Estudos relatam que os ácidos graxos livres estimulam a secreção de insulina através da ativação do GPR40. Diante destes fatos, investigamos a secreção de insulina, a expressão de proteínas da via do GPR40 nas células ß e a participação dos lipídios na resistência à insulina induzida por dexametasona, através do tratamento com o redutor de lipídios bezafibrato. Os grupos receberam gavagem uma vez ao dia durante 28 dias: Controle (CTL) e DEXA com goma arábica 5% (1 ml/kg, peso corpóreo); BEZA e BEZA-DEXA com bezafibrato (300 mg/kg, p.c.). Nos últimos 5 dias de tratamento os grupos receberam injeções intraperitoniais: CTL e BEZA de solução salina (1 ml/kg, p.c.); DEXA e BEZA-DEXA de dexametasona (Decadron® 1,0 mg/kg, p.c). A secreção de insulina estimulada por glicose aumentou nos grupos BEZA e DEXA. BEZA-DEXA exibiu diminuição dos níveis de ácidos graxos livres, triglicérides e de insulina, mas não houve elevação dos níveis de glicose no sangue. Além disso, houve melhora na resistência à insulina e restauração do padrão de secreção de insulina, em comparação ao grupo DEXA. Nas ilhotas dos animais BEZA-DEXA a expressão das proteínas GPR40, PLCß1 e PKCδ foi significativamente maior em relação aos valores obtidos em DEXA. Esta via permaneceu inalterada nas ilhotas de DEXA e BEZA. Em conclusão, o tratamento com bezafibrato melhorou a função das células ß e impediu a indução de resistência à insulina pelo tratamento com dexametasona, mas os mecanismos não são conhecidos. O aumento na secreção de insulina em DEXA aparentemente não está relacionado com a ativação do GPR40. Contrariando a literatura, apesar da redução na secreção de insulina, as ilhotas dos animais BEZA-DEXA apresentaram ativação da via do GPR40 / Increased glucose-stimulated insulin secretion is an adaptive mechanism exhibited by pancreatic islets from insulin resistant animal. Studies report that the free fatty acids stimulate the insulin secretion via GPR40. As such, we investigate the expression of GPR40 in ß-cells and the involvement of lipids in dexamethasone-induced IR, by lipid-lowering therapy with bezafibrate. Groups received once daily gavage for 28 days: Control (CTL) and DEXA with gum Arabic 5% (1.0 mg/kg, body weight); BEZA and BEZA-DEXA with bezafibrate (300 mg/kg, b.w.). In the last 5 days of the treatment groups received intraperitoneal injections: CTL and BEZA of saline (1.0 mg/kg, b.w.); DEXA and BEZA-DEXA of dexamethasone (Decadron® 1.0 mg/kg, b.w.). The glucose-stimulated insulin secretion increased in the DEXA and BEZA groups. BEZA-DEXA shows decrease in fatty acids, triglycerides and insulin levels, but not raised blood glucose levels. In addition, there was improved in insulin resistance and restoration the insulin secretory pattern, when compared to DEXA group. In BEZA-DEXA islets, GPR40, PLCß1 and PKCδ protein content was significantly higher than DEXA. This pathway remained unchanged in DEXA and BEZA islets. In conclusion, bezafibrate treatment improved ß-cell function and prevented dexamethasone-induced IR, but the mechanisms are not known. Augmented insulin secretion in DEXA appears to be unrelated to the activation of the GPR40. Contrary to the literature, despite the reduction in insulin secretion, BEZA-DEXA islets showed activation of the GPR40 pathway

Previous issue date: 2015-02-27 / FACEPE / BRAF é conhecido como um proto-oncogene que apresenta diversas mutações,
sendo a V600E a mais comumente encontrada em neoplasias humanas. A proteína
BRAF quinase oncogênica tornou-se um alvo para terapia específica em oncologia
por fazer parte de vias importantes como a Ras-MEK-ERK. A mutação BRAFV600E
tem sido encontrada em pacientes com melanoma, câncer de tireóide, colorretal e
neoplasias hematológicas. No entanto, a importância dessa molécula em carcinoma
tireoidiano papilar e Histiocitose das células de Langerhans ainda não é muito clara.
