Reproduction and its seasonal variation in the soupfin shark, Galeorhinus galeus

Thesis (MSc)--Stellenbosch University, 2001. / ENGLISH ABSTRACT: The soupfin shark, Galeorhinus galeus, is a circum-global shark species of great economical importance. Their reproductive biology is reasonably well understood, but intraspecific differences between global populations necessitate the study o f populations separately. For this study, 70 male and 74 female specimens were collected along the southwestern coast of South Africa between 34°8'S; 18°27'E and 34°24S; 21°25'E. Morphological changes were described and serum steroid hormone concentrations evaluated by enzyme-linked immunosorbent assay (ELISA) over an almost-complete reproductive cycle. Males display an annual spermatogenic cycle that starts with a significant peak in testosterone concentrations coinciding with the start o f spermatogenesis around April-May. Testosterone levels decrease to reach a minimum in late-winter after which spermiogenesis commences. The prevalence of spermiogenic cysts in the testis increase to reach a peak between February and April, after which mating occurs until about July. Females (aplacentally viviparous) are suggested to have a triennial cycle, similar to the Brazilian & Australian populations, but different from the Californian & Mediterranean populations. During the first year after parturition there is not much reproductive development. The following year sees increased vitellogenic activity in ovarian follicles and development of the oviducal glands and uteri. Ovulation then occurs towards the end of that year. Mating is suggested to precede ovulation by two to three months necessitating sperm storage by the females in their oviducal glands. A 12-15 month gestation ensues and ends in parturition the following summer. Testosterone and estradiol levels fluctuate together and are involved in the preparation for ovulation and oviducal gland development. Progesterone levels show two distinct peaks during the year, in both immature and mature females, and could not yet be functionally linked to any reproductive activity or condition. Impact of seasonal variation and growing environment on blackberry aroma

Blackberries have been enjoyed for their delicious flavor and high nutritional value for hundreds of years. In North America, blackberries were cultivated since 1850-1860. Blackberries have their characteristic and unique aromas depending on the cultivar and growing conditions. The objective of this study was to evaluate the impact of seasonal variation and growing environment on blackberry aroma. Volatile compositions of 'Marion' and 'Thornless Evergreen' blackberries from three growing seasons were analyzed using gas chromatography-flame ionization detection (GC-FED) and GC-mass spectrometry (GC-MS). Seasonal variations were observed for some volatile compounds in these two blackberry cultivars. In 'Marion', the compounds were mainly acids, while in 'Thornless Evergreen', they were mainly acids, alcohols, and a few terpenoids. Although seasonal variations were present, it was generally observed that the most abundant volatiles in 'Marion' blackberry were acetic, 2/3-methylbutanoic, hexanoic and decanoic acids and linalool, whereas the most abundant volatiles in 'Thornless Evergreen' were 2-heptanol, hexanol, octanol, α-pinene, nopol and/p-cymen-8-ol. Compared with 'Marion', 'Thornless Evergreen' contained significantly more total volatiles, especially in alcohols, terpenoids and phenols, whereas 'Marion' contained more organic acids. Odor Activity Values (OAVs) were used to determine the most potent odorants in each cultivar. The compounds with high odor activity values (OAV > 10) in 'Marion' were ethyl hexanoate, β-ionone, linalool, 2-heptanone, 2-undecanone, α-ionone and hexanal. The compounds with high odor activity values (OAV > 10) in 'Thornless Evergreen' were ethyl hexanoate, 2-heptanone, ethyl 2-methylbutanoate, 2-heptanol, 3-methylbutanal, α-pinene, limonene, p-cymene, linalool, t-2-hexenal, myrtenol, hexanal, 2- methylbutanal and sabinene. To investigate the influence of growing environment on blackberry aroma, aroma extraction dilution analysis (AEDA) was employed to characterize the aroma profile of 'Chickasaw' blackberries from two growing regions in United States: Oregon and Arkansas. Eighty-seven odorants were detected in the two berries, in which seventy-seven could be identified. Comparative AEDA analysis showed that the berries grown in these two regions had similar aroma compositions, however, those odorants had various aroma impacts in each region. The compounds with high FD factors in Oregon's 'Chickasaw' were ethyl butanoate, linalool, methional, trans,cis-2,6-nonadienal, cis-l,5-octadien-3-one, and 2,5-dimethyl-4-hydroxy- 3(2H)-furanone, whereas in the 'Chickasaw' grown in Arkansas, they were ethyl butanoate, linalool, methional, ethyl 2-niethylbutanoate, β-damascenone and geraniol. The flavor formation in 'Chickasaw' blackberries was largely influenced by their growing environment. / Graduation date: 2005

