Avaliação do comportamento biológico de materiais endodonticos utilizando duas linhagens celulares

A proposta deste estudo foi avaliar in vitro, em nível molecular, os possíveis efeitos citotóxicos e genotóxicos de materiais endodônticos sobre cultura de células pulpares humanas transformadas (tHPC) e fibroblastos V79. Os espécimes de hidróxido de cálcio (HC, Hydro C, Dentsply, Brasil), MTA cinza (Angelus, Brasil), MTA branco (Angelus, Brasil) e polímero da mamona (POM, Poliquil, Brasil) foram misturados e colocados em meio de cultura (91,6 mm2 superfície /ml) por 24h em 37°C. Vitrebond (3M ESPE) foi utilizado como grupo controle positivo. As tHPC foram expostas aos extratos e diluições dos materiais e após 24h, a sobrevivência celular foi determinada pelo teste de cristal violeta. A liberação de espécies reativas de oxigênio (ROS) em tHPC foram mensuradas após 1h de exposição aos materiais. ROS foram detectados pelo manchamento das células com H2DCF-DA e a fluorescência foi mensurada por citometria de fluxo (FACS). A genotoxicidade foi determinada pela formação de micronúcleos em células V79 após 24h de exposição ao HC, MTA cinza, POM e etilmetano sulfanato (controle positivo). O atraso do ciclo celular normal de células V79 foi analisado, após a exposição ao MTA cinza, HC, POM e TEGDMA (controle positivo) por FACS. As diferenças entre a taxa de sobrevivência celular, fluorescência, número de micronúcleos e distribuição de células no ciclo celular foram estatisticamente analisadas pelo teste de Mann-Whitney-U (p<0,05). A citotoxicidade em relação ao grupo controle decresceu na seguinte ordem Vitrebond > HC > MTA branco = MTA cinza >>> POM. O POM causou indução da proliferação celular, enquanto o HC diminuiu a sobrevivência celular significativamente (p<0,05)... / Endodontic materials could release toxic components that may interact with pulp or periapical tissues. The purpose of this study was evaluated the cytotoxicity and genotoxicity of endodontic materials on transformed human pulp-derived cells (tHPC) and fibroblasts V79. Specimens of calcium hydroxide (Hydro C, Dentsply- Brazil), MTA gray (Angelus-Brazil), MTA white (Angelus-Brazil), and castor oil bean (COB, Poliquil-Brazil) were mixed and subsequently extracted in culture medium (91,6 mm2 sample surface/ml) for 24h at 37°C. Vitrebond (3M ESPE) served as a positive control. Exposure of tHPCs to dilutions of the extracts was stopped after 24h, and cell survival was determined photometrically after staining the cells with crystal violet. The generation of reactive oxygen species (ROS) was determined after a 1h exposure of the cells to extracts. ROS were detected by staining the cells with H2DCF-DA, and fluorescence was measured by flow cytometry (FACS). The genotoxicity as indicated by the formation of micronuclei was determined in V79 cells after a 24h exposure period. The modification of the normal cell cycle was analyzed by FACS. Differences between cell survival rates and fluorescence levels were statistically analyzed (Mann-Whitney-U test, p<0.05). The ranking of the cytotoxic effects based on EC50 values was as follows: Vitrebond > Hydro C > MTA gray = MTA white >>> COB. Extract of COB even induced cell proliferation, but Hydro C decrease cell survival in a doserelated manner. ROS production was not detected with any experimental material (p>0,05). Vitrebond increased ROS production about sevenfold (p<0,05). The increase in the number of micronuclei and cell cycle delay in V79 cells were not detected with any experimental material in relation to control group.
