No. of bitstreams: 0 / A proteína jun é um dos principais integrantes do complexo AP-1 e está envolvido nos processos inflamatórios, diferenciação, apoptose e migração celular. Esta proteína pode formar homodímeros e heterodímeros por meio da dimerização que ocorre pelo sítio de sequências de leucinas. Existem evidências de que a proteína jun pode ser inibida pela proteína RPL10 mediante a ligação destas proteínas, no mesmo sítio de sequências de leucinas no núcleo celular, parando a progressão de tumores. O objetivo deste trabalho foi expressar, isolar e caracterizar a região de ligação das sequências de leucinas (região AP-1), para estudos posteriores de ligação com a proteína RPL10. O cDNA para proteína jun foi amplificado por PCR e clonado nos vetores de expressão pET 26b(+), pET 28a-c(+) e p1813 e expressa em E.coli BL21 (DE3). A proteína expressa em vetor pET28_AP1 foi eficientemente purificada pela técnica de cromatografia de afinidade a íons metálicos, por possuir uma sequência (poli)histidina que facilitou a purificação, apresentando um excelente grau de pureza. Analysis of Biogenic Amines by GC/FID and GC/MSNakovich, Laura 18 September 2003 (has links)
Low levels of biogenic amines occur naturally, but high levels (FDA sets 50 ppm of histamine in fish as the maximum allowable level) can lead to scombroid poisoning. Amines in general are difficult to analyze by Gas Chromatography (GC) due to their lack of volatility and their interaction with the GC column, often leading to significant tailing and poor reproducibility. Biogenic amines need to be derivatized before both GC and HPLC analyses. The objective of this research was to develop a relatively fast, reproducible method to derivatize and quantitate biogenic amines in fish at trace levels using GC/FID. The derivatizing reagent used in the experiments was propyl chloroformate, useful for aqueous samples. To confirm the identity of six derivatized biogenic amines GC/MS was used. To our knowledge no reference spectra for these derivatives has been published.
It was concluded that best results are obtained using a Cold-On-Column (C.O.C.) inlet with a short column (15 meters), thick film stationary phase (ZB-5, 1.00μm df), and with recommendations to cut 40 cm from the inlet end of the column every 25 injections when using C.O.C. Duplicate samples of Atlantic Salmon were analyzed on days 0, 3, and 5. Levels of histamine were below 50 ppm for days 0 and 3, but day 5 showed average levels of 160 pm (cadaverine), 1000 ppm (histamine), and 350 ppm (tyramine). Good precision of six amine stardards at 50 ppm was shown: heptylamine 5.2%, putrescine 5.6%, cadaverine 5.0%, histamine 9.9%, tyramine 5.1%, and spermidine 6.2% RSD. / Master of Science

A detector upgrade for phase-imaging ion cyclotron resonance measurements at the CPTMorgan, Graeme Edward Baglow 23 March 2016 (has links)
A position-sensitive microchannel plate (MCP) detector has been installed at the Canadian Penning Trap (CPT) mass spectrometer located at the CAlifornium Rare Isotope Breeder Upgrade (CARIBU) facility at Argonne National Laboratory in order to carry out Phase-Imaging Ion Cyclotron Resonance (PI-ICR) measurements. With this new measurement method, proof-of-principle mass measurements of five nuclei were made to a precision of $\delta m/m \approx 10^{-7}$. The PI-ICR results are found to be consistent with previous Time-of-Flight Ion Cyclotron Resonance (ToF-ICR) measurements.
The content of this thesis covers the entire mass measurement process beginning with beam production at CARIBU through to ion detection at the CPT and a comparison of the ToF-ICR and PI-ICR measurement methods. The future of mass measurements at the CPT with this new technique will also be discussed. / May 2016

Fatty acid and lipid profiles in models of neuroinflammation and mood disorders : application of high field NMR, gas chromotography and liquid chromotography-tandem mass spectrometry to investigate the effects of atorvaststin in brain and liver lipids and explore brain lipid changes in the FSL model of depressionAnyakoha, Ngozi Gloria January 2009 (has links)
Lipids are important for the structural and physiological functions of neuronal cell membranes. Alterations in their lipid composition may result in membrane dysfunction and subsequent neuronal deficits that characterise various disorders. This study focused on profiling lipids of aged and LPS-treated rat brain and liver tissue with a view to explore the effect of atorvastatin in neuroinflammation, and examining lipid changes in different areas of rat brain of the Flinders Sensitive Line (FSL) rats, a genetic model of depression. Lipids and other analytes extracted from tissue samples were analysed with proton nuclear magnetic resonance spectroscopy (1H-NMR), gas chromatography (GC) and liquid chromatography-tandem mass spectroscopy (LC/ESI-MS/MS). Changes in the lipid profiles suggested that brain and liver responded differently to ageing and LPS-induced neuroinflammation. In the aged animals, n-3 PUFA were reduced in the brain but were increased in the liver. However, following treatment with LPS, these effects were not observed. Nevertheless, in both models, brain concentration of monounsaturated fatty acids was increased while the liver was able to maintain its monounsaturated fatty acid concentration. Atorvastatin reversed the reduction in n-3 PUFA in the aged brain without reducing brain and liver concentration of cholesterol. These findings further highlight alterations in lipid metabolism in agerelated neuroinflammation and show that the anti-inflammatory actions of atorvastatin may include a modulation of fatty acid metabolism. When studying the FSL model, there were differences in the lipid profile of different brain areas of FSL rats compared to Sprague-Dawley controls. In all brain areas, arachidonic acid was increased in the FSL rats. Docosahexaenoic acid and ether lipids were reduced, while cholesterol and sphingolipids were increased in the hypothalamus of the FSL rats. Furthermore, total diacylglycerophospholipids were reduced in the prefrontal cortex and hypothalamus of the FSL rats. These results show differences in the lipid metabolism of the FSL rat brain and may be suggestive of changes occurring in the brain tissue in depression.
