Radiobiological end-points for the theoretical evaluation of the effectiveness of carbon ions and photons in treating tumours with dynamic hypoxiaLaura, Antonovic January 2014 (has links)
Tumours are characterised by unorganised vasculature, which often results in hypoxic regions. Hypoxia is a common cause for photon radiotherapy (RT) treatment failure, as hypoxic cells require up to 2-3 times higher doses compared to well-oxygenated cells for the same effect in terms of cell kill. The increase in dose that would be required to treat the tumours of cancer patients is limited by the radiation sensitivity of surrounding normal tissues. Using carbon ions instead of photons, the radiation dose can be conformed to the tumour to a much higher degree, resulting in an improved sparing of normal tissues. In addition, carbon ions have a much higher radiobiological effectiveness near the end of their range, which is positioned in the tumour. Also, the radiation modes of action leading to cell death when carbon ions interact with living tissues, are less sensitive to the oxygen status compared with the action modes of photons. The focus of this thesis lies in the development of models for the computation of the cell surviving fraction and tumour control probability (TCP) in hypoxic tumours after photon and carbon ion RT. The impact of fractionation was evaluated with regard to possible spatial changes in oxygenation, both for stereotactic body RT and for carbon ion RT. The feasibility of a method to determine and deliver the optimal photon dose for achieving a high TCP according to spatial variations in radiation sensitivity was evaluated in a treatment planning study. The radiobiological models were finally used for the theoretical quantification of the gain in using carbon ions instead of photons. The results show that there are great possibilities to increase the number of positive outcomes of radiation treatment of tumours if the key influential factors are taken into account, such as level and distribution of hypoxia, radiation quality and choice of fractionation schedule. / <p>At the time of the doctoral defence the following papers were unpublished and had a status as follows; Paper 3: Manuscript; Paper 4: Epubl ahead of print; Paper 5: Manuscript</p>

Previous issue date: 2013-12-13 / CNPq / Derivados de furano e tiofeno 2-substituídos são compostos bastante difundidos no meio ambiente e desempenham papel importante em processos bioquímicos. Além disso, estes compostos podem ser utilizados como building blocks na preparação de diversas moléculas bioativas. No entanto, reagentes prejudiciais ao meio ambiente e muitas etapas reacionais, são geralmente atribuídos às metodologias sintéticas que empregam estes derivados. Atualmente, poucos trabalhos têm reportado a utilização de enzimas para este tipo de sistema. Dessa forma, nosso grupo demonstrou interesse em propor uma metodologia quimioenzimática para preparação de derivados de furano e tiofeno 2-substituídos em suas formas enantiomericamente pura, e aplicá-los na preparação de moléculas bioativas enantiodependentes. A metodologia proposta emprega um número reduzido de etapas sintéticas e utiliza a enzima CAL-B na etapa chave para obtenção dos respectivos enantiômeros separadamente. Inicialmente, uma série de alcoóis secundários racêmicos contendo grupos heterocíclicos foi sintetizada em rendimentos que variaram de moderados a excelentes (40-98%) e submetidos à resolução cinética enzimática catalisada por lipase (RCE). Adicionalmente, um derivado dibromo vinilfurano 2-substituído foi empregado em estudos visando à síntese total da modiolida A, uma macrolactona de origem natural contendo 10 membros em seu anel. Este fragmento foi utilizado em uma reação do tipo Corey-Fuchs para obtenção de um intermediário avançado na síntese desta molécula. / Furan and Tiophene derivatives are widespread compounds in the environment and they also play important role on biochemical process. In addition these compounds can be employed as building blocks in the synthesis of bioactive molecules. However, environmentally harmful reagents and many reaction steps are generally assigned to synthetic methodologies that employ it. Nowadays, few works have been reported enzyme applications in this type of system. Here, we propose a chemoenzymatic methodology to prepare enantioenriched secondary alcohols containing furan and tiophene heterocyclic group and apply them in the preparation of bioactive enantiodependent molecules. The proposed methodology employs a reduced number of synthetic steps and CAL-B enzyme at the key step of deracemization. Initially, the racemic secundary alcohols containing heterocyclic groups were synthesized from moderate to good yields (40-98%) and each of them was submitted to lipase-catalyzed enzymatic kinetic resolution (EKR). Furthermore, a dibromovinylfuran 2-substituted has been employed in studies forward the total synthesis of Modiolide A, a 10-membered ring of natural lactone. This fragment has been used on Corey-Fuchs reaction in order to prepare an advanced intermediate to obtain the Modiolida A.

