DESCRIPTION DE PROJET DE RECHERCHE<p><p>Le canal sodium épithélial bloquable par l’amiloride (ENaC) est une protéine intégrale de la membrane apicale des épithéliums impliqués dans l’absorption du sodium. Deux fonctions majeures sont directement liées au fonctionnement d’ENaC. D’une part, la régulation de la balance sodée par le rein et donc de la pression artérielle et d’autre part, la clairance du fluide alvéolaire pulmonaire.<p>Le transport vectoriel de sel et d’eau à travers ces épithéliums à jonctions serrées repose sur un transport actif de sodium entraînant un flux osmotique d’eau. Ce transport de sodium s’effectue en deux étapes: l’entrée apicale, par diffusion, facilitée via ENaC, et la sortie basolatérale, active, par les pompes Na+/K+ ATPases.<p><p>Ces dernières années, un intérêt grandissant est porté sur les acides gras polyinsaturés à longues chaînes de type oméga 3 (PUFAs) et leurs implications dans divers processus physiologiques. Entre autres effets, les PUFAs modulent différents types de canaux ioniques (canaux Na+ dépendant du voltage, Ca++ L-type, K+).<p>Les études in vivo impliquant un effet à long terme des PUFAs décrivent des mécanismes inhibiteurs. Cependant, lors d’une étude précédente, axée sur la composition lipidique des membranes de cellules rénales en culture et l’influence de l’ajout d’acides gras saturés et insaturés sur le transport du sodium, nous avons constaté que les acides gras polyinsaturés à longues chaînes de type oméga 3 augmentaient la réabsorption du sodium. Ces résultats pourraient être intéressants, car les canaux sodiques de l’épithélium alvéolaire sont en contact direct avec le surfactant, dont la composition lipidique varie en fonction de l’apport alimentaire en PUFAs. Chez les prématurés humains, le syndrome de détresse respiratoire est une des causes les plus fréquentes de mortalité. Dans un certain nombre de cas, on peut restaurer une fonction pulmonaire satisfaisante par l’administration de surfactant.<p><p>Dans ce travail, nous avons opté pour une approche fondamentale des mécanismes de régulation du canal sodium épithélial par l’acide eicosapentanoïque (EPA, C 20:5, n-3). Des études électrophysiologiques, biochimiques et d’imagerie cellulaire ont été réalisées sur la lignée cellulaire A6 de rein d’amphibien, qui sert d’épithélium modèle pour l’étude d’ENaC depuis plus de 25 ans. Cette lignée exprime des canaux sodiques très sélectifs et possède des propriétés électrophysiologiques facilitant l’étude de leur régulation.<p><p>Ce travail nous a permis de mettre en évidence de nouveaux mécanismes fondamentaux dont la pertinence physiologique et /ou clinique ne pourra être établie qu’en transposant cette étude sur un modèle in vivo, comme nous le proposons dans les perspectives.<p><p>Dans le présent travail, nous avons étudié :<p><p>1.\ / Doctorat en Sciences biomédicales et pharmaceutiques / info:eu-repo/semantics/nonPublished

Effective Confinement and Bond Strength of Grade 100 Reinforcement

The primary reinforcement used for construction of structural concrete members has a yield strength of 60 ksi. This reinforcement grade was incorporated into construction over 50 years ago and remains the standard. Recent advances in material technology have led to the development of commercially available reinforcing steel with yield strengths of 100 ksi. While greater yield strengths can be utilized in design, it is essential that the bars can be properly anchored and spliced to fully develop their strength. Although design expressions are available for this purpose, they were established considering 60 ksi reinforcement. Therefore, the objective of this research program is to evaluate the development of high-strength reinforcing steel and establish a design expression for the development and splicing of this steel. Two phases of experimental tests were conducted. Phase I was performed by Glucksman (2018) and investigated the influence of splice length and transverse reinforcement on bond strength over four series of beam tests. This study (Phase II) was conducted following Phase I and consisted of reinforced concrete slab and beam testing over three series. An investigation was conducted on reinforcement development with a focus on the effect of splice length, concrete compressive strength, stress-strain relationships of the steel (ASTM A615 vs. ASTM A1035), and transverse reinforcement. Based on the results, the influences of test variables were identified, and a new confinement model was developed that estimates the transverse reinforcement contribution to bond strength. Finally, a design expression is provided for calculating the development and splice lengths of high-strength reinforcement.

The thesis is focused on the opinion of an office building from the perspective of two different standards and comparing their approaches. These standards is a currently valid European Standard EN 1992-1-1 (EC2) and standard according to US standards ACI 318-11. In this work were considered pillars in the second basement and on the first floor. Was also assessed locally supported slab above the 1st floor and staircase connects the individual floors. All the completed elements were assessed according to the 1st ultimate limit state.

Electrically Conductive Low Dimensional Nanostructures: Synthesis, Characterisation and Application

Miniaturization has become a driving force in different areas of technology including microelectronics, sensoric- and bio-technologies and in fundamental science. Because of the well-known limitations of conventional lithographic methods, newly emerging bottom-up approach, utilizing self-assembly of various nanoobjects including single polymer molecules and carbon nanotubes constitutes a very promising alternative for fabrication of ultimately small devices. Carbon nanotubes are attractive materials for nanotechnology and hold much promise to revolutionize fundamental science in a investigation of phenomena, associated with the nanometer–sized objects.It was found in this work that grafted chains of poly(2-vinylpyridine) form a shell covering the carbon nanotubes that makes them dispersible in organic solvents and in acidic water (CNTs-g-P2VP).The positively charged poly(2-vinylpyridine) shell is responsible for the selective deposition of carbon nanotubes onto oppositely charged surfaces. It was established that the deposition CNTs-g-P2VP from aqueous dispersions at low pH is an effective method to prepare ultra-thin films with a tunable density of carbon nanotubes.It was shown that poly(2-vinylpyridine) grafted to carbon nanotubes is a universal support for the immobilization of various nanoclusters at the carbon nanotube's surface. Prussian Blue nanoparticles were selectively attached to the surface of CNTs-g-P2VP.Conducting polymer nanowires are another very promising kind of nanomaterials that could be also suitable for applications in nanodevices and nanosensors. In this work was developed a simple method to control the conformation and orientation of single adsorbed polyelectrolyte molecules by co-deposition with octylamine. A simple chemical route to conductive polypyrrole nanowires by the grafting of polypyrrole from molecules of polystyrensulfonic acid was developed. The dc conductivity of individual polypyrrole nanowires approaches the conductivity of polypyrole in bulk.The conductivity can be described using variable-range hopping model.
