Studium optických vlastností tenkých vrstev prekurzorů pro přípravu monokrystalů perovskitů FAPbBr3 / Study of optical properties of thin films of perovskiteFAPbBr3 precursors

This thesis examines the preparation of thin layers of material for photovoltaic applications with focused on perovskites and determining their optical properties. Basic information about the photovoltaic panels, especially about the perovskites, and the preparation of thin layers is supplied in the theoretical section. This section includes description of optical properties and the main method of study of optical properties, spectroscopic ellipsometry. Experimental section is focused on the preparation of thin layers of perovskites FAPbBr3 and its precursors by spin-coating. Optical properties are evaluated using profilometry, UV VIS spectrometry and spectroscopic ellipsometry. The conclusion summarizes the results of this experiment with focused on comparison of ellipsometric spectrums of perovskites and its precursors.

Mechanické a transportní vlastnosti hybridních hydrogelových systémů / Mechanical and Transport Properties of Hybrid Hydrogel Systems

This master´s thesis deals with the study on mechanical and transport properties of hybrid hydrogel systems. Considering applications of hydrogels, especially in chemical industry, pharmacy, or eventually medical applications, for the study gellan and alginate-based hydrogels were selected. In order to compare individual characteristics physical and hybrid hydrogels were prepared. Gellan hydrogels were prepared in deionization aqua solution, calcium chloride dihydrate and tween 80 solution. Alginate hydrogels were prepared in calcium chloride dihydrate solution as well, and polyacrylamide with N,N´–methylenbisacrylamide. For the study of mechanical properties moisture analyser and rheology measurements were selected. Transport properties were studied using the diffusion experiments combined with UV-VIS spectroscopic detection. Concluding of this thesis is summarization of measured values, which provides comprehensive review of the problematics. It was discovered that the conveniently selected concentrations of structural components of hydrogel matrix and the additives can influence both the mechanical as well as the transport properties of studied hydrogels.

L'encapsulation de protéines pose de nombreuses difficultés liées à leurs propriétés physicochimiques, leur sensibilité aux conditions environnementales et opératoires. La structure 3D spécifique de chaque protéine étant directement liée à son activité biologique, un contrôle de l'intégrité protéique est indispensable pour assurer l'efficacité et la sécurité des systèmes formulés. Dans cet optique, l'encapsulation par émulsion multiple a été appliquée à une protéine modèle : la myoglobine (Mb). Le but de cette thèse a été l'étude du procédé d'encapsulation à travers 3 objectifs. Le premier fut d'étudier l'influence de paramètres de formulation sur les propriétés des particules obtenues et sur la conformation de la Mb. Les mesures en spectrométrie UV/Vis. et le calcul des principaux rapports d'absorbance ont constitué une méthode de contrôle fiable et rapide. Le second fut de valider la pertinence et de montrer les limites de cette méthode en la comparant à une seconde méthode utilisant des mesures conductimétriques. Enfin, l'étape de solidification des particules par élimination du solvant ainsi que le changement de solvant ont été étudiés.. Des microsphères creuses de 15μm avec un rendement d'encapsulation de 36% ont pu être obtenues tout en maintenant la conformation native de la Mb. L'emploi de polymère de masse moléculaire élevée et un taux d'élimination de solvant trop rapide sont 2 paramètres altérants significativement la protéine. Ces travaux ouvrent la voie au développement de transporteurs d'O2 utilisables par exemple dans le cas de pathologies musculaires / Protein encapsulation results in several problems related to their physicochemical properties, their sensitivity to environmental conditions and operating procedures. The specific 3D structure being directly linked to their biological activity, monitoring of protein integrity is crucial to ensure the efficacy and security of formulations. In this context, the multiple emulsion method was applied to a model protein: myoglobin (Mb). The aim of this thesis was to study the encapsulation process through 3 objectives. The first was to study the influence of formulation parameters on the particle properties and the conformation of Mb. Measurements by UV/Vis. spectrometry and calculation of key absorbance ratios established a reliable and rapid method for protein monitoring. The second was to validate the pertinence and show the limits of this method by comparing it to a second one using conductimetry measurements. Finally, the particle solidification by solvent removal and the solvent exchange were studied. The process maintains the Mb native conformation in Hollow microspheres of 15μm in diameter and an encapsulation efficiency of 36% were obtained, while keeping intact the Mb native conformation. The use of high molecular weight polymer and a fast solvent removal rate are 2 parameters inducing significant protein alteration. This work paves the way for the development of O2 carriers for muscular diseases for example

Kinetika koordinační polymerace 1-olefinů katalyzované diiminovými komplexy niklu / Kinetics of the Coordination Polymerization of -olefins Catalyzed by Nickel Diimine Complexes

This Ph.D. thesis is focused on kinetic study of propene and hex-1-ene coordination polymerizations initiated by complex [(2-tBuC6H4)N=C(1,8-naphtalenediyl)C=N(2-tBuC6H4)]NiBr2 activated by simple organoaluminium compounds and on product properties. In first three parts proper activators are chosen in model polymerizations. The attention is paid to the reproducibility of polymerizations and analytical methods. The fourth part is focused on propene polymerization kinetics with the aim to define optimal reaction conditions, especially polymerization temperature and time. The four part concerns also properties of polymers. The fifth and sixth parts deal with hex-1-ene polymerization kinetics conducted at various activator/initiator molar ratios and various catalytic precursor concentrations, to find out kinetic orders with respect to catalyst and cocatalyst concentration. Last part is oriented on detail mechanistic investigation of nickel diimine complexes activation process. Results of measurements in presence or absence of monomer enabled to propose new interpretation of UV-vis spectra based on concrete structures of absorbing species.

