ProduÃÃo de biossurfactantes por bactÃrias isoladas do solo, rizosfera e Ãgua do mangue do Rio Cocà (Fortaleza - CE) / Biosurfactants production by bacteria isolated from soil, rhizosphere and water Mangrove Coco River (Fortaleza - CE)

Mangroves are tropical and subtropical coastal ecosystems located in estuarine zones, important for the maintenance of biogeochemical cicles, marine fauna reproduction and the economy. However, these environments are under constant pressure, including contamination by oil and its derivatives threatens these environments. The use of microbial biosurfactants for environmental decontamination has been considered an eco-friendly alternative. The aim of this research was to collect and characterize biosurfactant-producing bacterial strains that emulsify oil products in Rio Cocà Mangrove, Fortaleza, CearÃ. One collection in rainy season and other one in dry season were made in two points, one of them in Cocà Ecological Park and another one in the river mouth. A total, 4 soil samples, 4 water samples and 4 Laguncularia racemosa seedlings were collected. The samples were homogenized and a fraction was added in Erlemeyers containing Bushnell-Haas broth added with kerosene and gasoline and incubated at 30 C for periods of 48 and 72 h. Inocula were made on the surface of TSA agar medium, incubated at 35ÂC for 48 h and then colonies were selected and isolated. The strains were characterized and verified the purity of cultures by Gram staining technique. Biosurfactant production capacity was assessed by drop collapse test and the detected emulsion was after assessed by volume and stability emulsification tests and emulsifier activity test. A total, 58 strains were isolated, 50 are Gram-negative and 8 are Gram-positive, separated according the source: 21 from water, 21 from sediment and 16 from the rhizosphere of seedlings. A total of 55 strains (95%) had positive results in the drop collapse test. All 10 strains with emulsification volume above 50% in 24 hours also exhibited stability above 50% up to 168 hours. Five strains showed emulsifying activity above 2,00 UAE, while 29 strains showed activity between 1,00 and 2,00 UAE. We concluded that it is possible to isolate biosurfactant-producing bacteria from Rio Cocà mangrove and that the most efficient strains in the tests were isolated from the point located in the Cocà Ecological Park. / Manguezais sÃo ecossistemas costeiros tropicais e subtropicais, localizados em zonas estuarinas, importantes para a manutenÃÃo dos ciclos biogeoquÃmicos, reproduÃÃo de espÃcies marinhas e para a economia. Apesar disso, sofrem constantemente pressÃo ambiental, incluindo a contaminaÃÃo por petrÃleo e derivados. O uso de biossurfactantes microbianos para descontaminaÃÃo ambiental tem sido considerado uma alternativa mais sustentÃvel. O objetivo da presente pesquisa foi coletar e caracterizar cepas bacterianas produtoras de biossurfactantes que emulsifiquem derivados de petrÃleo no Mangue do Rio CocÃ, Fortaleza, CearÃ. Foi realizada uma coleta no perÃodo chuvoso e outra no perÃodo seco em dois pontos no Rio CocÃ, sendo um deles no Parque EcolÃgico do Cocà e outro na foz do rio. No total, foram coletadas 4 amostras de solo, 4 de Ãgua e 4 de plÃntulas de Laguncularia racemosa. As amostras coletadas foram homogeneizadas e uma fraÃÃo foi acrescentada em Erlenmeyers contendo caldo Bushnell-Haas adicionado de querosene e gasolina e incubados em estufa a 30ÂC por perÃodos de 48 e 72 h. A partir do crescimento, visualizado pela turvaÃÃo do meio, foram feitos inÃculos sobre a superfÃcie do meio Ãgar TSA, que foram incubadas em estufa a 35ÂC por 48 h e posteriormente as colÃnias crescidas foram selecionadas e isoladas. As cepas foram caracterizadas e a pureza das culturas foi verificada pela tÃcnica de coloraÃÃo de Gram. A capacidade de produÃÃo dos biossurfactantes foi detectada pelo teste do colapso da gota e o emulsificante detectado foi posteriormente avaliado pelos testes de volume e estabilidade de emulsificaÃÃo e atividade emulsificante. Foram isoladas 58 cepas, sendo 50 delas Gram-negativas e 8 Gram-positivas, assim separadas de acordo com a origem: 21 de Ãgua, 21 de sedimento e 16 da rizosfera das plÃntulas. No total, 55 cepas (95%) tiveram resultados positivos no teste do colapso da gota. Todas as 10 cepas que apresentaram volume de emulsificaÃÃo acima de 50% em 24 horas tambÃm apresentaram estabilidade acima de 50% atà 168 horas. Cinco cepas apresentaram atividade emulsificante acima de 2,00 UAE, enquanto 29 cepas mostraram atividade entre 1,00 e 2,00 UAE. Conclui-se que à possÃvel isolar bactÃrias produtoras de biossurfactantes do Mangue do Rio Cocà e que as cepas mais eficientes nos testes realizados foram isoladas do ponto localizado no Parque EcolÃgico do CocÃ.

