Stanovení organických sloučenin v dehtu / Determination of organic compounds in the tar

The diploma thesis deals with determination of organic compounds in the tar. It is focussed on polycyclic aromatic hydrocarbons, BTEX and n-alkanes. The tar samples were collected by Faculty of Mechanical Engineering BUT Brno during two days and under diferent conditions of biomass gasification. The samples were filtrated and refined using column chromatography on silicagel sorbent. Gas chromatography with mass detection was chosen to determine polycyclic aromatic hydrocarbons and gas chromatography with flame ionisation detection was chosen to determine BTEX and n-alkanes.

Využití pasivního vzorkování při analýze hormonů v pitných vodách / Application of passive sampling for the analysis of hormones in drinking water

The thesis focuses on the use of passive sampling during an analysis of hormones in drinkable water. In the theoretical part there are described selected passive samplers and the sample of the type of POCIS is described at fuller length. The next chapter engages with steroid hormones and an input of estrogenic hormone into the environment. The thesis contains chapters dealing with possibilities of the determination of hormones in water. It is specialized on the analysis of drinkable and surface water. In the last chapter of the theoretical part there are introduced water works where samples for an experimental assessment were collected from. The experimental part of the thesis presents the specification of the extraction of analytes and an adjustment of used device. In the thesis there are specific calibrations for individual assessment of hormones. The results of the assessment for each collection point are ordered to a well-arranged table. Increased levels of targeted estrogens were not approved in most of sampling points.

Retardéry hoření a jejich průnik do terestrického ekosystému / Flame retardants and their penetration into the terrestrial ecosystem

This diploma thesis was focused on the analysis of four compounds from brominated flame retardants group, namely pentabromoethylbenzene, hexabromocyclododecane, hexabromobenzene and tetrabromobisphenol A. Matrices were represented by four soil samples and four seat of fire samples. In addition, bisphenol A was also determined. First of all, process of extraction of analytes from matrices was optimized. Three different extraction techniques were tested (pressurized solvent extraction, microwave-assisted extraction and ultrasound-assisted extraction). Next step was to verify stability of target compounds in strongly acidic conditions and to optimize clean-up of extract by column chromatography. Gas chromatography with electron capture detection (GC-ECD) was chosen as an appropriate analytical method for the determination of brominated flame retardants. Bisphenol A was determined by gas chromatography with mass spectrometric detection (GC-MS). The derivatization of phenolic analytes before their final analytical determination was also essential.

This diploma thesis deals with Bisphenol A, especially with its impact on the aquatic ecosystem. Information about its properties, production and current use are summarized here. Its harmful impact on the environment, especially on the aquatic ecosystem and the human body is characterized. Also the methods of its degradation within the aquatic environment have been described. A summary of the options of a determination of Bisphenol A in water samples is incorporated and the method of gas chromatography with mass spectrometry (GC/TOF MS) and comprehensive two-dimensional gas chromatography with mass spectrometry (GCxGC/TOF-MS) is compared in the experimental part. Analytical determination precedes the isolation of the analyte from the water samples by solid phase extraction (SPE) using SupelcleanTM ENVITM - 18 and derivatization using the silylation reagent, N,O-bis(trimethylsilyl)trifluoroacetamide (BSTFA). The result of this work is the analysis of a series of real samples from wastewater treatment plants Brno Modřice and Luhačovice by a two-dimensional gas chromatography with mass spectrometry (GCxGC/TOF-MS).

