This thesis describes the applied research of motivating factors conducted among paramedical staff in the Hospital "Nemocnice Rudolfa a Stefanie Benešov" in 2010. The objective was to study several dimensions of work motivation, with special emphasis on motivational factors and motivational potential of work. Purpose of the project also represented a compilation of the consultancy recommendations arising from the results and their providing to the hospital staff. These recommendations should contribute to more effective application of targeted motivating of subordinates by managers of paramedical staff in this hospital. We found out that the strongest motivating factors for respondents include patient satisfaction, good interpersonal relationships at work, expressions of recognition and work achievement. It was also suggested that the caregivers see the highest motivation potential of work in the importance of the performed tasks; that is the possibility to participate actively in influencing people's health. Adam Emmer: Potential hazardousness of selected moraine-dammed lakes in the Cordillera Blanca, Peru ABSTRACT: This work is devided into the two parts. Firs part reviews contemporary methods of assessment of potential hazardousness for moraine-dammed lakes (eight qualitative methods and three quantitative methods). Second part has two main aims: 1) Assessment of potential hazardousness of seven selected moraine-dammed lakes in the Cordillera Blanca, and 2) Analysis of suitability of these methods for use in this region. Required input data for potential hazardousness assessment were gained from analysis of remotely sensed photographs, research papers of INRENA/ANA institute (Huaráz), and from fieldwork realized in 2012. Ten methods of potential hazardousness assessment were applied on seven studied moraine-dammed lakes. There are no significant differences in results obtained in each method. These results showed, that potentially most hazardous lake is that of Arhueycocha, followed by Palcacocha. On the other hand potentially less hazardous lake is that of Rajucolta. The genus Blastocystis has recently attracted the attention of scientists, especially parasitologists. Similarly to the related opalines and proteromonads, Blastocystis is anaerobic and lives endobiotically in the intestine of various animals. This organism is also often found in humans, where it is associated with irritable bowel syndrome, though its pathogenic potential remains uncertain. The genus Blastocystis is remarkable for its rich genetic diversity. The taxonomy of Blastocystis is inconsistent and problematic. The strains isolated from homoiothermic vertebrates are divided into 17 subtypes, while strains from poikilotherms are either classified as separate species or are not considered in taxonomic studies at all. The aim of the study was to further examine the genetic diversity of the genus Blastocystis. We determined SSU rDNA sequences of 38 strains isolated from poikilothermic vertebrates and arthropods. The results of our phylogenetic analysis showed that Blastocystis is considerably diverse in these hosts, and we defined 21 new subtypes. The total number of known subtypes of Blastocystis has thus increased to 38. We also examined light-microscopical morphology of some strains. Analýza stabilitních podmínek při silných srážkách v ČR / Analysis of stability conditions during heavy rains in the Czech republic

In this thesis I describe the conceptual model of three kinds of instability in terms of precipitation. I describe ways of evaluating their presence in the atmosphere. They are: conditional instability, potential instability and symmetric instability. I have selected three events with strong precipitation in the Czech Republic so that the formation of each of them is with high probability involved in just one of the three kinds of instability. Events are first described using distance and station measurements. Through the NWP model COSMO are created prognostic fields of precipitation and several derived thermodynamic quantities for each event. On the horizontal (for the whole country) and vertical (for selected areas of the Czech Republic) distribution of these variables are demonstrated favourable conditions for the occurrence of the types of instability in the atmosphere. In the event of conditional instability the rainfall occurs in areas with high CAPE and negative or very small positive vertical lapse rate of potential temperature. The event with the potential instability is characterized by the occurrence of negative vertical lapse rate of equivalent potential temperature in the broad layers. Modifications de surface des nanodiamants : compréhension des mécanismes d’échanges électroniques et mise en évidence d’un effet thérapeutique / Nanodiamonds surface modifications : understanding of electron exchange mecanisms and evidence of a therapeutic effect

