Sex, estrogen and the role of cardiac vasoactive gene systems in the modulation of cardiac hypertrophy in ANP gene-disrupted mice

Sex dimorphism in the prevalence, onset, development and progression of cardiovascular disease (CVD) is well recognized. Sex-specific differences in adaptation to cardiac pathological progressions such as cardiac hypertrophy (CH), and the extent to which they are attributable to sex hormones requires further delineation. The objective of this dissertation was to determine which cardiac vasoactive systems are responsible for sex-specific differences in CH modulation using the atrial natriuretic peptide gene-disrupted (ANP-/-) mouse model. First, sex-specific differences in the expression of the cardiac natriuretic peptide (NP) and nitric oxide synthase (NOS) systems were evaluated. Next, the influence of 17β-Estradiol (E2) on the expression and signaling of the cardiac NP and NOS systems was determined in ovariectomized (OVX) female ANP+/+ and ANP-/- mice. Finally, sex-specific differences in cardiac adaptation to Angiotensin II (ANGII) pressure overload were elucidated in male and intact female ANP+/+ and ANP-/- mice. These studies revealed that males predominantly use the NP system and females predominantly use the NOS system. Sex-specific differences in the cardiac NOS system were further enhanced by E2 in OVX female ANP+/+ and ANP-/- mice. In the female ANP-/- mouse, E2 was found to signal through the NOS system to significantly increase plasma cGMP. Finally, male and female differences were demonstrated in the sex-specific patterns of cardiac vasoactive gene system expression and development of cardiac dysfunction in response to ANGII treatment. Sex dimorphism was observed in the expression of BNP and NPR-A in male and female ANP-/- mice treated with ANGII. Female ANP+/+ and ANP-/- ANGII-treated mice exhibited elevated E/E’ ratios that were not found to the same extent in genotype matched ANGII-treated male mice, demonstrating that female mice developed ANGII-mediated mild left ventricle diastolic dysfunction. Based on the results of this dissertation, we conclude that sex-specific differences do indeed exist in the cardiac adaptation to pathological stresses. Mapping the cellular mechanisms regulating atrial natriuretic peptide secretion

Abstract Atrial natriuretic peptide (ANP) and B-type natriuretic peptide (BNP) are cardiac hormones, which are involved in the regulation of blood pressure and fluid homeostasis. The major determinant for ANP and BNP release are atrial and ventricular wall stretch, but also some vasoactive factors such as endothelin-1 (ET-1) can enhance cardiac hormone secretion. The mechanical stretch rapidly activates multiple signal transduction pathways in cardiac cells, but the cellular mechanisms mediating stretch-induced ANP secretion are still unknown. The aim of the present study was to examine the cellular mechanisms of autocrine/paracrine factors and stretch-induced ANP secretion. Genistein, a potent protein tyrosine kinase (PTK) inhibitor, rapidly increased cardiac contractile force and ANP secretion in perfused rat heart. This effect of genistein may be unrelated to the inhibition of PTKs since this stimulation was blocked by a L-type calcium channel antagonist and Ca2+/calmodulin-dependent protein kinase II inhibitor. Pregnancy hormone relaxin increased heart rate and ANP secretion in perfused spontaneously beating heart, suggesting that relaxin may have a role in modulating cardiac function. Cellular mechanisms of atrial wall stretch-induced ANP secretion were also studied. This enhanced secretion was blocked by sarcoplasmic reticulum Ca2+-ATPase inhibitor thapsigargin and PTK inhibitor lavendustin A, indicating that thapsigargin sensitive Ca2+ pools and activation of PTK orPTK cascade have an important role in the regulation of stretch-secretion coupling. In addition, protein phosphatase inhibitor okadaic acid accelerated stretch-induced ANP secretion, suggesting that precise balance of protein kinase and phosphatase activity plays a role in mechanical stretch-induced ANP secretion. Finally interactions of endothelial factors regulating ANP exocytosis were studied. The potent nitric oxide synthase inhibitor L-NAME increased basal and atrial wall stretch-induced ANP secretion in the presence of ET-1, suggesting that nitric oxide may tonically inhibit ANP secretion.

