O objetivo deste estudo foi avaliar comparativamente a express?o das prote?nas Ki-67 e IMP-3 em QA e CELI a fim de contribuir com informa??es adicionais sobre a carcinog?nese em l?bio inferior. Foram estudados 33 casos de QA e 33 casos de CELI, sendo analisadas as caracter?sticas cl?nico-patol?gicas e a imunoexpress?o do Ki-67 e IMP-3. A an?lise imuno-histoqu?mica do Ki-67 se deu atrav?s da determina??o do ?ndice de prolifera??o (IP) e subsequente classifica??o dos casos de acordo com os escores : 0 (0% de c?lulas positivas), +1(?30%), +2 (>30% a ?60%) e +3 (>60%). Para aplica??o dos testes estat?sticos os casos foram classificados em: sem marca??o (escore 0), baixa express?o (escore +1) e alta express?o (escores +2 e +3). Para a express?o do IMP-3, foi estabelecido o percentual de c?lulas epiteliais imunomarcadas, sendo atribu?dos os escores: 0 (correspondeu a 0%), +1 (at? 30% das c?lulas positivas); +2 (entre 30% a 60% de c?lulas imunomarcadas) e +3 (acima de 60% das c?lulas positivas). Foram utilizados os testes estat?sticos Qui-quadrado de Pearson, Mann-Whitney e Wilcoxon. O n?vel de signific?ncia adotado foi de 5%. A maioria dos caos de QA foi do sexo masculino (78,8%), com m?dia de idade de 50 anos e dos casos de CELI tamb?m predominou o sexo masculino (69,89%) com m?dia de 62 anos. O Ki-67 se expressou em todos os casos de QAs bem como nos casos de CELI, predominando nas duas les?es o escore 2, correspondendo a 81,8% dos casos nas QAs e 54,5% nos CELI. A express?o do IMP-3 nas QAs ocorreu em 72,7% dos casos, com predomin?ncia em 36,3% dos casos do escore 1. J? nos CELI o IMP-3 se expressou em 60,6% dos casos, com predomin?ncia em 27,3% dos casos do escore 3. Estes resultados nos permite concluir que a express?o do IMP3 e da atividade proliferativa s?o eventos precoces na carcinog?nese de l?bio inferior independente do estado da altera??o. / Epithelial changes observed in actinic cheilitis (AC) and squamous cell carcinoma of the lower lip (LLSCC) are mainly caused by chronic exposure to ultraviolet rays (UV) and are studied using different immunohistochemical markers trying to evaluate the process of carcinogenesis. The objective of this study was to comparatively evaluate the expression of Ki-67 proteins and IMP-3 in AC and LLSCC to contribute with additional information on carcinogenesis in lower lip. A total of 33 cases of AC and 33 cases of LLSCC were studied, analyzed the clinical and pathological features and immunostaining of Ki-67 and IMP-3. Immunohistochemical analysis of Ki-67 was made through the determination of the proliferation index (PI) and subsequent classification of the cases according to the scores: 0 (0% positive cells) +1 (?30%) + 2 (> 30% and ?60%) and +3 (> 60%). For statistical tests cases were classified as unmarked (score 0), low expression (score +1) and high expression (scores +2 and +3). For the expression of IMP-3, the percentage of immunostained epithelial cells was established, and assigned scores: 0 (corresponding to 0%), +1 (up to 30% of positive cells); +2 (From 30% to 60% of immunostained cells) and +3 (over 60% of positive cells). Statistical tests chi-square test, Mann-Whitney and Wilcoxon were used. The significance level was 5%. Most AC chaos was male (78.8%) with mean age of 50 years and cases of LLSCC also were male (69.89%) with an average of 62 years. The Ki-67 was expressed in all cases of AC and in cases of LLSCC, predominantly in the two injuries the score 2, corresponding to 81.8% of cases in ACs and 54.5% in the CELI. The expression of IMP-3 in ACs occurred in 72.7% of cases, predominantly in 36.3% of LLSCC cases score 1. Already in the IMP-3 was expressed in 60.6% of cases, especially in 27.3% of the score of the cases 3. These results allow us to conclude that the expression of IMP3 and proliferative activity are early events in carcinogenesis independently lower lip state of change.