A partir de l'étude des effets de la chimie de surface des Nanodiamants (NDs) sur leurs propriétés électroniques, cette thèse a permis la mise en évidence d'un effet thérapeutique des NDs sur des cellules humaines. En particulier, il a été montré que les NDs de détonation peuvent générer des radicaux libres oxygénés sous radiation ionisante, ce qui pourrait améliorer l'efficacité de certains traitements de radiothérapie actuels. Les échanges électroniques entre le coeur des NDs et leur environnement sont en effet favorisés après des traitements de surface, notamment d'hydrogénation et de graphitisation de surface. Les conditions expérimentales permettant d'obtenir des NDs hydrogénées (NDs-H) sous plasma d'hydrogène ont été optimisées sous ultravide, puis ont été utilisées pour préparer de grandes quantités de NDs-H sous forme pulvérulente. La même procédure a été appliquée pour la graphitisation de surface des NDs, en utilisant des recuits sous vide à haute température. L'effet de ces traitements de surface sur les propriétés d'interactions électroniques des NDs a été étudié après exposition à l'air ambiant, puis en dispersion dans l'eau. Ces traitements de surface assurent notamment un potentiel Zeta positif aux NDs, dont l'origine a été discutée. Enfin, les interactions des NDs avec plusieurs lignées de cellules tumorales humaines ont été étudiées et l'efficacité des NDs pour radiosensibiliser des cellules radiorésistantes sous irradiation gamma a été montrée, ouvrant de nouvelles perspectives d'applications des NDs en nanomédecine. . / In this thesis, a therapeutic effect of nanodiamonds (NDs) has been evidenced by investigating the role of NDs surface chemistry on their electronic properties. More precisely, the generation of reactive oxygen species from detonation NDs under ionizing radiation, which could improve current radiotherapy treatments, has been demonstrated. To this end, surface treatments facilitating electron transfer from NDs to their environment, namely hydrogenation and surface graphitization, were developed. Experimental conditions ensuring an efficient hydrogenation by hydrogen plasma were determined under ultrahigh vacuum, before being used to prepare large quantities of NDs in powder phase. A similar procedure was applied to the surface graphitization of NDs, performed by annealing under vacuum at high temperature. The impact of such surface treatments on the electronic interaction properties of NDs has been investigated under ambient air and after dispersion in water. These surface treatments induce a positive Zeta potential to NDs in water, which origin has been discussed. Finally, their interactions with human tumor cells were observed. Radiosensitization of tumor cells using NDs under gamma irradiation was demonstrated, opening new perspectives for NDs in nanomedicine.

Studium migračního potenciálu testikulárních kmenových buněk u Xenopus tropicalis / Study of migration potential of testicular stem cells in Xenopus tropicalis

Because of their possible therapeutic potential stem cells are one of the most promising fields of study in biology. The aim of this thesis was to study the migration potential of testicular stem cells of Xenopus tropicalis, whose culture was established from the testes of juvenile males in the laboratory of my supervisor. Due to external fertilization and embryonic development Xenopus represents an ideal model organism for transplantation and microinjection experiments in-vivo. Transplantation of vital marked (PKH26) testicular stem cells into blastula and peritoneum of tadpoles showed their wide migratory potential including intestine (entoderm), heart, pronephros, genital groove (mesoderm) and epidermis (ectoderm). Based on my experiments, I found that the ideal number of cells for transplantation ranges between 250-500 per tadpole. To further characterize the stem cells, I constructed a plasmid vector carrying a gene for a red fluorescent protein. This plasmid was then used for preparation of frogs with whole-body expression of Katushka RFP under control of CAG promoter. The next aim is to gain the RFP positive offspring by crossing the transgenic individuals with the wild type. Flygindustrin fortsätter kontinuerligt att växa, med nyare och effektivare flygplan skapar flygbolagen en konkurrenskraft flygbolag emellan samtidigt som de säkrar deras egna framtid med vinster och besparingar i respektives ekonomi. Syftet med denna rapport är att bedöma potentialen för rörliga winglets på framtidens kortdistansflygplan. Kan detta koncept skära ner på flygbolagens ekonomi? Med oljeprisets historiska utveckling går det att fastställa att olika händelser runtom i världen kan påverka utveckling antingen negativt eller positivt. Genom konceptet rörliga winglets kan flygbolag förebygga de negativa konsekvenserna av oljeprisutvecklingen och i bästa fall öka de positiva konsekvenserna. För att bedöma potentialen fokuseras arbetet på följande två frågeställningar: Kan anpassningsbara winglets leda till en förbättring? Hur ser de mekaniska- och aerodynamiska krafterna ut på wingletsen vid förbättring?   Angreppsättet av frågeställningarna har gått till på sådant vis att krafterna som verkar på wingletsen har gjorts om till vektorer. Genom att vektorer användes för att undersöka frågeställningarna blev det lättare att illustrera samt matematiskt beräkna effekterna vid olika kantvinklar och anfallsvinklar. Resultatet visade att det finns en förbättringsmöjlighet på dagens winglets. Dock finns det mycket kvar att göra gällande andra aspekter som också berör potentialen.

B-Spline Boundary Element Method for Ships

The development of a three dimensional B-Spline based method, which is suitable for the steady-state potential flow analysis of free surface piercing bodies in hydrodynamics, is presented. The method requires the B-Spline or Non Uniform Rational B-Spline (NURBS) representation of the body as an input. In order to solve for the unknown potential, the source surface, both for the body as well as the free surface, is represented by NURBS surfaces. The method does not require the body surface to be discritized into flat panels. Therefore, instead of a mere panel approximation, the exact body geometry is utilized for the computation. The technique does not use a free surface Green's function, which already satisfies the linear free surface boundary conditions, but uses a separate source patch for the free surface. By eliminating the use of a free surface Green's function, the method can be extended to considering non-linear free surface conditions, thus providing the possibility for wave resistance calculations. The method is first applied to the double body flow problem around a sphere and a Wigley hull. Some comparisons are made with exact solutions to validate the accuracy of the method. Results of linear free surface conditions are then presented.