Kombinovaný nápoj na bázi vína a ovocné šťávy / Combined drink from wine and juice

This diploma thesis deals with preparation of combined drinks. These drinks consist of white wine and fruit concentrate base. This thesis was divided to 2 parts – senzory and chemical. As a fruit component was chosen aronia concentrate. The first sensory analysis was concentrated on a choice of a suitable white wine for this procedure. Second sensory analysis was focused on aditives. We added citric acid and ascorbic acid to the drinks. Evaluators had to choose the most tasty drinks. Next sensory analysis was dedicated to choice of the right drink composition. The last analysis was focused on choice of right fruit composition. Evaluators had to compare between the drink that was prepared from aronia concentrate or a drink prepared from aronia concentrate and other species of fruits. Their duty was to decide which one is more tasty. Chemical part was dealing with puting individual components of drink preparation to the test. Combined drinks had to undergo chemical tests too.

Studium extrakce biologicky aktivních látek do tukového základu / Study of extraction of biologically active substances into fatty base

This diploma thesis deals with macerates of comfrey (Symphytum officinale) in selected fats (food lard, cosmetic lard and almond oil). Selected fats were first characterized by dry matter content, saponification, acid, iodine number, peroxide value. At the same time, total and free fatty acids were determined using GC-FID. Macerates were prepared by extraction of comfrey roots with selected fats. In prepared macerates the change in acid and peroxide value was monitored, as well as the content of selected bioactive compounds. The content of total phytosterol and total carotenoid content was determined by UV-VIS spectrometry, phytosterols and carotenoids were also analysed using HPLC-DAD. Total phenolic content was measured using Folin-Ciocalteu reagent and antioxidant activity by ABTS assay. The measured properties were then compared with industrially produced comfrey ointment. In macerated fats the increase in acid and peroxide value was determined. Furthermore, an increase in the content of total phytosterols and total carotenoids was observed. Using HPLC-DAD the content of -sitosterol and stigmasterol was determined, but carotenoids were not detected. Of the tocopherols, only DL--tocopherol acetate was detected. During maceration, the content of total phenolic compound in fat increased, which caused a change in antioxidant activity.

Rozklady organických barviv metodami AOP / Organic dye decomposition by AOP´s methods

This diploma thesis is focused on the study of the influence of advanced oxidation processes on degradation of organic dyes. The field of AOP – very effective physically-chemical methods of wastewater treatment – includes application of strong oxidation agents, UV and ionizing radiation and electrical discharges. For this thesis, two of these methods were chosen – the application of oxidation agent (ozone) and UV radiation. Along with electrolysis, which is mentioned rather marginally in this thesis (it is the subject of the previous thesis), these phenomena are products of electrical discharge in water, where they participate in the processes of degradation in a different way. For all measurement series, two direct azo dyes were chosen as model substances – C.I. Direct Blue 106 and C.I. Direct Red 79. The ozonizer, in which either oxygen or synthetic or technical air were loaded as carrier gases, was used for degradation of dyes by ozone. The generated ozone was loaded into the bubbling vessel with dye solution of different initial concentration (10–130 mg.dm-3), which was followed by other bubbling vessel with KI solution for the next analytical determination of the amount of generated ozone. The other parameters changed were the gas flow (1–2 dm3.min-1), ozonizer output (minimal and maximal), type of dye, pH value of the solution (neutral or acid) and additional electrolyte (NaCl, Na2SO4 or any). The reactor for the study of the influence of UV radiation on degradation of dyes was an UV sterilizer into which the equivalent volume of dye solution was added. The possibilities of experimental settings were limited and only the type of dye, an additional electrolyte and pH value of the dye solution were adjusted (as in the case of ozone). Several series of samples were measured with various input conditions which more or less influenced the degradation of investigated dyes in this experiment. It was found that for both used methods the Direct Blue 106 dye was more degradable (with significantly better results for ozone than for UV radiation). The degradation of Direct Red 79 dye proceeded only by ozone treatment, in the case of the application of UV radiation no degradation occurred. By investigation of the influence of initial concentration of dye on its degradation, it was confirmed that with the initial concentration enhancement the final concentration rises as well, whereas in low concentrations (10–50 mg.dm-3) the initial concentration has no effect. The addition of an electrolyte had an accelerating effect on dye degradation in both methods (NaCl and also Na2SO4 showed similar results though the degradation proceeded in different ways). The addition of HCl accelerated the degradation only in the case of UV radiation; during the application of ozone the pH level of the system did not have any influence on the degradation. Oxygen and synthetic air had the strongest effect on ozone degradation (comparable results); in the case of technical air the final dye concentration was higher up to 30 %. The gas flow of 1.5 dm3.min-1 was stated as optimal with the ozonizer output 30 W (maximal). At minimal power the ozonizer produced very low amount of ozone.