Efeitos da laserterapia na atividade biol?gica de c?lulas de carcinoma epiderm?ide de l?ngua humano

It has been speculated that the biostimulatory effect of LLLT could cause undesirable enhancement of tumor growth in neoplastic diseases, since the malignant cells are more susceptible to proliferative stimuli. Within this context, this study evaluated the effect of LLLT on epidermoid carcinoma of the tongue cell line (SCC25) proliferation and invasion. Cultured cells were irradiated with an InGaAIP diode laser, 660nm, 30mW using two energy densities (0.5J/cm2 and 1.0J/cm2). Proliferative activity was assessed through trypan blue staining method and through cell cycle analysis using flow cytometry. The invasive potential was measured through cell invasion assay using matrigel. Cyclin D1, E-cadherin, -catenin and MMP-9 expressions were analyzed by immunofluorescence and flow cytometry and related to the investigated biological activities. Proliferation curve demonstrated that SCC25 irradiated with 1.0J/cm2 had the highest proliferative rate when compared to the control group and the group irradiated with 0.5J/cm2 (p<0.05). LLLT affected cell cycle distribution and energy density of 1.0 J/cm2 promoted a higher percentage of cells in S/G2/M phases, with statistically significant differences at 24h interval (p<0.05). LLLT, mainly with 1.0J/cm2, revealed significantly higher potential for invasion and influenced the expression of cyclin D1, E-cadherin, -catenin and MMP-9, promoting the malignant phenotype. In conclusion, our results indicate that LLLT has an important stimulatory effect on proliferation and invasion of SCC25 cells, likely due to altered expression of proteins associated with these processes / O aumento da prolifera??o celular ap?s utiliza??o do laser de baixa pot?ncia (LBP) tem sido observado em muitos tipos de c?lulas in vitro, incluindo ceratin?citos, fibroblastos, osteoblastos, c?lulas endotelias e c?lulas-tronco. Tem sido especulado que o crescimento de c?lulas malignas tamb?m pode ser estimulado pela irradia??o laser, uma vez que estas c?lulas s?o mais suscept?veis aos est?mulos proliferativos. Assim, em pacientes portadores de c?nceres de cabe?a e pesco?o, as c?lulas tumorais podem estar presentes no campo de irradia??o ou pr?ximas a ele, sendo a laserterapia n?o intencional um fator estimulante da progress?o tumoral. As c?lulas cultivadas foram irradiadas com um laser diodo (InGaAlP) com 30mW, 660nm e doses de 0.5 e 1.0J/cm2. A atividade proliferativa foi investigada atrav?s da curva de prolifera??o utilizando o m?todo de colora??o por azul de tripan e an?lise do ciclo celular atrav?s da marca??o por iodeto de prop?dio em citometria de fluxo. O potencial invasivo das c?lulas SCC25 foi verificado atrav?s da realiza??o de um ensaio de invas?o celular utilizando o matrigel. A an?lise da express?o da ciclina D1, E-caderina, -catenina e MMP-9, atrav?s da imunofluoresc?ncia e citometria de fluxo, foi relacionada ?s altera??es nas atividades biol?gicas estudadas. A curva de prolifera??o revelou maior crescimento das c?lulas irradiadas com dose de 1.0 J/cm2 (p<0.05). O LBP influenciou a distribui??o do ciclo celular, com destaque para a dose de 1.0J/cm2, a qual favoreceu a maior porcentagem de c?lulas na fase S/G2/M com diferen?a estatisticamente significativa no intervalo de 24 horas (p<0,05). O LBP, principalmente com dose de 1.0J/cm2, promoveu maior invas?o das c?lulas estudadas e foi capaz de influenciar a express?o da ciclina D1, -catenina, E-caderina e MMP-9, favorecendo o fen?tipo maligno. Dessa forma, nossos resultados indicam que a laserterapia teve um importante efeito estimulat?rio nas atividades de prolifera??o e invas?o das c?lulas SCC25, provavelmente por influenciar a express?o de prote?nas que possuem papel importante nestes processos.

