Neste caminho, o presente trabalho descreve o uso da macrófita, Typha sp, também conhecida como taboa, para remover as espécies metálicas Cu (II), Cr (III), Cd (II) e Zn (II) em amostras aquosas. Caracterizou-se inicialmente a macrófita Typha sp utilizando as técnicas analíticas como: espectroscopia na região do infravermelho (FT-IR), análise elementar de N e Ressonância magnética nuclear (RMN) de 13C, microscopia eletrônica de varredura (MEV), espectroscopia de energia dispersiva de raio X (EDS) e análise de área superficial utilizando adsorção de N2. As características adsortivas da Typha sp foram estabelecidas através de experimentos em batelada em função do pH, o tempo de contato e a concentração. As micrografias da Typha sp apresentou-se partículas com morfologia heterogênea, irregular em formato de bastão. O FT-IR e o RMN 13C confirmaram a presença de grupos funcionais como COOH, NH2 e S=C=N, que possuem pares de elétrons não ligantes e que coordenam com as espécies metálicas. A área superficial específica da Typha sp foi menor que 1 m2/g, característico de materiais não porosos. Os experimentos de adsorção mostraram que o pH 5,5 favorece a adsorção das espécies metálicas em estudo. (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: Biosorbents from lignocellulosic materials are low cost alternatives and more environmentally sustainable in the removal of metal species. In this way, the present work describes the use of the macrophyte, Typha sp, also known as Taboa, to remove the Cu (II), Cr (III), Cd (II) and Zn (II) metal species in aqueous samples. Typha sp macrophyte was initially characterized using analytical techniques such as infrared spectroscopy (FT-IR), elemental analysis of N and 13C nuclear magnetic resonance (NMR), scanning electron microscopy (SEM), energy spectroscopy dispersive X-ray diffraction (EDS) and surface area analysis using N2 adsorption. The adsorptive characteristics of Typha sp were established through batch experiments as a function of pH, contact time and concentration. The micrographs of the Typha sp presented particles with heterogeneous morphology, irregular in stick format. FT-IR and 13C NMR confirmed the presence of functional groups such as COOH, NH2 and S = C = N, which have non-binding electron pairs and coordinate with the metallic species. The specific surface area of Typha sp was less than 1 m2/g, characteristic of non-porous materials. The adsorption experiments showed that pH 5.5 favors adsorption of the metal species under study. The kinetics of adsorption is fast, being less than 15 min, adjusting to the experimental data of the kinetic model of pseudo-second order. Previous issue date: 2007 / Resumo: Neste trabalho foram preparadas redes poliméricas com propriedades elastoméricas a partir do poli(dimetilsiloxano) (PDMS), modificadas com íon piridínio ou etilenodiamina. Os materiais preparados foram caracterizados por espectroscopia no infravermelho, análise termogravimétrica, calorimetria diferencial de varredura e ressonância magnética nuclear de C e Si. Os resultados destas análises mostraram que ambos os materiais apresentam boa estabilidade térmica e uma temperatura de transição vítrea de ~ 160 K. Os dados de RMN indicaram que rede foi formada pela reticulação das cadeias lineares de PDMS por ¿clusters¿ de siloxanos do tipo T, provenientes da hidrólise/condensação de 3-cloropropiltrimetoxissilano e do 2-aminoetil-3- aminopropiltrimetoxissilano. Além disso, por ensaios de intumescimento foi calculada a massa molecular média entre os nós da rede polimérica e também a densidade de reticulação dos materiais. Os valores encontrados sugerem um alto grau de reticulação para ambos os materiais estudados. A rede modificada com etilenodiamina apresentou a maior densidade de reticulação. Com intuito de avaliar a potencialidade dos materiais como adsorventes de íons metálicos de uma solução etanólica, foram obtidas isotermas de adsorção. Para o material modificado com o íon piridínio os metais estudados foram o Co, Cu e Fe, que foram retirados da solução com boa eficiência, 0,25, 0,29 e 0,45 mmol g, respectivamente. Os haletos metálicos foram adsorvidos pela rede polimérica modificada na forma de complexos aniônicos FeCl4¨, CuCl4 e CoCl4. Os resultados de UV-Vis sugeriram que os complexos formados na interface têm uma simetria tetraédrica ou tetraédrica distorcida. Para a rede modificada com etilenodiamina foram estudados os metais Cu, Ni e Fe. Neste caso o material apresentou uma elevada capacidade de adsorção para os cloretos de Fe (1,35 mmol g) e Cu (1,25 mmol g), enquanto que, para o Ni a capacidade de adsorção foi consideravelmente menor, 0,36 mmol g. Os resultados de espectro eletrônico para este material sugeriram que o cobre é adsorvido pela rede polimérica modificada na forma do complexo, [Cu(en)], com uma estrutura tetraédrica distorcida, enquanto que para o níquel os resultados sugeriram formação de um complexo octaédrico. Os dados obtidos nas isotermas de adsorção foram usados para calcular