Factors Related to Student Persistence in a New Residential STEM High School: The Case of the Tennessee Governor’s Academy for Mathematics and Science

Public and private sectors are grappling with decreasing numbers of science, technology, engineering, and mathematics (STEM) professionals as the need for them rises. State-supported STEM residential high schools may be the premier conduit to educate and socialize students in order to prepare for STEM college majors and careers. Gaining understanding of how these schools can nurture students academically and affectively so that they successfully matriculate to university STEM settings is valuable; however, minimal research exists related to the affective domain in relationship to retention at state-supported STEM residential schools. This exploratory, mixed methods case study describes factors that contribute to student persistence in a residential school setting. This study had four purposes: a) to describe students’ meaning of belonging at a STEM residential school; b) to explore potential relationships between students’ personality traits, expectations fulfillment, and belonging; c) to explore potential relationships between belonging and persistence, expectation fulfillment and persistence, and personality traits and persistence, and d) to describe characteristics of persistors and non-persistors. Participants in the study were the student population of the Tennessee Governor’s Academy for Mathematics and Science (TGA) (n = 41). Following interviews of the population, qualitative analysis of the interviews included pattern coding and creation of an explanatory effects matrix; quantitative analysis of survey data utilized ANOVA, chi-square, and correlation. Results of analyses produced a student-generated, multidimensional definition of belonging and showed that students who perceived they belonged and expressed institutional commitment via ownership of TGA had the highest persistence rate (100%). Quantitative findings showed that students whose expectations for TGA were met or exceeded had a higher sense of belonging, and belonging was associated with higher persistence. The personality traits agreeableness, conscientiousness, openness, and work drive were significantly related to belonging, suggesting that sense of belonging may function as a moderator between personality traits and expectation fulfillment. For future study, a longitudinal design is recommended as well as incorporation of the variable educational practices in relation to the variables expectation fulfillment, personality, belonging, and persistence.

Characterization of Section 404 Permit Mitigation Plans, Coastal Margin and Associated Watersheds, Upper Texas Coast

A predicted loss of agricultural rice-wetlands and increasing urbanization and development threatens the remaining freshwater wetlands along the upper Texas coast. To avoid, minimize, and mitigate wetland loss, the U.S. Army Corps of Engineers (Corps) is directed to enforce Section 404 of the Clean Water Act (1975 amendment) by administering permits for development. Furthermore, a 1990 Memorandum of Agreement (MOA) between the Corps and the U.S. Environmental Protection Agency (EPA) proposed a national goal of no net wetland loss (NNL). My goals were to identify the frequency of occurrence of freshwater wetland loss due to dredge or fill, assess final plans to mitigate wetland loss, and verify the persistence of the created compensatory wetlands. I created a database of 96 individual, Section 404 permits issued from 1981 to 2001 in the counties of Chambers, Hardin, Jefferson, Liberty, Montgomery, Orange, and San Jacinto (Galveston District Office, U.S. Army Corps of Engineers). Descriptive statistics were calculated for permit characteristics in relation to issue date (pre- or post-NNL). Public comments received from national and state agencies were rank ordered against mitigation plan type to determine Spearman's Rank Order Correlation Coefficient. Visual identification (via site visits and 1996 aerial photos) was used to validate compensatory wetland persistence. Shoreline protection of private property and oil and gas drilling (64% of permit applicants and 59% of impacts) had the greatest effect on wetland loss in the region, particularly Chambers, Jefferson, and Montgomery counties. Overall, 79.3 ha of freshwater wetlands were gained; however, gain was overestimated due to large projects for habitat enhancement. Permits issued post-NNL were more likely to have formal mitigation plans (58% vs. 13% pre-NNL) and allowed no net wetland loss. Although agency comments recommending more formal mitigation plans increased after NNL, only a weak positive correlation was detected (Spearman's r less than or equal to 0.4). Six of seven created wetlands remained in existence through 2006 though they are freshwater ponds replacing more diverse aquatic systems. I recommend the development of a comprehensive method to track wetland loss, mitigation, and changes in watersheds over time.

