Meiosis and pollen stainability in Prunus avium L. cv. Lambert

Variation in pollen viability and the meiotic behaviour of sweet cherry Prunus avium L. cv. Lambert were studied. Pollen viability was estimated using aceto-carmine. Three types of pollen grain were observed; large, triangular; small, aborted; and intermediate. Only the first type were considered to have been viable at time of blossom harvest. No significant differences were detected between trees, between branches, or within branches, except for a North and South branch of one tree. Highly significant differences were detected among blossoms in a bud. The relative frequency of abnormal pollen was 42.26 per cent. Premeiotic budwood was stored at -5°C. Subsequent forcing at 10°, 15°, 20°, and 25°C to bring the buds to meiosis, and staining in alcoholic hydrochloric acid-carmine, revealed meiotic abnormalities at all temperatures except 15°C. Early meiotic division stages of additional material forced at this temperature were synchronous, but asynchrony developed after first metaphase. The first meiotic division was characterized by the frequent presence of a single pair of univalents, which appeared to arise from desynapsis of a bivalent in diplotene. Meiotic abnormalities associated with the univalents were observed at later division stages. The metaphase-anaphase division was typified by sequential disjunction of the bivalents, characteristic for sweet cherry meiosis. The sweet cherry crop has always been the number one fruit crop in Utah until the last three or four years when it shifted to second place due to severe spring frosts. However, the outlook for its continued success is still very good. Even though there are drawbacks such as virus infestations, spring frosts, nutritional disorders, cracking, doubling, and bird injury, new plantings are being made each year in each of the important fruit producing counties. Utah is also important in the national production of sweet cherries and is presently sixth in the nation.

Manipulation of the chilling requirement of sweet cherry trees

Thesis (MscAgric (Horticulture))--University of Stellenbosch, 2008. / Commercial production of sweet cherries has recently increased in South Africa, with more than 400 ha planted by 2006. Cherry, a high chilling fruit variety, is however not suited for the mild winter climate of South Africa. This was recognizable through common observed symptoms of delayed foliation and poor fruit set. In addition, cherry is exposed to long and hot summers in the postharvest period. The objective of this study was to evaluate cherry cultural practices that can manipulate (reduce) the trees chilling requirement under South African conditions. Cultural practices where aimed at increasing reserves (nitrogen, cytokinin and carbohydrates) in the tree. In addition, bud dormancy progression of cherry buds was quantified to determine the bud dormancy progression pattern under mild winter conditions. This was achieved through sampling of cherry shoots from different cherry production areas which was then forced in the growth cabinets. A model was developed to identify possible factors and groupings that can explain the cherry bud dormancy pattern. A model, comprising two joined straight lines, was fitted in order to characterize bud dormancy behaviour for sweet cherry cultivars under mild winter conditions. All cherry cultivars followed the expected pattern of entrance and exit from dormancy. Factor analysis showed that factors related to the entrance into dormancy primarily characterize bud dormancy behaviour. Bud dormancy patterns were also a function of environmental conditions within a year as shown by cluster analysis. In addition, buds entered dormancy in mid-summer and remained dormant until chilling accumulation commenced. Bud dormancy release was generally extended over a three to five-month period for all cultivars. Prior to spring budburst exit of both lateral and terminal buds occurred rapidly. Data indicate that there is no ecodormant phase for cherry under the prevalent climatic conditions in South Africa. Further experimentation was aimed at increasing reserves within the trees through cultural practices. In the nitrogen trials, fertilization in the postharvest period had no significant effect on field budburst or bud dormancy progression in one-year-old shoots. Time of flowering was advanced in N treatments during 2007 only. Yield was not significantly increased. Therefore, in this trial, N fertilization in the postharvest period did not significantly reduce the chilling requirement of mature sweet cherry trees under mild winter conditions. Application of particle films (Surround® and Raynox®) or ethylene inhibitors (Retain®) in the summer did not reduce the heat stress the trees experienced. Treatments had no significant effect on carbon assimilation, stomatal conductance, leaf surface temperature, fluorescence, bud dormancy, budburst, flowering and fruit set. Cytokinins sprays (benzyladenine) in autumn did not affect bud dormancy progression, spring budburst or flowering. Hydrogen cyanamide application in spring significantly advanced budburst, time to full bloom and increased yield. Promalin® and Retain®, however, had no significant effect on budburst, flowering or yield. It is therefore evident that cherry, due to its unexpected bud dormancy behaviour and its inability to be significantly influenced by several cultural practices, adapts poorly to South African climatic conditions through not reducing its chilling requirement significantly.

