross pathological findings were similar to the descriptions of affected animals by various authors throughout the world. The site of infection could generally be recognized at the gingival crevice, commonly between teeth, and excessive calculus formation associated with gingivitis and gingival recession was a common finding in affected animals. Penetration of the gingival mucosae by plant fibre was not found to be a significant predisposing factor in the initiation of the disease. (For complete abstract open document.)

Effects of sheep, kangaroos and rabbits on the regeneration of trees and shrubs in the chenopod shrublands, South Australia /Palisetty, Raghunadh. Unknown Date (has links)
After European settlement, Australian rangelands especially in South Australia underwent significant changes because of the main land use of pastoralism. Many studies have revealed that the plant communities are negatively effected by herbivory mainly by sheep. The main aim of this study is to separate the different effects of sheep, rabbits and kangaroos. This was examined by survey supported by experimental and modelling research. / A 32,000 km² area previously surveyed by Tiver and Andrew (1997) in eastern South Australia was re-surveyed to monitor populations of perennial plant species at sites of various intensity of grazing by sheep, rabbits and kangaroos (goats populations are low in the study area), the most important vertebrate herbivores. Plant population data were collected in both sheep paddocks and historically ungrazed by sheep (road reserves) by using the Random Walk method and analysed using Generalized Linear Modelling (GLM) to separate the effects of sheep and rabbits on plant regeneration and their regeneration in response to grazing. These data were also compared to similar data collected by Tiver and Andrew in 1992 (1997) to ascertain if the reduction in rabbit numbers through introduction of RCV had allowed increased regeneration. Regeneration of many species inside paddocks were negatively affected and species in roadside reserves did not significantly increase from 1992 to 2004. However, some species showed increase of populations in spite of sheep grazing, with some species being less susceptible than others. This research also indicates kangaroo grazing impact on some plant species. Reduction in rabbit numbers following the 1995 release of calicivirus has not been effective in restoring regeneration. Another experiment was conducted at Middleback Field Station near Whyalla to identify herbivore grazing pressure on the arid zone plant species Acacia aneura using unfenced, sheep fenced and rabbit fenced grazing exclosures. This experiment was set up with seedlings in exclosures, ten replicates of each treatment, at plots four different distances from the watering point to identify the survivorship of seedlings. Data were collected by recording canopy volumes of seedling over an 18 month period and analysed by Residual Maximal Likelihood (REML). Seedlings both near and far from the watering point were severely affected by large herbivores, either sheep, kangaroos or both, and in a separate experiment kangaroo grazing effects on the seedling were also identified. Seedlings browsed by the rabbits recovered better than the seedlings grazed by the large herbivores. Decreasing kangaroo activities has been noticed when the rabbit movements increased. Computer modelling was conducted to predict the future plant population structure over 500 years using a matrix population model developed by Tiver et al. (2006) and using data collected in the survey as a starting point. Extinction probabilities of populations of Acacia aneura near watering points, far from watering points and under pulse grazing scenarios were compared. Sheep grazing was found to cause eventual extinction of populations in all parts of sheep paddocks. Together, the results indicate that sheep are the major herbivore suppressing regeneration of perennial plant species. Kangaroo and rabbits have an identifiable but lesser effect. The results have implications for conservation and pastoral management. To achieve ecological sustainability of arid lands a land-use system including a network of reserves ungrazed by sheep and with control of both rabbit and kangaroo numbers will be required. / Thesis (PhD)--University of South Australia, 2006.

