This diploma thesis is focuses on the individualization of the education of pupils with autism spectrum disorder in the education. The diploma thesis is divided into two parts, a theoretical and practical part. The theoretical part of the thesis describes and defines autism spectrum disorders, nemely chilhood autism, atypical autism, Asperger's and Rett's syndrome and disintigration disorder in childhood. The theoretical part od the work is also focused on the principles and methods used in the education of pupils with ASD. The theoretical part also describes support measures that are closely related to the education of students with autism spectrum disorders. The practical part of this diploma thesis is focused mainly on the education of pupils with ASD in compulsory distance education, which occurred as a result of the epidemic of the spread of COVID-19. The theoretical part of this diploma thesis uses the research method of qualitative research. The strategy of semi-structured interviews was used as a research strategy for the processing of qualitative research. Characterizing the Function of PAS kinase in Cellular Metabolism and Neurodegenerative Disease

The second identified substrate of PAS kinase discussed is Pbp1. The human homolog of Pbp1 is ataxin-2, mutations in which are a known risk factor for amyotrophic lateral sclerosis (ALS). As diet and sex have been shown to be important factors regarding PAS kinase function, they also are strong contributing factors to ALS and are extensively reviewed herein. Pbp1 is known to be sequestered by PAS kinase under glucose depravation, and it can sequester additional proteins along with it to regulate different cellular pathways. To shed light on the pathways affected by Pbp1, we performed a yeast two-hybrid assay and mass spectrometry, identifying 32 novel interacting partners of Pbp1 (ataxin-2). We provide further analysis of the direct binding partner Ptc6, measuring mitophagy, mitochondrial content, colocalization, and respiration. This work elucidates novel molecular mechanisms behind the function of PAS kinase and yields valuable insights into the role of PAS kinase in disease.

Mitochondria is no longer viewed as merely a powerhouse of the cell. It is now apparentthat mitochondria play a central role in signaling, maintaining cellular homeostasis and cell fate.Mitochondrial dysfunction has been linked to many human diseases caused by cellular metabolicderegulation, such as obesity, diabetes, neurodegenerative disease, cardiovascular disease andcancer. Eukaryotic organisms have evolved an efficient way in sensing, communicating andresponding to cellular stress and regulating mitochondrial activity correspondingly through acomplex network of intercommunicating protein kinases and their downstream effectors. Thisdissertation focuses on the interplay of two of the master metabolic regulators in the cell: AMPKand PASK, and characterization of the functions of their downstream substrates: OSBP andMED13. AMPK is an energy sensing kinase that maintains energy homeostasis in the cell,whereas PASK is a nutrient sensing kinase that regulates glucose partitioning and respiration inthe cell. Both kinases play important roles in mitochondrial function and regulation, anddeficiency in either kinase has been found to associate with various human pathologies. Furthercharacterization of the cross-talk and molecular mechanisms of both kinases in controllingmitochondrial health and function may aid in the identification of new targets for treatingmetabolic diseases.

Characterizing Stress Granule Regulation by PAS Kinase, Ataxin-2 and Ptc6 and Investigating the Lifespan of Covid-19 Virus on Currency

The protein Ataxin-2 is a known positive regulator of stress granules in humans, mice and yeast (known as yeast PBP1). Due to the role that stress granules play in diseases including Amyotrophic Lateral Sclerosis (ALS) and cancer, this thesis investigates the role of Ataxin-2 and its protein binding partners in stress granule development and its effects on various metabolic phenotypes of the cell. PAS kinase is a sensory protein kinase, conserved from yeast to man, which regulates respiration and lipid biosynthesis. Our lab discovered that PAS kinase phosphorylates and activates Ataxin-2 in yeast, and that PAS kinase overexpression enhances localization of Ataxin-2 to stress granules. Our preliminary results from yeast show that PAS kinase positively regulates stress granule formation in response to metabolic stress. Ataxin-2 normally functions to promote stress granule formation and it has been specifically shown to sequester and inhibit mammalian target of rapamycin complex I (mTORC1), a major player in the regulation of cell growth, to stress granules in both yeast and mammalian cells. To build upon this knowledge we performed a large-scale yeast interactome to identify Pbp1 binding partners through yeast-two hybrid and mass spectrometry. We identified 32 novel putative binding partners. A protein of note was Ptc6, a known regulator of mitophagy with human homolog PPM1K, which is not known to be involved in stress granules. Through colocalization with Ppb1 we determined that Ptc6 is sequestered to stress granules under glucose depravation. Under Pbp1 overexpression, Ptc6 was shown to increase localization to a stress granule marker, Pab1, showing that Pbp1 may be actively promoting Ptc6 to stress granules. We investigated the effects of eliminating Pbp1 and Ptc6 in yeast cells, including on mitophagy, mitochondrial quantification, whole cell respiration and mitochondrial reactive oxidative species. In a separate project, due to the outbreak of a worldwide pandemic and early concerns that currency could be a potential SARS-CoV-2 fomite, we investigated whether the virus could survive on varying types of currency. We conducted environmental studies and found no viable virus on bank notes or money cards. In vitro studies with live virus suggested SARS-CoV-2 was highly unstable on banknotes, however SARS-CoV-2 displayed increased stability on money cards with live virus detected after 48 hours.