Managing Seasonality in Tourism : Challenges and Opportunities for the Tourism Industry in Húsavík, Iceland

Seasonality is a common phenomenon in the tourism industry around the world. Húsavík, a town on the northern coast of Iceland, has experienced tremendous growth in tourism in recent years and is sometimes recognised as the „Whale Watching Capital of Europe‟. However, Húsavík faces extreme seasonality with high demand mainly during the summer months and limited or no demand at other times of the year. As is emphasised in the tourism literature and widely in practice, seasonality is frequently seen as a problem for the tourism industry which has to be tackled. Academic research has never before been done on seasonality in Húsavík and the aim of this thesis is to understand the nature of seasonality within the tourism industry in Húsavík and discover potential ways to reduce seasonality in Húsavík tourism. In order to fulfil the aim, qualitative semi-structured interviews were undertaken with six tourism managers in Húsavík to investigate their perceptions of seasonality and if and how it could be managed. The results confirmed that the peripheral location of the destination stimulates seasonality and it poses major economic challenges for tourism businesses in Húsavík. Managers would prefer a longer tourism season. Several recommendations were provided for how to reduce seasonality such as develop other attractions than whale watching, improve accessibility, focus marketing on seasons outside the summer, and offer winter packages for foreign and domestic tourists.

The effects of the natural vertebrate steroid 17β-oestradiol and the xeno-biotic vertebrate oestrogen receptor agonist bisphenol-A on reproduction in selected temperate freshwater gastropods : the potential for (neuro-) endocrine disruption

Evidence of feminising effects, including additional or enlarged female organs, have been reported in the (sub)-tropical freshwater prosobranch Marisa cornuarietis exposed to vertebrate estrogen receptor agonists. The primary symptom is an increase in the number of eggs laid, but this is only observed when exposure occurs at relatively low temperatures. This research project exposed temperate freshwater prosobranchs and a pulmonate to 17β-oestradiol (10-200 ngL-l, nominal) in an outdoor mesocosm subject to natural seasons to determine whether similar effects occur in European native temperate freshwater gastropods. Laboratory exposures to 17β-oestradiol (1-100 ngL-l, nominal) and Bisphenol-A (0.2-20 ngL-l, nominal) were also carried out over a range of different temperatures and photoperiods to simulate natural seasons. In the mesocosm exposures, significant increases in reproduction were measured in Viviparus viviparus, Bithynia tentaculata and, if the mortality rate was not significantly increased, Planorbarius corneus. It was observed that increases only occurred after the onset of autumn. In the laboratory, the oviposition rate in P. corneus was constant at 20oC with a 16h photoperiod, but declined significantly at 15oC with a 12h photoperiod, except at 100 ngL-l 17β-oestradiol, when the rate remained constant. There were no similar effects from Bisphenol-A exposure. Small increases in reproduction were observed in all the prosobranch exposures to both compounds, but the interpretation of the data was confounded by several factors (test chemical degradation, high mortality rates and parasitized organisms) and there were no significant differences. In conclusion, there were indications that all of the assessed species were capable of increased reproduction, and in P. corneus this appears to be a perpetuation of summer oviposition rates in autumnal conditions. The consequences of this effect in annual semelparous pulmonates may not be detrimental at the population level, but the long-term fitness of iteroparous prosobranchs that practice ‘restrained reproduction’ in early breeding seasons may be adversely affected.

An analysis of the meroplankton assemblages of Station L4 and the development and application of molecular techniques to aid taxonomic resolution

Zooplankton data from the 1988-2007 Station L4 time-series were used to determine inter-and intra-annual patterns of meroplankton community change at Station L4, Plymouth, UK. Abundances were calculated for five groups: Cirripedia, Decapoda, Polychaeta, Echinodermata and Bivalvia. Analyses showed that, while there is some annual variability, seasonal variation accounts for the major changes in the meroplanktonic community composition throughout the time-series. Cirripedia were the only group to show any significant change in abundance over the time-series. Further sampling at the study site produced data at a finer taxonomic resolution allowing for the analyses of the seasonal cycles of abundance of previously unresolved families and genera from the 1988-2007 time-series. A similar pattern was shown to that seen in the earlier time-series with no evidence of major changes in the meroplanktonic community being found. Comparison with historical data taken from Lebour (1947) showed little evidence of major variation in meroplanktonic species composition. The limitations of traditional methods of taxonomic resolution were highlighted during these analyses and led to the investigation of molecular techniques as a viable aid to identification. Bivalve larval samples were identified to species in many cases using PCR and sequencing reactions focussing on the 18S rRNA gene. Larvae of Phaxas pellucidus were shown to be the most common, and further analysis revealed the presence of two groups of species within the data over the time sampled, comprising the larvae of several hard-substrata species. Development of an RFLP technique focussing on the mtCOI gene, allowed for the successful discrimination of porcellanid larvae to species that were unable to be resolved morphologically. Long-term data-sets play an important role in revealing the long-term patterns of community composition and abundance of meroplanktonic larvae and should be continued to identify those patterns not evident over the time-period studied. Molecular techniques were valuable in aiding the taxonomic resolution of meroplanktonic larvae, allowing previously unknown patterns of species diversity and richness to be ascertained.