Agrégation de la convection dans un modèle de circulation générale : mécanismes physiques et rôle climatique / Aggregation of convection in a general circulation model : physical mechanisms and role in climateCoppin, David 20 February 2017 (has links)
Cette thèse porte sur l'agrégation de la convection dans le modèle de circulation générale LMDZ5A à l'équilibre radiatif-convectif (RCE). L'instabilité du RCE mise en évidence nous permet d'étudier les mécanismes d'initiation de l'agrégation et leur dépendance à la température de surface océanique (SST). A basse SST, l'agrégation résulte d'un couplage entre la circulation grande-échelle et les effets radiatifs des nuages bas. A haute SST, elle provient d'un couplage entre la circulation de grande-échelle et les flux turbulents à la surface. Le couplage de l'atmosphère avec une couche de mélange océanique rend l'initiation de l'agrégation moins dépendante de la SST et des mécanismes d'initiation, à l'exception des effets radiatifs des nuages hauts. L'impact de l'agrégation sur la sensibilité climatique et la température de surface est aussi analysé. En favorisant la formation de zones ciel clair sèches, l'agrégation refroidit fortement le système climatique. Toutefois, cet effet est limité par l'effet des changements de gradients de SST et de fraction de nuages bas qui tendent au contraire à faire augmenter la sensibilité climatique. Aux plus courtes échelles temporelles, en revanche, le couplage entre océan et agrégation de la convection est à l'origine d'une boucle de rétroaction stabilisatrice qui contrôle l'agrégation et renverse complètement son effet. Ainsi, l'effet de l'agrégation sur la sensibilité climatique est assez faible par rapport à ce que laissent penser les simulations où le couplage océan-atmosphère est absent. Ces résultats montrent l'importance de considérer le couplage océan-atmosphère dans l'étude du rôle de l'agrégation dans le climat. / This thesis focuses on the study of convective aggregation in LMDZ5A general circulation model, used in Radiative-Convective Equilibrium (RCE) configuration. The instability of the RCE allows us to look at the mechanisms controlling the initiation of convective aggregation and its dependence on sea surface temperatures (SST). At low SSTs, a coupling between the large-scale circulation and the radiative effects of low clouds is needed to trigger self-aggregation. At high SSTs, the coupling between the large-scale circulation and the surface fluxes controls this initiation. When the atmosphere is coupled to a slab ocean mixed layer, SST gradients facilitate the initiation of convective aggregation. Except for the high-cloud radiative effects, triggering mechanisms are less crucial. Convection also becomes less dependent on the SST.The impact of convective aggregation on the climate sensitivity and surface temperature is also analyzed. Convective aggregation is found to increase the area of dry clear-sky zones. Thus, it tends to cool the system very efficiently. However, the negative feedback associated with an increase in aggregation is generally balanced by offsetting changes in SST gradients and low clouds that tend to increase the climate sensitivity. In contrast, at shorter timescales, the coupling between ocean and convective aggregation also controls the strength of convective aggregation and overturn its effect. Thus the impact of convective aggregation may not be as strong as what can be inferred from experiments with uniform SSTs.These results emphasize the importance of considering ocean-atmosphere coupling when studying the role of aggregation in climate.

