O trabalho teve como objetivo estudar a eficiência de diferentes métodos na avaliação do potencial fisiológico de sementes de rúcula, buscando indicar opções para utilização em programas de controle de qualidade adotados por instituições produtoras de sementes dessa espécie. Para tanto, o estudo foi conduzido utilizando cinco lotes de sementes das cultivares Rúcula Cultivada e Rúcula Gigante. Foram realizados os testes de germinação, primeira contagem de germinação, emergência de plântulas em casa de vegetação e estudos específicos para os testes de deterioração controlada (graus de umidade de 18%, 21% e 24%, a 45ºC, durante 24 e 48 horas), de envelhecimento acelerado, com e sem solução salina (períodos de 48, 72 e 96 horas, a 38ºC, 41ºC e 45ºC), de condutividade elétrica (temperaturas de 20ºC, 25ºC e 30ºC, volumes de 25mL, 50mL e 75mL de água, 25 e 50 sementes, e períodos de 1, 2, 4, 6, 8, 12, 18 e 24 horas) e de lixiviação de potássio (temperaturas de 25ºC e 30ºC, volumes de 50mL e 75mL de água, 50 e 100 sementes, e períodos de 30 minutos, 1 h, 1,5 h, 2 h, 2,5 h, 3, 4 e 5 h). Concluiu-se que o teste de deterioração controlada e o teste de envelhecimento acelerado (procedimento tradicional) não são eficientes para avaliação do vigor das sementes de rúcula. O teste de condutividade elétrica utilizando 50 sementes/50mL, a 25ºC por 4 horas; de lixiviação de potássio utilizando 50 sementes/50mL, a 30ºC por 2 horas; e de envelhecimento acelerado (com solução salina) utilizando a combinação 41ºC por 72 horas, apresentaram sensibilidade suficiente para avaliação do potencial fisiológico das sementes de rúcula. / Abstract: The quality of the seed used in the process of agricultural production is one of the main factors to be considered for the implantation of the culture, having consensus between all the segments, on the importance of the vigor of the seeds and the necessity to evaluate it. The work had as objective to study the efficiency of different methods in the evaluation of the physiological potential of seeds of arugula, searching to indicate options for use in programs of quality control adopted by producing institutions of seeds of this specie. Five seed lots each from the Rucula Cultivada e Rucula Gigante were used. The following tests were performed: germination, germination first count and seedling emergence. Specific studies were also conducted to the following tests: controlled deterioration (seed moisture content of 18%, 21% and 24%, at 45oC, for 24 and 48 hours), traditional accelerated aging, saturated salt accelerated aging (for 48, 72 and 96 hours, at 38oC, 41oC and 45oC), electrical conductivity (25 or 50 seeds imbibed in 25 mL, 50 mL or 75 mL in water, at 20oC, 25oC and 30oC, for 1, 2, 4, 6, 8, 12, 18 and 24 hours) and potassium leachate (50 or 100 seeds imbibed in 50 mL or 75 mL in water, at 25oC and 30oC, for 30 minutes, 1 h, 1,5 h, 2 h, 2,5 h, 3, 4 e 5 h). It was concluded that controlled deterioration and accelerated aging (tradicional procedure) tests, does not constitute an efficient option for the vigor evaluation of arugula seeds. The electrical conductivity using 50 seeds/50mL, at 25oC for 4 hours; the potassium lixiviation using 50 seeds/50mL, at 30oC for 2 hours; and the saturated salt accelerated aging using 41oC for 72 hours, showed to be sufficiently sensitive for the evaluation of the physiological potential of arugula seeds. / Doutor

Analytic properties of the scattering amplitude for interaction via nonlocal potentials

The derivation of a partial-wave amplitude for scattering by a separable, nonlocal potential given by MCMillan in Nuovo Cimento 29, 4153 (1963) is reviewed. Using his results, an exact expression for the amplitude is derived for a potential of the form -g V(r)V(r¹), where V(r) = ra e-μr , and its analytic properties are studied. The asymptotic behaviour of the amplitude as |ℓ| → ∞ (where ℓ is the usual angular-momentum parameter) is derived, and is shown to permit a Sommerfeld-Watson transformation to be performed on the series expression for the total scattering amplitude in terms of the partial-wave amplitudes. By means of this transformation, a double-dispersion relation is derived for the total amplitude in both the complex-energy and complex-cos θ planes. Explicit forms are derived for the weight functions, and the convergence of the integrals involved is studied. In addition to the usual branch cuts along the positive, real energy and cos θ axes, an extra cut along the negative , real energy axis is found which is not present for the local case. Its origin is traced to the fact that the Wronskian of two solutions of the nonlocal radical Schroedinger equation is not necessarily a constant, as it is in the purely local case; and to the conditions necessary to ensure convergence of the extra integral in the nonlocal Schroedinger equation. / Science, Faculty of / Physics and Astronomy, Department of / Graduate