Managing overabundant and mobile wildlife: social and institutional dimensions of kangaroo harvest in South Australia.Thomsen, Dana Arlene January 2007 (has links)
In South Australia, overabundant kangaroo populations are managed through commercial harvest. Kangaroo harvest rates over the past decade have averaged only 40% of the harvest quota despite strong demand for the product. With kangaroo populations increasing, the problem of low kangaroo harvest rate in South Australia requires research attention. Previous research regarding kangaroo harvest has focused on questions of biology and ecology and little attention has been directed towards advancing understanding of the human dimensions of kangaroo management. This research sought to fill this gap in knowledge. Qualitative research methods were most appropriate due to the focus on social and institutional dimensions of kangaroo management. Data were collected during interviews with people involved in commercial kangaroo harvest: landholders, harvesters and meat processors. The main topics covered were regnlations and policy, economic issues, the rights and interests of various industry stakeholders and South Australian harvest rates. The views of Aboriginal people were also sought including the significance of kangaroos to Aboriginal people, access to kangaroos for subsistence harvest, kangaroo management and the kangaroo industry. The main findings of this research are presented as a series of peer-reviewed articles: • Article I introduces the research topic and presents preliminary findings of this study. • Article 2 establishes that an increase in South Australia's low harvest rate is needed if kangaroo harvest is to make greater contributions to regional communities, and recommends institutional reform to meet this goal. • Article 3 examines the management regime for kangaroos in South Australia and shows how the informal rules in use are often incongruent with the formal rules established by management administrators. • Article 4 describes the undervalued position of landholders in the kangaroo industry and the obstacles to landholders deriving income from kangaroo harvest. • Article 5 discusses the cultural basis of Aboriginal perspectives on kangaroo harvest and includes suggestions for appropriate ways for Aboriginal people to contribute to kangaroo management. The main findings of this research were applied in a comparative study of kangaroo management with that of moose management in Finland. This study found that similar social and institutional factors impact on the management of moose and kangaroos. The broad lessons for wildlife management drawn from the comparative study are: • mobile wildlife resources require flexible management systems • stakeholder involvement is critical to management • hunters/harvesters are conservative of their resource base • declining harvester numbers need to be addressed through support, incentives and training. These lessons can be applied in part, or in whole, to other overabundant and mobile wildlife species. Strategic management of artificial watering points for biodiversity conservationMontague-Drake, Rebecca, School of Biological, Earth & Environmental Sciences, UNSW January 2004 (has links)
Since pastoralism began in Australia???s rangelands, the number of artificial watering points (AWPs) has increased dramatically, such that today, few areas of rangeland are further than 10 km from water. This increased availability of water has caused many ecological impacts. Unfortunately, such impacts are poorly understood in the context of an Australian conservation reserve, thus hindering strategic management. This study examined the spatial distribution of vertebrate (kangaroos, small mammals, lizards and avifauna) and vegetative variables around open AWPs as well as AWPs that have been closed since pastoralism (sheep-grazing) ceased nearly thirty years ago in Sturt National Park, arid New South Wales. The study also examined vertebrate use of AWPs, with a particular emphasis on kangaroos and avifauna. The study revealed that most variables showed few differences in spatial distribution with distance from open and closed AWPs, thus suggesting that the observed piospheric impacts were primarily attributable to historical sheep-grazing. Indeed, piospheric patterns were weak suggesting some recovery over the last thirty years. That kangaroos did not exhibit water-focused grazing is no surprise, since despite their regular use of AWPs, particularly during hot, dry times, the current spatial arrangement of AWPs facilitates regular travel to, and from, such resources allowing kangaroos, like much other fauna, to distribute themselves in relation to food and shelter preferences rather than in relation to water supply. In contrast, the majority of avifaunal groups (excluding ground-dwelling species) were clustered around open AWPs, often irrespective of season, because of food and water requirements. Such spatial concentrations of avifauna are thought to cause a range of interspecific effects. Experimental AWP closure and GIS modelling showed that whilst closure of AWPs will increase the average distance to water, which will have key benefits, the majority of areas in Sturt National Park would still be accessible to most water-dependent species even if all unused AWPs were closed. Strategic retention of AWPs to replace water sources lost since European settlement, aid threatened and migratory species??? conservation and enhance nature-based tourism opportunities is thus recommended and an example of a strategic management and monitoring plan outlined.