Conformations of semiflexible polymers and filaments

The biological function and the technological applications of semiflexible polymers, such as DNA, actin filaments and carbon nanotubes, strongly depend on their rigidity. Semiflexible polymers are characterized by their persistence length, the definition of which is the subject of the first part of this thesis. Attractive interactions, that arise e.g.~in the adsorption, the condensation and the bundling of filaments, can change the conformation of a semiflexible polymer. The conformation depends on the relative magnitude of the material parameters and can be influenced by them in a systematic manner. In particular, the morphologies of semiflexible polymer rings, such as circular nanotubes or DNA, which are adsorbed onto substrates with three types of structures, are studied: (i) A topographical channel, (ii) a chemically modified stripe and (iii) a periodic pattern of topographical steps. The results are compared with the condensation of rings by attractive interactions. Furthermore, the bundling of two individual actin filaments, whose ends are anchored, is analyzed. This system geometry is shown to provide a systematic and quantitative method to extract the magnitude of the attraction between the filaments from experimentally observable conformations of the filaments. / Die biologische Funktion und die technologischen Anwendungen semiflexibler Polymere, wie DNA, Aktinfilamente und Nanoröhren aus Kohlenstoff, werden wesentlich von deren Biegesteifigkeit bestimmt. Semiflexible Polymere werden charakterisiert durch ihre Persistenzlänge, mit deren Definition sich der erste Teil dieser Arbeit befasst. Anziehende Wechselwirkungen, wie sie z.B. bei der Adsorption, der Kondensation und der Bündelung von Filamenten auftreten, können die Konformation eines semiflexiblen Polymers verändern. Die Konformation ist dabei abhängig von der relativen Größe der Materialparameter und kann durch diese gezielt beeinflusst werden. Im Einzelnen werden hier die Morphologien semiflexibler Polymerringe, wie z.B. DNA oder ringförmiger Nanoröhren, untersucht, die auf drei verschieden strukturierten Substraten adsorbieren: (i) Ein topographischer Kanal, (ii) ein chemisch modifizierter Streifen und (iii) ein periodisches Muster topographischer Oberflächenstufen. Die Ergebnisse werden mit der Kondensation von Ringen durch anziehende Wechselwirkungen verglichen. Des Weiteren wird die Bündelung zweier Aktinfilamente, deren Enden verankert sind, untersucht. Diese Systemgeometrie liefert eine systematische Methode, um die Stärke der Anziehung zwischen den Filamenten aus experimentell beobachtbaren Konformationen zu berechnen.

Characterization and persistence of potential human pathogenic vibrios in aquatic environments

Vibrio spp., natural inhabitants of aquatic environments, are one of the most common causes of bacterial gastroenteritis in the world, being spread to humans via the ingestion of seafood, contaminated drinking water or exposure to seawater. The majority of Vibrio spp. are avirulent, but certain strains may sporadically be human pathogenic. Vibrio cholerae may cause cholera and fatal wound infections, Vibrio parahaemolyticus may cause gastroenteritis and Vibrio vulnificus may cause wound infections and sepsis. To expand current knowledge of the occurrence, ecological niche and persistence of potential human pathogenic Vibrio spp. in aquatic environments, occurrence and laboratory studies were performed. The seasonal variation of Vibrio spp. in clams and mussels from Mozambique and Sweden were studied, with isolated strains characterized and compared with those isolated from water samples collected in India. Results showed that the numbers of Vibrio spp. in Mozambican clams peaked during the warmer rainy season and that the dominating species was V. parahaemolyticus. Biochemical fingerprinting and virulence screened by PCR revealed a high similarity among strains from the different aquatic environments. However, isolate functional hemolytic analyses and antibiotic resistance patterns differed between strains; Swedish and Indian strains were less sensitive to the tested antibiotics and had a lower hemolytic capacity than those from Mozambique. Molecular analysis of bacterial DNA from Swedish mussels showed the presence of the three Vibrio spp. most commonly linked with human illness, as well as their associated virulence genes. The strains isolated from marine and clinical environments were equally and highly harmful to the tested eukaryotic cells. The persistence of clinical V. cholerae in aquatic environments was investigated in vivo. Strains were exposed to mussels, with bacterial uptake and elimination then examined. The mussels were able to avoid the most potent strain by complete closure of shells. The less potent strain was accumulated in mussel tissue in low levels and one marine control strain to a higher degree. Mussels eliminated the pathogenic strain less efficiently than they did the marine strain. One clinical and one marine strain were then exposed to 4°C for 21 days, with the temperature then increased to 20°C. The clinical strain was more prone to lose culturability than the marine strain at 4°C, the former performed significantly better in regaining culturability after the temperature up-shift. Subsequently, the persistence of the clinical strain in natural bottom sediment, incubating as above, was studied and results showed a similar decrease in culturable numbers in the sediment as in the water. As the clinical V. cholerae strains did not carry any of the standard set of virulence genes, the ability to change from non-culturable to culturable may be of great importance to strain pathogenicity. The results also show that natural bottom sediment may be a potential reservoir of human pathogenic Vibrio spp.