Plasma N-terminal Proatrial Natriuretic Peptide Concentration in Cats with Hypertrophic Cardiomyopathy

Objective: We sought to determine N-terminal proatrial natriuretic peptide concentrations [Nt-proANP] in plasma from cats with hypertrophic cardiomyopathy (HCM). Secondarily, we wished to evaluate the relationship between [Nt-proANP] and echocardiographic variables. Methods: Venous blood samples were obtained from seventeen cats with HCM and from nineteen healthy cats. Plasma [Nt-proANP] was determined using an ELISA assay. The relationship between plasma [Nt-proANP] and M-mode, 2-dimensional and Doppler echocardiographic variables was evaluated. Cats that were hyperthyroid or had evidence of renal disease were excluded from the study. Results: The mean plasma [Nt-proANP] was higher in cats with HCM (3.81 +/- 1.23 pmol/l) than in control cats (3.08 +/- 1.41 pmol/l); however, this difference was not statistically significant (p=0.17). There was a significant correlation between plasma [Nt-proANP] and left ventricular posterior wall thickness (r = 0.42; p=0.01). Additionally, plasma [Nt-proANP] was correlated with left atrial size (r = 0.35; p=0.03). A linear regression model was developed to further explore these relationships. LAs2D and LVPWd had an interactive effect on plasma [Nt-proANP] (R2 = 0.2737; p= 0.02). There was no correlation between any other echocardiographic variable and plasma [Nt-proANP]. There was no correlation between plasma [Nt-proANP] and heart rate (HR), body-weight, or age. Conclusions: Cats with HCM do not have significantly higher plasma [Nt-proANP] than normal cats. Placental insufficiency and fetal heart: Doppler ultrasonographic and biochemical markers of fetal cardiac dysfunction

Abstract The first aim of this study was to investigate the relationship between Doppler ultrasonographic parameters and biochemical markers of human fetal cardiac dysfunction and myocardial cell damage in pregnancies complicated by placental insufficiency and/or fetal growth restriction. Our second aim was to examine fetal central and peripheral hemodynamic characteristics associated with retrograde aortic isthmus net blood flow. Fetuses with significant myocardial cell damage (cTnT > 0.10 ng/ml) had increased pulsatility in the blood velocity waveforms of ductus venosus, left hepatic vein and inferior vena cava, and had more often atrial pulsations in the umbilical vein. Their umbilical artery NT-proANP concentrations were higher than in fetuses without myocardial cell damage. The proportion of left ventricular cardiac output of the combined cardiac output was greater and the corresponding proportion of the right ventricle was less than in fetuses with only increased NT-proANP levels ( > 1145 pmol/l). Tricuspid regurgitation was present more often and the right ventricular fractional shortening was less in fetuses with myocardial cell damage than in fetuses with normal umbilical artery cTnT levels. In fetuses with placental insufficiency and/or growth restriction (n = 48), umbilical artery NT-proANP concentrations showed a significant positive correlation with ductus venosus, left hepatic vein and inferior vena cava pulsatility index values for veins. Fetuses with placental insufficiency and antegrade aortic isthmus net blood flow demonstrated a shift in their right ventricular cardiac output from the pulmonary to the systemic circulation, and foramen ovale volume blood flow made up the majority of the left ventricular cardiac output. Fetuses with retrograde aortic isthmus net blood flow failed to demonstrate these changes, and they had signs of increased left atrial pressure. In addition, right ventricular fractional shortening was decreased and the pulsatility in the ductus venosus blood velocity waveforms was increased. In conclusion, human fetal myocardial cell damage was associated with a rise in systemic venous pressure, a change in the distribution of cardiac output towards the left ventricle and a rise in right ventricular afterload. Fetuses with retrograde aortic isthmus net blood flow failed to rearrange the distribution of the cardiac output and they had signs of increased left atrial pressure. In addition, right ventricular afterload and pulsatility in the ductus venosus blood velocity waveforms were increased.

