Surface Roughness Effects on Separated and Reattached Turbulent Flows in Open Channel

An experimental research was performed to study the effects of surface roughness on the characteristics of separated and reattached turbulent flows in an open channel. A backward facing step was used to induce flow separation. The rough surfaces comprised wire mesh grit-80 and sand grains of average diameter 1.5 mm. In each experiment, the Reynolds number based on the step height and freestream velocity of approach flow was fixed at 3240 and the Reynolds number based on the approach flow depth and freestream velocity was kept constant at 25130. Particle image velocimetry (PIV) technique was used to measure the flow velocity. The results showed that roughness effects on the mean and turbulent quantities are evident only in the recovery region. Moreover, roughness effects on the flow dynamics are dependent on the specific roughness element.

Com o presente estudo constatou-se que as moléculas, N-alquilbenzenossulfonamidas, produzem efeitos inibitórios sobre KV3.1. Através da técnica de whole cell patch clamp, observou-se que os valores de IC50 para as moléculas que bloquearam o canal foram 13,5 μM, 16,9 μM, 25,9 μM, 34,2 μM, 34,9 μM e 60 μM, respectivamente, para 4-cloro-3-nitro-N-butilbenzenossulfonamida (SMD2), 4-cloro-3-nitro-N-furfutilbenzenossulfonamida (SMD3), 4-[N-(3’aminopropil)-2-pirrolidona]-3-nitro-N-butilbenzenossulfonamida (SMD2_APP), 4-[N-(3’aminopropil)-2-pirrolidona]-3-nitro-N-furfurilbenzenossulfonamida (SMD3_APP), 4-cloro-N-butilbenzenossulfonamida (SMD2_SN) e 4-cloro-N-furfurilbenzenossulfonamida (SMD3_SN). O efeito de todas as moléculas mostrou-se reversível quanto à ligação com o canal e todas atuaram como bloqueadores de canal aberto. Em SMD2, molécula que mostrou o menor valor de IC50, observou-se um deslocamento de -8 mV em relação ao controle, nas curvas de condutância versus voltagem, nas cinéticas de ativação e na recuperação a partir da inativação em relação à voltagem. O SMD2 não alterou as constantes de tempo de desativação, embora tenha mudado as constantes de ativação e inativação, além de ter induzido o fenômeno de tail crossover. Observou-se que para potenciais mais despolarizados, ocorreu o alívio do bloqueio (Block Relief). Não foi observado o efeito da dependência do pH para o bloqueio e SMD2 não mudou a seletividade do canal. Constatou-se que pulsos despolarizantes de curta duração induzem efeitos menos intensos, ao passo que pulsos despolarizantes mais longos, produzem efeitos mais intensos de SMD2 sobre o canal. Além disso, foi observado que, quanto mais o canal é usado, ou seja, aberto, mais ele é bloqueado por SMD2. Todos esses dados sugerem que SMD2 não interage com o estado fechado e nem com o estado inativado do canal, mas sim com seu estado aberto, apresentando também um efeito dependente de uso. De um ponto de vista farmacológico, isso indica que SMD2 pode ser uma molécula importante na modulação da atividade dos canais KV3.1, presentes em células com altas frequências de disparos de potencial de ação, podendo constituir uma nova classe de moduladores farmacológicos desses canais. / This thesis had the aim of studying the organic molecules (N-alkylbenzenesulfonamides) that block KV3.1 potassium channel heterologously expressed in L-929 cells. It was found that N-alkylbenzenesulfonamides have restrained effects on KV3.1. Through the whole cell patch clamp technique, it was observed that the values of IC50, for molecules that block the channel, were 13,5 μM, 16,9 μM, 25,9 μM, 34,2 μM, 34,9 μM and 60 μM, respectively 4-Chloro-3-nitro-N-butylbenzenesulfonamide (SMD2), 4-Chloro-3-nitro-N-furfurylbenzene-sulfonamide (SMD3), 4-[N-(3′-Aminopropyl)-2-pyrrolidone]-3-nitro-N-butylbenzenesulfona-mide (SMD2_APP), 4-[N-(3′-Aminopropyl)-2-pyrrolidone]-3-nitro-N-furfurylbenzene-sulfonamide (SMD3_APP), 4-Chloro-N-butyllbenzenesulfonamide (SMD2_SN) e 4-Chloro-N-furfurylbenzenesulfonamide(SMD3_SN). The effect of all molecules was reversible regards to the linking with the channel and all act as open channel blocker. In SMD2, molecule which showed the smallest value of IC50, it was observed a displacement of -8 mV compared to control, for conductance curves versus voltage, for the kinetics of activation and for the recovery from inactivation in relation to voltage. SMD2 did not change the deactivation of time constants, although it changed the activation and inactivation constants, and more, SMD2 have induced tail crossover phenomenon. It was observed that, for more depolarized potentials, there was a block relief. It was not observed the effect of pH dependence for the block and SMD2 did not change the channel selectivity. It was observed that, short duration depolarizing pulses prompt less intense effects, whereas long duration depolarizing pulses prompt more intense effects of SMD2 on the channels. Furthermore, it was observed that the more the channel is used, in an open state, the more it is blocked by SMD2. All of these data suggest that SMD2 does not interact neither with the closed state nor the inactivated state of channel, but with its open state presenting an use-dependent manner, also showing a use-dependent effect. In a pharmacological point of view, this indicates that SMD2 may be an important molecule in the modulation of the activity in the KV3.1 channels, presents in cells with high frequency of firing of action potential and may constitute a new class of pharmacological modulators.