Para isso, modelos murinos para essas doenças foram desenvolvidos para testar a
hipótese de que a mutação V600E do gene BRAF em camundongos é suficiente
para induzir características fenotípicas semelhantes a observadas em pacientes com
câncer tireoidiano papilifero (PTC) e Histiocitose das células de langerhnas (LCH).
Nos modelos de PTC e LCH, o gene BRAF mutado é expresso nas células
foliculares da tireóide (animal chamado TCB) e nas células do compartimento
mielóide CX3CR1+ (animal chamado FRBRAF), respectivamente. Após 90 dias de
administração de tamoxifeno para induzir a mutação, histologicamente, os animais
TCB apresentam tireóide com hiperplasia folicular, protusões papilares e
recrutamento leucocítico; enquanto os animais FRBRAF apresentam
hepatoesplenomegalia com infiltrados inflamatórios no fígado, pulmão e baço.
Análise de citometria de fluxo demonstrou aumento no número de linfócitos T
predominantemente T CD4+ com presença de células Tregs Foxp3+ e Th17 na
tireóide transformada em camundongos TCB. Mesma analises, em camundongos
FRBRAF revelou a expansão do compartimento mielóide. Expressão de citocinas e
quimiocinas são características comuns aos dois modelos animais como é o caso de
CCL2, 5, 6, 8, 9/10, 17, 22 CXCL-1, 2 na tireóide de camundongos TCB e TNFα, IL-
1α, M-CSF, IL-7, CCL17 no fígado de animais FRBRAF. Com destaque, observamos
que no modelo de LCH, células CD11b+MHCII+Langerin+, marcador comum da
lesão em humanos, também foram encontradas no fígado desses animais. Dessa
forma, concluímos que a expressão de BRAFV600E em camundongos é capaz de
gerar fenótipo semelhante ao de pacientes com câncer tireoidiano papilifero com
infiltrado inflamatório e Histiocitose das células de langerhnas no figado. Esses
resultados implicam diretamente na importância de BRAF nesses doenças. / BRAF is known as a proto-oncogene that has multiple mutations, V600E is
commonly found in human cancers. The oncogenic protein kinase BRAF has
become a target for specific therapy in oncology because of their involvemnt in the
Ras-MEK-ERK pathway. The BRAFV600E mutation has been found in patients with
melanoma, thyroid, colorectal cancer and hematologic malignancies. However, the
importance of this molecule in papillary thryoid cancer and Langerhans cell
histiocytosis is still not very clear. For this, murine models for these diseases have
been developed to test the hypothesis that the BRAF V600E mutation in mice is
sufficient to induce a similar phenotypic characteristics observed in patients with
papillar thyroid cancer (PTC) and langerhnas cell histiocytosis (LCH). In these murine
models of PTC and LCH, BRAF activacted is expressed in the thyroid follicular cells
(TCB mouse), and in the myeloid cells CX3CR1+ (FRBRAF mouse), respectively.
After 90 days of administration of tamoxifen to induce mutation, histologically, TCB
thyroid had follicular hyperplasia, papillary protrusions and leukocyte recruitment;
while FRBRAF mice had hepatosplenomegaly with inflammatory infiltrates in the
liver, lung and spleen. Flow cytometric analysis demonstrated an increase in the
number of T lymphocytes CD4+ predominantly Foxp3+ regulatory T cells and Th17
cells inside the TCB thyroid. Similar analyse in FRBRAF mice showed an expansion
of the myeloid compartment. Expression of cytokines and chemokines are common
features in two models such as CCL2, 5, 6, 8, 9/10, 17, 22 CXCL-1, 2, in TCB
thyroid and TNF, IL-1α, M-CSF, IL-7, CCL17 in FRBRAF liver. Remarkably,
CD11b+MHCII+ Langerin+ cells, typically found within LCH lesions in humans, were
also present in the liver infiltrates. Thus, we conclude that the expression of
BRAFV600E in mice is able to resemble human PTC and LCH. These results directly
implicate BRAFV600E in both diseases.

Previous issue date: 2007 / Objetivo: Comparar a densidade das Células de Langerhans (CL) no colo uterino de
mulheres HIV-soropositivas em diferentes graus de Neoplasia Intraepitelial Cervical (NIC).