Dexamethasone Recruits Atrial Natriuretic Peptide Secretory Cells in the Rat Left Atrium and Apex of the Ventricle

The response of the atrial natriuretic peptide (ANP) secreting cells from both rat atria and the apex of the ventricles to dexamethasone (DEX) was analyzed by the plaque assay. In right atrial cardiocytes, 25% of the cells secreted ANP basally; DEX treatment did not alter this percentage. However, in the left atrial secretory population, a discordance between the basal (15%) and DEX stimulated (25%) percent plaque formation was found. ANP secreting cells from the ventricular apex responded similarly to DEX exposure (26%), with 8% of the cells basally releasing the hormone. These data suggest that in both the left atria and apex of the rat ventricles, exposure to DEX recruits ANP secretory cells from a non-secreting population. Consequently, the release of ANP from these tissues would increase after glucocorticoid stimulation.

Atrial natriuretic peptide (ANP): A Novel Mechanism for Reducing Ethanol Consumption and Seeking Behaviors in Female Alcohol Preferring (P) Rats

Atrial Naturietic Peptide (ANP) is a neuropeptide that regulates function of the hypothalamic-pituitary-adrenal (HPA) axis, immune and neuroimmune system, and epigenetic factors. Research has indicated that ANP may mediate alcohol intake, withdrawal, and craving like behaviors. ANP receptors are present in the mesocorticolimbic (MCL) reward pathway of the brain, which includes the nucleus accumbens (Acb) and the ventral tegmental area (VTA). The objectives of the present study were to examine the effects of ANP microinjected into Acb subregions (Shell (Sh), Core (Co), ventral to AcbSh) on operant ethanol (EtOH) self-administration and into posterior VTA (pVTA) on EtOH-seeking behavior of female alcohol-preferring (P) rats. In the first experiment, ANP (0, 10 μg, or 100 μg) was microinjected into subregions of the Acb to determine its effects on EtOH self-administration. In the second experiment, ANP was microinjected into pVTA to determine its effects on Pavlovian Spontaneous Recovery (PSR) of responding, a measure of context-induced EtOH-seeking behavior. Administration of ANP directly into the AcbSh significantly reduced EtOH self-administration compared to vehicle, whereas ANP into the AcbCo or areas directly ventral to the AcbSh did not alter responding for EtOH. Microinjection of ANP into the pVTA significantly reduced responding on the EtOH-associated lever during the PSR test. The data indicate that activation of ANP systems in the (a) AcbSh can inhibit EtOH intake, and (b) in the pVTA can inhibit EtOH-seeking behavior. The results suggest that manipulations of the ANP system could be a potential target for pharmacotherapeutic intervention to treat alcohol use disorder. Novel Insights into PKG Activation and cGMP Signaling in Response to Nitric Oxide and Atrial Natriuretic Peptide in Vascular Smooth Muscle Cells

Cyclic 3',5'-guanosine monophosphate (cGMP) is a key signaling molecule involved in a myriad of physiological processes, including vascular smooth muscle (VSM) tone, water- and electrolyte homeostasis, platelet aggregation, airway smooth muscle tone, smooth muscle proliferation and bone formation. Increased occurrence of vascular disorders including erectile dysfunction, hypertension, stroke and coronary artery disease, have made it increasingly important to study the dynamic interplay between cGMP synthesis and hydrolysis in VSM cells. This dissertation examines the spatial distribution of intracellular cGMP, [cGMP]i, in response to NO and atrial natriuretic peptide (ANP) in VSM cells. To investigate the spatial patterning of [cGMP]i, we have developed a new generation of non-FRET (fluorescence resonance energy transfer) cGMP biosensors that are suitable to monitor [cGMP]i in response to physiological (low-nanomolar) NO and ANP concentrations and that qualify for real-time, confocal imaging techniques. We have termed these indicators FlincGs, for green fluorescent indicators of cGMP. For the development of FlincGs, we made use of the specific cGMP binding characteristics of PKG. We utilized site-specific mutagenesis, kinetic cGMP binding, dissociation and kinase assays, as well as crystallography, in order to investigate PKG activation and cGMP binding dynamics in greater detail. Based on these studies, our novel, non-FRET cGMP biosensors were designed by attaching cGMP binding fragments of PKG to the N-terminus of circular permutated green fluorescent protein. We applied FlincGs in cultured VSM cells as well as in intact tissue to determine whether two spatially distinct populations of guanlylyl cyclase (cytosolic versus membrane bound) underlie the generation of spatiotemporally-specific patterns of [cGMP]i formation.

