Transmisión de fitoplasmas por el cicadélido Bergallia valdiviana Berg 1881 / Phytoplasmas transmission by the leafhopper Bergallia valdiviana Berg 1881

Memoria para optar al Título Profesional de Ingeniero Agrónomo / Bergallia valdiviana Berg 188, perteneciente a la familia Cicadellidae, es uno de los insectos más comunes en los viñedos chilenos. En algunos individuos de B. valdiviana se ha detectado la presencia de un fitoplasma perteneciente al subgrupo ribosomal 16SrIII-J. Sin embargo, no hay informaciones acerca de su capacidad de trasmitir fitoplasmas. Durante octubre de 2012 a Junio de 2013, en un viñedo infectado con fitoplasmas ubicado en la localidad de Casablanca, Región de Valparaíso, se capturaron individuos adultos de B. valdiviana para realizar pruebas de transmisión en jaulas entomológicas utilizando plantas de vinca (Cantharanthus roseus L.) obtenidas desde semillas. Las capturas se realizaron utilizando una red entomológica. Dos de catorce pruebas resultaron exitosas en la transmisión del fitoplasma 16SrIII-J (relacionado con „Candidatus Phytoplasma pruni‟; X-disease group) a las plantas de vinca, las cuales presentaron fuertes amarilleces en hojas y desordenes en las flores. La detección e identificación del fitoplasma se realizó a través de PCR anidado, secuenciación y RFLP in silico. Los resultados del presente trabajo indican, por primera vez, que B. valdiviana es vector del fitoplasma 16SrIII-J. / Bergallia valdiviana Berg 1881, belonging to the family Cicadellidae, is one of the most common insects in Chilean vineyards. Several individuals of B. valdiviana have been previously reported to be carriers of a phytoplasma belonging to ribosomal subgroup 16SrIII-J. However, there is no information about its capacity to transmit this phytoplasma to plant hosts. From October 2012 to June 2013, a wide number of B. valdiviana adults were captured in phytoplasma infected vineyards located in the locality of Casablanca, in Valparaiso region, in order to perform transmission trials in entomological cages. Non infected periwinkles (Cantharanthus roseus L.) obtained from seeds were used as indicator plants. Catches were made using an entomological net. In 2 out of 14 trials was observed transmission of 16SrIII-J phytoplasma (related to 'Candidatus Phytoplasma pruni'; X-disease group) to periwinkle plants, which showed strong yellowing, disorders in leaves and flowers. Detection and identification of the phytoplasma was carried out by nested PCR, sequencing and in silico RFLP. The results of this study indicate for the first time, that B. valdiviana is vector of the phytoplasma 16SrIII-J.
