A taxonomic study of Senecio speciosus, Senecio macrocephalus and possible hybrid populations using morphological data, toxicty tests and chromatography

The variation between populations of Senecio speciosus Willd., Senecio macrocephalus DC. and intermediate plants was investigated in a comparative study of morphological characters, toxicity of plant extracts to brine shrimps (Artemia salina) and chromatography of plant extracts. Specimens were collected at 18 localities in the Eastern Cape Province. All these specimens were examined morphologically and chemical extracts were tested for toxicity and by comparative chromatography. The collection of Senecio speciosus and Senecio macrocephalus specimens in the Selmar Schonland Herbarium (GRA) was also examined morphologically. Six geographical areas were represented in the combined collections. Analysis of morphological data separated typical Senecio speciosus and Senecio macrocephalus at either end of hybrid index histograms and principal components analysis diagrams. The intermediate populations displayed morphological characters of both Senecio speciosus and Senecio macrocephalus. Some specimens were intermediate between these two species, falling within the range of variation of these species while others fell outside this range. The Brine Shrimp Assay was used to test for toxicity and to investigate the possibility of using toxicity data as a genetic marker in taxonomic studies. As Senecio speciosus extracts were less than 1% toxic and Senecio macrocephalus extracts were at least 95% toxic to the brine shrimps it is suggested that in this case toxicity can be used as a genetic marker. Toxicity can even be described as a good taxonomic character as discontinuity is very sudden and complete. The intermediate plants in the Grahamstown area were at least 92% toxic to the brine shrimps linking them to Senecio macrocephalus. Thin layer and paper chromatography were used as comparative techniques to study the chemical profiles of the specimens. Alkaloids, terpenoids and flavonoids were studied. Thin layer chromatography to separate the alkaloid components of the plant extracts showed Senecio speciosus and Senecio macrocephalus to have distinct chemical profiles suggesting that they are separate species. The intermediate plants- were found to contain chemical compounds matching either or both Senecio speciosus and Senecio macrocephalus suggesting that they may have arisen by hybridisation. In a preliminary investigation Senecio speciosus extracts showed a complete lack of terpenoid compounds whereas extracts from Senecio macrocephalus and the intermediate specimens tested gave a terpenoid colour reaction in the basal spot only. This links the intermediate populations with Senecio macrocephalus. Paper chromatography to separate the flavonoid constituents of the plant extracts also showed typical Senecio speciosus and Senecio macrocephalus to be distinct. The intermediate populations contained flavonoid compounds from one or both of these species. The populations in the Grahamstown area show morphological features close to and in some cases indistinguishable from Senecio speciosus. Chemically these specimens show some similarities with Senecio macrocephalus. In the East London area specimens show a similar mixture of characters but appear morphologically to be closer to Senecio macrocephalus. However, in the Amatole Mountains, despite both species being present in the same locality it appears that no hybridisation has occurred. It is therefore suggested that at some of the localities where the geographical ranges of Senecio speciosus and Senecio macrocephalus overlap in the Eastern Cape Province hybridisation between these two species occurs.

Radiation pasteurization of raw and chlortetracycline-treated shrimp

Pasteurization radiation offers a new means of extending the refrigerated storage life of foods. This procedure avoids many of the undesirable changes which result when sterilization radiation is used and still eliminates most of the spoilage organisms. Shrimp was treated with 5 ppm chlortetracycline and subjected to pasteurization levels of gamma radiation and stored at 38°F. The levels of irradiation used were 0.5 and 0.75 megarad. These levels were based on the flavor threshold of irradiation intensity. The quality of the stored irradiated shrimp was determined by subjective evaluation, chemical analyses and microbiological examinations. The storage life of the shrimp irradiated at 0.5 megarad was extended to 5 weeks, compared with 1 week for the unirradiated samples held at the same temperature. Throughout the ten week storage period, the samples which received doses of 0.5 megarad and CTC and those which received 0.75 megarad with and without CTC remained in good condition. / Graduation date: 1963

A sustentabilidade neste trabalho é baseada principalmente em duas dimensões: ambiental, e econômica. O presente trabalho teve como objetivo avaliar a sustentabilidade ambiental e a viabilidade econômica em pequena escala de produção de pós-larvas do camarão de água doce da espécie Macrobrachium amazonicum, por meio de um conjunto indicadores de sustentabilidade. Foram considerados os ciclos de produção durante um ano de cultivo de pós-larvas do camarão-da-amazônia. A capacidade produtiva considerada foi de 290 milheiros de pós-larvas/mês. A sustentabilidade ambiental foi medida com base na utilização de recursos naturais, eficiência no uso destes recursos e poluição liberada ao ambiente pelo meio de cultivo. A sustentabilidade econômica foi medida com base nos aspectos sobre a eficiência no uso dos recursos financeiros e a capacidade de gerar recursos para reinvestimentos na atividade. Os resultados sugerem que há viabilidade econômica para o produtor que realiza larvicultura em sistemas de fundo de quintal (backyard hatchery). Além disso, os dados obtidos também mostraram que embora a larvicultura seja uma atividade bastante intensificada, há eficiência na utilização dos recursos naturais, sendo mais sustentável do que outras fases de produção, como o berçário e crescimento final da mesma espécie. In this work, sustainability is based on environmental and economic dimensions. The objective of the study is to evaluate the environmental and economic sustainability of a small scale Macrobrachium amazonicum post-larvae production using a set of indicators for sustainability. Production cycles during one year of the Amazon Prawn hatchery was considered. The considered productive capacity was 290 thousand post larvae/month. Environmental sustainability measurements were based on the use of natural resources, efficiency in the use of resources and pollution released to environment from the production facility. Economic sustainability measurements were based on the aspects of efficiency in the use of financial resources and the capacity to generate resources for reinvestments in the activity. Results show that there is economic feasibility for the 'backyard' hatcheries system for freshwater prawn farmers. In addition, data indicate that in spite of hatchery being a highly intensified activity, there is efficiency in using natural resources, and it is more sustainable than other production phases, such as nursery and grow-out phase of the same species. THE USE OF CHEMICAL CUES BY SARGASSUM SHRIMPS LEANDER TENUICORNIS AND LATREUTES FUCORUM IN ESTABLISHING AND MAINTAINING A SYMBIOSIS WITH SARGASSUM ALGAE

A mutualistic symbiosis exists between the alga Sargassum spp. and two shrimp species, L. tenuicornis and L. fucorum. But little is known about how the shrimp locate their host alga. Both visual and chemical cues are potentially available. Visual cues would be presumably restricted at night but chemical cues are potentially available continuously. Additionally, a previous study has looked at both cue variables with results that are mixed. This current research elaborates on the previous study in an attempt to fully understand Sargassum shrimp chemoreception. A y-maze and four-chambered apparatus were used to test if the shrimp were able to detect Sargassum cues, conspecific cues, and Dimethylsulfoniopropionate (DMSP) cues. Neither shrimp species showed a strong directional response to any of the chemical cues, but the Sargassum and DMSP cues did cause more shrimp to exhibit searching behavior. Additionally, several differences in response between male and female shrimp were found for each cue. A weaker dilution of DMSP was tested in an attempt to determine sensitivity of L. fucorum shrimp to the chemical cue. This weaker dilution also caused L. fucorum to exhibit searching behavior, but the sensitivity to the cue was not found and further research is needed to fully answer this question.
