A resistência aos antibióticos é um problema de saúde pública global com sérias consequências para o tratamento de várias infecções bacterianas. Os fatores de transcrição da família MarR têm sido descritos controlando resistência a antibióticos e vários outros processos em bactérias. Neste trabalho, estudamos mecanismos de resistência a antibióticos em Chromobacterium violaceum, uma bactéria Gram-negativa ambiental que pode atuar como um patógeno oportunista em humanos. A estratégia envolveu a varredura de um painel de treze linhagens mutantes de fatores de transcrição da família MarR de C. violaceum disponíveis, por testes de susceptibilidade a 24 antibióticos. Estes ensaios revelaram que apenas o mutante ?emrR apresentou resistência aumentada ao antibiótico ácido nalidíxico em relação à linhagem selvagem. Esta resistência aumentada do mutante ?emrR ao ácido nalidíxico foi revertida em uma linhagem complementada deste mutante, conforme verificado por ensaios de viabilidade, ensaios de difusão em disco e concentração inibitória mínima (MIC). O fenótipo de diminuída produção de violaceína deste mutante, observado em meio líquido, também foi complementado. Além disso, foi realizado o isolamento de mutantes espontâneos de C. violaceum resistentes a ácido nalidíxico com mutação pontual em emrR. Os ensaios de microarranjo de DNA mostraram que EmrR reprime algumas dezenas de genes, incluindo o operon emrCAB, o qual codifica a bomba de efluxo EmrCAB. Os ensaios de Northern blot confirmaram que o EmrR reprime o operon emrCAB, e que a expressão desta bomba é induzida por salicilato, mas não outros compostos, como ácido nalidíxico ou brometo de etídeo. Os ensaios de alteração de mobilidade eletroforética (EMSA) mostraram que a proteína EmrR purificada se liga diretamente às 8 regiões promotoras de emrR, emrCAB e vários outros genes do regulon EmrR, para exercer uma regulação negativa direta sobre esses genes. Um mutante nulo ?emrCAB foi obtido, mas a ausência desta bomba de efluxo não tornou C. violaceum mais susceptível ao ácido nalidíxico, sugerindo que ela é importante somente em condições nas quais é induzida. Estas condições indutoras talvez incluam estresse oxidativo, uma vez que enzimas antioxidantes são parte do regulon de EmrR e a proteína EmrR formou dímeros covalentes na presença de agentes oxidantes in vitro. MarR family transcription factors have been described controlling antibiotic resistance and several other processes in bacteria. In this work, we studied mechanisms of antibiotic resistance in Chromobacterium violaceum, an environmental Gramnegative bacterium that can act as a human opportunistic pathogen. The strategy involved a screening of an available collection of thirteen C. violaceum mutant strains of MarR family transcription factors, by susceptibility testing for 24 antibiotics. These assays revealed that only the ?emrR mutant showed increased resistance to the antibiotic nalidixic acid in relation to the wild-type strain. This increased resistance of the ?emrR mutant to nalidixic acid was reversed in a complemented strain of this mutant, as verified by viability, disk diffusion, and minimal inhibitory concentration (MIC) assays. The phenotype of decreased violacein production of this mutant, observed in a liquid medium, was also complemented. In addition, it was performed the isolation of spontaneous mutants of C. violaceum resistant to nalidixic acid with a point mutation in emrR. DNA microarray assays showed that EmrR represses a few dozen of genes, including the emrCAB operon, which encodes the EmrCAB efflux pump. Northern blot assays confirmed that EmrR represses the emrCAB operon and that the expression of this pump is induced by salicylate, but not other compounds, such as nalidixic acid or ethidium bromide. Electrophoretic mobility shift assays (EMSA) showed that the purified EmrR protein binds directly to the promoter regions of emrR, emrCAB and several other genes of the EmrR regulon, to exert a direct negative regulation of these genes. A ?emrCAB null mutant strain was obtained, but the absence of this efflux pump did not make C. violaceum more susceptible to nalidixic acid, suggesting that it is important only under conditions in which it is induced. These inducing conditions may include 10 oxidative stress since antioxidant enzymes are part of the EmrR regulon and the EmrR protein has formed covalent dimers in the presence of oxidizing agents in vitro. Therefore, our data reveal that point mutations or molecules such as salicylate abolish the repressive activity of the EmrR transcription factor on the emrCAB operon, causing overexpression of the EmrCAB efflux pump and increasing the resistance to nalidixic acid in C. violaceum.

