Single CdSe/CdS dot-in-rods fluorescence properties / Propriétés de fluorescence de nanocristaux de CdSe/CdS coeur-bâtonnets uniques

Les nanocristaux colloïdaux synthétisés par voie chimique sont des sources prometteuses de lumière non-Classique à température ambiante. Ce travail est consacré à l'étude des propriétés optiques d'un type particulier de nanocristaux colloïdaux, appelé coeur-Bâtonnet, dans lequel un noyau de Seleniure de Cadmium (CdSe) sphérique est entouré d'une coquille de Sulfure de Cadmium (CdS) de forme cylindrique. En étudiant des particules de type coeur-Bâtonnet à température ambiante avec un microscope confocal, une caractérisation complète des propriétés optiques de ces émetteurs est réalisée. Nous étudions d'abord la statistique de clignotement de ces émetteurs. Nous montrons que les émetteurs coeur-Bâtonnet avec des coquilles épaisses se caractérisent par un clignotement réduit sur des échelles de temps courts, inférieurs à quelques millisecondes. Ensuite, une caractérisation détaillée de la statistique de photons des émetteurs coeur-Bâtonnet tenant compte du phénomène de clignotement est réalisée. La polarisation de l'émission est également étudié. Nous montrons que la polarisation d'émission peut être controlée en changeant la géométrie de la structure.Enfin, nous présentons également des expériences de couplage de ces émetteurs avec des dispositifs photoniques. Nous montrons la possibilité d'exciter un émetteur coeur-Bâtonnet en utilisant un nanofil d'oxyde de Zinc (ZnO). Nous montrons aussi que nous sommes en mesure d'orienter efficacement des nanoémetteurs uniques en utilisant la formation controlée de défauts dans des cristaux liquides. / Wet-Chemically synthesized colloidal nanocrystals are promising room temperature non-Classical light sources. This work is devoted to the study of the optical properties of a particular type of colloidal nanocrystals, called dot-In-Rods, in which a spherical Cadmium Selenide (CdSe) core is surrounded by a rod-Like Cadmium Sulfide (CdS) shell. By studying single dot-In-Rods at room-Temperature with a confocal microscope, a complete characterization of the optical, and especially quantum optical, properties of dot-In-Rods is provided for several geometrical parameters. We first study the blinking statistics of such emitters. We show that dot-In-Rods with thick shells are characterized by a reduced blinking that happens on fast timescales, typically on millisecond timescales. We then go on with a detailed characterization of the photon statistics of dot-In-Rods. A complete description of the photon statistics taking into account the blinking process is realized. The polarization of the emission is also investigated. We show that the emission polarization can be tuned by engineering the geometry. Finally, we also present experiments where we couple dot-In-Rods with various photonic devices. We demonstrate the possibility of excitation of a single emitter using a Zinc Oxyde (ZnO) nanowire. Using defects in liquid crystals, we also show that we are able to efficiently orientate single nanoemitters.

Estudos prévios do nosso laboratório têm mostrado a presença e importância das GTPases Rho, proteínas de baixo peso molecular que são ativadas por diversos sinais extracelulares, em neurônios da retina adulta, durante o desenvolvimento e na degeneração induzida por luz. O objetivo deste trabalho foi determinar por meio de imunocitoquímica a distribuição de alguns membros desta família (RhoA, Rac1 e Cdc42) nos fotorreceptores da retina de camundongos BalbC em situações normais e de adaptação ao claro e ao escuro. Os resultados mostraram que Rac1, Cdc42 e RhoA são expressos principalmente no segmento interno e nas possíveis terminações de cones e bastonetes na camada plexiforme externa. Em condições diferentes de estimulação não ocorreu translocação das GTPases, como ocorre com outras proteínas envolvidas na fototransdução. Os resultados sugerem que elas possam desempenhar papéis importantes na reorganização do citoesqueleto ou no processo de fototransdução, regulando outras proteínas e/ou protegendo contra danos induzidos pela luz. / Previous studies from our laboratory have shown the presence and importance of the Rho family of small GTPases, proteins that are activated by different extracellular signals, in neurons of the adult retina, during development and light-induce degeneration. The aim of the present work was to determine by immunocytochemistry the distribution of RhoA, Rac1 and Cdc42 in photoreceptors of the BalbC mice retina in different conditions of light/dark adaptation. Our results showed that Rac1, RhoA and Cdc42 are mainly expressed in the inner segments and presumptive axon terminals of cones and rods in the outer plexiform layer. In addition, there was no evidence of Rho GTPases translocation under different conditions of stimulation, as shown for some phototransduction proteins. The results suggest that Rho GTPases may play important roles in the reorganization of the cytoskeleton or in the phototransduction process, regulating other proteins and/ or protecting against light-induced damage.

