Topologically conserved hydrophobic residues of the thioredoxin C-subdomain stabilise GSTs

The thioredoxin-like fold is a well conserved fold that is present in many families of proteins. One such superfamily of proteins include the GSTs which are involved in phase II detoxification. GSTs primarily catalyse the metabolism of xenobiotics but are also involved in transporting non–substrate ligands and reactive compounds. The GST fold comprises an N-terminal thioredoxin domain and an all alpha helical C-terminal domain and is present in at least 18 classes of proteins. The N-terminal thioredoxin domain is characterised by the βαβαββα topology and can be further divided into two structural motifs, an N-terminal (βαβ) and a C-terminal (αββα) motif. A well conserved hydrophobic network exists between these two motifs and the role of the C-terminal motif is elucidated in this study using class Alpha GST as a model protein. A topologically conserved valine (Val58) and an isoleucine (Ile75) located on β3 and α3, respectively, were mutated to alanine. Secondary and tertiary structural characterisation as well as ligandin function of the mutant enzymes displayed no major structural alteration with respect to the wild-type enzyme. This was confirmed with high resolution crystal structures obtained. Enzymatic activity was maintained indicating that no structural alterations have occured that affects the active site dynamics and the domain interface as a result of the induced mutations. Thermal denaturation studies, however, indicated a slight destabilisation in the enzyme in the case of the valine mutation, but a large destabilisation was witnessed as a result of the isoleucine mutation. This is further observed in denaturant-induced equilibrium studies where the thermodynamic stability of proteins can be determined. Furthermore, as a result of the isoleucine mutation, the enzyme unfolds via a populated intermediate in contrast to the wild-type which globally unfolds via a two-state mechanism with no stable intermediates being populated. Pulse-proteolysis was employed as an additional probe for thermodynamic stability where the enzyme was digested by thermolysin at varying denaturant concentrations. Pulse-proteolysis results were in agreement with the thermal and denaturant-induced stability studies further confirming that the isoleucince substitution causes a large destabilisation. Thus these conserved hydrophobic residues of the thioredoxin C-subdomain play a crucial stabilising role in the GST fold.

Dynamique non-linéaire d'un dirigeable flexible / Non-linear dynamics of a flexible airship