Testování a optimalizace podmínek tlakového modulátoru pro GCxGC analýzy / Testing and optimization of pressure modulator conditions for GCxGC analyses

Pulse (fluid) pressure modulator was constructed for comprehensive gas chromatography. Optimal conditions of its connection to the gas chromatograph with a flame ionization detector were found for the analysis of selected mixture of volatile solvents. Pressure interdependences in the system and relations between dimensions of modulator capillaries and columns have been evaluated with respect to the length of the modulation period and pulse duration. Two non-polar columns (15 m × 0.25 mm DB-5MS, 10 m × 0.15 mm CP-Sil 5 CB) of the same stationary phase were tested to compare the separation efficiency in the first dimension and the DB-5MS column was selected. The Supelcowax 10 columns of the lengths 1, 2 and 3 m were compared in the second dimension. It was found that using of 3 m length column lead to the extension of analysis time, however the increase in separation efficiency in the second dimension was very evident. The experimental measurements have shown that the selected parameters for the evaluation of the separation efficiency (the peak capacity and total resolution product) were inappropriate. Gasoline, kerosene and diesel fuel were analyzed as examples of real samples. Amount of added ethanol as a biofuel additive was determined in the gasoline sample. Às vezes tem-se urgência no resultado de determinadas análises e isto fica prejudicado pela forma como as análises são feitas. Assim, a quimiometria, associada à espectroscopia molecular (NIR e MIR em particular) vem gerando métodos alternativos para a caracterização e avaliação de propriedades físicas e químicas de petróleos e seus derivados com elevada exatidão, confiabilidade e rapidez. Para melhorar o desempenho previsor têm sido utilizados procedimentos apropriados para a seleção das regiões espectrais associadas com a propriedade de interesse. Desta forma, face às suas aplicabilidades, foi proposto neste trabalho a utilização das ferramentas quimiométricas com seleção de variáveis (método dos mínimos quadrados parciais por intervalos, iPLS, e por sinergismo de intervalos, siPLS; método de eliminação de variáveis não informativas por mínimos quadrados parciais, UVE; e algoritmo genético, GA), associada ao MIR e ao NIR, para a determinação das seguintes propriedades em frações de petróleo: Grau API, Índice de cetano, Índice de refração (a 20°C), Teor de Enxofre (%m/m), Ponto de fuligem (mm), Ponto de anilina (°C), Ponto de congelamento (°C), Ponto de entupimento (°C), Ponto de névoa (°C) e Ponto de fluidez (°C), avaliando, assim, a performance dos modelos obtidos, bem como as técnicas utilizadas na seleção de variáveis. Essa avaliação se deu pela determinação e análise do coeficiente de determinação (R2), de diversos erros calculados para os conjuntos de calibração e previsão. Os modelos foram, ainda, submetidos a testes estatísticos (α=0,05), e tiveram suas figuras de mérito calculadas. Os melhores modelos para a previsão do Grau API e do ponto de névoa foram criados aplicando-se iPLS a dados de MIR, enquanto que para a previsão do teor de enxofre e pontos de refração, de fuligem e de anilina foram criados aplicando-se siPLS também ao MIR. Já para a previsão do índice de cetano e do teor de enxofre e do ponto de entupimento, os melhores modelos foram criados aplicando-se iPLS a dados de NIR. Nesse contexto, o melhor modelo para a predição do ponto de fluidez foi o GA. Finalmente, para a previsão do ponto de congelamento, nenhum método de seleção de variáveis melhorou a capacidade preditiva, quando comparados ao modelo criado aplicando-se PLS a dados de MIR. Dessa forma, conclui-se que houve um melhor desempenho dos modelos criados a partir de dados de MIR. Quanto aos métodos de seleção de variáveis, iPLS e siPLS obtiveram o melhor desempenho. / Petroleum products are, in general, highly complex and a considerable effort is needed to characterize their chemical and physical properties, though sometimes the results of several analyses are urgent and this is compromised by the way the analyses are carried out. Thus, chemometrics associated with molecular spectroscopy (particularly NIR and MIR) has good potential as a tool in analytical chemistry, creating alternative methods to characterize and evaluate physical and chemical properties of petroleum and its derivates with high precision, reliability and rapidity. To improve the predictor performance, appropriate procedures are being used to select spectral regions associated with the property of interest. In face of their applicabilities, this work proposes the use of chemometric tools, with variable selection (Interval Partial Least Square, iPLS and Sinergism Interval Partial Least Square, siPLS; Elimination of Uninformative Variables, UVE and Genetic Algorithm, GA), associated with mid infrared (MIR) and near infrared (NIR) spectroscopies to determine the following properties in petroleum fractions: API gravity, Cetane index, Refractive index (at 20°C), Sulfur content (%m/m), Smoke point (mm), Aniline point (°C), Freezing point (°C), Plugging point (°C), Cloud point (°C) and Pour point (°C), enabling the evaluation of performance of the obtained models, as well as the techniques used in variable selection. This evaluation was performed by determination and analyses of the following requirements: coefficient of determination (R2), several calculated errors for the calibration and prediction set. The models were also subjected to statistical tests (α=0,05), and the figures of merit were calculated. The best models to predict API gravity and cloud point were created by applying iPLS to the MIR data, whereas for prediction of sulfur content, refractive index, and smoke and aniline points the models were created by applying iPLS to NIR data. In this context, the best model to predict the pour point was the GA. Finally, to predict freezing point, none of the variable selection methods improved the predictive capability when comparing to the model created using only PLS in MIR data. Thus, the conclusion is that a better performance was obtained for the models created from MIR data. Regarding efficiency of variable selection methods, the iPLS and siPLS methods resulted in a best performance.