Some factors affecting the digestible energy requirements and dry matter intake of mature donkeys and a comparison with normal husbandry practices

The purpose of this study was to compile practical feeding guidelines for donkeys in the UK. Current guidelines are to feed 0.75 of horse feeding recommendations on a body weight basis. However, the superior digestive efficiency of donkeys, compared to horses, may render the use of horse recommendations inappropriate. The formulation of guidelines specific to donkeys would enable owners to calculate their donkey‟s requirements with greater accuracy and prevent overfeeding. A postal survey, used to gain information on the body condition score of donkeys in the UK, and the husbandry and feeding practices used to manage them, indicated that approximately 24% of donkeys in the UK are overweight. Feeding practices indicated that although owners were aware of their donkey‟s requirement for fibrous forages, the practice of feeding unnecessary concentrates, chaffs and high energy forages, in addition to grazing, was the likely cause of donkeys becoming overweight. The finding that the majority (85 – 90%) of donkeys were kept as non-working companion animals also reduced the need for owners to feed higher energy foods to their donkeys. Results also suggested that owners were unsure of how to adjust their donkey‟s diet to account for seasonal changes in requirements and pasture availability, as most owners‟ adjusted grazing access, and not the feeding of supplementary feeds. From a study of dry matter (DM) and digestible energy (DE) intakes by 20 mature donkeys maintaining weight during each UK season, the maintenance DE requirements of donkeys were calculated. Results showed no effect of sex on DM or DE intake. Season significantly (P<0.001) affected DM and DE intakes, implying increased requirements in winter compared to spring, summer and autumn. Dry matter intakes (DMI) increased from 51g/kg BW0.75 in spring, summer and autumn to 66g/kg BW0.75 in winter. Digestible energy requirements increased from 0.32MJ/kg BW0.75 in spring, summer and autumn to 0.43MJ/kg BW0.75 in winter. Comparison of results with horse recommendations showed considerably reduced requirements by donkeys. Horse recommendations overestimated DE requirements in summer and winter by 82 and 30%, respectively, making horse recommendations unsuitable for calculating donkey energy requirements. Husbandry practices commonly used by owners to manage their donkeys grazing access (grazing time, grazing area, strip grazing), were assessed for their effect on DMI by grazing donkeys in summer and autumn, using n-alkanes. The effect of grazing time was assessed by restricting donkeys to 8, 12 or 23 hours grazing per day. Season significantly affected food intake with donkeys in the 8 and 23 hour grazing groups eating more during summer when pasture availability was greater. Donkeys responded to the poorer quality summer pasture by grazing more intensively but less selectively, increasing the rate at which food was consumed. Grazing time was only influential over grass intake in summer, when pasture was more abundant. Restricting donkeys to 12 hours or less grazing per day significantly (P<0.001) reduced their grass intake compared to that of donkeys with 23 hours access. When grazing sparse pastures (autumn), grazing time did not influence grass intake, indicating an effect of herbage mass on grazing behaviour. Herbage mass was the most influential factor over diet composition (percentage of grass and straw consumed) in a second grazing study assessing the affect of strip grazing and set stocking systems on intake by grazing donkeys during summer and autumn. Herbage mass per donkey was higher in the set stocking system during both seasons, resulting in higher grass intakes. Determining if either grazing system was more effective at regulating grass intake was prevented due to differences in pasture availability between study sites. It is concluded that donkeys have lower DMI and maintenance DE requirements than horses, requiring donkey feeding guidelines to be formulated. Excess body weight in donkeys is caused in part, by the feeding of energy dense feeds in addition to low energy forages. Most owners place little nutritional importance on pasture, despite its potential to provide a large percentage of daily DM, DE and nutrient intake. Therefore nutritional guidelines must include advice on how to manage access to grazing, and how to feed donkeys with access to pasture. Restricting grazing time to 8 hours a day did reduce grass intake by donkeys, but was only effective when grazing abundant pastures. Providing ad libitum straw to grazing donkeys allows them to satisfy their DM and dietary fibre requirements without consuming excess energy.