Bien que déjà opérationnels depuis quelques dizaines d'années, les engins volants autonomes intéressent de plus en plus la communauté des mécaniciens et des automaticiens.Les recherches approfondies sur ce sujet se sont intensifiées. D'une part le développement des technologies permet d'envisager des concepts innovants, et d'autre part il s'agit d'étendre les capacités opérationnelles de ces engins pour pouvoir réaliser des missions plus complexes.Néanmoins, certaines difficultés techniques doivent être maitrisées, notamment l'étude de l'effet de la flexibilité structurelle, la considération des phénomènes aérodynamiques et la conception des lois de commande adéquates.Les ballons dirigeables autonomes font partie de cette classe d'engins promise à un bel avenir notamment dans le domaine du développement durable ou de la surveillance.La conception de dirigeables a souvent péché par son classicisme. Les chercheurs évitaient souvent les complexités mathématiques inhérentes aux formes non ellipsoïdales. Mais afin d'optimiser les performances de ces engins, différentes formes originales ont été proposées ces dernières années, notamment par le réseau DIRISOFT-FRANCE dont le dirigeable MC500 représente le support de mes travaux.Dans ce contexte, le présent manuscrit porte sur la modélisation et commande d'un dirigeable flexible MC500 ayant la forme d'une aile volante.Nous étudierons, dans un premier temps, la modélisation cinématique et dynamique du dirigeable sous sa forme " rigide " puis en tenant compte de sa flexibilité, en prenant en compte les efforts aérodynamiques. Nous admettons que ces engins volants subissent de grands déplacements et de petites déformations élastiques. Le phénomène des masses ajoutées est également pris en compte. Une étude analytique de ce phénomène a été développée pour le cas du dirigeable rigide et similairement pour le cas du corps flexible en grands mouvements. Elle est fondée sur la notion de potentiel des vitesses du fluide, et du développement de l'énergie cinétique du fluide sous l'effet d'un mouvement global du dirigeable. Cette méthode a permis de rechercher des solutions au problème de Dirichlet à travers la recherche de fonctions satisfaisant l'équation de Laplace pour les fluides interagissant avec une structure aux formes complexes pouvant être approchée dans un premier temps par un tronc de cône de section elliptique. L'étude du potentiel des vitesses du fluide autour du dirigeable a ainsi conduit à des équations de Lamé qu'il a fallu résoudre en tenant compte de certaines hypothèses. Un modèle dynamique non-linéaire complet du dirigeable a ainsi pu être établi.Une étude sur la loi de commande est effectuée sur le modèle " rigide " du dirigeable par deux approches : la première est linéaire se basant sur le linéarisé tangent, la deuxième est multi-modèle se basant sur la représentation quasi-LPV (linear parameter varying): cette approche nous permet de mettre le dirigeable dans un polytope bien défini par ses sommets. On a ainsi obtenu, par cette approche, un espace de convergence du dirigeable vers un point désiré. Cette approche q-LPV nous permet aussi d'exprimer le modèle dynamique non-linéaire du dirigeable au moyen de sous-systèmes linéaires, et on résout ces sous-systèmes par la formulation des LMI (linear matrix inequality). L'étude de la robustesse des commandes choisies par l'approche q-LPV a été établie et prouvée pour ce système en présence d'incertitudes sur les paramètres du dirigeable ainsi que de bruit de mesures des états de cet engin volant. Des simulations numériques seront présentées justifiant les résultats théoriques obtenus, et une analyse critique de ces deux approches sera présentée. / The autonomous flying machines interest more and more the community of the mechanics and the control engineers.The researches on this subject is intensified in these last decades. In one hand the development of technologies makes it possible to consider innovating concepts, and on the other hand it is a question of extending operational capabilities of these machines to be able to carry out more complex missions.Nevertheless, certain technical difficulties must be mastered, in particular the study of the effect of structural flexibility, consideration of the aerodynamic phenomena, and the design of adequate control laws.The autonomous airships are part of this class of machines promised a bright future in particular in the field of sustainable development or the monitoring.The design of airships is often characterized by its classicism. The researchers often avoided the inherent mathematical complexities of the non-ellipsoidal shapes. But in order to optimize the performances of these machines, various original forms were proposed these last years, in particular by the network DIRISOFT-FRANCE whose airship MC500 represents the support of my work.In this context, this manuscript focuses on the modeling and control of a flexible airship characterized by its flying wing shape.We will study initially the kinematic and dynamic modeling of the airship in its “rigid” form, and in a second step, we will include the study of its flexibility taking into account, in both cases, the aerodynamic efforts. We admit that these flying machines undergo large displacements and small elastic deformation.The phenomenon of the added masses is also taken into account. An analytical study of this phenomenon was developed for the case of the rigid configuration and similarly for the case of the flexible configuration in large displacements. The study is based on the concept of the velocity potential of the fluid, and the development of the kinetic energy of the fluid under the effect of an overall motion of the airship. This method made it possible to seek solutions with the problem of Dirichlet through the search for functions satisfying the Laplace equation for fluids interacting with a structure. We assume that the complex shape of the airship is able to be initially approximate by a truncated cone of elliptic section. Consequently, the study of the velocity potential of the fluid around the airship led to the famous equations of Lamé that have to be solved by taking account of certain assumptions. A complete non-linear dynamic model of the airship could therefore be established.A study on the control law is carried out on the “rigid” model of the airship by two approaches: The first is linear. It is based on the linearized tangent model. The second is multi-model and based on the quasi-LPV (linear parameter varying) representation. This approach enables us to put the airship in a polytope well defined by its summits. We thus obtained, by this approach, a space of convergence of the airship towards a desired point.This q-LPV approach also enables us to express the non-linear dynamic model of the airship by means of linear subsystems, and we solve these subsystems by the LMI formulation (linear matrix inequality).The study of the robustness of the control laws chosen by the approach q-LPV was drawn up and proven for this system in the presence of uncertainties on the parameters of the airship as well as noise of measurements of the states of this flying machine. Numerical simulations are presented justifying the theoretical results obtained, and a critical analysis of these two approaches are presented.

I denna uppsats har en studie genomförts med målet att ur ett företagsekonomiskt perspektiv beskriva och sedan analysera hur mindre optikerföretag styrs mot value added selling filosofin. Studien har genomförts med en abduktiv ansats och med en kvalitativ metod. Empiriskt material har samlats in via semi-strukturerade intervjuer med Hagelin & Co, Björkegrens Optik AB, Optiker Tottie och Specsavers. Vi författare har kommit fram till att value added selling filosofin finns mer eller mindre implementerat de flesta mindre företag i någon form av norm eller företagskultur. Det är också företagskulturen vi författare identifierar som det viktigaste styrmedlet för value-added selling.  I optikerbranschen som vi studerat är detta speciellt intressant av den anledningen att respondenterna visat flera tecken på hur value added selling filosofin fungerar, vilket vi författare anser gör studien relevant för mindre företag i allmänhet som verkar på en konkurrensintensiv marknad.