Modelo computacional para análise de transiente hidráulico em canais / Computational model for the study unsteady open-channel flows

Este trabalho representa a continuidade de estudos envolvendo a problemática dos escoamentos com superfície livre, contemplando a análise do fenômeno transiente em canais, a partir do modelo matemático unidimensional de Saint-Venant. Para tanto, é desenvolvido um modelo computacional em linguagem FORTRAN, capaz de avaliar o comportamento do escoamento não permanente. As equações hidrodinâmicas completas são discretizadas por um esquema completamente implícito de diferenças finitas e aplicadas no modelo computacional para a avaliação de dois casos. O modelo é previamente testado para um caso simples, cujos resultados são analisados viabilizando o modelo. No primeiro caso, o modelo é aplicado ao canal de alimentação da Usina Hidrelétrica Monjolinho em São Carlos-SP, para avaliar a necessidade de vertedouro quando se dá o fechamento brusco da turbina, e a ocorrência da entrada de ar na mesma quando da sua abertura repentina. No segundo caso, procurou-se avaliar o desenvolvimento do escoamento no Canal do Trabalhador, responsável pelo abastecimento da cidade de Fortaleza-CE. Com manobras de enchimento e esvaziamento do sistema, é possível determinar o tempo de antecedência de liga-desliga do sistema de recalque a partir das alturas dágua e velocidades de ocorrência, permitindo também a automação para as operações de controle. Em ambos os casos o modelo reproduziu resultados que ilustram com coerência os conceitos pré-estabelecidos, constituindo numa ferramenta útil para análise do fenômeno transiente nos escoamentos em condutos livres. / This work presents a computational model developed in FORTRAN language for the study of unsteady open-channel flows with the use of Saint-Venant one-dimensional equation. The discretization of hydrodynamic equations are presented in a completely implicit method of finite differences and applied in the model for the investigation of two cases, besides the one used previously to test the model. In the first case, the model is applied for a channel that supplies the Monjolinho hydroelectric plant in Sao Carlos SP, aiming to evaluate the need of a spillway when the turbine is closed and the flow abruptly stopped, as well as the occurrence of air entering the turbine when it is opened instantaneously. In the second case, the model simulates the development of the flow in the Trabalhador channel, responsible for the water supply in the city of Fortaleza - CE, in order to make possible the automation of operational control, based on data of flow velocity and water level. In both cases the model is presented as a useful tool for the analysis of unsteady open-channel flows, showing results and coherency with theory.

A numerical study of flow hydrodynamics and mixing processes at open channel confluences

River confluences - locations where rivers join one another - are fundamental components of natural drainage networks. Differences in topography, geology, soils, land use, and human activities within watersheds upstream of confluences can produce differences in thermal or chemical properties of river flows and in the materials transported by these flows. Mixing is initiated along the mixing interface (MI) that develops between the two incoming streams with different properties. Therefore, the understanding of fluvial processes at confluences is important for determining river mixing both at and downstream of individual confluences and at the scale of drainage networks. The primary goal of this thesis is to describe the main mechanisms that control mixing and transport at river confluences and the role played by the complex flow structures in the flow and how they change with planform geometry and other flow and geometrical parameters. The study is carried out using Computational Fluid Dynamics modeling based on the state of the art Detached Eddy Simulation approach and High Performance Computing. By starting with a mixing layer between parallel streams with simple geometry, the model is validated based on laboratory experiment data. Moreover, some hypotheses regarding the growth of the mixing layer are amended with the extensive data provided by the model, which is a valuable supplement to the experiment. By performing a detailed parametric study in very long and wide domains for simplified cases one can focus on the spatial development of the MI and the large scale coherent structures forming within and in the vicinity of the MI without the complications of other factors. More specifically, the influence of velocity and density difference of the two streams, flow depth, inflow conditions and angles between the two streams on the spatial development of the MI is analyzed. The data resulting from these simulations conducted in simple geometries is a unique set of data which can be used to test and improve theoretical models used to predict global parameters describing flow and mixing at natural river confluences. In particular, this research uses for the first time well resolved Large Eddy Simulation based techniques to investigate how density differences between the incoming streams affect the spatial development of the mixing interface and mixing downstream of the confluence apex. In order to investigate flow dynamics, mixing processes and effects of temperature stratifications at natural river confluences with discordant bed, a series of simulations is performed for the confluence of the Ebro and Segre Rivers in Spain, which is one of the most studied confluences in Europe. With the detailed survey data of the confluence bed and flow conditions data provided, the goal is to understand the main mechanisms responsible for mixing at a confluence with a large bed discordance and how the velocity ratio between the two incoming streams affects mixing. Besides, more insights are provided that if temperature stratification effects affect significantly flow structure and mixing based on real conditions recorded at a natural confluence. The study provides a comprehensive set of flow data in the confluence including velocity, temperature distribution etc. It serves as important supplement to the field measurements, which are generally more difficult to obtain. It also allows estimating scale effects between field conditions and conditions at which laboratory experiments of confluence flow and mixing are conducted.

