Com o objetivo de desenvolver novos revestimentos mais protetores e quantificar a resistência à corrosão resultante, estudou-se a adição de caulinita em novos revestimentos híbridos a base de silanos sobre a liga de alumínio AA7075. Os revestimentos com silanos aqui estudados diferem dos relatados anteriormente na literatura, os quais usualmente contém cargas minerais apenas para a liberação de inibidores de corrosão, enquanto que os revestimentos desenvolvidos no presente estudo contém caulinita lamelar com a finalidade de barreira de difusão para espécies corrosivas. Para tanto, estudou-se primeiramente a esfoliação da caulinita e produção de filmes finos (2-3μm) por diferentes rotas de preparação pelo processo sol-gel, obtendo-se os melhores resultados com o processo envolvendo acidificação final. Os revestimentos e as partículas de caulinita processada foram caracterizados por microscopia ótica, microscopia de varredura eletrônica e difração de raios-X. A resistência à corrosão foi estudada por voltametria cíclica e cronoamperometria, em potencial aplicado entre o potencial de pite e o de repassivação. A maior resitência à corrosão por pites foi obtida para os revestimentos contendo 3% em massa de caulinita, seguido dos com 33% em massa. Estudou-se também preliminarmente a permeação de H2O no revestimento por medidas rápidas de capacitância, após exposição cíclica em soluções contendo cloreto seguidas de secagem, sendo esta metodologia empregada até o momento apenas para tintas bem mais espessas (≈ 300μm). Observou-se que o método é aplicável para medidas no primeiro ciclo de absorção de H2O, concluindo-se que após a primeira exposição, o cloreto continua difundindo, diminuindo o tempo de resistência ao pite. / Aiming to develop more protective new coatings and to quantify the resulting corrosion resistance, new hybrid coatings based on silanes with the addition of kaolinite were produced and applied on the AA7075 aluminum alloy. The studied silane coatings differ from those reported previously in the literature that usually contain mineral fillers to release corrosion inhibitors, since we here developed silane coatings contain lamellar kaolinite as a barrier against the diffusion of corrosive species. For this, we first studied the exfoliation of kaolinite, and production of thin coating (2-3μm) by different preparation routes of the sol-gel process, obtaining the best results with the process of final acidification. Coatings and processed kaolinite particles were characterized by optical microscopy, scanning electron microscopy and X-ray diffraction. The corrosion resistance was evaluated by cyclic voltammetry and chronoamperometry at potentials between that of the repassivation and of the pitting of the substrate alloy. The greatest pitting resistance was obtained for coatings with 3% kaolinite addition, followed by the ones with 33%. The permeation of H2O into the coating was also preliminarily studied by fast capacitance measurements after cyclic exposure to solutions containing chloride followed by drying, a methodology that up to date was only used for much thicker paints (≈ 300μm). It was concluded that in chloride media this method is only applicable for measurements in the first cycle, as after the first exposure, chloride ions continuously permeate into the coating during the drying phase, greatly reducing the time to pit nucleation.

Onion Root Anatomy and the Uptake of Sulphate and Phosphate Ions

Ions in the soil solution traverse many layers (epidermis, exodermis, central cortex, and endodermis) within the root to reach the stele. The endodermis is present in almost all vascular plants while the exodermis is found only in majority of angiosperm roots tested. The maturation of the exodermis and the death of epidermis alter the plasma membrane surface areas (PMSA) potentially available for ion uptake. Do these changes reduce the ion uptake in proportion to the loss of absorptive surface areas? To answer this question onion (Allium cepa L cv. Wolf) adventitious root segments representing above features: Immature Exodermis Live Epidermis (IEXLEP), Mature Exodermis Live Epidermis (MEXLEP), Mature Exodermis Dead Epidermis (MEXDEP) were excised. Using a compartmental elution technique, radioactive sulphate and phosphate present in various internal compartments were quantified. Quantities of ions moved across the plasma membrane, a summation of quantities in the cytoplasm, ‘vacuole’, and ‘bound’ compartments, indicated that the maturation of the exodermis reduces the uptake of sulphate but not phosphate. In contrast, epidermal death reduced the movement of both ions across the plasma membranes. Although there is a reduction in the available PMSA with the maturation of the exodermis and death of the epidermis, these events do not necessarily reduce the ion movement into the plasma symplast. The endodermal cells of onion roots deposit suberin lamellae as secondary walls. As seen in cross-sections some cells remain without these lamellae and are known as ‘passage cells’. What is the pattern of suberin lamella deposition along the root? Is the suberin lamella a continuous layer? To answer these questions, endodermal layers isolated from onion adventitious roots were used in the present study. These layers were observed using four stains (Sudan Red 7B, Fluorol yellow 088 [Fy], berberine, and Nile red) and three microscopes (compound-white light, compound-epifluorescence and confocal scanning). In differentiating cells with and without suberin lamellae in endodermal layers Sudan Red 7B served the best results for compound-white light microscope, Fy for compound-epifluorescence microscope and Nile for confocal laser scanning microscope (CLSM). Suberin lamellae deposition initiated almost in a random manner; they continued to be deposited resulting in the production of longitudinal files alternating with files with passage cells, and were ultimately deposited in almost all cells at a distance of 255 mm from the tip. The suberin lamellae are perforated with pores, a consistent feature even as far as 285 mm from the tip. These pores may serve as portals for water, ions, and pathogen movement.

