The Role of Farnesyltransferase β-subunit in Neuronal Polarity in Caenorhabditis Elegans

Little is known about the molecular components and interactions of the planar cell polarity pathway that regulate neuronal polarity. This study uses a prkl-1 induced backwards locomotion defect as an array to perform a prkl-1 suppressor screen in C. elegans looking for new components of the planar cell polarity pathway involved in the neuronal polarization of VC4 and VC5. The screen discovered twelve new alleles of vang-1, one new allele of fntb-1 and five new mutations in unknown polarity genes. fntb-1 encodes for the worm ortholog of Farnesyltransferase β-subunit and is important for neuronal polarization. Acting cell and non-cell autonomously, fntb-1 regulates the function and localization of prkl-1 through the recognition of a CAAX motif. Therefore, fntb-1 modifies prkl-1 to regulate the neuronal polarity of VC4 and VC5.

A Role for the Planar Cell Polarity Pathway in Neuronal Positioning Along the AP Axis of C. elegans.

We sought to investigate the role of the Planar Cell Polarity (PCP) pathway in neuronal positioning along the Anterior-Posterior (AP) axis of C. elegans, and chose the worm’s DD-type motor neurons as a model. The six DD neurons (DD1-DD6) are evenly spaced in the ventral nerve cord of wild type animals. Here we showed that mutations in core PCP genes caused DD neuron spacing and positioning defects. prkl-1 double mutant combinations with vang-1 and fmi-1 showed a suppression of the more severe prkl-1 single mutant defects, which was evidence of genetic interactions between these PCP components. We also conducted a candidate screen of Frizzled, Dishevelled, Wnt, and ROCK genes, and found that dsh-1/Dishevelled, mom-2/Wnt and let-502/ROCK also played roles in DD neuronal positioning. Both vang-1 and prkl-1 were found to function within the nervous system to guide DD neuronal positioning, and prkl-1 was further identified as playing a cell autonomous role. The origins of observed DD neuron anterior positioning defects were investigated during embryogenesis, in which 1.5 fold stage prkl-1(ok3182) embryos displayed delayed intercalation of the DD neurons. This represents a novel role for the PCP pathway in mediating DD neuronal intercalation.

An Optical Biosensor Towards Urinary Tract Infection Diagnosis

We explore a new laboratory technique in the field of urinalysis promising a combination of speed and selectivity in support of urinary tract infection diagnosis. Laboratory experimentation demonstrates long range surface plasmon polaritons (LRSPP) waveguides as a useful biosensor to selectively detect gram negative bacteria or gram positive bacteria in human urine. The biosensor can detect bacteria at concentration of 105 CFU/ml, the internationally recommended threshold for diagnostic of urinary tract infection (UTI). Using a negative control solution at bacterial concentration 1000x higher than the targeted bacteria in urine with a weak concentration of constituents, the power ratio between the negative control signals to the target bacteria signal is measured to be 5.4. Thus we report a conclusive demonstration of the LRSPP waveguide biosensor selectivity to the gram of bacteria in human urine. In addition, the biosensor may prove useful as an alternative urinalysis test method to determine the urine specific gravity, to estimate proteinuria, and to detect biofilm formation on surfaces.

Le développement des dispositifs embarqués visant des applications telles que la téléphonie mobile, l’avionique ou encore les véhicules électriques est en très forte croissance. Les contraintes liées à ces applications sont principalement la réduction du volume, du poids, et ainsi que le coût de production des composants actifs et passifs. C’est ainsi que le Laboratoire Telecom Claude Chappe de l’Université Jean Monnet de Saint Etienne, s’est positionné sur le développement de composants passifs magnétiques dans le but de réduire leur dimension tout en optimisant leur performance. Ce travail de doctorat avait pour but de développer des modèles « type circuit » comportant peu de paramètres pour des composants inductances planaires à couches magnétiques. Les modèles développés doivent prendre en compte les propriétés des couches magnétiques, à savoir la tangente de perte (tanδ) et la perméabilité relative (μr) en fonction de la fréquence, ainsi que les phénomènes haute fréquence : effets de peau et de proximité dans les conducteurs d’une part, couplage capacitif entre spire et entre spires et plan de masse d’autre part. L’étude bibliographique a montré qu’il existe plusieurs modèles d’inductances, avec et sans couche de matériau magnétique. Ces modèles traduisent bien le comportement de l’inductance, mais ne prennent pas en compte certaines propriétés telles que l’évolution de la perméabilité relative et de la tangente de perte en fonction de la fréquence. La simulation nous a permis de mettre au point nos modèles. Nous avons simulé des structures à air, avec une couche de YIG dont la perméabilité relative variait en fonction de la fréquence (avec et sans perte) et enfin des structures à deux couches avec une perméabilité relative et une tangente de perte variable en fonction de la fréquence. Le logiciel nous a également permis de vérifier ainsi le bon comportement des modèles. Afin de valider et de compléter les résultats obtenus en simulation nous avons réalisé les mêmes dispositifs que ceux simulés en utilisant les techniques de micro-électronique. La caractérisation des dispositifs réalisés a été conduite en basse fréquence à l’aide d’un LCRmétre et en haute fréquence à l’aide d’un analyseur vectoriel de réseaux. Les paramètres des modèles ont été obtenus à partir de la simulation ou de la mesure en utilisant un programme d’optimisation. Afin de vérifier l’exactitude des valeurs obtenues, nous avons recalculé les paramètres Yij à partir des éléments extraits et les avons comparés avec les paramètres Yij de mesures ou de simulation. Une bonne corrélation entre les différents paramètres Yij a été constatée sur une large bande de fréquence. Ce travail de thèse a permis de mettre au point trois modèles : modèle d’inductance à air ; modèle d’inductance à une couche et à deux couches tout en prenant en compte les propriétés magnétiques constituant l’inductance / The development of embarked devices used in mobile phone, avionics or electrical vehicles is in very strong growth. Constraints connected to these applications are mainly the reduction of volume, weight and as well as the production cost of the active and passive components. So, Telecom Claude Chappe Laboratory of the Saint Etienne University is positioned on the development of magnetic passive components with the aim of reducing their size while optimizing their performance. This PhD thesis work aimed at developing models "like circuit" containing few parameters for planar inductor components with magnetic layers. The developed models must take into account the magnetic layer properties, as the loss tangent (tanδ) and the relative permeability (μr) according to the frequency, as well as the skin and proximity effects in the conductors, capacity between turn and capacities between turns and ground plan. The bibliographical study showed that there are several inductors models with and without magnetic layer. These models translate well the inductor behavior but do not take into account some properties such as the variation of the loss tangent and the magnetic permeability according to the frequency. The simulation allowed to work out our models. Inductors structures with air, with one magnetic layer and with two magnetic layers when the magnetic permeability and the loss tangent varied according to the frequency have been simulated. The software allowed to verify the good behavior of the models. In order to validate results obtainedin simulation, devices have been realized by using microelectronics techniques. The devices characterization has been carried out at lower frequency by using an LCRmeter and at high frequency by using a vector network analyzer. The model parameters are obtained from simulation and measurement by using an optimization algorithm. To verify the values accuracy, Yij parameters are been recalculated from the extracted parameters and compared with the Yij simulated or measured parameters. A good correlation between everyYij parameters has been noticed on a broad band frequency. This PhD thesis allowed to work out three inductors models: with air, with one magnetic layer, with two magnetic layers while taking into account the magnetic properties constituting the inductor

