A Structure/Function Analysis of Nhsl1b in Facial Branchiomotor Neurons

The goal of this study was to identify critical regions of a novel gene, Nance-Horan syndrome-like 1b (nhsl1b). It was previously discovered that C-terminal truncation of the Nhsl1b protein in nhsl1b mutants resulted in a loss of migration in the facial motor neurons of the hindbrain (Walsh et al. 2011). As nhsl1b expresses many isoforms, multiple targets were investigated in order to determine which transcript bears the largest impact on the motor neurons. Using confocal microscopy to observe immunostained embryos, we examined a mutation in an nhsl1b transcript that encodes a WHD, a domain that is known to function within the actin nucleation and polymerization pathways. In situ hybridization and injection of antisense morpholino oligonucleotides indicate that it is not the WHD but another transcript (ex1bnhsl1b) that is necessary for migration. The control experiments for rescuing the mutant phenotype have successfully been performed, but inducing expression of full length nhsl1b in the nhsl1b mutants is proving difficult.

Malliavin calculus and its applications to mathematical finance

(Applied Mathematics)) -- University of Limpopo, 2020 / In this study,we consider two problems.The first one is the problem of computing hedging portfolios for options that may have discontinuous payoff functions.For this problem we use the Malliavin property called the Clark-Ocone formula and give some examples for diferent types of pay of functions of the options of European type.The second problem is based on the computation of price sensitivities (derivatives of the probabilistic representation of the pay off functions with respect to the underlying parameters of the model) also known as`Greeks' of discontinuous payoff functions and also give some examples.We restrict ourselves to the computation of Delta, Gamma and Vega.For this problem we make use of the properties of the Malliavin calculus like the integration by parts formula and the chain rule.We find the representations of the price sensitivities in terms of the expected value of the random variables that do not involve the actual direct differentiation of the payout function,that is, E[g(XT ) ] where g is a payoff function which depend on the stochasticdic differential equation XT at maturity time T and is the Malliavin weigh tfunction. Cell Cycle Regulation of Retinal Progenitors; a role for the Nance-Horan Syndrome Protein in Retinogenesis

The Nance-Horan syndrome gene (NHS) plays a role in lens, eye and brain development. To date, the function of NHS remains unclear. Recent evidence showed that p53 isoform, Δ113p53, inhibits abnormal cell growth during organogenesis. We show that NHS is expressed in the retinas of Danio rerio and Xenopus tropicalis during key stages of retinogenesis, and that knockdown of the gene resulted in a small eye phenotype in both species. Initially, knockdown of nhsb in zebrafish had no visible defects at 24hpf. But examination of the retina at 48hpf, we see a marked difference in size compared to control embryos. Cell proliferation is a major feature of the developing retina from 24 hpf to 48 hpf. Differentiation of neurons was delayed, while the total number of cells that makes up the volume of the retina was markedly reduced. Here we show that the small retina in nhsb knockdown embryos are due to p53-dependent cell cycle arrest with specific induction of p53 target gene, Δ113p53 and p21. Δ113p53 protects nhsb- knockdown cells from p53-mediated apoptosis. We hypothesize that nhsb overcomes a proliferation restriction in retina progenitor cells during retinogenesis, while knockdown of nhsb increases expression of Δ113p53 and p21, lengthening the cell cycle.

A Retrospective Cohort Study of Fixed Space Maintainers and a Survey of their use by Pediatric Dentists and Orthodontists

Purpose: This study was conducted to evaluate the success of fixed space maintainers (fspms) placed by a sole dental practitioner in a private practice setting. The survey was conducted to evaluate the use of fspms by pediatric dentists and orthodontists. Methods: 899 fspms placed in 531 patients were evaluated. An online survey was sent to 1000 pediatric dentists and 1000 orthodontists. Results: The clinical success rate was 86.6%. The response rate for pediatric dentists surveyed was 43.0% but for orthodontists was only 7.0%. The vast majority of responding pediatric dentists (99.8%) and orthodontists (97.0%) placed fspms. The most common reason cited for failure by the pediatric dentists was cement loss (47.3%) and breakage by the orthodontists (30.4%). Conclusions: Fixed space maintainers had a high clinical success rate (86.6%) and were placed by the vast majority of responding pediatric dentists (99.8%) and orthodontists (97.0%).