Ability of ADV Measurements to Detect Turbulence Differences Between Angular and Rounded Gravel Beds of Intermediate - Roughness Scale

A set of laboratory experiments was carried out to distinguish flow characteristics(bed origin, shear velocity, turbulence intensity, turbulent kinetic energy) between beds of differing gravel angularity. Ten vertical profiles of velocity measurements were taken from angular and rounded fixed gravel beds with a 16 MHz micro acoustic Doppler velocimeter (ADV) sampling at 50 Hz. Both gravel beds had a bottom slope of 0.2% and were in the intermediate - roughness scale. Shear velocities were calculated using three common methods: St Venant, Reynolds stress, and Clauser. The Reynolds stress method resulted in the closest visual match to turbulence distributions proposed by others. The bed origin was found to be on average 0.24D50 and 0.21D50 for the angular and rounded gravel beds respectively. These differences, however, were not statistically significant. Turbulence intensity within 20% of the bed showed considerable scatter. The difficult measuring conditions likely prevented the ADV to detect significant differences of turbulence intensity in the longitudinal and transverse directions between the two gravel beds. But the ADV measurements in the vertical direction may well resolve turbulence even in difficult flow conditions (determined by acoustic Doppler performance curve formulation). For the vertical direction, the angular gravel bed showed an increase in TImax that extended throughout the profile. The increased turbulence intensity had a concomitant effect of increasing the turbulent kinetic energy for the angular bed.

Reservoir Applications of Arced Labyrinth Weirs

In recent years, magnitudes of flood estimates used in hydraulic design have increased for many reservoirs. Consequently, many existing spillways are now deficient as they do not meet current discharge capacity requirements. To rehabilitate existing, fixed-width spillways, labyrinth weirs are often viable solutions. For reservoir applications, arcing labyrinth weirs into the reservoir increases hydraulic efficiency. This results from better cycle orientation to the approaching flow field. This study supplements available arced labyrinth weir hydraulic data by observing flow characteristics of three laboratory-scale physical models and two numerical (CFD) models. Physical model results provide head (energy)-discharge data and empirical coefficients for hydraulic design. Results also show that increasing the arc angle improves efficiency at H/P<0.3, where H/P is upstream piezometric head divided by weir height; after which, efficiency improvements diminish as downstream submergence also increases. The purpose of the CFD analysis was to assess the appropriateness of CFD as a design tool for arced labyrinth weir head-discharge relationship development. The CFD model results found good agreement with the physical model data indicating CFD’s usefulness as a hydraulic design tool; however, it is recommended that CFD models be calibrated to reliable laboratory or field data. This study’s data may be used, with sound engineering judgement, to aid in hydraulic design of arced labyrinth weirs

Effects of Submergence in Montana Flumes

As part of a continued research project for the Utah Water Research Laboratory and the State of Utah, a study of flow measurement devices is being conducted throughout the state. Initially the project included only measurement devices associated with high-risk dams, but has since been broadened to any measurement structure of interest for water users in the state. The physical dimensions, relative elevations, and flow accuracy were documented for each included device. After visiting sixteen sites, it was found that fourteen of the measuring devices had incorrect geometries. Of these fourteen, thirteen of them were originally Parshall flumes. A large percentage of Parshall flumes with geometry inaccuracies was also found from previous data collected for this project. One reoccurring issue was that the flumes had not been well maintained and had damage to the walls or floor. Some of these Parshall flumes did not have a diverging downstream section and are referred to as Montana flumes. In these cases, a standard Parshall rating curve was used to determine flow where it did not apply. Some of the flumes that were tested operated regularly under submerged conditions, and no adjustments were made for submergence. The objective of this research is to determine if Montana flumes (Parshall flumes without a diverging section) operate similarly to fully constructed Parshall flumes under both free-flow and submerged conditions. Laboratory tests were performed in the Utah Water Research Laboratory to determine corrections for submergence. Flow 3D, a computational fluid dynamics (CFD) software program, was also used to develop corrections for a submerged Montana flume. The laboratory results were compared to the computational fluid dynamics results. By using Flow 3D, a reliable numerical process was developed to determine the flow rate in a submerged Montana flume in an effort to expand the results to other seized flumes.