Development of a Si-Based Resonant-Cavity-Enhanced Infrared PhotodetectorGagnon, Adrian J. 04 1900 (has links)
<p>Resonant-cavity-enhanced (RCE) photodetectors have recently attracted attention due to their wavelength selectivity and high efficiency in comparison to conventional photodetectors. The goal of this ongoing research initiative is to develop a Si-based RCE infrared photodetector using inductively coupled plasma chemical vapor deposition (ICP-CVD) as the primary fabrication method. At the current stage of the project, wavelength-selective optical structures have been successfully fabricated using Si/SiO<sub>2</sub> layer pairs. These structures demonstrate sharp transmission peaks at their intended wavelength, making them potentially useful for efficient photodetection. The next phase of the photodetector development process involves using ion implantation to introduce dopants and create the bias.The project also explores the temperature sensing capability of the resonant-cavity structures. The temperature sensitivity tests indicate that the specific type of structure fabricated in this project may be relevant for fiber-optic temperature sensing applications. Additional testing is required to evaluate the performance characteristics of such structures as Fabry-Perot sensors capable of wavelength-encoded temperature measurement.</p> / Master of Applied Science (MASc)

Les sources d'ions de type RCE sont issues de la recherche sur la fusion et ont, en quelques sorte, révolutionnées le domaine des sources d'ions multichargés. Ces équipements ont permis à la fois d'augmenter les performances des accélérateurs actuels (énergies, type d'ions, intensités) permettant par la même l'exploration de nouveaux domaines de Physique (matériaux, agrégats etc...) comme d'ouvrir de nouvelles voies plus applicatives : hadronthérapie, traitement de surface, etc... Ce mémoire expose les développements réalisés au GANIL / CIMAP sur les développements techniques et théoriques de ce type d'équipement afin de proposer de nouvelles versions soit plus performantes soit plus versatiles dans leurs applications / utilisations. Dans ce mémoire trois exemples d'utilisation de sources RCE sont développés : utilisation des ions multichargés comme sonde pour l'étude Physique des agrégats de fullerènes ; production de faisceaux d'ions multichargés mais de taille micrométriques et la modification des propriétés physiques d'une surface de Ti-6Al-4V grâce à l'implantation d'ions multichargés multiénergies. Gageons que toutes les études et développements autour de ces sources verront voir l'apparition de nouvelles applications prometteuses aussi bien dans le domaine de la Physique fondamentale que dans celui d'applications sociétales.

O novo cenário da macroeconomia e os freqüentes alertas daqueles que estudam o meio ambiente trazem a luz outros fatores que devem ser respeitados num processo de tomada de decisão para qualquer investimento a ser realizado. Neste sentido vem se consolidando o Mercado de Crédito de Carbono no mundo, que se por um lado viabiliza a implantação de um empreendimento por admitir a compensação ambiental, por outro pode viabilizá-lo disponibilizando recursos para a sua instalação e operação. A geração de Crédito de Carbono, isto é, Redução Certificada de Emissões (RCE) e o aporte de recursos no sistema empreendimento em função da negociação dos créditos no Mercado de Carbono, pode se caracterizar em fonte de funding de empreendimentos de geração de energia elétrica no Brasil. Sendo assim, o objetivo desta pesquisa é a montagem de um protótipo de Pequena Central Hidrelétrica - PCH, tendo por base o project finance, enquanto estratégia de investimento e estrutura de negócio e adequado aos padrões do MDL no Brasil visando verificar a viabilidade de utilização das RCEs para compor o funding desses empreendimentos e, consequentemente, a validação de investimentos em geração de energia. Dessa forma, fornecerá as referências necessárias para a análise econômica e financeira do investimento em empreendimentos dessa tipologia. Após a análise dos indicadores da qualidade do protótipo é possível afirmar que com a criação do MDL e outros mecanismos promovidos por ele, os empreendimentos de geração de energia o tipo PCH tornaram-se atrativos para investidores típicos que buscam renda estável e de longo prazo, e o setor elétrico brasileiro sofreu mudanças em sua matriz, onde o