Desenvolvimento somático, neurocomportamental e fertilidade da plole de ratos exposta ao antimoniato de meglumina pela via transplacentária e leite materno / Somatic development, and neurobehavioral plole fertility of rats exposed to meglumine antimoniate transplacentally and breast milk

No. of bitstreams: 0 Previous issue date: 2010 / Embora os antimoniais pentavalentes (antimoniato de meglumina, AM, e estiboglucanato de sódio) tenham sido introduzidos no mercado em meados dos anos 1940, eles ainda são medicamentos de primeira linha no tratamento das leishmanioses. Sendo um fármaco antigo para uma das doenças mais negligenciadas, há importantes lacunas na base de dados não clínicos de segurança disponível para o AM (e.g. estudos de toxicidade reprodutiva). Neste estudo investigamos se o tratamento com AM durante a gestação e lactação prejudicaria o crescimento pós-natal, a maturação neurocomportamental e a fertilidade da prole na vida adulta. Ratos Wistar foram tratados com AM (0, 75, 150 and 300 mg Sb+5/kg peso corporal/dia, via sc) durante a gravidez (dia 1 de gravidez em diante), parto e lactação até o desmame (dia 21 pós-natal, 21PN). A prole das mães tratadas foi avaliada quanto ao crescimento pós-natal, maturação somática e neurocomportamental (dias nos quais marcos do desenvolvimento apareceram, e teste do campo aberto nos dias 25 e 60 PN). O protocolo do estudo foi aprovado pelo Comitê de Ética de Pesquisas do Uso de Animais da FIOCRUZ (CEUA, L0016/08). As ratas tratadas com AM não exibiram deficits de ganho de peso ou outros sinais de toxicidade materna. A duração da gravidez, o tamanho da ninhada, e o peso dos filhotes ao nascimento não foram alterados pelo tratamento com AM. O peso da prole nos grupos tratados com as duas maiores doses foi reduzido nas dias 50 e 60 PN. Os marcos do desenvolvimento físico e reflexos, a maturação sexual e a fertilidade na vida adulta não foram afetados. A prole feminina das mães tratadas com as duas maiores doses apresentou uma exploração vertical (frequência de levantar) reduzida no teste do campo aberto realizado no dia 60 PN. O peso do fígado discretamente menor foi achado em machos e fêmeas mortos na idade adulta. Considerados em conjunto, os resultados desse estudo sugeriram que, exceto por alguns poucos efeitos de menor monta em relação ao peso corporal e desempenho no teste do campo aberto no dia 60 PN, notados apenas na prole feminina, o tratamento materno com AM, no intervalo de doses testado, durante toda a gestação e lactação, não afetou o desenvolvimento, maturação e fertilidade da prole exposta. / Although having been introduced in the market in mid 1940s, pentavalent antimony drugs (meglumine antimoniate, MA, and sodium stibogluconate) are still first choice drugs for treatment of leishmaniasis. Being a rather old drug for one of the most neglected diseases, important gaps are found in the non-clinical safety data base available for MA (e.g., reproductive toxicity studies). In this study we investigated whether maternal treatment with MA during gestation and lactation impaired the postnatal growth, neurobehavioral maturation and fertility of the offspring in the adulthood. Wistar rats were treated with MA (0, 75, 150 and 300 mg Sb+5/kg body wt/day by sc route) during pregnancy (from gestation day 1 onwards), parturition and lactation until weaning (postnatal day 21, 21PN). The offspring of treated dams was evaluated regarding postnatal growth, somatic and neurobehavioral maturation (day on which developmental landmarks appeared and open field test on 25 and 60 PN). The study protocol was approved by the Ethical Committee on the Use of Animals of FIOCRUZ (CEUA, L0016/08). Dams treated with MA did not show body weight gain deficits or other signs of maternal toxicity. Duration of pregnancy, litter size and pup body weight at birth were not altered by maternal treatment with MA. Pup body weight in groups exposed to the two highest doses was reduced on 50 e 60PN. The milestones of physical and reflexes development, sexual maturation, and fertility in adulthood were not affected. Female offspring from mothers treated with the two highest doses showed a decreased vertical exploration (rearing up frequency) in the open field test on 60 PN. A slightly lower liver weight was found in males and females euthanized in adulthood. Taken together, results from this study suggested that, except for a few minor effects in female body weight and open field behavior on 60 PN, maternal treatment with MA, in the dose range tested, during the whole pregnancy and suckling periods, did not affect the development, maturation and fertility of the exposed offspring.