Harmony search method based optimum topology design algorithm is presented for single layer lamella domes. The harmony search method is a numerical optimization technique developed recently that imitates the musical performance process which takes place when a musician searches for a better state of harmony. Jazz improvisation seeks to find musically pleasing harmony similar to the optimum design process which seeks to find the optimum solution. The optimum design algorithm developed imposes the behavioral and performance constraints in accordance with LRFD-AISC. The optimum number of rings, the height of the crown and the tubular cross-sectional designations for dome members are treated as design variables. The member grouping is allowed so that the same section can be adopted for each group. The design algorithm developed has a routine that build the data for the geometry of the dome automatically that covers the numbering of joints, and member incidences, and the computation of the coordinates of joints. Due to the slenderness and the presence of imperfections in dome structures it is necessary to consider the geometric nonlinearity in the prediction of their response under the external loading. Design examples are considered to demonstrate the efficiency of the algorithm presented.

Composition of lignin in outer cell-wall layers

The composition of lignin in the outer cell-wall layers of spruce and poplar has been studied and the data obtained have been compared with those of the mature reference wood in which the secondary cell wall predominates. Materials with exclusively or predominantly outer cell-wall layers were examined. Accurate data relating to the lignin monomer composition and the number of β-O-4´ bonds were obtained from pure middle lamella/primary cell wall lignin. Firstly, a 10 000 year old white spruce material, with most of the secondary cell wall missing, was studied. The aged lignin was composed of guaiacyl units only, and was slightly more condensed but otherwise similar to the reference lignin. Secondly, the developing xylem of a Norway spruce clone was analyzed during a growth season. In spring and early summer, growth is very rapid and the intention was to sample tissues in which the secondary cell-wall layers had not yet lignified, but where the outer layers at least had started to lignify. Microscopy, Klason lignin and carbohydrate analyses showed that the lignin in the developing xylem of samples from mid-June was located exclusively in the middle lamella. The lignin was more condensed, was composed of guaiacyl units only and contained more end-groups than the reference Norway spruce wood. Thirdly, the cambial tissues of a Balsam poplar clone were surveyed during a growth season. Both the phloem side and the xylem side of the cambial region were examined. The Klason lignin content and carbohydrate monomer distribution showed that in June and August the tissues on the phloem side contained material with mainly middle lamella/primary walls. In June, the xylem side in the cambial region contained mainly middle lamella/primary walls, and in August the secondary cell wall carbohydrates were being deposited. Both tissues contained lignin that was more condensed and had more end-groups than the reference lignin. In mid-June, the developing xylem had a ratio of syringyl to guaiacyl units of 0.6, whereas the ratio for the reference wood was 1.3. In the final study, lignin from the primary cell walls from a hybrid aspen cell suspension culture was investigated. O presente trabalho teve como objetivo o estudo da potencialidade de aplicação da flotação por ar dissolvido (FAD) lamelar (de alta taxa) e convencional como parte final de sistema de tratamento sequencial de esgoto sanitário, constituído de reatores anaeróbios de manta de lodo (UASB) seguidos de tanque de aeração. O trabalho, desenvolvido na Estação de Tratamento de Esgotos Jardim das Flores da cidade de Rio Claro, foi dividido em três etapas. Na primeira etapa, realizada com equipamento Flotateste, foram investigados os valores mais adequados de dosagens de polímero sintético catiônico, amido de araruta, cloreto férrico e associações entre cloreto férrico e polímero ou amido de araruta, de razão ar/sólidos (A/S), bem como de tempo e gradiente de mistura rápida e de floculação. Nessa etapa, os melhores desempenhos da FAD foram obtidos com o emprego de polímero catiônico, tanto isoladamente quanto associado ao cloreto férrico, sendo esta última condição imprescindível para maior remoção de fósforo. A flotação de liquor misto utilizando apenas o cloreto férrico como coagulante, apresentou desempenho extremamente insatisfatório, com a formação de flocos com características ruins de flotabilidade. Além disso, observou-se que a flotação desse tipo de efluente requisitou condições mais amenas de mistura, sendo que, em muitos casos, os melhores resultados foram obtidos mediante a eliminação da mistura rápida ou da floculação. Nas segunda e terceira etapas, realizadas com unidades piloto de