as constantes de equilíbrio para a adsorção dos íons metálicos. Estes resultados permitiram verificar que no material modificado com o íon piridínio a adsorção de CoCl2 e CuCl2 segue o modelo descrito por Langmuir. Para a adsorção do FeCl3 outro modelo, em que centro adsorvedor é composto por dois grupos ligantes, foi necessário para descrever o processo de adsorção. Neste caso, há um efeito cooperativo entre as espécies formadas na superfície. Para o material modificado com etilenodiamina, os cálculos mostraram que a adsorção dos três íons metálicos pode ser descrito adequadamente pelo modelo de Langmuir / Abstract: In this work elastomeric polymeric networks based on poly(dimethilsiloxane) PDMS, modified with pyridine ion or ethylenediamine, were prepared. These materials were characterized by infrared spectrometry, thermogravimetric analysis, differential scanning calorimetry and nuclear magnetic resonance (NMR) of Si and C. The results showed that both materials presented a good thermal stability and a glass transition temperature at ~ 160 K. The NMR data indicated that the polymeric network was formed by PDMS linear chain crosslinked with siloxane clusters of T Si units formed from hydrolyses/condensation of 3-chloropropyltrimethoxysilane or 2-aminoethyl-3- aminopropyltrimethoxysilane. Using the swelling measurements, the average molecular weight between crosslinking points and the polymeric network crosslinking density were calculated. The results obtained suggested a high degree of crosslinking for the prepared materials, which was higher for the ethylenediamine. To evaluate the potential use of the prepared materials as adsorbents, adsorption isotherms were obtained from an ethanolic solution. For the polymeric network modified with pyridine ion the metallic ions studied were Cu, Co and Fe, which were adsorbed with high efficiency of 0.25, 0.29 e 0.45 mmol g, respectively. Metal halides were adsorbed by the modified polymeric network as an anionic complex, FeCl4¨, CuCl4 and CoCl4. The UV-Vis data suggested that the formed complexes at surface presented a tetrahedral symmetry or distorted tetrahedral symmetry. For The polymeric network modified with ethylenediamine, the metallic ions studied were Cu, Ni and Fe. In this case, the material showed a higher capacity of adsorption for Fe (1.35 mmol g) and Cu (1.25 mmol g), while, to Ni that capacity was significantly lower, 0.36 mmol g. The electronic spectra data suggested that the copper was adsorbed by the modified polymeric network as [Cu(en)], with a distorted tetrahedral symmetry, while the nickel was adsorbed with a octahedral symmetry. From the isotherm data, the equilibrium constants of the metallic ion adsorption were calculated. These results allowed verifying that the adsorptions of CoCl2 e CuCl2 are appropriate described by Langmuir equation. For the FeCl3 another model, which two adsorptions centers are envolved was necessary to describe the adsorption process. In this case, there was a cooperative effect among the adsorbed species. An investigation into the efficacy of eggshells as a low cost adsorbent for the removal of potentially toxic inorganic elements from aqueous solutionsMashangwa, Terence Dambudzo 09 1900 (has links)
Excessive release of metals and metalloids into the environment is a major global environmental concern and there is need for environmentally friendly and cost effective methods for their removal. The present study investigated the adsorptive removal of zinc, lead, copper and nickel ions from synthetic aqueous solutions and various metals from three acid mine drainage (AMD) sites using chicken eggshells. Process parameters including the initial metal concentration were evaluated and the optimum conditions obtained were pH 7, adsorbent dose of 7 g and contact time of 360 minutes (for the removal of 100ppm metal ions). Under these conditions the percentage adsorptions were; 97% for lead, 95% for copper, 94% for nickel and 80% for zinc. Aluminium, iron, potassium, nickel and zinc ions all had percentage adsorptions above 75% in AMD Sample 1. Potassium had a 98.78% adsorption while magnesium, strontium and zinc had 72.33; 68.75 and 53.07% adsorption respectively in Sample 2. Arsenic, chromium, copper, iron, antimony and tellurium ions were above 75% for Sample 3. The study demonstrated the efficacy of chicken eggshells and presents it as a viable low-cost adsorbent for bioremediation. / Environmental Science / M. Sc. (Environmental Science)

Bioprocess development for removal of nitrogenous compounds from precious metal refinery wastewaterManipura, Walappuly Mudiyanselage Janakasiri Aruna Shantha Bandara January 2008 (has links)
