Mitjançant una anàlisi cronològica de l'obra assagística de Christa Wolf -una de les autores més reconegudes de l'Alemanya socialista des del final dels anys 60 fins avui- i dels seus diaris, aquesta tesi presenta un seguiment dels principals punts de reflexió de l'autora pel que fa a diversos conceptes lligats a la realitat de la República Democràtica Alemanya, especialment els que fan referència al paper de l'intel·lectual i de la dona i als canvis experimentats en el seu propi compromís polític, a mesura que la postura oficial del govern s'anava radicalitzant i, especialment, durant i després de la reunificació. Per comprovar el reflex d'aquests temes en la seva obra, així com la seva evolució, s'han escollit quatre relats separats en el temps i alhora significatius quant a temàtica. L'anàlisi i comentari dels textos assagístics i literaris conformen els quatre capítols centrals de la tesi, que els pren com a font d'informació per definir la particular recerca d'una opció alternativa davant la realitat. Es parteix de la hipòtesi que aquesta recerca ve definida per una crítica al dualisme originat per una mentalitat racionalista radical, i té en el llenguatge el seu principal instrument manipulador. La comprovació d'aquesta hipòtesi ve a confirmar l'obra de Wolf com a exemple de coherència i reflexa un esforç constant per assimilar vida i literatura amb un marcat tarannà pacifista i conciliador. / By way of a chronological analysis of the essays of Christa Wolf -one of the best known authors from socialist Germany since the 60s until now- and of her diaries, this thesis goes in pursuit of the main matters of reflection of the author related to the reality in the Democratic Republic of Germany. It particularly points at those that directly deal with the role of intellectuals and women in society and with the changes that she experienced in her own political engagement as the government position became more and more radical, especially during and after German Reunification. Four narrations, quite different in time and subject, have been selected to verify the reflection of these themes in her literary work, as well as their evolution in time. The analysis and commentary of the essays, diaries and narrations shape the four central chapters of the thesis, which takes them as a source of information in order to define the particular search of an alternative option in front of reality. As hypothesis, it is assumed that this search is defined by the criticism to dualism originated by inherited extreme rationalist mentality, and it is based on the language as its main manipulating instrument. The verification of this hypothesis confirms the work of Wolf as an example of coherence and it reflects a constant effort to assimilate life and literature with a deep pacifist and conciliating will.

Influence of management practices on weed communities in organic cereal production systems in Saskatchewan

Management practices on organic farms in Saskatchewan are largely unstudied, as are their effect on weed populations and soil quality. The objective of this study was to document what management practices are used on organic farms, classify those practices into management systems and determine if those management systems affect weed populations and soil properties. During the 2002 growing season 73 organic fields in the province of Saskatchewan were surveyed. Three components comprised the data set for each field: a management questionnaire, weed counts, and soil samples that were collected and analyzed for various soil properties. Classification of the management practices identified farming systems: the diverse cropping system, the diverse cropping system using green manure, the low diversity cropping system using summerfallow, and the moderately diverse cropping system using perennials in rotation. Ordination of weed data and the four systems was done with redundancy analysis. It determined that the farm management systems only accounted for 5% of the variation in the weed populations. The only system that affected the weed populations was the moderately diverse cropping system using perennials in rotation. Soil properties were compared among the different management systems. Soil properties were not different between the diverse cropping system using green manure, and the low diversity cropping system using summerfallow. The system that included perennials in rotation had significantly lower pH, electrical conductivity, soil organic matter, phosphorous and potassium levels. The nutrient levels in all systems were low, underscoring the importance of nutrient additions to export farming systems.

A principal porta de entrada deste patógeno é pela via respiratória, disseminando-se posteriormente e atingindo principalmente o sistema nervoso central, provocando a meningite criptocóccica. A primeira parte deste estudo teve como objetivos determinar, nos 106 isolados clínicos de C. neoformans obtidos de dois Estados (São Paulo e Rio de Janeiro), (1) as variedades e (2) os mating-types por PCR (Polymerase Chain Reaction), (3) analisar a diversidade genética por PCR-fingerprinting com a seqüência iniciadora específica para regiões microssatélite (GACA)4, (4) por RAPD (Random Amplification of Polymorphic DNA) com o iniciador 6 e (5) por PCR-RFLP (Restriction Fragment Length Polymorphism) do gene da fosfolipase B (PLB1) digerido com a enzima de restrição AvaI e (6) determinar a sensibilidade a quatro antifúngicos (fluconazol, itraconazol, 5-fluorocitosina e anfotericina B) seguindo o método de referência (documento M27-A2) do CLSI (Clinical and Laboratory Standards Institute). A segunda parte teve como objetivo, analisar transcritos diferencialmente expressos durante a infecção pulmonar de C. neoformans, pela técnica de RDA (Representational Difference Analysis). Entre os 106 isolados, 104 foram identificados como C. neoformans e apenas dois foram C. gattii (=C. neoformans var. gattii), todos MATa. O tipo molecular VNI (C. neoformans var. grubii, sorotipo A) foi o mais prevalente entre os isolados (97/106), seguido do tipo VNII (C. neoformans var. grubii, sorotipo A) (7/106) e VGII (C. gattii, sorotipos B ou C) (2/106) quando analisados por PCR-fingerprinting e PCR-RFLP. Homogeneidade alta foi obtida com o iniciador (GACA)4, com a maioria dos isolados apresentando correlação em torno de 0,9. Os resultados do RAPD, por sua vez, revelaram maior heterogeneidade com número maior de perfis moleculares. / Abstract: Cryptococcus neoformans is an important pathogen, mainly in immunocompromised patients. The pathogen penetrates mainly by respiratory way, disseminate afterward and reach specially the central nervous system causing the cryptococcal meningitis. The first part of this study had the objective to determine, in the 106 clinical isolates of C. neoformans obtained from São Paulo and Rio de Janeiro State, (1) the varieties and (2) mating-types by PCR (Polymerase Chain Reaction), (3) analyze the genetic diversity by PCR-fingerprinting with specific primer to microsatellite regions (GACA)4, (4) by RAPD (Random Amplification of Polymorphic DNA) with primer 6 and (5) by PCR-RFLP (Restriction Fragment Length Polymorphism) of the phospholipase B gene (PLB1) digested with restriction enzyme AvaI and (6) to determine the susceptibility to four antifungal (fluconazole, itraconazole, 5-flucytosine and amphotericin B) following the reference method (document M27-A2) from CLSI (Clinical and Laboratory Standard Institute). The second part had the goal to analyze differentially expressed transcription during pulmonary infection of C. neoformans, by RDA (Representational Difference Analysis) technique. Among 106 isolates, 104 were identified as C. neoformans and only two were C. gattii (=C. neoformans var. gattii), all MATa. The molecular type VNI (C. neoformans var. grubii, serotype A) was the most prevalent among the isolates (97/106), followed by VNII (C. neoformans var. grubii, serotype A) (7/106) and VGII (C. gattii, serotypes B ou C) (2/106) when analyzed by PCR-fingerprinting and PCR-RFLP. High homogeneity was obtained with the primer (GACA)4, with most of the isolates showing correlation around 0.9.