flotação lamelar de alta taxa (FADAT) e convencional, respectivamente, e empregando apenas polímero como auxiliar de floculação/flotação (dosagem em torno de 1 g de polímero/kg de SST), foram investigados os valores mais adequados de Taxa de Aplicação Superficial (TAS), Taxa de Aplicação de Sólidos (TS) e quantidade de ar fornecida à flotação, calculada através da razão A/S ou da dosagem de ar (Dar), em g de ar/'M POT.3' de esgoto afluente, dependendo da concentração de sólidos suspensos totais afluentes à flotação (SSTafl). Através dos resultados dos ensaios pôde-se observar que a razão A/S é mais adequada para o cálculo da quantidade de ar na flotação de suspensões concentradas (SSTafl acima de 1100 mg/L no presente estudo), ao passo que Dar é mais representativo no caso da flotação de suspensões diluídas. Empregando a unidade FADAT, alimentada com liquor misto de tanque de aeração em boas condições de biofloculação, foi possível obter resultados satisfatórios para TAS de até 350 'M POT.3'/'M POT.2'.dia e TS entre 120 e 250 kg de SST/'M POT.2'.dia, desde que fornecida quantidade adequada de ar ao processo (Dar na faixa de 15,0 a 18,0 g de ar/'M POT.3' de afluente), para SSTafl entre 800 e 1100 mg/L. Na flotação lamelar de alta taxa, constatou-se que o bom desempenho esteve diretamente relacionado a baixos valores de SSTafl. As recomendações para a flotação com unidade de FAD convencional tiveram que ser mais conservadoras, devido às condições ruins de biofloculação apresentadas pelo liquor misto do tanque de aeração durante a realização da terceira etapa do trabalho. Nesses ensaios, os resultados mais satisfatórios foram obtidos com o emprego de TAS em torno de 200 'M POT.3'/'M POT.2'.dia, TS menor que 200 kg de SST/'M POT.2'.dia e A/S em torno de 0,020 (Dar de 21,0 g de ar/'M POT.3' de afluente), para SSTafl em torno de 1400 mg/L / The aim of this research was the study of the lamella design (high rate) and conventional dissolved air flotation (DAF) process potentiality of application as the final part of a domestic sewage sequential treatment system made up anaerobic reactors (UASB) followed by aeration tank. The work, developed at the Jardim das Flores WWTP of Rio Claro city, was separated in three phases. In the first phase, using the flotatest apparatus, the most suitable values of chemical (synthetic cationic polymer, arrow-root starch, ferric chloride and ferric chloride combined with polymer or arrow-root starch) dosages, air to solids ratio (A/S), as well as rapid mix and flocculation time and mean velocity gradient values, were investigated. In this phase, the best DAF performances were obtained with the application of cationic polymer, alone and associated to ferric chloride. The use of ferric chloride was essential to obtain better phosphorus removal results. The mixed liquor flotation using ferric chloride alone as primary coagulant presented extremely unsatisfactory performance, with the formation of flocks showing bad characteristics of flotability. Moreover, it was observed that flotation of this kind of effluent had requested gentler mixture conditions. In many cases, the best results were obtained by means of rapid mix or flocculation step elimination. In the second and third phases, using the lamella design and conventional flotation units respectively and cationic polymer as the only flocculation/flotation aid (dosage around 1 g of polymer/kg TSS), the most suitable values of Overflow Rate (OR), Solids Application Rate (SAR) and amount of air supplied to the flotation process, expressed as the A/S ratio or air dosage (AD, in g of air/'M POT.3' of influent) depending on influent total suspended solids concentration (TSSinfl), were investigated. The flotation essays results showed that the A/S ratio is more suitable for the air supply estimation in flotation of concentrated suspensions (TSSinfl above 1100 mg/L in this study). Contrarily, the parameter AD is more representative in cases of flotation of diluted suspensions. Using the lamella design unit, fed with aeration tank mixed liquor presenting good bio-flocculation conditions, it was possible to obtain adequate results applying OR as high as 350 'M POT.3'/('M POT.2'.day) combined with SAR values between 120 and 250 kg of TSS/('M POT.2'.day), since the adequate amount of air has been provided for the process (AD values between 15,0 and 18,0 g of air/'M POT.3' of influent), for TSSinfl between 800 and 1100 mg/L. For the lamella flotation, it was noticed that the best performance has been directly associated to lower values of TSSinfl. The recommendations for flotation using the conventional DAF unit had to be more conservative, taking into account the poor bio-flocculation conditions presented by the mixed liquor during the third phase of work. In theses essays, the more adequate results were obtained by applying OR around 200 'M POT.3'/('M POT.2'.day), SAR under 200 kg of TSS/('M POT.2'.day) and A/S ratio around 0,020 (AD of 21,0 g of air/'M POT.3' of influent), for TSSinfl around 1400 mg/L