Cette thèse porte sur deux sujets d’algorithmique des graphes. Le premier sujet est l’inférence de réseaux. Quelle est la complexité pour déterminer un graphe inconnu à partir de requêtes de plus court chemin entre ses sommets ? Nous supposons que le graphe est de degré borné. Dans le problème de reconstruction, le but est de reconstruire le graphe ; tandis que dans le problème de vérification, le but est de vérifier qu’un graphe donné est correct. Nous développons des algorithmes probabilistes utilisant une décomposition en cellules de Voronoi. Ensuite, nous analysons des algorithmes de type glouton, et montrons qu’ils sont quasi-optimaux. Nous étudions aussi ces problèmes sur des familles particulières de graphes, démontrons des bornes inférieures, et étudions la reconstruction approximative. Le deuxième sujet est l’étude de deux problèmes d’optimisation sur les graphes planaires. Dans le problème de classification par corrélations, l’entrée est un graphe pondéré, où chaque arête a une étiquette h+i ou h-i, indiquant si ses extrémités sont ou non dans la même catégorie. Le but est de trouver une partition des sommets en catégories qui respecte au mieux les étiquettes. Dans le problème d’augmentation 2-arête-connexe, l’entrée est un graphe pondéré et un sous-ensemble R des arêtes. Le but est de trouver un sous-ensemble S des arêtes de poids minimum, tel que pour chaque arête de R, ses extrémités sont dans une composante 2-arête-connexe de l’union de R et S. Pour les graphes planaires, nous réduisons le premier problème au deuxième et montrons que les deux problèmes, bien que NP-durs, ont un schéma d’approximation en temps polynomial. Nous utilisons la technique récente de décomposition en briques. / This thesis focuses on two topics of graph algorithms. The first topic is network inference. How efficiently can we find an unknown graph using shortest path queries between its vertices? We assume that the graph has bounded degree. In the reconstruction problem, the goal is to find the graph; and in the verification problem, the goal is to check whether a given graph is correct. We provide randomized algorithms based on a Voronoi cell decomposition. Next, we analyze greedy algorithms, and show that they are near-optimal. We also study the problems on special graph classes, prove lower bounds, and study the approximate reconstruction. The second topic is optimization in planar graphs. We study two problems. In the correlation clustering problem, the input is a weighted graph, where every edge has a label of h+i or h−i, indicating whether its endpoints are in the same category or in different categories. The goal is to find a partition of the vertices into categories that tries to respect the labels. In the two-edge-connected augmentation problem, the input is a weighted graph and a subset R of edges. The goal is to produce a minimum-weight subset S of edges, such that for every edge in R, its endpoints are two-edge-connected in the union of R and S. For planar graphs, we reduce correlation clustering to two-edge-connected augmentation, and show that both problems, although they are NP-hard, have a polynomial-time approximation scheme. We build on the brick decomposition technique developed recently.

Philosophiae Doctor - PhD / The organic-inorganic hybrid perovskites such as methyl ammonium lead iodide (MAPbI3) or mixed halide MAPbI3-xClx (x is usually very small) have emerged as an interesting class of semiconductor materials for their application in photovoltaic (PV) and other semiconducting devices. A fast rise in PCE of this material observed in just under a decade from 3.8% in 2009 to over 25.2% recently is highly unique compared to other established PV technologies such as c-Si, GaAs, and CdTe. The high efficiency of perovskites solar cells has been attributed to its excellent optical and electronic properties. Perovskites thin film solar cells are usually deposited via spin coating, vacuum thermal evaporation, and chemical vapour deposition (CVD).

This diploma thesis deals with a design of antennas based on a flexible substrate. There is also a design of a transmission line between a flexible and rigid substrate with FLEX- RIGID technology. In the first part of thesis, there is the technology, advantages and disadvantages of using this technology in design of antennas mentioned. There are also basic antenna parameters mentioned. In second part, the design of a transmission line and four antennas is presented. Two of these antennas are manufactured and measured. Finally, the measured parameters are compared with the simulated results.