Dieta hiperlipídica com ou sem adição de sal modula diferentemente a produção de peptídeo natriurético atrial e a expressão de renina em camundongos / Diets rich in saturated fat and/or salt differentially modulate atrial natriuretic peptide and renin expression in C57Bl/6 mice

Fundação de Amparo à Pesquisa do Estado do Rio de Janeiro / Estudo dos efeitos de uma dieta rica em sal e / ou gordura saturada em grânulos de peptídeo natriurético atrial (ANP), hipertensão, expressão da renina e ultraestrutura cardíaca em camundongos C57Bl / 6. Camundongos machos adultos jovens foram separados em quatro grupos (n = 12) e alimentados com uma das seguintes dietas por 9 semanas: dieta padrão para roedores (Grupo P), dieta hiperlipídica (Grupo HL), dieta hipersódica (Grupo HS) e dieta hiperlipídica e hipersódica simultaneamente (HL-HS). Foram examinados: alterações no ANP sérico, ultra-estrutura dos cardiomiócitos produtores de ANP, estrutura do ventrículo esquerdo, pressão arterial sanguínea, expressão da renina no rim, taxa de filtração glomerular (TFG), eficiência alimentar, parâmetros lipídicos e glicídicos. Os animais alimentados com dieta hiperlipídica mostraram um pequeno aumento na produção de ANP, discreta hipertrofia ventricular esquerda, aumento da eficiência alimentar, dislipidemia e hiperglicemia. Animais alimentados com dieta hipersódica tiveram um grande aumento na produção de grânulos de ANP e correspondente elevação do seu nível sérico, hipertrofia ventricular esquerda, hipertensão arterial, diminuição dos níveis de renina e aumento da TFG. A combinação das duas dietas (HL-HS) teve um efeito aditivo. A ingestão de uma dieta com alto teor de sal e lipídeos induz alterações ultraestruturais dos cardiomiócitos, aumento da produção de ANP e elevação de seu nível sérico e reduz a quantidade de renina no rim. / To study the effects of a diet rich in salt and/or saturated fat on atrial natriuretic peptide (ANP)-granules, hypertension, renin expression and cardiac structure in C57Bl/6 mice. Young adult male mice were separated into four groups (n=12) and fed one of the following for 9 weeks: standard chow/normal-salt (SC-NS), high-fat chow/normal salt (HF-NS), standard chow/high-salt (SC-HS) and high-fat chow/high-salt (HF-HS). Alterations in the serum ANP, ultrastructural analysis of cardiomyocytes that produce ANP, structural analysis of the left ventricle, blood pressure, renin expression, glomerular filtration rate (GFR), feed efficiency and lipid and glucose parameters were examined. The HF-NS diet showed a small increase in ANP production and left ventricular hypertrophy, increased food efficiency and abnormal lipid and glucose parameters. The SC-HS diet showed a large increase in ANP granules in myocytes and corresponding elevation in ANP serum levels, left ventricular hypertrophy, hypertension, decrease of renin levels and increase in GFR. The combination of the two diets (HF-HS) had an additive effect. The incorporation of a high-fat high-salt diet induced ultrastructural changes in cardiomyocytes, increased the production of ANP and increased its serum level, and reduced the amount of renin in the kidney.
