O tratamento desta doença pode ser realizado através da quimioterapia, uma alternativa muito importante. Desde a descoberta da atividade antitumoral da cisplatina, a investigação a respeito do uso de complexos metálicos na quimioterapia do câncer cresceu de forma relevante, isto porque a cisplatina possui elevada atividade, mas em contrapartida, severos efeitos colaterais. Portanto, visando à melhoria do espectro de atividade antitumoral, diminuição dos efeitos colaterais e a resistência celular, a síntese de complexos de platina têm sido de grande interesse em pesquisas científicas. Neste cenário, o presente trabalho visa obter novos complexos de platina com potencial atividade biológica, sua incorporação em lipossomas, visando um aumento desta potencial atividade. Os complexos apresentam ligantes diaminados N-alquilados com cadeia carbônica longa. Uma série de complexos apresenta um grupo hidroxila terminal no ligante e a outra série consiste de complexos dinucleares de platina. Os compostos sintetizados foram caracterizados por espectroscopia na região do IV, RMN de 1H, de 13C e de 195Pt e análise elementar e tiveram sua citotoxicidade investigada em células tumorais e células não tumorais. Tendo em vista que nosso grupo de pesquisa tem investigado complexos de platina com ligantes diaminados N-alquilados com cadeias carbônicas longas, nosso objetivo é estudar a atividade biológica destes novos compostos, já que a presença da hidroxila terminal poderá afetar a polaridade do complexo e, conseqüentemente, sua interação com biomomoléculas e a membrana celular. / Cancer is considered a public health problem, accounting for about 12% of all causes of death around the world. The treatment of this disease can be achieved using chemotherapy, a very important alternative. Since the discovery of the antitumor activity of cisplatin, studies involving the use of metal complexes as drugs have grown substantially, since cisplatin shows high activity, but on the other hand, severe side effects. Therefore, aiming to improve the spectrum of antitumor activity, reduce these side effects and cellular resistance, the synthesis of platinum complexes have been of great interest in scientific research. Within this context, the principal objective of the present work is to obtain novel platinum complexes with potential biological activity, to incorporate these complexes into liposomes, and hoping for a potential increase in their biological activity. The complexes present long alkyl chain diamine as ligands. The first series of complexes incorporates a terminal hydroxyl substituent and the second series are constituted of dinuclear complexes. The synthesized complexes were characterized by IR, 1H, 13C and 195Pt NMR spectroscopies, elemental analysis and their cytotoxicities have been investigated in both normal and tumor cells. Our research group has previously studied complexes employing long chain, N-alkyl derivatives of diamines as ligands. We are interested in investigating the impact of the new ligands on the activity of the complexes, considering that the presence of the hydroxyl group could affect the polarity of the complex and consequently, its interaction with biomolecules and the cellular membrane.