crescimento de investimentos em fontes alternativas de energia é uma realidade. / The new scenery of macroeconomics and the alerts frequents of those that study the environment brought up other factories that must be respected in a decision process in any investment to be carried through. Therefore, the objective of this research is the assembly of a prototype of small hydroelectric - PCH, based on project finance, as investment strategy and business structure and appropriate standards of the CDM in Brazil to check the feasibility of using CERs to compose the funding of these projects and hence the validation of investment in power generation. Thus, it will provide the necessary references to the economic and financial analysis of investment in projects of this type. Etudes expérimentales de sources d'ions RCE à 2,45GHz pour la production de courants intensesColy, Arona 15 December 2010 (has links) (PDF)
Cette thèse s'inscrit dans le cadre d'une collaboration industrielle avec la société Pantechnik. Elle a consisté au développement d'un nouveau banc de test dédié à la caractérisation de sources d'ions RCE fonctionnant à 2,45 GHz en vue de la production de faisceaux intenses pour des applications industrielles. Deux sources RCE avec des structures magnétiques différentes sont testées sur la ligne avec le même système d'injection HF. Une nouvelle source avec aimants permanents, présentant une extraction dipolaire (SPEED) a été conçue et testée. Une étude de l'extraction des faisceaux d'ions dans le champ magnétique dipolaire est proposée. Les premières expériences ont montré une densité de courant ionique total de ~10 mA/cm2 à 20 kV et une puissance HF de 900 W. Les tests en plasma d'hydrogène ont montré que l'espèce dominante est la molécule H2+. Des propositions sont faites pour modifier la structure magnétique de la source d'ions afin améliorer ces performances et produire principalement des ions H+. La source d'ions MONO1000 a été testée à forte puissance HF avec un système de couplage par guide d'onde. De très intenses densités de courant d'ions hydrogène ont été mesurées (~95 mA/cm2) avec une extraction de type diode et une efficacité de transport de l'ordre de 70%. Les premiers tests utilisant un système d'extraction amélioré à cinq électrodes sont présentés.

Previous issue date: 2013-08-29 / Universidade Federal de Sao Carlos / Lotic systems are highly influenced by the landscape through which they flow, and changes in the landscape are the main threat to their ecological integrity. Evaluating the ecological "health" of lotic systems is essential and of great importance to the management of water resources worldwide. In this study, we evaluated the integrity of streams in southeastern Brazil using the protocol WHEBIP (Watershed Habitat Evaluation and Biotic Integrity Protocol), which combines information about the coverage of riparian vegetation, geomorphology, land cover and human activities to assess the physical characteristics of the sub-basin and to evaluate the integrity of streams. We compared this protocol with the protocol RCE (Riparian, Channel and Environmental), already widely used in tropical regions, to directly assess stream physical condition. As a measure of the functional integrity of streams, we estimated leaf breakdown rates, an important process in lotic systems. Furthermore, we sorted and identified the macroinvertebrates that colonized leaves in the 2nd and 28th days of the decomposition experiment and calculated environmental quality metrics related to these communities to test their relationship with WHEBIP. The protocols WHEBIP and RCE were significantly correlated to each other, with greater differentiation among streams especially when RCE was high. Possibly, the WHEBIP, by incorporating spatial variables on a large scale in the landscape, best differentiates these streams. None of the protocols tested were correlated with leaf breakdown rates. However, when calculating variation of decay rates (standard error), we observed a positive relationship with the protocol WHEBIP. Thus, we grouped the metrics of the protocol WHEBIP in a Principal Component Analysis (PCA), noting the trend to a significant correlation between changes in leaf breakdown rates and the first axis of the PCA, which indicates that the increase of forest cover in the microbasin could reduce variation in leaf breakdown rates in streams. The WHEBIP and RCE showed no