Ceratitis fasciventris, Ceratitis anonae and Ceratitis rosa are polyphagous agricultural pests originating from African continent. Their behaviour is heavily altered by pheromones. Insect chemical communication channels are species-specific, represents taxonomic and reproduction barriers. Taxonomy of this group (so-called Ceratitis FAR complex) is unclear. Therefore new chemical approaches along with genetic tests for identification of entities within the cryptic species FAR complex are being developed. To study multi-component mixtures of male volatiles originating from the three mentioned fruit fly species, comprehensive two-dimensional gas chromatography with time- of-flight mass spectrometer (GC×GC-TOFMS) was used. A number of compounds were identified, out of which 23 were found to be distributed and shared among the studied species, 11 out of these were present in all three species. Analyses of male pheromone volatiles, using gas chromatography combined with electroantennographic detector (GC-EAD) revealed 4 common compounds with antennal activity shared among three studied species: methyl (E)-hex-3-enoate, 6-methylhept-5-en-2- one, linalool, and methyl (2E,6E)-farnesoate. Insects have developed many strategies of defence against predators in the course of evolution. The evolutionarily oldest and most widely used type of defence is chemical defence, followed by acoustic or optical defence. However, many species of insects use simultaneously multiple types of warning signals, which affect different sensory receptors of the given predator. Such a complex method of warning signals is called multimodal method. It may consist of a combination of simultaneous chemical and optical signals, or a combination of acoustic and optical signalling. The combination of chemical and optical signalling used against a predator is probably the most common form of multimodal signalling. The presented work deals with the analysis of biologically active substances, which participate in the defence mechanisms of a widespread species of insects - true bugs (Heteroptera). Pterin derivatives represent a large group of natural compounds derived from pteridin, bicyclic heterocycle, and they are found in virtually all living organisms from bacteria to vertebrates. In insects, they primarily serve as pigments, resulting for example in striking coloration of cuticles of Heteroptera. Dissertation thesis is a 5-publications' collection concerning characterization and application potential of cyclodextrins, polysaccharides and macrocyclic antibiotics based chiral stationary phases. The effects of stationary phase and mobile phase are studied. This approach ensures the complex insight into separation systems studied. Systems with different nature of chiral selector were studied by HPLC. Namely, macrocyclic antibiotics and derivatized polysaccharides were used for experiments. Former ones provided better results for enantioseparation of non-coded amino acids than latter ones. Dynamic coating procedure was used for preparation of a new chiral stationary phase. Characterization of new cationic cyclodextrin based chiral stationary phase was performed. Linear free energy relationship method was used for characterization of two different separation systems, i.e. newly prepared stationary phase and commercially available stationary phase. Based on results obtained, newly prepared stationary phase showed better results for separation of different achiral groups of analysts. New stationary phase prepared by dynamic coating was compared with chromatographic system, in which the chiral selector was used as a mobile phase additive. This thesis is focused on protective and antioxidant properties of bile pigment bilirubin. Bilirubin is a potent scavenger of free radicals and reactive oxygen species generated by increased oxidative stress. Mildly elevated systemic levels of bilirubin are actually associated with lower incidence of oxidative stress-mediated diseases, such as cardiovascular disease, atherosclerosis, diabetes and certain types of cancer. The main aim of this thesis was to prepare bilirubin oxidation products in vitro, characterize their physical-chemical properties and thein determine biological impact.

Liquid crystals are widely used in electronics, medicine and other fields. Analytical separations are important in the development of new liquid crystals to control the purity of synthesized substances. The sample analysis is important for detection of impurities formed during synthesis and for determination of chiral purity of the substance. Liquid crystal-forming substances cannot be separated by capillary zone electrophoresis due to the absence of readily ionizable groups. Electrokinetic chromatography is a method in which a suitable surfactant is added to the background electrolyte. The uncharged substances then interact with the electrically charged surfactant to obtain an effective charge. Separation can occur if they interact differently with the added surfactant. Another problem complicating the analysis is the very low solubility of analytes in water. Separations in this work were therefore carried out under non-aqueous conditions in acetonitrile. However, under these conditions a poor repeatability of the migration times of the substances was observed. Therefore, capillaries with differently coated inner walls were used in subsequent measurements. Surface modification should improve the repeatability of migration times. The theoretical part of the Master´s thesis describes steroids, their features, and the methods for their determination in waters. In experimental part three steroid hormones, namely 1,4-androstadiene-3,17-dione (ADD), 1,4-androstadiene-3-one-17(beta)-ol (boldenone) and 17(beta)-estradiol were selected. The compounds were concentrated from water by LLE and SPE extraction and determined by gas chromatography coupled to mass spectrometry. Several experiments were conducted to assess the recovery of specific extraction for studied compound.
