Postnatal vitamin D supplementation normalizes neonatal bone mass following maternal dietary vitamin D deficiency in the guinea pig

Since vitamin D deficiency is common at birth, the objective of this study was to test if postnatal vitamin D supplementation would normalize bone mineralization. Forty guinea pigs were randomized to receive a diet with or without vitamin D3 during pregnancy. Newborn pups were randomized to receive 10 IU of vitamin D3 or a placebo daily until d28. Measurements at birth and d28 included whole body and regional bone mass, osteocalcin and deoxypyridinoline, plus biomechanical testing of excised tibias and femurs. Offspring from deficient sows had lower body weight, whole body and tibia bone mineral content (BMC) and lower osteocalcin and biomechanical integrity. By d28 this group had lower whole body bone density and femur BMC, unless supplemented. Interactions with gender showed males continued to have low 25(OH)D despite supplementation. Therefore, neonates born to sows with dietary vitamin D deficiency require supplemental vitamin D to support normal bone mineral accretion.

Vitamin B-6 and pyrimidine deoxynucleoside metabolism in the rat

Serine transhydroxymethylase (STHM), a pyridoxal 5'- phosphate requiring enzyme is indirectly involved in pyrimidine deoxynucleotide metabolism. A decrease in the activity of this enzyme could lead to altered deoxycytidine (dC) metabolism. This study was undertaken to determine if a vitamin B-6 deficiency affects dC metabolism. The effect of a vitamin B-6 deficiency on the activity of STHM in liver, thymus, spleen and bone marrow was examined. In addition, the effect of a vitamin B-6 deficiency on urinary excretion of dC was examined. The effect of a vitamin B-6 deficiency on the urinary excretion and tissue retention of ³H label from ip injected ³H-dC was monitored. Rats were assigned in groups of six to one of four treatment groups: ad libitum control (ALC), pair fed control (PFC), ad libitum deficient (ALD) or meal fed deficient (MFD). At the end of weeks 2 and 6, rats from each treatment group received an ip injection of ³H-dC. Urines were collected for 24 hours following the ip inhibited due to lack of cofactor, then dTMP levels would fall. In an attempt to increase the concentration of dTMP, enzymes active in the conversion of dC and dCMP to dUMP would be expected to increase. Thus, dC salvage pathways would increase and dC synthesis would decrease as metabolism shifts toward production of deoxythymidine triphosphate (dTTP). The result would be lower urinary dC excretion. The present study was undertaken to explore the relationship between vitamin B-6 and pyrimidine deoxynucleotide metabolism. There were four hypothesis tested: Vitamin B-6 deficient rats will excrete less urinary dC than either ad libitum or pair fed controls; vitamin B-6 deficient rats will excrete a lower percentage of labeled dC in urine than control rats; vitamin B-6 deficient rats will incorporate less labeled dC into DNA than control rats but may retain more label in tissues as dC metabolites; activity of STHM from tissues of vitamin B-6 deficient rats will be lower than that from the control rats. / Graduation date: 1990

Prevalence of anaemia, deficiencies of iron and vitamin A and their determinants in rural women and young children: a cross-sectional study in Kalale district of northern Benin

Objective: To identify the magnitude of anaemia and deficiencies of Fe (ID) and vitamin A (VAD) and their associated factors among rural women and children. Design: Cross-sectional, comprising a household, health and nutrition survey and determination of Hb, biochemical (serum concentrations of ferritin, retinol, C-reactive protein and alpha(1)-acid glycoprotein) and anthropometric parameters. Multivariate logistic regression examined associations of various factors with anaemia and micronutrient deficiencies. Setting: Kalale district, northern Benin. Subjects: Mother-child pairs (n 767): non-pregnant women of reproductive age (15-49 years) and children 6-59 months old. Results: In women, the overall prevalence of anaemia, ID, Fe-deficiency anaemia (IDA) and VAD was 47.7, 18.3, 11.3 and 17.7%, respectively. A similar pattern for anaemia (82.4 %), ID (23.6%) and IDA (21.2%) was observed among children, while VAD was greater at 33. 6%. Greater risk of anaemia, ID and VAD was found for low maternal education, maternal farming activity, maternal health status, low food diversity, lack of fruits and vegetables consumption, low protein foods consumption, high infection, anthropometric deficits, large family size, poor sanitary conditions and low socio-economic status. Strong differences were also observed by ethnicity, women's group participation and source of information. Finally, age had a significant effect in children, with those aged 6-23 months having the highest risk for anaemia and those aged 12-23 months at risk for ID and IDA. Conclusions: Anaemia, ID and VAD were high among rural women and their children in northern Benin, although ID accounted for a small proportion of anaemia. Multicentre studies in various parts of the country are needed to substantiate the present results, so that appropriate and beneficial strategies for micronutrient supplementation and interventions to improve food diversity and quality can be planned.

Clinical Practice Recommendations for Screening Patients with Type-2 Diabetes for Vitamin D Deficiency: An Integrative Literature Review

Type-2 diabetes is a nationally growing health concern. Previous literature has implicated that vitamin D deficiency and type-2 diabetes are interconnected. At this time there are no guidelines in place to guide the evaluation or treatment of vitamin D deficiency in type-2 diabetic patients. In order to create up to date guidelines for the treatment and evaluation of vitamin D deficiency in type-2 diabetic patients an integrative literature review was conducted using EMBASE, PubMed, and CINAHL. The literature review resulted in 44 articles that met the inclusion and exclusion criteria. The literature review resulted in the creation of five clinical recommendations. The most significant change to current clinical standards includes screening all type-2 diabetic patients for vitamin D deficiency. Vitamin D supplementation is not recommended for use in treating type-2 diabetes, diabetic complications, or preventing the progression from prediabetes to diabetes. However, type-2 diabetic patients with vitamin D deficiency should be treated with vitamin D supplementation to improve indirect health outcomes and prevent morbidity and mortality.