correlation with the environmental quality metrics related to macroinvertebrate communities. But we tested the hypothesis that higher values of WHEBIP decreases the variation in leaf breakdown rates due to higher stability of macoinvertebrate communities, we found a significant correlation to withdraw one of the streams of the analysis. This stream differed greatly from the rest, because even getting down in value in WHEBIP, showed a differentiation between communities, due to the dominance of certain taxa in the days analyzed. This is the first study in the tropics that relates the variation in leaf breakdown rates in streams and forest cover of the microbasin within they are inserted. Our results suggest that WHEBIP protocol can be used to assess the integrity of streams in tropical regions. This protocol showed significant relationships with the variation in leaf breakdown rates, which may prove to be an interesting metric for use in monitoring, and the stability of the macroinvertebrate community, which is greater in less impacted streams, as suggested by other studies. / Os sistemas lóticos são extremamente influenciados pela paisagem através da qual fluem, e as transformações na paisagem são a principal ameaça à sua integridade ecológica. Avaliar a saúde ecológica dos sistemas lóticos é fundamental e de grande importância para o manejo dos recursos hídricos em todo o mundo. Neste estudo, avaliamos a integridade de córregos no sudeste brasileiro utilizando o protocolo WHEBIP (Watershed Habitat Evaluation and Biotic Integrity Protocol), que combina informações sobre a cobertura de vegetação ripária, geomorfologia, cobertura da terra e atividades humanas para avaliar as características físicas da sub-bacia e estimar a integridade de córregos. Comparamos esse protocolo com o protocolo RCE (Riparian, Channel and Environmental), já bastante utilizado em regiões tropicais, para avaliação física direta dos córregos. Como medida da integridade funcional dos córregos, estimamos a taxa de decomposição foliar, importante processo nos sistemas lóticos. Além disso, triamos e identificamos os macroinvertebrados que colonizaram as folhas referentes ao 2º e ao 28º dias do experimento de decomposição e calculamos métricas de qualidade ambiental relacionadas às comunidades desses organismos, para testar sua relação com o WHEBIP. Os protocolos WHEBIP e RCE foram significativamente correlacionados entre si, havendo maior diferenciação entre os córregos principalmente quando o RCE foi alto. Possivelmente o WHEBIP, por incorporar variáveis espaciais em uma escala maior na paisagem, diferencia melhor esses córregos. Nenhum dos protocolos testados apresentou correlação com as taxas de decomposição. No entanto, ao calcularmos a variação das taxas de decomposição (erro padrão), observamos uma relação positiva com o protocolo WHEBIP. Assim, agrupamos as métricas do protocolo WHEBIP em uma Análise de Componentes Principais (PCA), observando tendência a uma correlação significativa entre a variação das taxas de decomposição e o primeiro Eixo da PCA, o que indica que o aumento da cobertura florestal na microbacia poderia reduzir a variação nas taxas de decomposição foliar nos córregos. O WHEBIP e o RCE não apresentaram correlações com as métricas de qualidade ambiental relacionadas às comunidades de macroinvertebrados. Mas ao testarmos a hipótese de que os maiores valores do WHEBIP diminuiriam a variação das taxas de decomposição devido a maior estabilidade das comunidades de macroinvertebrados, encontramos uma correlação significativa ao retirarmos um dos córregos das análises. Esse córrego diferenciava-se bastante do restante, pois mesmo obtendo baixo valor no protocolo WHEBIP, apresentou uma baixa diferenciação entre as comunidades, devido à dominância de alguns táxons nos dias analisados. Este é o primeiro trabalho para os trópicos que relaciona a variação nas taxas de decomposição foliar nos córregos e a cobertura florestal das microbacias em que estão inseridos. Nossos resultados sugerem que o protocolo WHEBIP pode ser utilizado para avaliar a integridade de córregos em regiões tropicais. Esse protocolo apresentou relações significativas com a variação nas taxas de decomposição, que pode vir a ser uma métrica interessante para o uso em monitoramento, e com a estabilidade da comunidade de macroinvertebrados, que é maior em córregos menos impactados, conforme sugerido por outros estudos.

