Alignment of LC-MS Data Using Peptide Features

Integrated liquid-chromatography mass-spectrometry(LC-MS) is becoming a widely used approach for quantifying the protein composition of complex samples.In the last few years,this technology has been used to compare complex biological samples across multiple conditions. One challenge in the analysis of an LC-MS experiment is the alignment of peptide features across samples. In this paper,we proposed a new method using the peptide internal information (both LC-MS and LC-MS/MS information) to align features from multiple LC-MS experiments.We defined Anchor points which are data elements that are highly confident we have identified and are shared by both samples. We chose one sample as template data set, find Anchor points in this sample, then apply alignment to modify another sample, find Anchors in modified sample, these Anchors should line up with one another. One advantage of our method is that it allows statistical assessment of alignment performance. Use anchor points to perform alignment between samples, and labeling an objective performance in LC-MS.

There were made an analysis of Object-oriented programming, CASE – computer aided system engineering, Component modeling, MS Access. To understand business environment I created Business Interaction Model, Workflow models, Use Case Model. For this purpose I used ProVision Workbench package. I created data base project. I used MS Access XP and Visual Basic, because of its stability, multipurpose, potential, good working with Windows 9*, Windows 2000, Windows XP operational systems. There were made a User Interface, including forms, reports, and queries.

Antigenic induction of nerve growth factor (NGF) in experimental autoimmune encephalomyelitis (EAE), an animal model of multiple sclerosis (MS)

Nerve growth factor (NGF) represents a new therapeutic strategy for multiple sclerosis (MS) because of its immunomodulatory and neuroprotective activity. To analyze changes in NGF expression experimental autoimmune encephalomyelitis (EAE) was induced in Lewis rats. In the dorsal root ganglia (DRG) of animals with EAE, NGF mRNA and protein increased between 18 - 24 days post induction (dpi) during complete neurological recovery. In the spinal cord (SC) of animals with EAE, NGF mRNA and protein expression increased at 15 dpi and 12 dpi, respectively, to reduce EAE-induced disability. We identified the 25 kDa pro-NGF as a biologically active isoform during EAE. EAE SC axons demonstrate a loss or thinning of myelin which correlated with maximal neurological disability. NGF plays a role in minimizing EAE-induced inflammation and myelin damage to promote neurological recovery. NGF may be an “off switch” for a cytokine-neurotrophin signaling triad to govern the extent of myelin damage.
