Variabilidade genética e evolução do gene PRNP em veados cinzas (Mammalia; Cervidae; Mazama)

Este segundo grupo é formado por M. gouazoubira e M. nemorivaga. O gene PRNP codifica a proteína Príon envolvida, quando em sua forma patogênica, na susceptibilidade a um conjunto de doenças neurológicas fatais e incuráveis. A Doença da Debilidade Crônica em Cervídeos (CWD) é um dos males neurológicos associados aos Príons, sendo epidêmica na América do Norte, porém nunca relatada em indivíduos Sul-americanos. A fim de entender qual curso evolutivo que esse gene percorreu nas espécies da família Cervidae, realizou-se uma análise de quais os mecanismos evolutivos que ele vem sendo submetido. Para tanto, foram analisadas as sequencias de nucleotídeos do éxon 3 do gene PRNP em amostras de 42 espécimes do gênero Mazama (M. gouazoubira - n: 36; M. nemorivaga - n: 06) de diversas regiões do Brasil, em comparação com outras espécies da família Cervidae. A inferência do mecanismo evolutivo do PRNP entre os cervídeos analisados neste estudo através do Teste de Neutralidade (dN-dS) sugere que nos Veados do Velho Mundo o gene vem passando por deriva genética. Já nos Veados do Novo Mundo a evolução ocorre em alternância de processos de deriva genética e seleção positiva. / The Mazama Genus consists of South American deer and is divided, according to the fur color in red deer and brocket deer. This second group is formed by M. gouazoubira and M. nemorivaga. The PRNP gene encoding the prion protein involved when the pathogenic form in susceptibility to a group of incurable and fatal neurological diseases. The Chronic Wasting Disease (CWD) is one of these neurological malady associated with prions and epidemic in North America, but never reported in South American individuals. In order to understand which evolutionary course that ran gene in the species of the Cervidae family was held an analysis of which of the possible evolutionary mechanisms that it are being submitted. Therefore, we analyzed the sequences of nucleotides of exon 3 of the PRNP gene in samples from 42 specimens of the genus Mazama (M. gouazoubira - n: 36; M. nemorivaga -n: 06) from different regions of Brazil. In comparison with other species of the Cervidae family, the conservation of the gene was observed in aligning sequences. The inference of the evolutionary mechanism of PRNP among cervids analyzed in this study through the Neutrality Test (dN-dS) suggests that Old World Deer in the gene has undergone genetic drift. Already in the evolution of New World deer occurs in alternation of positive selection and genetic drift processes.