Removal of nitrogenous compounds from precious metal refinery (PMR) wastewater is important in terms of avoiding eutrophication (environmental protection), metal recovery (increased overall process efficiency and value recovery) and reuse of treated water (maximum use of natural resources). Extreme pH conditions (4 to 13 depending on the wastewater stream), high chemical oxygen demand (> 10,000 mg/I), numerous metals and high concentrations of those metals (> 20 mg/l of platinum group metals) in the wastewater are the main challenges for biological removal of nitrogenous compounds from PMR wastewater. Nitrogenous compounds such as NH₄⁺-N and N0₃-N are strong metal ligands, which make it difficult to recover metals from the wastewater. Therefore, a bioprocess was developed for removal of nitrogenous compounds from carefully simulated PMR wastewater. A preliminary investigation of metal wastewater was carried out to determine its composition and physico-chemical properties, the ability to nitrify and denitrify under different pH conditions and denitrification with different carbon Source compounds and amounts. Even at pH 4, nitrification could be carried out. A suitable hydraulic retention time was found to be 72 hours. There was no significant difference between sodium acetate and sodium lactate as carbon sources for denitrification. Based on these results, a reactor comparison study was carried out using simulated PMR wastewater in three types of reactors: continuously stirred tank reactor (CSTR), packed-bed reactor (PBR) and airlift suspension reactor (ALSR). These reactors were fed with 30 mg/l of Rh bound in an NH₄⁺ based compound (Claus salt: pentaaminechlororhodium (III) dichloride). Total nitrogen removal efficiencies of > 68 % , > 79 % and > 45 % were obtained in the CSTR, PBR and ALSR, respectively. Serially connected CSTR-PBR and PBR-CSTR reactor configurations were then studied to determine the best configuration for maximum removal of nitrogenous compounds from the wastewater. The PBR-CSTR configuration gave consistent biomass retention and automatic pH control in the CSTR. Ammonium removal efficiencies > 95 % were achieved in both reactors. As poor nitrate removal was observed a toxicity study was carried out using respirometry and the half saturation inhibition coefficients for Pt, Pd, Rh and Ru were found to be 15.81, 25.00, 33.34 and 39.25 mg/l, respectively. A mathematical model was developed to describe the nitrogen removal in PMR wastewater using activated sludge model number 1 (ASMl), two step nitrification and metal toxicity. An operational protocol was developed based on the literature review, experimental work and simulation results. The optimum reactor configuration under the set conditions (20 mg/I of Rh and < 100 mg/I of NH₄⁺-N) was found to be PBR-CSTR-PBR process, which achieved overall NH₄⁺-N and N0₃⁻-N removal efficiencies of > 90 % and 95 %, respectively. Finally, a rudimentary microbial characterisation was carried out on subsamples from the CSTR and PBRsecondary. It was found that the CSTR biomass consisted of both rods and cocci while PBRsecondary consisted of rods only. Based on these experimental works, further research needs and recommendations were made for optimisation of the developed bioprocess for removal of nitrogenous compounds from PMR wastewater.

Genomic characterisation and antimicrobial resistance profiles of Listeria monocytogenes isolated from pig farmsMasemola, Puseletso Maselepe 07 1900 (has links)
Listeria monocytogenes is a zoonotic foodborne pathogen, transmissible from the natural
agricultural environment to animals and humans. In recent years, the pig production industry has
experienced a series of monetary losses as a result of the L. monocytogenes outbreak which
threatened the economy of South Africa. This outbreak also had a detrimental effect on the health
system of the country. In South Africa however, there is limited information regarding the genomic
diversity of L. monocytogenes. Therefore, an overview of the genomic diversity of L.
monocytogenes strains circulating at different levels of the pork production chain needs to be
determined so as to be able to identify routes of contamination of the pathogen and thus improve
meat safety. This study was aimed to determine the antimicrobial resistance patterns and
population structure of L. monocytogenes isolated from pig farms in South Africa. Based on wholegenome
sequence analysis, 77 isolates of L. monocytogenes were differentiated into four molecular
serogroups with IIa (45.5%) being the most prevalent followed by IIc (26.0%), IVb (22.1%) and IIb (6.5%). Overall, 11 clonal complexes (CCs) were identified in this study, with the
predominance being observed from; CC204 (23.4%), CC1 (19.5%) and CC2 (16.9%). Genetic
elements associated with biocide, antimicrobial and heavy metal resistance were noted in 24.7 %,
48% and 11.7% of the isolates, respectively. Listeria pathogenicity island 1 and 3 that harbored
clusters of virulence genes were present in 38.8% of the isolates. Five different plasmids were
found in 68.9% of the isolates. This study has given baseline data on the genomic diversity of L.
monocytogenes strains that are associated with biocides, heavy metal and antibiotics resistance
genes. The data again demonstrated the genotypes of L. monocytogenes that are prone to