Express?o imunoistoqu?mica de GLUT-1 e marcadores de prolifera??o e apoptose em anomalias vasculares orais

This study aimed by histopathology and immunohistochemical expression of human glucose transporter protein (GLUT-1), correctly identify and classify oral vascular anomalies, besides analyzing the immunoexpression of markers proliferation and apoptosis (Ki-67 and Bcl-2). All cases diagnosed as "oral hemangiomas" belonging to the archives of the Service of Pathological Anatomy from the subject of Oral Pathology of the Department of Dentistry (DOD), of the Federal University of Rio Grande do Norte (UFRN) were reviewed, totalizing 77 cases. Immunohistochemical analysis for GLUT-1 showed that only 26 (33.8%) of the specimens were true infantile hemangiomas (IHs). The 51 (66.2%%) GLUT-1 negative specimens were then reclassified as pyogenic granulomas (PGs) and vascular malformations (VMs) from their histopathologic characteristics,totalizing 26 (33.8%) cases of IHs, 20 (26.0%) of PGs and 31 (40.2) cases of oral VMs. The cases analyzed by the marker Ki-67 showed different median IH (13,85), PG (33,70) and VM (4.55) with statistically significant differences between them (p <0.001). In relation to the protein Bcl-2, the groups also showed different median of the established scores IH (1.00), PG (1.50), VMs (0.0) demonstrating statistically significant differences between them (p<0,001). No statistically significant correlation between the indexes of positivity for Ki-67 and the scores of immunoexpression of Bcl-2 were observed in any group. Thus, we can conclude that it is necessary a careful and parameterized review of cases of vascular anomalies making use of auxiliary tools such as GLUT-1, since the histopathological findings alone, sometimes, are not sufficient to differentiate some anomalies. Furthermore, analysis of the expressions of markers involved in the levels of proliferation of lesions is important for a better understanding of its biological behavior aspect / As anomalias vasculares constituem um grupo de les?es distintas, mas que podem apresentar caracter?sticas cl?nicas e histopatol?gicas semelhantes, que podem levar a equ?vocos diagn?sticos.Este estudo objetivou por meio da histopatologia e da express?o imuno-histoqu?mica daprote?na humana transportadora de glicose (GLUT-1), identificar e classificar corretamente as anomalias vasculares orais, al?m de analisar a imunoexpress?o de marcadores de prolifera??o e apoptose (Ki-67 e Bcl-2).Todos os casos diagnosticados como hemangiomas orais pertencentes aos arquivos do Servi?o de Anatomia Patol?gica da disciplina de Patologia Oral do Departamento de Odontologia (DOD) da Universidade Federal do Rio Grande do Norte (UFRN) foram revisados, totalizando 77 casos. A an?lise imuno-histoqu?mica para GLUT-1 revelou que apenas 26 (33,8%) dos esp?cimes tratavam-se de hemangiomas da inf?ncia (HIs) verdadeiros. Os 51 (66,2%%)esp?cimes GLUT-1 negativos foram ent?o reclassificados em granulomas piog?nicos (GPs) e malforma??es vasculares (MVs) a partir de suas caracter?sticas histopatol?gicas, totalizando 26 (33,8%) casos de HIs, 20 (26,0%) de GPs e 31 (40,2) casos de MVs orais. Os casos submetidos ? an?lise do marcador Ki-67 apresentaram medianas diferentes HI (13,85), GP (33,70) e MV (4,55) com diferen?as estatisticamente significantes entre elas (p<0,001). Em rela??o ? prote?na Bcl-2, os grupos tamb?m apresentaram diferentes medianas dos escores estabelecidos HI (1,00), GP (1,50), MVs (0,0), demonstrando diferen?as estatisticamente significantes entre elas (p<0,001). N?o foi observada correla??o estatisticamente significante entre os ?ndices de positividade para o Ki-67 e os escores de imunoexpress?o de Bcl-2 em nenhum grupo.Dessa maneira, podemos concluir que se faz necess?rio uma revis?o criteriosa e parametrizada dos casos de anomalias vasculares utilizando ferramentas auxiliares, como a GLUT-1, uma vez que os achados histopatol?gicos sozinhos, ?s vezes, n?o s?o suficientes para diferenciar algumas anomalias.
