Previous issue date: 2010 / Resumo: Na planta modelo Arabidopsis thaliana o desenvolvimento do fruto foi amplamente estudado e uma série de genes foram relacionados com a ontogênese e o amadurecimento. A identidade dos tecidos dos frutos secos de Arabidopsis é determinada principalmente pelos genes FRUITIFULL (FUL) e SHATTERPROOF (SHP), pertencentes à família multigênica MADS-box. Homólogos a estes genes foram encontrados em frutos carnosos, no entanto a expressão destes homólogos e o papel biológico dos mesmos durante o desenvolvimento dos frutos carnosos são desconhecidos. No banco de dados de genes expressos em Citrus (CitEST), sequências homólogas a FUL e SHP foram encontradas. Uma vez que os frutos de Citrus são carnosos e não apresentam as mesmas estruturas dos frutos de Arabidopsis, decidiu-se caracterizar detalhadamente o desenvolvimento dos frutos em Citrus sinensis e Citrus reticulata e estudar as possíveis funções dos homólogos de FUL e SHP durante o desenvolvimento dos frutos em C. sinensis. Técnicas de microscopia óptica e eletrônica de varredura foram utilizadas para caracterização do desenvolvimento dos frutos. Resultados de RT-PCR mostraram a expressão diferencial dos homólogos de Citrus de FUL e SHP em frutos maduros. Os resultados de hibridização in situ mostraram que o homólogo de FUL expressou-se diferencialmente durante o desenvolvimento do fruto, principalmente nas células secretoras das cavidades oleíferas e das vesículas de suco, nos tecidos do epicarpo, mesocarpo e endocarpo. O homólogo de SHP também se expressou no epicarpo, mesocarpo, endocarpo e nas vesículas de suco. Os resultados obtidos poderão auxiliar num melhor entendimento do desenvolvimento do fruto em Citrus. / Abstract: Fruit development is extensively studied in the model plant Arabidopsis thaliana and a number of genes is being related to the control of fruit ontogenesis and ripening. The FRUITFULL (FUL) and SHATTERPROOF (SHP) genes, which belong to the MADS-box multigenic family are, in large part, responsible for determining the identity of the Arabidopsis dry fruit tissues. Homologs to these genes have been described for species harbouring fleshy fruits, but the expression patterns and biological functions for these genes during fruit development are unknown. Sequences showing similarity to FUL and SHP were found at the database of the CitEST Project (expressed sequence tags in Citrus). Since Citrus fruits are fleshy and do not have the same structures of the Arabidopsis fruits, we decided to characterize in detail the development of fruits in Citrus sinensis and Citrus reticulata and to study the possible functions of FUL and SHP homologues during the development of C. sinensis fruits. These sequences were characterized by bioinformatic's tools and phylogenetic analysis. The use of light and scanning electron microscopy (SEM) techniques allowed the morpho-anatomical characterization of Citrus fruit development. The RT-PCR results showed a preferential expression of FUL and SHP in developing fruits. The in situ hybridization results showed that the C. sinensis homolog of the FUL gene is differentially expressed in the secretory cells of the oil cavity as well as in the juice vesicles, epicarp, mesocarp and endocarp tissues. The homolog of the SHP gene also expressed in epicarp, mesocarp, endocarp and juice vesicles. All the results taken together might contribute to a better understanding of the processes involved in Citrus fruit development. / Mestrado / Mestre em Biologia Vegetal

Characterization of sterile tassel silky earl: A Homeotic B-Class Gene Involved in Specification of Floral Organ Identity In Zea maysWilliams, Steven Keith 12 December 2012 (has links) (PDF)
Specification of floral organ identity in angiosperm flowers is accomplished by the coordinated activity of A-, B-, C-, and E-class MADS-box genes. In the eudicots, B-class genes specify petal and stamen identity. This eudicot B-class function depends on the simultaneous expression of genes from two paralogous B-class lineages (the DEFICIENS/APETALA3 lineage and the GLOBOSA/PISTILLATA lineage). Proteins produced by genes from these two lineages interact as obligate heterodimers and together regulate the transcription of various downstream targets. These obligate heterodimers also positively regulate the transcription of the B-class genes themselves, thereby mediating a unique B-class autoregulatory feedback loop. There is compelling evidence that B-class function at the phenotypic and molecular level is highly conserved among the eudicots. The degree to which B-class homeotic function, obligate heterodimerization, and autoregulation are conserved in non-eudicot, however, remains a topic of debate. Here we describe loss of function in Sterile tassel silky ear1 (Sts1) a maize ortholog of GLOBOSA/PISTILLATA formerly known as Zmm16. Mutation in Sts1 results in homeotic transformation of lodicules and stamens into bract-like organs in male inflorescences. Female inflorescences are affected in a similar manner. Stamens in these inflorescences are, however, transformed into carpels instead of into bract-like organs. This mutant phenotype suggests that Sts1 has a B-class homeotic function. Using qRT-PCR we also demonstrate that Sts1 participates in positive transcriptional regulation of all of the maize B-class genes. These findings suggest a high degree of B-class functional conservation between the monocots and the eudicots. Analysis of tasselseed1/sts1 and grassy tillers1/sts1 double mutants suggests that maize B-class genes also play a role in the sex determination process.