CoordenaÃÃo de AperfeiÃoamento de Pessoal de NÃvel Superior / Conselho Nacional de Desenvolvimento CientÃfico e TecnolÃgico / Este trabalho teve por objetivos avaliar frutos de muricizeiros (Byrsonima dealbata), oriundos de diferentes genÃtipos, por meio da caracterizaÃÃo fÃsica e fÃsico-quÃmica. Os frutos utilizados no experimento foram provenientes de campos experimentais da Embrapa AgroindÃstria Tropical. Analisou-se 18 genÃtipos de muricizeiros, dos quais sete foram oriundos do municÃpio de Pacajus (PAC0, PAC1, PAC2, PAC3, PAC5, PAC6, PAC7), quatro do municÃpio de Fortaleza (EM1, EM3, EM4, EM5) e sete do municÃpio de Paraipaba (PAR3, PAR4, PAR5, PAR7, PAR 9, PAR 11 e PAR 12), todos no Estado do CearÃ. Para avaliaÃÃo do potencial comercial dos genÃtipos foram realizadas anÃlises estatÃsticas uni e multivariadas. O genÃtipo EM4 apresentou maior peso. Para todos os genÃtipos de muricizeiros estudados, exceto EM3, observou-se que o diÃmetro à maior que o comprimento, caracterizando frutos oblongos. A mÃdia geral para vitamina C foi de 61,58 mg/100 g. De modo geral, o fruto à Ãcido, contÃm elevado teor de aÃÃcares solÃveis totais, amido, pectina total e solÃvel e baixo teor de carotenÃides e flavonÃides amarelos. As caracterÃsticas fÃsico-quÃmicas analisadas apresentaram altos coeficientes de determinaÃÃo e coeficientes de repetibilidade de intermediÃrios a altos, tendo, portanto, baixo nÃmero de mediÃÃes necessÃrio para prediÃÃo do valor real da caracterÃstica. Os elevados valores das estimativas de repetibilidade sÃo indicativos de que existe variabilidade genÃtica observada nesse germoplasma. A anÃlise de agrupamento, feita por meio da otimizaÃÃo de Tocher, a anÃlise de componentes principais e a dissimilaridade dos genÃtipos, poderÃo ser utilizados para orientar a seleÃÃo de genÃtipos promissores para possÃveis programas de melhoramento genÃtico. / The objective of this work was to evaluate fruits of nance (Byrsonima dealbata), from different genotypes, by means of the physical and physical-chemical characterization. Fruits of nance were collected of from genotypes of the three experimental station of EMBRAPA - Empresa Brasileira de Pesquisa AgropecuÃria, in CearÃ, Brazil. Eighteen genotypes of nance, of which seven had been deriving of the Pacajus city (PAC0, PAC1, PAC2, PAC3, PAC5, PAC6, PAC7), four of the Fortaleza city (EM1, EM3, EM4, EM5) and seven of the Paraipaba city (PAR3, PAR4, PAR5, PAR7, PAR 9, PAR 11 and PAR 12), all in the State of the CearÃ. To evaluate the commercial potential of the genotypes joined and multivaried statistical analyses had been carried through. The genotype EM4 presented the greatest weight. For all the studied genotypes of nance, except EM3, were observed that the diameter is greater than the length, characterizing oblong fruits. The general average for vitamin C was of 61.58 mg/100 g. In general, the fruit is acid, with high levels of total soluble sugars, starch, total and soluble pectin and low levels of carotenoids and yellow flavonoids. The physical chemical characteristics analyzed presented high coefficients of determination and medium to high coefficients of repeatability therefore, needing a low number of measurements for prediction of the real value of the characteristics. The high values of the estimates of repeatability indicates that there is a genetic variability in this germplasm. The group analysis made by means of the Tocher Optimization, the analysis of main components and the dissimilarity of the genotypes, could be used to guide the election of promising genotypes on programs of genetic improvement.

Non-concave and behavioural optimal portfolio choice problems

Our aim is to examine the problem of optimal asset allocation for investors exhibiting a behaviour in the face of uncertainty which is not consistent with the usual axioms of Expected Utility Theory. This thesis is divided into two main parts. In the first one, comprising Chapter II, we consider an arbitrage-free discrete-time financial model and an investor whose risk preferences are represented by a possibly nonconcave utility function (defined on the non-negative half-line only). Under straightforward conditions, we establish the existence of an optimal portfolio. As for Chapter III, it consists of the study of the optimal investment problem within a continuous-time and (essentially) complete market framework, where asset prices are modelled by semi-martingales. We deal with an investor who behaves in accordance with Kahneman and Tversky's Cumulative Prospect Theory, and we begin by analysing the well-posedness of the optimisation problem. In the case where the investor's utility function is not bounded above, we derive necessary conditions for well-posedness, which are related only to the behaviour of the distortion functions near the origin and to that of the utility function as wealth becomes arbitrarily large (both positive and negative). Next, we focus on an investor whose utility is bounded above. The problem's wellposedness is trivial, and a necessary condition for the existence of an optimal trading strategy is obtained. This condition requires that the investor's probability distortion function on losses does not tend to zero faster than a given rate, which is determined by the utility function. Provided that certain additional assumptions are satisfied, we show that this condition is indeed the borderline for attainability, in the sense that, for slower convergence of the distortion function, there does exist an optimal portfolio. Finally, we turn to the case of an investor with a piecewise power-like utility function and with power-like distortion functions. Easily verifiable necessary conditions for wellposedness are found to be sufficient as well, and the existence of an optimal strategy is demonstrated.