The thesis is divided into two parts, theoretical and practical. The theoretical part provides a comprehensive introduction to the basic terms of talent management and individual steps to implement talent management program. It is based on specialized literature and theoretical knowledge. It also analyzes the problems that may arise during implementation. The practical part starts with an analysis of HR activities in relation to talent management and suggests various measures. The following chapter contains a proposal of talent management for the company. One section of the proposal contains evaluation of the most important competencies for individual programs.

The anticancer potential of CITme, a quinoline derivative

3-[3-(7-chloro-quinolin-4-yl amino) phenyl]-1-(4-methoxy-phenyl) prop-2-enone citrate “CITme” is a substituted quinoline derivative, synthesized as a potential antitumour agent. The aim of this study was to investigate CITme with regard to antitumour activity, toxicity and pharmacokinetics. In vitro screening for neoplastic cytotoxic effects using standard cell culture techniques revealed cytotoxic activity against HeLa, DU-145, MCF-7, Jurkat and CoLo 320 human derived cancer cell lines at low concentrations. Toxicity was significantly less in either normal resting and stimulated lymphocytes or primary chicken fibroblast cells. CITme showed highest cytotoxic specificity for DU-145 with an IC50 of 11 μM compared to 38.2 ìM for lymphocytes after a three-day incubation period and 2.5 μM, after a seven-day incubation period. CITme exhibited poor solubility in aqueous solutions and this had to be addressed prior to in vivo acute toxicity studies being performed. CITme was found to be insoluble in many biocompatible and acceptable formulating solvents. 2-methyl pyrrolidone and Cremophor EL showed promise as potential solvents but the solubility proved to be too low to enable therapeutic CITme dosing while avoiding solvent/carrier toxicity. An oral microcrystalline cellulose suspension, two combinations of 2-methyl pyrrolidone with plasma protein complexation and a propheroid formulation were also tested. These were tested in pilot acute toxicity studies using BALB/c mice or Sprague Dawley rats. No plasma concentrations were observed with the cellulose suspension and both the 2-methyl pyrrolidone formulations elicited toxic responses in vehicle control and experimental groups despite working well below the published LD50 of 2-methyl pyrrolidone. A unique formulation developed by the Department of Pharmacy of NWU referred to as propheroid drug delivery system, provided sufficient solubility for acute and chronic toxicity studies of CITme. In vivo acute and chronic toxicity study of the propheroid delivery system formulation alone and the CITme propheroid at 30 mg/kg/day and 60 mg/kg body weight (acute toxicity study), and 10mg/kg/day (chronic toxicity study) demonstrated safety after seven and forty-five (six weeks) days administration by oral gavage. The potential anti-tumour activity of CITme could not be determined in a nude mouse tumour model, using the most susceptible in vitro cell line, DU-145 due to the low success rate of tumour induction and the long lag period required for the development of subcutaneous prostate tumours after transplantation of either a DU-145 cell suspension or DU-145 tumour tissue blocks. An LC-MS/MS method was developed to separate and quantify CITme from plasma and organ homogenates. The LC-MS/MS method was used to determine the pharmacokinetic parameters of I.V. administered CITme. A pharmacokinetic study of CITme in mice indicated that CITme is absorbed after an oral administration and exhibits a very rapid distribution (metabolism) and/or elimination after IV administration. CITme could be found at relatively high concentrations 24 hours after oral dosing and to a smaller extent 10 minutes after I.V. administration in both the liver and kidney tissues. Despite the fact that the efficacy and oral pharmacokinetics properties of CITme could not be determined, this study has proved that CITme is a selective and potent anti-tumour agent in vitro with low toxicity both in vitro and in vivo and therefore this compound warrants further scientific evaluation. / Dissertation (MSc)--University of Pretoria, 2011. / Pharmacology / unrestricted