Previous issue date: 2011 / Resumo: A família dos genes MADS-box codifica fatores de transcrição que atuam como reguladores importantes em muitas etapas no desenvolvimento de diversos organismos. Em plantas, estes genes estão envolvidos na determinação da identidade dos meristemas reprodutivos e dos órgãos florais, bem como no controle de diversos processos durante o desenvolvimento. O presente trabalho teve como objetivo estudar o padrão de expressão dos prováveis ortólogos dos genes do modelo ABC (APETALA1, APETALA3, PISTILATA e AGAMOUS de Arabidopsis thaliana, e do gene TM6 de Solanum lycopersicum) em Coffea arabica L. Estes genes pertencem à família MADS-box e estão relacionados à determinação da identidade dos órgãos florais na planta-modelo A. thaliana. A partir do banco de dados de sequências expressas de cafeeiro (CAFEST), foram identificados 23 possíveis homólogos de genes MADS-box em cafeeiro. Perfis de expressão por RT-PCR indicaram que a maioria destes genes são expressos em flor e fruto. A análise dos dados gerados pelo uso de microscopia óptica e de varredura permitiu estabelecer uma sequência de desenvolvimento para estabelecimento dos órgãos florais em cafeeiro, facilitando a identificação dos locais de expressão dos ortólogos do modelo ABC pela técnica de hibridização in situ. Sendo C. arabica uma espécie relativamente recente e com características peculiares, foi proposto um mecanismo de atuação dos genes do modelo ABC. Dessa forma, os resultados obtidos contribuem para a compreensão do estabelecimento dos órgãos florais em C. arabica. Adicionalmente, pela caracterização de um número elevado de genes da família MADS, foram identificados outros genes potencialmente envolvidos em outros processos de desenvolvimento, que futuramente poderão ser utilizados para incremento da indústria cafeeira. / Abstract: The MADS-box gene family encodes transcription factors that act as key regulators in many steps in the development of various organisms. In plants, these genes are involved in determining the identity of reproductive meristems and floral organs as well as in controlling several processes during development. This work aimed to study the expression patterns of putative orthologs of the ABC model genes (APETALA1, APETALA3, AGAMOUS and PISTILATA from Arabidopsis thaliana, and TM6 from Solanum Lycopersicum) in Coffea arabica L. These genes belong to the MADS-box family and are related to the determination of floral organ identity in the model plant A. thaliana. From the CAFEST database of expressed sequence tags, 23 MADS-box gene sequences were identified in coffee. Expression profiles of these genes, determined by RT-PCR, indicated that most of these genes are expressed in flowers and fruits. The analysis of data from optical microscopy and scanning electron microscopy allowed the establishment of a developmental sequence for the establishment of floral organ, facilitating the characterization of the spatial expression patterns of orthologs of the ABC genes by in situ hybridization. A diversified role of conserved genes of the ABC model was proposed for the relatively recent and peculiar specie that is C. arabica. The obtained results aid the understanding of the establishment of floral organs in C. Arabica.