Tropospheric carbon monoxide : satellite observations and their applications

Carbon monoxide (CO) is present in the troposphere as a product of fossil fuel combustion, biomass burning and the oxidation of volatile hydrocarbons. It is the principal sink of the hydroxyl radical (OH), thereby affecting the concentrations of greenhouse gases such as CH4 and O3. Consequently, CO has an atmospheric lifetime of 1-3 months, making it a good tracer for studying the long range transport of pollution. Satellite observations present a valuable tool to investigate tropospheric CO. The Atmospheric InfraRed Sounder (AIRS), onboard the Aqua satellite, is sensitive to tropospheric CO in ~50 of its 2378 channels. This sensitivity to CO, combined with the daily global coverage provided by AIRS, makes AIRS a potentially useful instrument for observing CO sources and transport. An optimal estimation retrieval scheme has been developed for AIRS, to provide CO profiles from near-surface altitudes to 150 hPa. Through a validation study, using CO profiles from in-situ aircraft measurements, this retrieval scheme has been shown to provide CO observations with strong correlations to in situ measurements. Compared to the operational AIRS v4 CO product this retrieval scheme is shown to provide total column CO retrievals with a reduced bias relative to the in situ measurements (~ -10% to ~ -1%). In addition, the optimal estimation retrieval is shown to provide improved estimation and characterization of the retrieval errors. Further validation work has been carried out through comparison with the established CO observations from the MOPITT instrument, onboard the Terra satellite. Good agreement (correlation coefficient > 0.9, and bias < 1.0 ppbv) between the instruments is observed in the mid-troposphere. At this level, the optimal estimation scheme is shown to remove a positive bias of ~10 ppbv, relative to MOPITT, that is present in the AIRS v4 CO product. The AIRS instrument is also shown to be less sensitive to CO in the lower troposphere than MOPITT. AIRS is also demonstrated to provide fewer pieces of independent information about the vertical structure of CO at tropical latitudes, where higher thermal contrast increases the sensitivity of MOPITT. Through time series analysis, the capability of AIRS to detect seasonal trends in CO is demonstrated. The potential of AIRS to be used to track, both horizontal and vertical, CO transport is explored. AIRS is shown to be capable of tracking horizontal transport, and to have potential to track vertical transport when combined with another satellite sensor.

Spatio-temporal modelling of climate-sensitive disease risk : towards an early warning system for dengue in Brazil

The transmission of many infectious diseases is affected by climate variations, particularly for diseases spread by arthropod vectors such as malaria and dengue. Previous epidemiological studies have demonstrated statistically significant associations between infectious disease incidence and climate variations. Such research has highlighted the potential for developing climate-based epidemic early warning systems. To establish how much variation in disease risk can be attributed to climatic conditions, non-climatic confounding factors should also be considered in the model parameterisation to avoid reporting misleading climate-disease associations. This issue is sometimes overlooked in climate related disease studies. Due to the lack of spatial resolution and/or the capability to predict future disease risk (e.g. several months ahead), some previous models are of limited value for public health decision making. This thesis proposes a framework to model spatio-temporal variation in disease risk using both climate and non-climate information. The framework is developed in the context of dengue fever in Brazil. Dengue is currently one of the most important emerging tropical diseases and dengue epidemics impact heavily on Brazilian public health services. A negative binomial generalised linear mixed model (GLMM) is adopted which makes allowances for unobserved confounding factors by including spatially structured and unstructured random effects. The model successfully accounts for the large amount of overdispersion found in disease counts. The parameters in this spatio-temporal Bayesian hierarchical model are estimated using Markov Chain Monte Carlo (MCMC). This allows posterior predictive distributions for disease risk to be derived for each spatial location and time period (month/season). Given decision and epidemic thresholds, probabilistic forecasts can be issued, which are useful for developing epidemic early warning systems. The potential to provide useful early warnings of future increased and geographically specific dengue risk is investigated. The predictive validity of the model is evaluated by fitting the GLMM to data from 2001-2007 and comparing probabilistic predictions to the most recent out-of-sample data in 2008-2009. For a probability decision threshold of 30% and the pre-defined epidemic threshold of 300 cases per 100,000 inhabitants, successful epidemic alerts would have been issued for 94% of the 54 microregions that experienced high dengue incidence rates in South East Brazil, during February - April 2008.