Neste trabalho, identificamos de modo global os fatores de transcrição da família MarR envolvidos na virulência do patógeno oportunista de humanos Chromobacterium violaceum. Usando mutagênese por troca alélica, geramos mutantes nulos não polares para doze dos quinze reguladores da família MarR encontrados no genoma de C. violaceum. Em ensaios de virulência, quando injetados por via intraperitoneal em camundongos BALB/c, os mutantes ?CV_0210 (?ohrR), ?CV_0577 e ?CV_2726 foram menos virulentos, enquanto o mutante ?CV_1776 foi mais virulento, quando comparados à linhagem selvagem. The CV_1810 is a repressor of a regulon that comprised two divergent operons encoding enzymes that possibly metabolize aromatic compounds, but catabolic products of these compounds did not function as ligands capable of antagonizing the repression of CV_1810 on the CV_1801 gene. The regulon of the activator CV_2726, defined as fourteen differentially expressed genes commonly found in assays in the absence and overexpression of the CV_2726 gene, revealed few genes (cstA) with potential to be involved in the phenotype of lower virulence of the ?CV_2726 mutant strain. Regulators CV_0577 and CV_1776 were allocated in the urate-responsive UrtR subfamily and probably afect the virulence of C. violaceum with overlapping regulons. The CV_1776 regulon contains dozens of genes, many of them related to amino acid catabolism, but there are few candidates for classical virulence factors (pecM, escU). Some genes related to catabolism/utilization of purine (CV_0578 and CV_3771) were 12 regulated by both CV_1776 and CV_0577 and responded to the presence of urate. The transcriptional profile of the adaptive response of C. violaceum to CHP, a ligand that oxidizes the OhrR regulator, revealed the upregulation of genes related to the detoxification of peroxides (antioxidant enzymes and thiol-reducing systems), degradation of the aromatic moiety of CHP (oxygenases), and protection against other secondary stresses (DNA repair, heat shock, iron limitation, and nitrogen starvation responses). The OhrR regulon was shown to be small, including two upregulated genes, CV_0209 (ohrA) and CV_0208 (putative diguanylate cyclase), and three downregulated genes (hemolysin, chitinase, and collagenase) in the ?ohrR mutant. Thus, the attenuated virulence of the ?ohrR mutant might be related to the increased production of the second messenger cyclic di-GMP (c-di-GMP) and the decreased expression of extracellular enzymes required for tissue dissemination, in this mutant strain. In conclusion, we have defined the transcriptional response to CHP, identified potential virulence factors such as diguanylate cyclase as members of the OhrR regulon, and shown that C. violaceum uses the transcription factors of the MarR family CV_0577, CV_1776, CV_2726 and OhrR to modulate its virulence.

This paper describes two methods for constructing a depth map from images. Each method has two stages. First, one or more needle maps are determined using a pair of images. This process employs either the Marr-Poggio-Grimson stereo and shape-from-shading, or, instead, photometric stereo. Secondly, a depth map is constructed from the needle map or needle maps computed by the first stage. Both methods make use of an iterative relaxation method to obtain the final depth map.

Interested in acquiring tracts of land from the Canadian Government, the Temperance Colonization Society hoped to one day establish a new colony free from the temptations of alcohol and the troubles associated with older colonies. By 1884, a settlement was established along the south shore of the South Saskatchewan River. This was the beginning of Saskatoon.<p>
As Saskatoon grew from a small settlement founded on temperance ideals to a recognized municipal corporation, the meaning of the material culture associated with this transition also changed. Two archaeological sites pertaining to this transition, the Marr Residence at 326 11th Street East (FaNp-5) and the 11th Street Privy site (FaNp-31), currently comprise the only excavated privy assemblages in the city and hold rich potential for shedding light on urban consumption behaviour at the turn of the 20th century. This study will analyze the archaeological assemblages recovered from these excavations under the scope of consumer behaviour. By orienting the essence of this study towards an archaeology of consumerism, information regarding the dimensions of everyday life in the Saskatoon area at the turn of the 20th Century can be ascertained.

Effects of Work Exposure on Maximum Acceptable Repetition Rates in a Manual Torquing TaskKant, Ravi 30 August 2007 (has links)
Repetitive and forceful exertions have been dentified as an important risk factor for occupational injuries. One method used to determine appropriate exposures to these and other risk factors is psychophysics, which is based on individual perceptions of task demands and/or risk. Effects of work exposure have been indicated as of potential importance, but have not been well studied. Indications from an earlier study related to psychophysical limits for a repetitive manual torquing task were that five days of work conditioning had minimal effects on resulting Maximum Acceptable Repetition Rates (MARR). However, it is unknown whether and how longer work exposure durations might influence MARRs. The current study investigated the effects of work exposure on MARR and adjustment time over 10 working days (two weeks) with two days of rest after five days. Ten participants (five males and five females) performed a manual torquing (45 Nm load) task at mid-chest level in the coronal plane for a one hour test session. Starting repetition rate for each participant was set at single high and low rate on alternate days. Temporal (exposure) effects were determined, where day of exposure was the independent variable, and MARR and adjustment time were the dependent variables. Final MARRs were relatively lower during the first few exposure days (14 - 15 repetitions/min) and increased for days 5 - 7 (16 - 18 repetitions/min). On average participants made four adjustments to reach MARR . Day was not found to significantly affect MARR, though week affected both MARR and adjustment time. Thus, an exposure of two weeks may be needed to obtain stable and valid psychophysical limits for manual torquing and, perhaps, related tasks. / Master of Science