Método: Foram avaliadas 55 mulheres infectadas pelo HIV com alterações colposcópicas e
histocitológicas para detecção de NIC e densidade das CL entre março de 2005 e março de
2006. As CL foram identificadas utilizando coloração imunohistoquímica com proteína S-
100 . A reação em cadeia de polimerase foi utilizada para detectar o HPV-DNA cervical. A
associação entre a densidade das CL, achados histológicos, contagem de linfócitos T CD4+,
carga viral do HIV, detecção do HPV-DNA e tabagismo foram efetuados utilizando a
comparação de médias pelo teste de Kruskall-Wallis e coeficiente de correlação de
Spearman.
Resultado: A média de CL nas mulheres infectadas pelo HIV (n=51) foi de 1,90
células/mm2. Não foi possível corar a lâmina de quatro mulheres e uma das mulheres foi
excluída do estudo devido a diagnóstico de Carcinoma Cérvico-uterino Invasivo. A média
das CL por campo microscópico foi de 1,94 células/mm2 nas NIC de baixo grau (n=24) e
2,23 células/mm2 nas de alto grau (n=11), porém esta diferença não foi estatisticamente
significante (p=0,65). A biópsia resultou negativa/metaplásica em 20 casos. Não houve
associação significante entre as médias das CL quando comparadas entre os diversos tipos
de HPV (p=0,68), tabagismo (p= 0,58) e carga viral do HIV (p= 0,77). Ocorreu um maior
número das CL quando a contagem de linfócitos T CD4+ foi menor que 200 células/mm2,
sem diferença estatisticamente significante (p=0,08), mas o valor do p foi próximo ao
ponto de corte (0,05).

Conclusão: Existe um maior número de CL no colo uterino de mulheres HIV-soropositivas
com NIC de alto grau quando comparadas com as com NIC de baixo grau. O aumento
destas células com o agravamento das NIC sugere que existe resposta do sistema imune
contra células com atipias mais acentuadas em mulheres HIV soro-positivas

An investigation of Langerhans' cell function in aged skinOgden, Stephanie January 2013 (has links)
With increasing age, aspects of the innate and adaptive immune systems show functional decline. In the skin this is associated with an increased incidence of epidermal malignancies and infections, a decreased incidence of contact allergy, and the development of autoimmunity. The mechanisms underlying these clinical effects in aged skin are poorly understood. Langerhans’ cells (LCs), which are members of the wider family of dendritic cells (DCs), reside in the epidermis where they act as sentinels of the immune system by processing and presenting antigen and inducing T cell responses. Previous investigations have suggested that the number of epidermal LCs is reduced, and that the motility of LCs is impaired in aged skin. A series of investigations was performed to characterise the mechanistic basis for the reduced frequency and restricted mobility of epidermal LCs in the skin of the elderly. Initially LC-like cells were cultured from circulating monocyte precursors and characterised using flow cytometry. The ability of precursors to differentiate into LC-like cells was not impaired in the aged; furthermore there were no age-associated differences in expression of markers of LC activation at baseline or upon stimulation. The phenotype of epidermal LCs was assessed using flow cytometric analysis of epidermal cell suspensions and did not appear altered in aged individuals. In addition, using the same techniques with dermal cell suspensions the dermal DC population was not altered with age. Langerhans’ cell migration from epidermal explants prepared from the skin of aged individuals was impaired but could be restored with exogenous interleukin (IL)-1β. There was no age-related reduction in the epidermal levels of IL-1β or caspase-1 (IL-1β converting enzyme which converts pro-IL-1β to the active form) or the expression of the IL-1 receptor I (IL-1RI), to account for this observation. However, the amount of IL-1 receptor antagonist was reduced in aged skin suggesting a change in the overall local cytokine balance. Based on previous reports that topical retinoic acid (RA) can increase cutaneous IL-1 production, a 4-day patch test assay was performed using 0.025% all-trans RA cream to explore whether this could restore LC migration in the aged. There was no effect on LC migration from epidermal explants prepared after treatment with RA in the aged.These data demonstrate that changes in LC function in the elderly may not be associated with changes in systemic DC biology. Age related changes in the cutaneous microenvironment are likely to be more relevant.