The Life Course of Single Welfare-Reliant Mothers: Experiences in Seeking Access to and Persisting in Post-Secondary Education

Welfare reform, as a social policy, has implications for higher education policy, because it restricts welfare-reliant women from seeking sufficient post-secondary education for economic mobility. The 2006 Deficit Reduction Act was reinterpreted in the 2008 final rule, allowing welfare-reliant mothers to pursue up to 12 months of post-secondary education. However, this is not sufficient for mothers to persist toward completion of associate's or bachelor's degrees. Recent scholarship has not adequately investigated the impact of this expanded access to post-secondary education. The purpose of this study was to investigate the essence of single welfare-reliant mothers' experiences in their life course from poverty to post-secondary education. To better understand the essence of their experience, this study utilized a transcendental phenomenological approach to investigate experiences that influenced their (a) pathways and social roles, (b) perspectives as sole providers, and (c) decisions to access and persist in post-secondary education. The three major findings of this study were presented in the context of a developmental life course framework supported by social role theory and women's adult The three major findings of this study were presented in the context of a developmental life course framework supported by social role theory and women's adult identity development concepts. The first major finding elucidated two distinct pathways to adulthood for six single welfare-reliant mothers. The three teen mothers experienced transitions to adulthood that were premature, truncated, and compacted. As a result they missed their developmental task of exploring possible selves in their transitions to adulthood. During their identity development as sole providers they returned to the task of exploring possible selves in their choices to access post-secondary education. The second major finding elucidated that single welfare-reliant mothers' perspectives as sole providers were experienced as an evolving adult identity, beginning with the birth of their first child and evolving throughout the era of early adulthood as mothers persisted in post-secondary education. The third major finding elucidated a recurring pattern of negotiating between role conflict and role salience experienced by single welfare-reliant mothers that resulted in critical junctures and recurring commitments to their decisions to persist toward post-secondary education goals. This study determined that commitment toward their adult identity as sole providers had a direct link to their commitment toward persisting in post-secondary education.

Pathobiology of African relapsing fever Borrelia

Relapsing fever (RF) is a disease caused by tick- or louse-transmitted bacteria of the genus Borrelia. It occurs worldwide but is most common in Africa where it is one of the most prevalent bacterial diseases. The main manifestation is a recurring fever which coincides with massive numbers of bacteria in the blood. Severity ranges from asymptomatic to fatal. RF is usually considered a transient disease. In contrast, B. duttonii causes a persistent, residual brain infection in C57BL/6 mice which remains long time after the bacteria are cleared from the blood. The host gene expression pattern is indistinguishable from that of uninfected animals, indicating that persistent bacteria are not recognized by the immune system nor do they cause noticeable tissue damage. This is probably due to the quite low number of bacteria residing in the brain. The silent infection can be reactivated by immunosuppression allowing bacteria to re-enter the blood. To investigate if the residual infection is in a quiescent state or if the bacteria are actively dividing, mice with residual brain infection were treated with the cell-wall disrupting antibiotic ceftriaxone, which is only active against dividing bacteria. Since all mice were cured by ceftriaxone we conclude that the bacteria are actively growing in the brain rather than being in a latent, dormant state. The brain is used as an immunoprivileged site to escape host immune defence and probably as a reservoir for bacteria. RF is a common cause of pregnancy complications, miscarriage and neonatal death in sub-Saharan Africa. We established a murine model of gestational relapsing fever to study the pathological development of these complications. B. duttonii infection during pregnancy results in intrauterine growth retardation as well as placental damage and inflammation. Spirochetes cross the maternal-foetal barrier, resulting in congenital infection. Further, pregnancy has a protective effect, resulting in milder disease during pregnancy. A clinic-based study to investigate the presence of RF in Togo was performed. Blood from patients with fever were examined for RF by microscopy, GlpQ ELISA and PCR. About 10% of the patients were positive by PCR and 13% had antibodies to GlpQ. Many RF patients originally had a misdiagnosis of malaria, which resulted in ineffective treatment. The inability of microscopic analysis to detect spirochetes demonstrates the need for tests with greater sensitivity. To provide simple, fast, cheap and sensitive diagnostics using equipment available in small health centres, a method based on enrichment of bacteria by centrifugation and detection by Giemsa staining was developed which detects <10 spirochetes/ml. To study the phylogeny of RF, IGS and glpQ were sequenced and neighbor joining trees were constructed. B. persica and B. hispanica were distant from the other species iswhereas B. crocidurae appeared to be a heterogeneous species. B. duttonii is polyphyletic in relation to B. recurrentis suggesting that the two species may in fact be the same or have a polyphyletic origin.

