Studium vlivu požárů skládek komunálních a průmyslových odpadů na kontaminaci životního prostředí, produkty hoření pryže a halogenovaných polymerů / Study of the influence of fires of municipal and industrial waste landfills on the enviromental contamination, the combustion products of rubber and halogenated polymers

This thesis deals with the issue of fires of municipal and industrial wastes, particularly of rubber and halogenated polymers. When these materials burn, significant amount of harmful substances liberate into environmental compartments. Analysis of samples subjected to thermal decomposition in laboratory conditions explored formation of characteristic compounds in dependence on conditions of combustion. Samples of air, water and soil taken from real fires that occurred between the years 2007 – 2011 in the region of Western Bohemia monitored spreading of these substances into the environment. Majority of these samples were analysed by the means of gas chromatography with mass spectrometric detection. Additionally the effect of extinguishing methods and use of fire extinguishers on formation and spreading of combustion products was studied, especially in soil and in water sources.

Diploma thesis is focused on determination of macrolide antibiotics in wastewater. Environmental contamination with drugs currently represents a major problem. To determine the level of contamination is necessary to develop the appropriate analytical techniques. From the group of macrolide antibiotics were selected four representatives: erythromycin, clarithromycin, azithromycin and roxithromycin due to their frequent use in Czech Republic. For their isolation from wastewater was selected solid phase extraction and for analysis high performance liquid chromatography with mass spectrometric detection was chosen. Under optimized conditions, the ten days analysis of wastewater from the WWTP Brno-Modřice, two days analysis of wastewater from the WWTP Mikulov and one day analysis of wastewater from WWTP of University of Veterinary and Pharmaceutical Sciences Brno was performed.

Charakterizace chemického složení dehtu po zplyňování biomasy / Characterization of the chemical composition of the tar produced in the gasification of biomass

Biomass is a renewable source of energy and represents an acceptable alternative to fosil fuels. Usable methods of energetic use of biomass are combustion and gasification. Main interest is focused on gasification in last years. Gasification is based on conversion of organic material to usable gaseous product called syngas, which can be used as a fuel for energy production. Large amount of tar is formed as a by-product of incineration and gasification of biomass. Tar is composed by various mixture of organic substances, has a negative effect on operating conditions and is a potential threat to environment. Experimental part of this study was focused on analysis of tar obtained by gasification of various kind of biomass. Selected groups of hydrocarbons and oxygen compounds were identified in analysed samples. Individual biomass samples were compared regarding to determined relative content of selected substances. Analysis was done by complete twodimensional gas chromatography with mass spectrometric detection (GCxGC/TOF-MS).

BioSealing is an innovative method of soil body reconstruction, which uses stimulation of soil bacterial activity to seal the leakage. The method was developed in the Netherlands and it was used in conditions of the Czech Republic in years 2015 and 2016 on the Hornice water work. This earth dam was monitored by electrical impedance spectrometry by apparatus with device Z-meter IV in this period and the data are processed and assessed in this diploma thesis.

This diploma thesis deals with preparation of combined drinks. These drinks consist of white wine and fruit concentrate base. This thesis was divided to 2 parts – senzory and chemical. As a fruit component was chosen aronia concentrate. The first sensory analysis was concentrated on a choice of a suitable white wine for this procedure. Second sensory analysis was focused on aditives. We added citric acid and ascorbic acid to the drinks. Evaluators had to choose the most tasty drinks. Next sensory analysis was dedicated to choice of the right drink composition. The last analysis was focused on choice of right fruit composition. Evaluators had to compare between the drink that was prepared from aronia concentrate or a drink prepared from aronia concentrate and other species of fruits. Their duty was to decide which one is more tasty. Chemical part was dealing with puting individual components of drink preparation to the test. Combined drinks had to undergo chemical tests too.

Surveillance of c-allocation in microalgal cells

When microalgae are exposed to changing environmental conditions, e.g., light-dark cycles or oscillations in nutrient availability (CO2, nitrogen, phosphate or silicate) they respond with metabolic changes in the carbon allocation pattern. Short time regulations in the time range of few seconds to minutes can be mirrored best by mass spectroscopy based metabolomics. However, these snap shots do not reflect the alterations in the carbon flow to the cellular macromolecules like protein, carbohydrate or lipid. In this review it is shown how the combination of FTIR spectroscopy and Chla-in-vivo-fluorescence based electron transport rates can reveal changes in the metabolic flux rates of carbon during a shift of the environmental conditions. The review will demonstrate in which time range FTIR spectroscopy can deliver significant information and how FTIR spectroscopy data can synergistically support metabolome analysis by mass-spectroscopy.

Studium komplexů tebukonazolu s ionty kovů / Study of tebuconazole complexes with metal ionts

Tebuconazole belongs to widely utilized plant fungicides, mostly in vineyards. Its traces are present in plant tissues, wine as well as in soil. The aim of the study was to determine stoichiometries and stabilities of metal (Cu, Zn, Cd, Ca and Ag) complexes with tebuconazole (1) by means of electrospray ionization mass spectrommetry (ESI-MS). It was found that the complexes of tebuconazole with copper ions are particularly stable ([Cu(1)]+ has appearance energy 224 kJ·mol-1 , corresponding with the dissociation of the water molecule from the tebuconazole molecule), while the stability of another tebuconazole complexes is lower, about 150 kJ·mol-1 . In real environmental samples, the detection of 1 itself is hampered by its spreading into various species. The method involved here is based upon a hyphenation of electrochemistry (amperometry) and electrospray-ionization mass spectrometry (ESI-MS). It uses the fact that Cu creates very stable complexes with 1. We demonstrate that at appropriate potentials (at higher than 500 mV) Cu cations can be generated in an electrochemical cell and subsequently form complexes with 1 present in the sample. Příprava rekombinantních forem extracelulární domény myších leukocytárních receptorů z rodiny NKR-P1. / Preparation of recombinant forms of the extracellular part of mouse leukocyte receptors from NKR-P1 family.

Mouse NK cell receptors belonging to NKR-P1 family plays role in activation, inhibition and cytokine secretion by these cells. Aim of this thesis is preparation of extracellular parts of C57BL/6 mouse strain activating receptors mNKR-P1A and mNKR-P1C. Production vectors with coding sequences of both proteins were prepared. Next, optimization of production in E. coli was done and appropriate in vitro refolding and purification protocol were developed. Purified proteins were characterized by mass spectrometry and labeled by a fluorescent dye. Primary screening for potential ligand was performed. Further work will involve structural characterization of the receptors and identification of their ligands. These data may help to clarify the function of NK cells.
