Movement Patterns and Catch-and-Release Impacts of Striped Bass in a Tidal Coastal Embayment in MassachusettsTyrrell, Heather M 01 January 2014 (has links) (PDF)
An investigation into the spatial ecology and effects of catch-and-release angling on the physiology and behavior of striped bass was conducted. Fine-scale behavior was assessed by tagging fish with acoustic transmitters equipped with pressure and tri-axial accelerometer sensors and tracking them within a fixed array (n=34 receivers) in a Massachusetts estuary. Activity space changed significantly over the course of the season and increased with water temperature. Striped bass most frequently exhibited low levels of locomotory activity representing 67% of total activity measurements, with occasional high activity and burst swimming, often within the upper 3 m of the water column. Depth distribution of striped bass remained shallower when temperatures peaked at over 21 oC. Diel vertical migration was present with shallowest depths observed during the day and greatest depths during high tide. To investigate catch-and-release consequences, 102 striped bass were angled and blood sampled between July and November 2011. A subsample of 35 striped bass (July n=11, August n=11, September n=13) were implanted with tri-axial acoustic accelerometers to assess relative behavior and survival post-release. Results from principle component analyses produced five factors describing 72.7% of the variance for blood physiology parameters, total length, and water temperature. Subsequently, only eigenvalues from PC1, with high loading for blood lactate, plasma sodium and chloride, and total length, were significantly correlated with fight time. Eight individual fish were detected within 12 hours of release and exhibited their greatest mean daily activity space estimate within that time (1.5 km2 ± 0.6, 50%; 5.6 km2 ± 2.2, 95%). Depth ranged from 0-6.15 m (1.89±1.3 m) and acceleration ranged from 0.095-3.51 ms-2 (0.95±0.33). In summary, no observed mortality suggests that fish were able to recover from the physical and physiological impacts of angling. This thesis has increased the understanding of striped bass ecology and will help promote future conservation and management initiatives for striped bass and facilitate additional research.

The uptake, distribution and depuration of a water soluble, mono-aromatic hydrocarbon contained in petroleum and refined products was studied in two species of marine fish, Nature northern anchovy (Engraulis mordax) and juvenile striped bass (Horone saxatilis) were exposed to sub-lethal concentrations of 14c benzene for 48 hours. Residues in tissues exhibiting a high lipid content (non-metabolic pathway) or representing apparent major metabolic sites were measured during the exposure and afterwards when the fish were transferred to clean seawater. Fish exhibited a rapid uptake over a wide range of benzene concentrations in the water column. Accumulation in anchovy was considerably greater than in striped bass. Results indicate that the pathway of hydrocarbons through the liver, gall bladder, intestines and colon is a major depuration route. Residues were depurated rapidly after cessation of exposure; in striped bass tissues. most residues were undetectable by seven days,

Utilization of DNA vaccines in aquaculture has been gaining interest and recent efforts have been focused on methods of delivering DNA vaccines to fish. In the present study, a methodology was sought that could protect DNA vaccines such that they could be orally administered. The main objective of the study was to determine if a DNA vaccine could be effectively compounded into an orally administrable formulation with chitosan or polylactide-co-glycolide (PLG). The immune response of hybrid striped bass (Morone saxatilis x Morone chrysops) following oral delivery of a DNA vaccine containing Mycobacterium marinum Ag85A plasmid in either chitosan or PLG nanoparticle encapsulation was evaluated. Hybrid striped bass were divided into four experimental groups: IM immunization of the DNA vaccine as a positive control, oral delivery of uncomplexed DNA vaccine, oral delivery of chitosan or PLG alone as a negative control, and oral delivery of complexed chitosan or complexed PLG DNA vaccine. Fish were bled at regular intervals and an ELISA was used to evaluate antibody levels in individual fish. While the chitosan /plasmid DNA complex containing the Mycobacterium marinum Ag85A gene failed to produce a significant antibody response, the PLG/plasmid DNA matrix stimulated humoral immune response in the fish. / Master of Science