Previous issue date: 2017-11-13 / The Enterprise Cooperation Networks (ECN) were created to increase competitiveness
and protect Medium and Small Businesses (MSB) of large competitors. Many ECNs
find it difficult to remain active in the market. The life cycle model contributes to
overcome these problems, taking into account the characteristics of MSBs and broadens
the understanding of their changes over time. In this scenario, the present research aims
to identify, in this life cycle model, the stage that some selected networks in the State of
Goiás find, replicating the study of the same theme carried out in the networks of the
South of Brazil. This research aims to broaden the understanding of four different points
of view, involving ECNs, namely: theoretical, managerial, regional development and
public management point of view. It is a study of multiple cases, of descriptive
character, also involving, bibliographical survey. This model was applied in two ECNs
located in the State of Goiás, identifying its level of development. Subsequently, the
results obtained in the State of Goiás were compared with those obtained in the research
conducted in the South of the country. Semi-structured and structured interviews were
conducted with qualitative and quantitative data approaches, developed by content
analysis techniques. As a result, Rede 2000 was part of the Decline phase and the Real
Estate Network in the transition from the Development to Maturity / Consolidation
phase. Comparing the results with those of the South, the importance of external
influence on the ECNs was verified in both studies. It was noted that in the southern
survey there were thirteen ECNs available and only two in Goiás. In the South, ECNs
receive public support from programs specifically geared to them, and in Goiás this
does not happen, with programs focused only on cooperatives and Local Productive
Arrangements (LPA). Nowadays, in Goiás, members access social networks intensively
through computers and smartphones, and this has provided an informal exchange of
information among them, strengthening the link between them, helping to develop and
maintain network consolidation. / As Redes de Cooperação Empresarial (RCEs) foram criadas para aumentar a
competitividade e proteger as Médias e Pequenas Empresas (MPEs) dos concorrentes de
grande porte. Muitas RCEs encontram dificuldades para se manterem ativas no
mercado. O modelo de ciclo de vida contribui para superar estes problemas, tendo em
conta as características das MPEs e amplia a compreensão de suas mudanças ao longo
do tempo. Neste cenário, a presente pesquisa tem como objetivo identificar, neste
modelo de ciclo de vida, o estágio que se encontram algumas redes selecionadas no
Estado de Goiás, replicando o estudo do mesmo tema realizado nas redes do Sul do
Brasil. Esta pesquisa visa ampliar o entendimento sobre quatro pontos de vista
diferentes, envolvendo RCEs, a saber: ponto de vista teórico, gerencial,
desenvolvimento regional e gestão pública. É um estudo de múltiplos casos, de caráter
descritivo, envolvendo também, levantamento bibliográfico. Este modelo foi aplicado,
em duas RCEs localizadas no Estado de Goiás, identificando seu nível de
desenvolvimento. Posteriormente, foram analisados, comparativamente, os resultados
obtidos no Estado de Goiás com os obtidos na pesquisa realizada no Sul do País. Foram
realizadas entrevistas semiestruturadas e estruturadas, com abordagens qualitativa e
quantitativa dos dados, desenvolvida por técnicas de análise de conteúdo. Como
resultados, a Rede 2000 se enquadrou na fase de Declínio e a Rede de Imobiliárias na
transição da fase de Desenvolvimento para Maturidade/Consolidação. Comparando os
resultados com os do Sul, verificou-se, nos dois estudos, a importância da influência
externa nas RCEs. Notou-se que na pesquisa do Sul havia a disponibilidade de treze
RCEs e em Goiás apenas duas. No Sul as RCEs recebem apoio público de programas
voltados especificamente para elas e em Goiás isto não acontece, existindo programas
voltados apenas para cooperativas e Arranjos Produtivos Locais (APLs). Atualmente,
em Goiás, os associados acessam intensivamente redes sociais, por meio de
computadores e smartphones, e isso tem proporcionado troca de informações informais
com maior frequência entre os mesmos, fortalecendo o vínculo entre eles, auxiliando no
desenvolvimento e manutenção da consolidação da rede.
