Darüber hinaus wurden neue siliciumhaltige Synthesebausteine für weitere Wirkstoffsynthesen synthetisiert. / Silicon-containing analogs of well-known drugs and derivatives of several odorants were prepared. Siliciumhaltige Analoga von Loperamid und Penfluridol wurden hergestellt, Versuche zu einer alternativen Synthese von Sila-Haloperidol wurden unternommen. Siliciumhaltige Sigma-Liganden des 1,4´-Silaspiro[tetralin-1,4´-piperidin]-Typs wurden hergestellt und kristallographisch sowie pharmakologisch mit ihren Kohlenstoff-Analoga verglichen. / Silicon-containing analogs of loperamide and penfluridol were synthesized, an alternative synthesis of sila-haloperidol was tried. Silicon-containing sigma ligands of the 1,4´-silaspiro[tetralin-1,4´-piperidinium] type were synthesized and their x-ray structures as well as their pharmacological properties were compared with their respective carbon analogs.

Im Rahmen der vorliegenden Arbeit wurden siliciumhaltige Derivate des allosteren Modulators W84 und Sila-Analoga der Wirkstoffe Meptazinol und Trifluperidol synthetisiert. Weiterhin wurden potentielle holzige Riechstoffe dargestellt. Zudem wurde im Zuge der Synthese des Sila-Analogons von Trifluperidol ein Beitrag zur Etablierung der 2,4,6-Trimethoxyphenyl-Einheit als Schutzgruppe für das Silicium-Atom geleistet. Ebenso wurde die Anwendbarkeit der Olefin-Metathese-Reaktion und der Gold(I)-katalysierten intramolekularen Hydroaminierung als präparative Methoden zum Aufbau von 3-Silazepan- bzw. 3-Silapyrrolidin-Derivaten untersucht. / In this work, silicon-containing derivatives of the allosteric modulator W84 and sila-analogues of the drugs meptazinol and trifluperidol were synthesized. In addition, compounds with potentially woody scent were prepared. In the course of the synthesis of the silicon analogue of trifluperidol, a contribution was made to the establishment of the 2,4,6-trimethoxyphenyl moiety as protecting group for the silicon atom. Furthermore, the utilization of the olefin-metathesis reaction and the gold(I)-catalyzed hydroamination reaction as preparative tools for the synthesis of 3-silazepane or 3-silapyrrolidine derivatives was investigated.

Die vorliegende Arbeit beschreibt die Synthese von siliciumhaltigen Derivaten des Retinoid-Agonisten Bexarotene und berichtet in diesem Zusammenhang über die Darstellung von neuen siliciumhaltigen Synthesebausteinen. Außerdem wurden siliciumhaltige Derivate der bekannten Riechstoffe Phantolide, Okoumal, Galaxolide und (3E,5E)-5-tert-Butyl-7,7-dimethylocta-3,5-dien-2-on synthetisiert und hinsichtlich ihrer olfaktorischen Eigenschaften untersucht. Die Charakterisierung der Zielverbindungen sowie aller auftretenden Zwischen¬stufen erfolgte durch NMR-Spektroskopie (1H, 11B, 13C, 29Si) und Elementaranalyse. Zusätzlich konnte in einigen Fällen eine strukturelle Charakterisierung durch Einkristall-Röntgenstrukturanalyse realisiert werden. / This doctoral thesis describes the syntheses of silicon-containing derivatives of the retinoid agonist bexarotene and in this context reports on the preparation of new silicon-containing building blocks. Furthermore, silicon-containing derivatives of the known odorants phantolide, okoumal, galaxolide, and (3E,5E)-5-tert-butyl-7,7-dimethylocta-3,5-dien-2-one were synthesized and studied for their olfactory properties. The characterization of all target compounds and their precursors was performed by NMR spectroscopy (1H, 11B, 13C, 29Si) and elemental analyses. In some cases, a structural characterization by single-crystal X-ray diffraction could be realized.

Siliciumhaltige Analoga der Histaminantagonisten Terfenadin und Fexofenadin sowie diverser Riechstoffe (Majantol, Linalool) wurden dargestellt. Desweiteren wurden siliciumhaltige alpha-Aminosäuren synthetisiert und auf einer Kieselgeloberfläche immobilisiert. / Silicon-containing analogs of the histaminic antagonists terfenadine and fexofenadine and of several fragrances (majantol, linalool) were prepared. Silicon-containing alpha-amino acids were synthesized and immobilized on a silica surface.

Die vorliegende Arbeit beschreibt die Synthese siliciumhaltiger Analoga (C/Si-Austausch) der Wirkstoffe Panamesine, Niceritrol und Hepronicate sowie die Darstellung dieser Wirkstoffe selbst. Darüber hinaus wurden siliciumhaltige Derivate eines selektiven N-Typ-Calciumkanal-Antagonisten sowie siliciumhaltige Synthesebausteine vom allgemeinen Formeltyp Si(CH2X)4 dargestellt (X = SAc, SH, OAc, OH, Br, I, N3). Die Charakterisierung der Zielverbindungen sowie aller auftretenden Zwischenstufen erfolgte durch NMR-Spektroskopie (1H, 13C, 15N, 19F, 29Si) und elementaranalytische Untersuchungen. / This work describes the syntheses of panamesine, niceritrol, hepronicate and their sila-analogues (C/Si exchange). Silicon-containing derivatives of a selective N-type calcium channel antagonist and tetrafunctional silanes of the formula type Si(CH2X)4 (X = SAc, SH, OAc, OH, Br, I, N3) were synthesized as well. All these compounds and all the intermediates were characterized by NMR studies (1H, 13C, 15N, 19F, 29Si) and elemental analyses.