Modeling and Analysis of Population Dynamics in Advective Environments

We study diffusion-reaction-advection models describing population dynamics of aquatic organisms subject to a constant drift, with reflecting upstream and outflow downstream boundary conditions. We consider three different models: single logistically growing species, two and three competing species. In the case of a single population, we determine conditions for existence, uniqueness and stability of non-trivial steady-state solutions. We analyze the dependence of such solutions on advection speed, growth rate and length of the habitat. Such analysis offers a possible explanation of the "drift paradox" in our context. We also introduce a spatially implicit ODE (nonspatial approximation) model which captures the essential behavior of the original PDE model. In the case of two competing species, we use a diffusion-advection version of the Lotka-Volterra competition model. Combining numerical and analytical techniques, in both the spatial and nonspatial approximation settings, we describe the effect of advection on competitive outcomes. Finally, in the case of three species, we use the nonspatial approximation approach to analyze and classify the possible scenarios as we change the flow speed in the habitat.

Does normal developmental expression of psychosis combine with environmental risk to cause persistence of psychosis? A psychosis proneness-persistence model

Background. Research suggests that low-grade psychotic experiences in the general population are a common but transitory developmental phenomenon. Using two independent general population samples, the hypothesis was examined that common, non-clinical developmental expression of psychosis may become abnormally persistent when synergistically combined with developmental exposures that may impact on behavioural and neurotransmitter sensitization such as cannabis, trauma and urbanicity. Method. The amount of synergism was estimated from the additive statistical interaction between baseline cannabis use, childhood trauma and urbanicity on the one hand, and baseline psychotic experiences on the other, in predicting 3-year follow-up psychotic experiences, using data from two large, longitudinal, random population samples from the Netherlands [The Netherlands Mental Health Survey and Incidence Study (NEMESIS)] and Germany [The Early Developmental Stages of Psychopathology (EDSP) study]. Results. The 3-year persistence rates of psychotic experiences were low at 26% in NEMESIS and 31% in EDSP. However, persistence rates were progressively higher with greater baseline number of environmental exposures in predicting follow-up psychotic experiences (χ2=6·9, df=1, p=0·009 in NEMESIS and χ2=4·2, df=1, p=0·04 in EDSP). Between 21% and 83% (NEMESIS) and 29% and 51% (EDSP) of the subjects exposed to both environmental exposures and psychotic experiences at baseline had persistence of psychotic experiences at follow-up because of the synergistic action of the two factors. Conclusion. The findings suggest that environmental risks for psychosis act additively, and that the level of environmental risk combines synergistically with non-clinical developmental expression of psychosis to cause abnormal persistence and, eventually, need for care.

The phytotoxic effect of ALS inhibiting herbicide combinations in prairie soils

The objective of this study was to determine if the presence of two ALS inhibiting herbicide residues in three Saskatchewan soils would result in an additive, synergistic, or antagonistic interaction. This was determined through field trials where herbicides were applied sequentially over the course of two years and through dose-response modelling. The herbicides examined in these experiments were imazamethabenz, flucarbazone-sodium, sulfosulfuron, and florasulam, each in combination with imazamox/imazethapyr. The phytotoxicity and persistence of the herbicides in soil was assessed using an Oriental mustard root inhibition bioassay. The determination of herbicide interaction was made through the comparison of the experimentally observed values to theoretically expected values derived from a mathematical equation.<p>The dose response curves created by placing incremental concentrations of these herbicides in soil were compared using the I50 parameter, which is the concentration resulting in a 50% reduction in root length. It appeared that soil organic matter followed by soil pH had the greatest effect in reducing herbicide residue phytotoxicity in the tested soils. Based on the bioassay analysis of sequentially applied ALS inhibiting herbicides, it is proposed that the phytotoxic effect of herbicide residues in soil result in additive injury effects rather than synergistic or antagonistic interactions.

Comparing the Use of Abundance and Consistent Occupancy Measures to Predict Local Species Persistence

I compared the utility of two continuous time-series data measures for applied conservation biology by investigating how well each could predict future local persistence of a diverse set of bird species. I used 37 years of data from the North American Breeding Bird Survey to calculate abundance from yearly point-counts and permanence (i.e., consistent occupancy over time) from yearly presence-absence data in the early portion of the study period, then used the later portion of data to empirically evaluate how well each measure predicted persistence two decades into the future. I found that permanence could only match the ability of abundance to accurately predict local species persistence if multiple within-year repeated observations contributed to its calculation. Neither measure was effective at predicting persistence for regionally rarer species. I suggest the yearly and within-year repeated collection of abundance estimating data for use in applied conservation biology to best ensure biodiversity persistence.
