<p>This thesis work consisted of the development of a virtual instrument that automates enzyme activity measurements and spectrum measurements with the spectrophotometer UVmini-1240. The purpose was to expand the functionality of the instrument, to eliminate the human error and to decrease the amount of time spent on measurements. A PC was connected to the UVmini-1240 via a RS-232C interface and the cell position and temperature was regulated with a CPS-240A controller. The new interface allows all the parameters to be set in the same place. It allows a visualization of the continuous monitoring of the sample absorbance and the option to save the data for post-processing. Also a module for measuring the spectrum of a sample in the wavelength range of 190 nm to 1100 nm is included. The graphical programming language LabView was used to develop the virtual instrument. This thesis work also contained measurement series of the catalase enzyme activity. These were carried out to determine the best storage temperature for the catalase solution and to determine the optimal surrounding temperature for the highest activity in the catalase solution. The conclusions were that the activity does not change considerably the first week of storage, not matter the temperature, and that the activity goes down when the surrounding temperature reaches above 30° C. These measurements were part of a bigger project to develop an ultrasonic method for measuring enzyme activity at the Institute of Acoustics at C.S.I.C in Madrid.</p>

The development of resistance measurement techniques is very important for characterization of future nanoelectronics. Pulsed IV measurement techniques are very useful for accurate resistance measurements on nanoscale samples because of the efficient removal of e.g. EMF errors. In the project we have designed a pulsed IV-setup based on a state-of-the art current source (6221) and nanovoltmeter (2182A) from Keithley, and used the setup for resistance measurements on ferromagnetic samples. Two different samples were investigated using the pulsed IV system – ferromagnetic wires with a central nanoconstriction and amorphous microwires. We have tested the pulse delta system with different pulse widths, duty cycles and voltage levels. The results show a successful integration of the setup. From the measurement results we confirm that the pulse delta system provides accurate measurements with a low noise of about 0.02Ω. The resistance of the samples increases approximately quadratically with bias which is interpreted as a heating effect due to the very high current density of about 107A∙cm-2.

Project Completion Report, OWRT Project No. A-040-ARIZ / Agreement No. 14-31-0001-4003 / Project Dates: July, 1972 - June, 1974 / Acknowledgement - The work upon which this report is based was supported by funds provided by the United States Department of the Interior, Office of Water Research and Technology, as authorized under the Water Resources Research Act of 1964. / Hydraulic and physicochemical measurements were made on treated sewage effluent releases at established locations within the channel of an ephemeral stream, the Santa Cruz River of Southern Arizona. Water quality samples were taken in sequence so that incremental flows at different hydrograph stages could be traced as the effluent moved downstream. Hydrographs obtained from two H -L flumes were used to calibrate a modified kinematic wave model. Hydraulic parameters from the kinematic model and physicochemical measurements from water quality samples were combined together into a statistical-empirical kinetic model of nitrogen transformations which may occur in sewage effluent releases. There was fair agreement between the measured data and the nitrogen species values calculated with the model. Measured nitrogen species values indicated that the rate of nitrification in sewage effluent releases is related to flow distance and physical characteristics of the stream.