An immunohistochemical and histological evaluation of the African buffalo (Syncerus caffer) retina

Vision studies and visual acuity investigations are an ongoing and progressive field in veterinary ophthalmology. These independent studies all help to contribute to a combined and collective knowledge in our understanding of this truly complex matter. Understanding retinal morphology and physiology is an integral factor in piecing together overall function of the eye. Many of these studies have been done in both medical and veterinary ophthalmology using behavioural factors, electrophysiology, special staining and scanning techniques on a histological level. In the veterinary field many species have been studied pointing out similarities or differences among them. This study hopes to contribute to the understanding of the retinal ultrastructure of the African buffalo (Syncerus caffer). Twenty-five pairs of African buffalo eyes were obtained, but only forty-eight eyes were included in this investigation. The globes of one donor appeared to have chronic intraocular disease and were phthisical. Since this is a descriptive study of normal anatomy and function, these eyes were excluded. Globe dimensions were recorded and statistically analysed, revealing an average horizontal diameter of 32.91mm and a vertical diameter of 33.04mm. The median age of the donor group was 4 years with Using scanning electron microscopy it was established that African buffalo retinas, like other domestic species, have a specialised region a few millimetres dorsolateral to the optic disc, synonymous to the well described area centralis. In this region a higher concentration of cones is found as opposed to other rod-rich regions. In a concurrent investigation, the contralateral globes were processed for immunohistochemical antibody staining. Colour specific anti-bodies were used to identify the cone population present in the African buffalo retina. Characterization of Selected Bacteria from the North Arm of the Great Salt Lake

Thirteen bacterial cultures were isolated from the North arm of Great Salt Lake during the months of January and February of 1973. Eleven isolates were gram-negative pleomorphic rods which lysed in hypotonic solution. The remaining two were gram-positive cocci. All isolates and one known strain of Halobacterium salinarium were subjected to examination of morphological, cultural, physiological, and biochemical characteristics. A numerical taxonomic analysis was applied to the compiled characters to compute a coefficient of similarity for each individual isolate as compared to all other isolates. A comparative analysis was included in the similarity computation using characters assembled from those reported in the literature for six taxonomically accepted species of halophilic bacteria. The lake isolates proved to be extreme halophiles with relative high levels of similarity between each other and the known bacteria included in the numerical analysis.

En particulier, les tiges élastiques telles que cables électriques, flexibles hydrauliques et pneumatiques, apparaissent dans de nombreux contextes industriels. Du fait d'une modélisation complexe et d'un grand nombre de degrés de liberté, l'extension des méthodes de planification de mouvement à de tels corps est un problème particulièrement difficile. En se basant sur les propriétés des configurations à l'équilibre statique, cette thèse propose plusieurs approches au problème de planification de mouvement pour des tiges élastiques. / The motion planning problem has been broadly studied in the case of articulated rigid body systems but so far few work have considered deformable bodies. In particular, elastic rods such as electric cables, hydraulic or pneumatic hoses, appear in many industrial contexts. Due to complex models and high number of degrees of freedom, the extension of motion planning methods to such bodies is a difficult problem. By taking advantage of the properties of static equilibrium configurations, this thesis presents several approaches to the motion planning problem for elastic rods.