The Effects of Pre-Emergence Applications of Simazine and Post - Emergence Applications of Dalapon on Physio-Chemical Changes in Apple, Sour Cherry, Sweet Cherry, and Prunus Mahaleb

Simazine and dalapon have been used to control weeds in many crops during the last few years, but in 1963, when this study was begun, their uses in fruit orchards were limited. To study the effects of simazine and dalapon both on the control of weeds and on various metabolic changes in apple (Malis pumila), Montmorency sour cherry (Prunus cerasus), sweet cherry (Prunus avium), and Prunus mahaleb root s tock, experiment s were conducted from 1964 through 1966 in both the field and in the greenhouse. The application of a low do sage of simazine (4 lb/A) to soil around twelve-year-old Montmorency sour cherry trees in the field resulted in 60 to 70 percent weed control, while tree growth, chlorophyll content of the leaves, fruit y i e ld, and fruit quality were enhanced. Simazine applied at the higher dosages (8 and 12 lb/A) gave very good weed control (up to 100 percent) but reduced tree growth, chlorophyll content, and fruit yield and quality. In the greenhouse , the application of simazine at all three levels caused severe damage to R· mahal eb and sweet cherry trees and hence reduced the water uptake , but young apple trees treated with simazine showed no visible damage , although water uptake was reduced. In young sour cherry trees treated with 4 lb/A simazine, slight chlorophyll increase s we re observed , while at the higher dosages , decreases were observed. Simazine applied to the soil surface was absorbed readily by all trees. It accumulated in large amounts in the l eaves compared with the other parts of these plants. Sour cherry trees accumulated more simazine than apple trees. Results from both field and greenhouse trial sindicate that apple trees were comparatively resistant, and sweet cherry and ~ · mahaleb trees were very sensitive. Enzyme analys i s indicated that simazine caused an increase in the activity of peroxidase and polyphenol oxidase in all trees. The activity of catalase and cytochrome oxidase was decreased in f. mahaleb, sweet cherry , and sour cherry trees, but apple trees showed an increase in both catalase and cytochrome oxidase activity. Compared to the other trees , the most tolerant , untreated trees (apple) had relatively high peroxidase and low polyphenol oxidase activity, with moderate catalase and cytochrome oxidase activity . The moderately resistant species ( sour cherry) had moderate peroxidase, catalase, and cytochrome oxidase activity but very low polyphenol oxidase activity , while the sensitive trees (~ . mahaleb and sweet cherry) showed a wide range of enzyme activities . ~ · mahaleb leaves had low peroxidase and catalase activities and high polyphenol oxidase and cytochrome oxidase activities, but sweet cherry leaves had moderate peroxidase and polyphenol oxidase , high catalase, and l ow cytochrome oxidase activities. The movement of simazine in the soil was s low. Regardless of applied amounts, most of the herbicide stayed in the top 0-6 inches of soil after surface application, but movement in t he soil increased as the rate of application increased. The post-emergence application of dalapon to a Montmorency sour cherry orchard reduced weed growth in general, but complete weed control was not observed. Tree growth, chlorophyll content of the l eaves, fruit yield, and fruit quality of dalapon-treated Montmorency sour cherry trees were reduced compared with the untreated control. These reductions became greater as t he dalapon dosage increased .

Background: Low-grade chronic inflammation has been implicated as a risk factor in prostate-related pathologies including benign hyperplasia and cancer. Sweet cherry containing the bioactive anthocyanin (ACN), has demonstrated tumor inhibitory action in model systems, specifically inhibition of inflammatory molecules and prostaglandin biosynthesis. Objective: To assess the urinary and plasma concentrations of ACN from the daily consumption of 3 cups of sweet cherries for 4 weeks and test the relationship of ACN levels and cherry consumption to inflammatory biomarkers in an at risk population. Results: Prostaglandin E2 Metabolite (PGEM) levels were reduced with cherry consumption in men with elevated baseline values. Conclusion: We conclude that 1c (142g) of sweet cherries 3 times daily for 4 weeks significantly reduced the COX-2 metabolite, PGEM, in men with elevated baseline levels. This was the first study to examine the chronic effects of daily sweet cherries on COX-2 inhibition in an at risk population.

