Adhezní struktury leukemických buněk a jejich regulace kinázami rodiny Src / Adhesion structures of leukemia cells and their regulation by Src family kinasesObr, Adam January 2018 (has links)
Adhesion signaling is a field of cell biology studied mostly on adherent cell types. However, hematopoietic cells grow in suspension, and use adhesion to the extracellular matrix (ECM) only in their early development, or - in case of differentiated cells - to perform the tasks they are specialized for. Peripheral leukemic cells are derived from more or less immature hematopoietic precursors that have, among other alterations, defects in adhesion to the bone marrow microenvironment. On the other hand, leukemic stem cells (LSC) use adhesion to the bone marrow ECM as a mean to evade chemotherapy, and are a source of the minimal residual disease, and of the disease relapses. Kinases of the Src family (SFK) are known regulators of adhesion signaling in adherent cell types, and their overexpression and/or hyperactivation is often seen in malignant diseases. They are also involved in hematooncologic disease progression and resistance to therapy, particularly in several types of leukemias. In the present work, we used a variety of methods including microimpedance measurement, fluorimetric measurement of adhered cell fraction, immunoblotting, confocal microscopy, and interference reflection microscopy. Our results indicate that active Lyn kinase, a hematopoietic SFK, is present in adhesion structures of...

Vývoj tribometru pro měření adheze na reálné kolejnici / Development of a tribometer for the adhesion measurement on a real railVodehnal, Michal January 2019 (has links)
The submitted master’s thesis describes the development of a tribometer used to measure the adhesion of a real rail. The thesis contains an overview of the basic terms concerning wheel and rail contact. Following that, there is an overview of devicesthat are able to measure the adhesion on a real track. The author's solution includes a design of the tribometer, the proposed measurement methodology and also a description of the individual components. The result of this thesis is a functional device, verified in laboratory conditions and on a real track.

Konstrukce experimentálního zařízení pro studium pískování kolejových vozidel / Design of Experimental Stand for the Study of Railway Vehicles SandingGalas, Radovan January 2013 (has links)
Presented master’s thesis describes the design and the implementation of an experimental device used for the study of tribological aspects of wheel-rail contact. In this work, the reader will first learn the fundamentals such as adhesion, slip, traction curve, etc. Subsequently, the work is describing devices that are used for the study of wheel-rail contact around the world. This section is followed by the author proposed solution. The result is a twin disc device in the scale of 1:3. The device allows determining traction characteristics for different environmental conditions (presence of water, sand and oil in contact for different temperatures) and various operating parameters (contact pressure and speed). A part of this work are also the validation experiments and complete design documentation.

Vliv klimatických podmínek na adhezi v kontaktu kola a kolejnice / The effect of climate conditions on wheel-rail contact adhesionNohel, Rostislav January 2017 (has links)
Presented master’s thesis is focused on the experimental research of influence of climate conditions on wheel-rail adhesion, where climate conditions are meant two parameters: ambient air temperature and air humidity. These parameters always affect wheel-raid contact and their values change significantly during the year. For presented experimental research was used tribometer Mini Traction Machine (MTM) in configuration „ball on disc“. For determination of influence climate conditions was first found out the influence of air humidity on adhesion at certain temperatures, which varied between (1 to 50) °C. These experiments were performed for two types of discs, where one disc was exposed to leaf extract contamination. After that was assembled influence of both parameters (air humidity and ambient air temperature) on adhesion in the form of a 3D map, where were distinctioned states, when condensation has occured and when didn‘t. Obtained results in this thesis will be used for optimalization of rail vehicle control systems.

Studium flexibility a adheze cementových lepidel při různém stupni modifikace polymerním pojivem / Study of deformation and adhesion of cement adhesives at different stages of polymeric modificationKozielová, Silvie January 2019 (has links)
The content of the diploma thesis deals with the study of the flexibility and adhesion of cement tile adhesives, which are modified by a polymeric additive. The effects of the quantity and type of redispersible polymer powder used on the properties of the adhesives, in particular on the deformability of adhesives and their adhesion, are monitored. The theoretical part summarizes the basic data concerning classification of the ceramic tiles, information about the adhesives, the processed part concerning the polymer modified adhesives and the effects of the polymeric additives on their properties. The practical part deals primarily with the study of adhesion and flexibility of polymer modified adhesives and study of microstructure of selected materials.

This diploma thesis takes into account the possibility of repairing high-strength steels using coldspray method. The theoretical part is focused on the coldspray method, austenite transformation, bainitic steels and Rmat contact fatigue tests. In the experimental part evaluate the hardness, the structure of the supplied substrate. The hardness, adhesion and porosity of the test sprays are ranked. Furthermore, tests for contact fatigue are evaluated. Finally, the results of the experiments are summarized and measures are proposed to improve adhesion, or to continue this issue.

Vývoj maziva pro temeno kolejnice / Top of rail lubricant developmentSkurka, Šimon January 2021 (has links)
Friction modification within the wheel-rail interface is an important way of achieving ecologically friendly transportation of both persons and goods. This thesis aims to develop a new TOR lubricant, which will be able to maintain suitable frictional conditions while securing minimal adhesion required for traction. All measurements were carried out on tribometer MTM in the ball-on-disc configuration. In the first step, individual components were examined. More complex compositions were measured after that and the three best of them were compared with commercial TOR lubricants. The results show a good ability of developed compositions to maintain required adhesion, reduce wear, and all of them had resistivity against over-lubrication. Lastly, the process of lubricant verification before its application in real traffic was discussed.

Vliv povrchových úprav skleněných vláken na mechanickou odezvu GF/polyester kompozitu / The influence of surface modification of glass fibers on mechanical response of the GF/polyester compositeJaneček, Pavel January 2008 (has links)
The diploma thesis is aimed at an influence of surface modifications of fibrous reinforcements on mechanical properties at the fiber-matrix interface in fiber reinforced composites. Glass fibers were used as reinforcements and polyester resin was the matrix. The sample of composite consisted of polymer matrix in a form of cylindrical body placed on bundle of glass fibers. The design of the sample was constructed with respect to results of Finite Element Analysis (FEA). The tensile test using a materials testing machine (Zwick) was employed to evaluate adhesion at the fiber-matrix interface. Untreated glass fibers, fibers with commercial sizing, and fibers modified by deposition of plasma polymer film using Plasma-Enhanced Chemical Vapor Deposition (PECVD) at different deposition conditions were used for fabrication of composite samples. The fibers and composite samples were analyzed by Scanning Electron Microscopy (SEM) and Confocal Laser Scanning Microscopy (CLSM).