Sorghum pericarp pigments are associated with the contents of carotenoids and provitamin A

However, the correlation of sorghum intake and vitamin A deficiency is inconsistent. The objective of this study was to identify and quantify the carotenoids and pro-vitamin A in the selected sorghum accessions with various pericarp pigments by using LC-MS. Among of total five carotenoids (α-carotene, β-carotene, lutein, zeaxanthin, and β-cryptoxanthin) that were identified and quantitated, three (α-carotene, β-carotene and β-cryptoxanthin) are precursors of vitamin A. The highest content of total carotenoids was detected in the sorghum accessions with yellow pericarp (PI656096, PI585374, PI563448, PI585351), while the highest β-carotene content was found in the accessions with brown or yellow pericarp (PI655996, PI656096, PI585374, PI563448, PI585351). The lowest carotenoids were found in the accessions with white pericarp (PI533943, PI656112, PI565121, PI560493). The pro-vitamin A was 584.9 ± 38.9 ng/g DW in yellow pericarp, 250.6 ± 28.9 ng/g DW in brown pericarp, and 89.0 ± 12.3 ng/g DW in white pericarp, respectively. It appeared the phenotypic diversity of sorghum pericarp colors was strongly associated with the contents of carotenoids and pro-vitamin A, indicating a different impact of various sorghum varieties on vitamin A deficiency and suggesting a possible prevention of vitamin A deficiency by breeding certain sorghum varieties with pericarp pigments.

Introdução A hipovitaminose A é reconhecida mundialmente como um dos problemas nutricionais mais importantes, causando impacto negativo na saúde pública por afetar alguns setores da população e pelas sérias conseqüências à saúde. Seus efeitos deletérios justificam o compromisso do governo brasileiro no controle e erradicação da deficiência desta vitamina. Objetivo Comparar o teor de vitamina A de alimentos comuns para a população brasileira com aqueles desenvolvidos com a substituição parcial do óleo/gordura utilizados na sua preparação por azeite de dendê, e avaliar suas características sensoriais e sua aceitação. Metodologia Foram desenvolvidas as seguintes preparações: sequilho salgado, sequilho doce e biscoito de polvilho. Para a caracterização das amostras dos produtos desenvolvidos, com e sem dendê, foram realizadas análises de umidade, atividade de água, composição centesimal, estabilidade oxidativa, cor, textura e perfil de ácidos graxos. A vitamina A foi determinada pela quantificação de -caroteno presente nas amostras. Todas as análises foram realizadas quando o alimento foi elaborado (T0) e após 30 dias (T30). As preparações desenvolvidas foram submetidas à análise sensorial. Resultados Os resultados das análises de umidade, atividade de água, composição centesimal e estabilidade oxidativa dos biscoitos sem dendê não diferiram significativamente das amostras com dendê, excetuando-se o biscoito de polvilho, onde possivelmente uma vedação deficitária da embalagem alterou algumas destas variáveis no T30. Quanto à cor, à textura e ao perfil de ácidos graxos, observou-se que as alterações ocorridas nos biscoitos com dendê foram em função da composição do próprio azeite. Com relação ao teor de vitamina A, todos os produtos com dendê apresentaram concentrações significativas deste nutriente quando comparados aos biscoitos sem dendê. No T30, o biscoito de polvilho não apresentou boa retenção da vitamina A, provavelmente em função da tecnologia adotada, que promoveu a aeração do produto, e pela vedação deficitária. Mesmo assim o produto pode, de acordo com a ANVISA, ser considerado fonte de vitamina A e, portanto, capaz de atender parcela importante das necessidades nutricionais. Todos os produtos tiveram boa aceitação no T0, porém foram rejeitados no T30. Conclusão- o azeite de dendê é uma potencial fonte de vitamina A e pode ser utilizado no enriquecimento de alimentos visando o controle da hipovitaminose A. / Introduction Vitamin A deficiency is recognized as one of the most important nutritional problems, and its causing negative effects in public health because it affects some population sections as well as the serious consequences to health. Its deleterious effects justify the Brazilian government commitment to control and vitamin deficiency eradication. Objective Compare the vitamin A content from the brazilian population ordinary foods with those developed with partial substitution of the oil/fat normally used for palm oil, and assess its sensorial characteristics and acceptance. Methodology The follow prepares were developed: salt starchy biscuit, sweet starchy biscuit and cookies made from cassava flour. To describe the samples of the developed products with and without palm oil, moisture, water activity, centesimal composition, oxidative stability, color, texture and fatty acids profile were performed. The vitamin A was determined by -carotene quantification in samples. All analysis were done when the food was made (T0) and after 30 days (T30). All the developed prepares were submitted to a sensorial analysis. Results The biscuits with no palm oil, contents of moisture, water activity, centesimal composition, oxidative stability, results were not significantly different from biscuits with palm oil. It has excepted the cookies made from cassava flour, a possible pack defective seal changed some of these variables during T30. The color, texture and fatty acids profile changes on biscuits with palm oil happened because of the own oil composition. All products with palm oil have shown significantly vitamin A content concentrations compared with biscuits with no palm oil. The cookies made from cassava flour didnt show a good vitamin A retaining during T30. It happened probably because of the technology used that organized the product aeration and by the defective seal. According to ANVISA the product could be considerate vitamin A source anyway, and, therefore it is able to reach important part of nutritional necessities. All products had good acceptation during T0, however they were rejected during T30. Conclusion- The palm oil is a vitamin A powerful source and can be used to foods fortification aimed at vitamin A deficiency control.