PAS Kinase and TOR, Controllers of Cell Growth and Proliferation

Nutrient sensing kinases lie at the heart of cellular health and homeostasis, allowing cells to quickly adapt to changing environments. Target of Rapamycin (TOR) and PAS kinase (PASK, or PASKIN) are two such nutrient kinases, conserved from yeast to man. In yeast, these kinases each have paralogs. The two TOR paralogs in yeast mimic the mammalian TORC1 and TORC2 complexes, except both Tor1 and Tor2 may contribute to TORC1 or TORC2 function. The two PAS kinase paralogs are paired with the TOR paralogs, meaning that both Psk1 and Psk2 regulate TORC1, while Psk2 suppresses a temperature-sensitive allele of Tor2. Herein we review the evolutionary models for these paralogs, their function in yeast and mammalian cells, as well as the overlapping function of PAS kinase and TOR. We also use Rice University’s Direct Coupling Analysis algorithms to analyze co-evolutionary relationships and identify potential interaction sites between PAS kinase and several of its substrates.

Le présent travail compte deux parties. La première (section création), intitulée Léon et le vent du diable, consiste en l'adaptation dramatique que j'ai faite du roman de Bruno Hébert C'est pas moi, je le jure ! Cette adaptation a donné lieu à un exercice présenté devant public au LANTISS (Laboratoire des nouvelles technologies de l'image du son et de la scène) de l'Université Laval, en janvier 2006. La deuxième partie (section essai) consiste en un retour sur l'ensemble de l'expérimentation théâtrale (adaptation, mise en scène et création) menée dans le cadre de ma recherche de maîtrise. Il s'agit d'une réflexion personnelle sur mon parcours de création, intitulée « De l'adaptation dramatique de C'est pas moi, je le jure ! à la représentation scénique de Léon et le vent du diable ». Une des représentations publiques a été captée sur DVD. Ce DVD accompagne et complète le travail.

Investigating Security Measures in Common Data Environments: Insights from AEC Industry Case Studies

Data exchange is a vital aspect of the construction industry, which means there is need for a consistent platform to manage documents that can be relied on. An important digital information management system in the Architectural, Engineering, and Construction (AEC) sector is Building Information Modeling (BIM). However, problems exist regarding secure and compatible systems for data sharing. The study explores why adaptable and tailored security measures are needed to suit project specifications. Through this examination of centralized versus decentralized Common Data Environments (CDEs), it emerges that open BIM systems are impractical when compared to closed ones. The findings highlight the crucial role that standardization and customization play towards efficient, safe and flexible BIM implementations. It also recommends further research for future studies as well as emphasizes transparency in implementing CDE-based security protocols.

Notre recherche-création porte sur la problématique que représente l’adaptation d’un texte narratif au théâtre lorsqu’il s’agit d’y incorporer les possibilités offertes par les médias numériques, et d’adopter la forme d’écriture élargie qui en découle. Pour ce processus d’adaptation qui s’éloigne de la façon de procéder traditionnelle, nous optons pour une approche intermédiale. Nous nous penchons plus particulièrement sur le théâtre hypermédia, ce qui nous permet de nous interroger sur les connexions qui surviennent entre les écritures visuelles, sonores, spatiales et textuelles, ainsi que sur leurs dynamiques de coprésence dans la scène théâtrale. Une partie essentielle de la méthodologie de cette recherche-création repose sur l’élaboration d’une série de laboratoires de recherche scénique dans lesquels ont été créés des dispositifs vidéographiques, sonores et spatiaux. Ces dispositifs ont été intégrés dans la création dramaturgique, présentée sous la forme d’une écriture intermédiale. Les chapitres 1 et 2 touchent le cadre conceptuel. Dans le premier chapitre, nous présentons les concepts d’intermédialité, de théâtre comme hypermédia, d’écriture intermédiale ainsi que d’autres notions qui nous permettent de discuter l’adaptation et sa relation avec les médias. Dans le deuxième chapitre, nous étudions la vidéo, le son, l’espace et le texte en relation avec les médias numériques. De plus, nous réfléchissons à la composition dramaturgique de chacun de ces éléments expressifs et aux points de repère qui peuvent contribuer à concevoir la dramaturgie. Les chapitres 3 et 4 discutent le volet pratique et la création. Dans le troisième chapitre, nous décrivons les dispositifs scéniques créés dans les quatre laboratoires de recherche scénique et les processus de création. Nous en faisons une analyse intermédiale. Nous présentons également un modèle de protocole de recherche en création qui a émergé de ces expériences pratiques réalisées au Laboratoire des Nouvelles Technologies de l'Image, du Son et de la Scène (LANTISS). Enfin, dans le quatrième chapitre, nous présentons l’œuvre dramaturgique intitulée Le monde n’arrête pas et son processus de création. / Our research-creation addresses the problematic of adapting a narrative text to the theater when it comes to incorporating the possibilities offered by the digital media, and adopting the expanded text that follows. For this adaptation process, which is moving away from the traditional process, we opt for an intermedial approach. We focus more particularly on the hypermedia theater, which allows us to question the connections that occur between the visual, sonic, spatial and textual writings, as well as on their dynamics of co-presence in the theater scene. An essential part of the methodology of this research-creation rests on the development of a series of scenic research laboratories in which videographic, sound and spatial devices have been created. These devices have been integrated into the dramaturgical creation presented in the form of an intermedial script. Chapters 1 and 2 touch on the conceptual framework. In the first chapter, we present the concepts of intermediality, theater as hypermedia, intermedial writing and other concepts that allow us to discuss adaptation and its relationship with the media. In the second chapter, we study video, sound, space and text in relation to digital media. In addition, we reflect on the dramaturgical composition of each of these expressive elements and the points of reference that can contribute to the conception of dramaturgy. Chapters 3 and 4 discuss the practical and creative aspects. In the third chapter, we describe scenic devices in the four scenic research laboratories and creative processes. We make an intermedial analysis. We also discuss the working protocol that emerged from these practical experiences in the Laboratory of New Technologies of Image, Sound and Scene (LANTISS). Finally, in the fourth chapter, we present the dramaturgical work entitled “The World Does Not Stop” and its process of creation.