The aim of the research. To evaluate the genetic diversity in sweet and sour cher-ry populations, to characterize the injuries to blossoms and fruits caused by blossom blight brown rot, to identify the species composition of Monilinia patho-gens and to analyse the DNA polymorphism within and among pathogens popula-tions. Main tasks: 1. To analyse the genetic diversity of sweet and sour cherry cultivars and wild cherry population, growing in west Lithuania, using molecular marker methods (AFLP and SSR). 2. To determine factors affecting injuries caused by blossom blight. 3. To investigate characteristics of brown rot agent M. fructigena development in sweet and sour cherry cultivars differing in resistance to brown rot. 4. To identify species composition of Monilinia spp. in IH LRCAF stone fruit collection based on PCR methood and to evaluate inter- and intraspecific DNA polymorphism of Monilinia spp. based on AFLP method. / Tyrimų tikslas. Įvertinti trešnės ir paprastosios vyšnios populiacijų ge-netinę įvairovę, ištirti trešnės ir paprastosios vyšnios žiedų bei vaisių užsikrėtimo kaulavaisinių monilioze mechanizmus, nustatyti šios ligos sukėlėjų Monilinia rūšinę sudėtį bei išanalizuoti jų tarprūšinį ir vidurūšinį DNR polimorfizmą. Rapporten innehåller en bakgrundsdel med fakta om fågelbär som trädslag. Den innehåller en marknadsplan som studerar marknaden för körsbärsträ i Sverige och Europa, främst i Tyskland. I studien finns också en skötselplan för fågelbärsparcellerna i lövskogsförsöket på Mellomgården, Lerdala. Marknadsanalysen är gjord utifrån fakta från svensk och tysk litteratur och skötselplanen är ett resultat av mätningar och observationer i fält på den aktuella platsen. Marknadsanalysen visar att marknaden för fågelbär är begränsad i Sverige och att en generell prisnivå inte finns. Däremot är marknaden större i Tyskland och därifrån finns det flera prisexempel. Skötselplanen påpekar att en gallring omgående är nödvändig i försöket. Den huvudsakliga slutsatsen är att fågelbär med rätt skötsel kan uppnå en bra kvalitet och på så vis matcha kraven på marknaden.

<p>Mrka trulež, čiji je prouzrokovač Monilinia laxa, spada u najče&scaron;če i najznačajnije bolesti ploda ko&scaron;tičavih voćaka, naročito tre&scaron;nje. Cilj ovog rada je bio da se utvrde razlike u reakciji&nbsp; devet sorti plodova tre&scaron;nje (različitih pomolo&scaron;kih osobina) prema mrkoj truleži, uzorkovanih sa oglednog dobra Departmana za voćarstvo i vinogradarstvo, Poljoprivredni fakultet Novi Sad na Rimskim &Scaron;ančevima (koordinate: 45&deg;20&acute;N, 19&deg;51&acute;E). S obzirom da se u prirodnim uslovima plod tre&scaron;nje može zaraziti od strane vi&scaron;e patogena, biohemijski parametri su analizirani i na ve&scaron;tački inokulisanim plodovima. Ispitivane sorte su pokazale značajne razlike u pojavi oboljenja na plodu, kako u uslovima ve&scaron;tačke inokulacije, tako i u prirodnoj infekciji. U oba slučaja kod sorti su zabeležene različite ocene intenziteta zaraze, zavisno od sortimenta. Biohemijskom analizom plodova zabeležene su značajne razlike u sadržaju rastvorljivih proteina (SP), aktivnosti supereksid dismutaze (SOD), aktivnosti gvajakol peroksidaze (GPx), aktivnost pirogalol peroksidaze (PPx), lipidnoj peroksidaciji (LP). Utvrđene su značajne razlike u sadržaju &scaron;ećera, organskih kiselinih i sekundarnih metabolita (fenoli, tanini, proantocijanidini, flavonoidi, antocijanini). Takođe rezultati su pokazali razlike u antioksidantnoj aktivnost (DPPH, FRAP, ABTS i TRC testovi). Na sadržaj sekundarnih biomolekula kao i na antioksidantnu&nbsp; aktivnost&nbsp; uticali su sorta, intenzitet zaraze ploda, kao i interakcija između ova dva faktora.
