Virulencia de mutantes exoenzimaticos de Metarhizium anisopliae var. anisopliae e seus revertentes a Rhodnius prolixus

Também procurou-se caracterizar os mutantes e revertentes exoenzimáticos, através de eletroforese para os padrões de a-esterase e aspectos citológicos como comprimento de conídios e número de núcleos por conídio. Por fim, determinou-se a virulência dos mutantes e revertentes à ninfas de 3º estadio de Rhodnius prolixus. Para isso, empregaram-se as linhagens E9 e V14 de K.anisopliae var. anisopliae.. A mutante amilase- e lipase-apresentaram índices enzimáticos iguais a um, o que não ocorreram com os mutantes protease-. Todos os revertentes apresentaram fenótipo exoenzimáticos semelhante aos tipos selvagens. Não houve alteração na germinação, comprimento de conídios e número de núcleos por conídio dos mutantes e revertentes. O padrão eletroforético de a ¿esterase mostrou-se diferente somente nos mutantes lipase-, entretanto não houve alteração para os revertentes correspondentes lipase+. A virulência dos mutantes amilase- e lipase- diminuiu significativamente, enquanto os mutantes protease-, não mostraram redução significativa. Os revertentes restabeleceram a virulência em níveis semelhantes aos tipos selvagens / Abstract: The present work was conducted in order to study by means of enzimatyc index the obtainment of mutants of Metarhizium anisopliae var. anisoplae strains E9 e V14 wich lost or had some reduction in their capability to produce extracellular enzymes namely amylases, lipases and proteases as well their capability to produce extracellular enzymes namely amylases, lipases and proteases as well their reversion induced by ultraviolet light. The mutants and revertants were characterized electrophoretically for a -esterase pattern and cytologically for conidia size as well as number of nuclei. The virulence of mutants and revertant strains were evaluated in bioassays against third instar nymphae of Rhodnius prolixus. The mutant strains for amylase and lipase showed phenotypically similar to their parental strains, and it was not observed variation in length, germination and number of nuclei per conidia for both mutants and their revertants. The electrophoretic pattern for a ¿esterases was similar for most of the studied strains except for the mutant showing deficiency for lipolytic activity. The virulence of amylolytic and lipolytic mutants were reduced when compared to their parental strain, while the proteolytic mutants did not show any significant reduction.