The effects of ozone treatment on chemical parameters of a recirculating aquaculture system producing hybrid striped bassHerbst, Jennifer Lynn 17 January 2009 (has links)
The hypothesis tested in this study is that the application of ozone to reuse aquaculture water provides more favorable conditions for hybrid striped bass at production levels. Evaluation of water quality in fish culture systems being treated with ozone was made during a production trial (1200 fish/14,OOO 1). Originally, a control and four ozonated systems with doses of 3, 13, 25, and 45 g O3/kg feed/day supported fish for experimentation but, due to mechanical failure, only the control, the 13 and 25 g O3/kg feed/day treatments completed the full 35 week study. On an equal feeding basis, concentrations of dissolved organic carbon (DOC), carbonaceous biochemical oxygen demand (CBODs), and chemical oxygen demand (COD) were lowest in the most heavily ozonated systems. Ozone increased the biodegradation of DOC. Total suspended solids, particle size and distributions were impacted by ozone's flocculating properties. The percentages of particles in the smallest size range, 5-20 microns, were greater in the control system than the ozonated systems during weeks of equal feeding rates. When feeding rates differed, the control system maintained a larger number total particles per kilogram of feed administered. The pH of the ozonated systems was lower than the control which favored the ionized (less toxic) form of ammonia. Enhanced biodegradation of organics in the ozonated systems may have increased carbon dioxide production and caused a decline in pH. Greater nitrification in the ozonated systems may have also decreased the pH. Higher total ammonia nitrogen (TAN) levels were produced in the control on an equal feeding basis. Average weekly TAN t unionized ammonia (NH3), nitrite and nitrate concentrations remained at acceptable levels across treatments and throughout the production trial. Overall, the major indication of improved system conditions due to ozone treatment was that fish raised in the ozonated systems fed steadily throughout the growth trial while the control system's culture experienced periods of reduced feeding. / Master of Science

Cobia (Rachycentron canadum) is a tropical marine fish, with increasing commercial aquaculture importance worldwide. One of the major limitations to intensive aquaculture is disease. Diseases spread rapidly in an aquatic environment and pose a major threat to the development and introduction of new species, such as cobia, in aquaculture. This is due to the necessity to use wild caught broodstock, which pose a greater threat to introducing disease to a facility. Bacteria of the genus Vibrio play a major role in the diseases of cultured cobia and other species of marine fish. The goal of this study is to reduce the incidence of disease in a population, by either eliminating the potential pathogen or increasing the resistance of the host. To reach that goal, a disinfection assay to evaluate the effectiveness of nine common aquaculture chemical disinfecting compounds was evaluated against two bacterial pathogens (Vibrio anguillarum and V. ordalii). Both bacterial species were susceptible to a variety of common disinfecting compounds including Chloramine-T®, chlorine, ethanol, iodine, Lysol®, Roccal®-D Plus, and Virkon-S®. In addition, both species showed a resistance to disinfection with formalin and tap water. The humoral immune response of cobia to vaccination with a commercially-available vaccine for Vibrio spp. was evaluated by an ELISA. There was a significant difference between control and vaccinated groups (P<0.0001), showing significant antibody production resulting from vaccination. / Master of Science

Hypervariable DNA markers and population structure in three fish speciesLaughlin, Thomas Fain 06 June 2008 (has links)
The utility of hypervariable DNA polymorphisms as a general population genetics method was studied in three fish species by the use of multilocus DNA fingerprinting. Laboratory lines and field caught specimens from Belize and Florida of the clonal species <i>Rivulus marmoratus</i> were examined to determine the relative contributions of mutation and migration to genetic variation in the species. Specimens of <i>Poecilia latipinna</i>, the sailfin molly, from Florida and Georgia were used to explore the properties of hypervariable markers in the context of an outbred and abundant species that exhibits typical levels of genetic variability at nuclear loci. The results were compared to those of a previous allozyme survey of the same populations. Samples of <i>Morone saxatilis</i> from the Chesapeake Bay system were used to investigate the utility of hypervariable markers in the description of genetic variation of an outbred species depauperate in other measures of genetic variation.
The results of this study indicate that variation observed among among <i>R. marmoratus</i> clones characterized by hypervariable loci may be the result of natural selection; based on the analyses of mutation rates and population structure. Results from the work with <i>P.latipinna</i> showed that hypervariable loci could have general utility as a method for studying population structure. This utility was demonstrated in the examination of Chesapeake Bay populations of <i>M. saxatilis</i>. Large degrees of interindividual variation at hypervariable loci permitted the characterization of population structure within Chesapeake Bay populations of this species. / Ph. D.
