Využití polarimetrie a refraktometrie ke stanovení koncentrace invertního cukru v invertních sirupech / Use of polarimetry and refractometry for determination of invert sugar concentration in invert syrups

Invert syrup is a liquid sweetener, which is produced by sucrose hydrolysis to form fructose and glucose. Important qualitative parameter of invert syrup is the sucrose inversion – weight percentage of invert sugar (sum of fructose and glucose) in invert syrup dry mass. This diploma thesis presents a possibility of sucrose inversion determination by polarimetry in combination with refractometry in invert syrups of 70–78 % dry mass produced at 80 °C and pH 2,4. New relationship describing the sucrose inversion as a function of polarization and refractometric dry mass was determined. The results of sucrose inversion determined using this new relationship were compared to results obtained by HPLC/RI, which was used as a reference method. The difference between these 2 determination methods was 0,1 ± 0,5 %. The new relationship was detemined with reference to the fact, that under the processing conditions, more glucose than fructose is formed. The fructose : glucose concentration ratio formed under processing conditions was observed to be equal 0,96 ± 0,02.

Možnosti eliminace sulfonamidů z vodního ekosystému / Possibilities of elimination of sulfonamides from the aquatic ecosystem

This diploma thesis focuses on the determination of sulfonamide antibiotics especially the possibility of elimination of these substances from the aquatic ecosystem. Nowadays, environmental contamination of the pharmaceuticals and their residues is a serious concern. Main sources of this contamination are wastewater treatment plants (WWTPs), where these compounds are not effectively removed by contemporary conventional technology. For this reason, new methods are being developed and tested that could eliminate the number of contaminants entering the environment in this way. There is a possibility to use the potential of the enzymatic system of wood-decay fungi, especially white rot fungi. Six representatives of sulfonamide antibiotics were selected and isolated from the aquatic matrix via solid phase extraction. The final identification and quantification method was high performance liquid chromatography with mass spectrometric detection. Monitoring of the concentration level of selected sulfonamide antibiotics at the inflow and effluent at the Brno-Modřice WWTP was carried out weekly. Moreover, the effectiveness of elimination of selected antibiotics from the aquatic ecosystem by the use of Trametes versicolor wood-decay fungi cultured on a suitable carrier was verified.

This diploma thesis deals with the stability and basic properties of combined beverages and fruit concentrates. The theoretical part describes the production and use of fruit concentrates. Furthermore, the work deals with anthocyanic pigments and phenolic substances. The principle and instrumentation of liquid chromatography and electron paramagnetic resonance were described. In the next subchapter, the methods of determination of total anthocyanins and phenolic substances were described. The experimental part of the thesis described the determination of soluble solids, viscosity and antioxidant activity. The total content of phenolic compounds were determined by the Folin-Ciocalteua method and the total anthocyanins were determined by the pH-differential method. In the samples were determined cyanidine-3-glucoside and cyanidin-3-galactoside by liquid chromatography.

Vliv hydroxykyselin na rychlost transdermální absorpce vybraných farmaceuticky aktivních ingrediencí / Influence of hydroxyacids on transdermal absorption of selected active pharmaceutical ingredients

Diploma thesis deals with the experimental study of transdermal transport of individual hydroxyacids and their combination with water-soluble vitamins. During this work the literature review was prepared, which was focused on the use of hydroxyacids in the cosmetic and pharmaceutical industries. At the same time the current state of use of water-soluble vitamins for topical applications was studied. Series of experiments was performed using Franz diffusion cells to simulate transdermal transport in vitro across cutaneous, synthetic and artificial Strat M ® membranes. The methodology of sample preparation and analysis of their transport properties was invented and optimized. The optimized methodology was subsequently used in a series of diffusion experiments in which the penetrated amount of hydroxyacids was analyzed by ion chromatography. The concentration of water-soluble vitamins was determined by HPLC. All examined active substances demonstrate positive penetration properties. The best results were obtained with combination of glycolic acid with pyridoxine.

Studium autenticity kávy různého geografického původu / Studying the authenticity of coffee of various geographical origins

This diploma thesis researches coffee authenticity problematice, mainly focusing on the authenticity of geographic origin. In the theoretical part of this work, botanical classification is described as well as production technology and processes. The work also includes chemical composition of coffee, describing the major components and changes during production phases. It describes major production areas of the world, in terms of general description and brief history. Problematics with coffee fraud and its identification are also described. Theoretical part also includes general geological description of 17 studied coffee growing regions. Experimental part is devoted to trace amount analysis of selected elements and volatile compounds. The element analysis was conducted using mass spectrometry or optical emission spectrometry, volatile compounds were determined using gas chromatography combined with mass spectrometry detection. Results were statistically described and analyzed, resulting in several discrimination models based on geographic origin.

Analýza látek uvolněných z kompozitního zubního materiálu / Analysis of substances released from composite dental material

This master's thesis deals with a present problem of alternative dental fillings, which should replace amalgam fillings. Although there are health concerns about these alternative materials, especially resin composite fillings raise concerns, because they release potentionally harmful substances into the oral cavity. Accordingly even this medical device subjects various tests before releasing to the commercial market and one of these tests was carried out according to ČSN EN ISO 10993 and available scientific literature in the experimental part of this thesis. The amalgam controversy, substances used for manufacturing of resin dental fillings and analytical methods are described in the theoretical part of the thesis. In the experimental part of the thesis, 30 days long cumulative extraction experiments were carried out into various extraction mediums in order to assess suitability of new dental resin filling from ADM, a.s. by comparison with commercially available dental material from company GC EUROPE N.V. These samples were analysed by LC-MS and the amount of eluted substances from dental composites to extraction medium was quantified.

This diploma thesis is focused on the study of structure and physicochemical properties of organic fractions of humic substances, which were obtained by the method of thin-layer chromatography. Humic substances, which were used in this study, were isolated from a sample of Slovak alginite based on the procedure of the International Humic Substances Society (IHSS). The following analytical techniques were selected for the characterization of isolated humic substances: thermogravimetric and elemental analysis, molecular absorption spectroscopy (UV/Vis), Fourier transform infrared spectroscopy (FTIR) and steady-state fluorescence spectroscopy. Each organic fraction of humic substances were characterized by molecular absorption spectroscopy (UV/Vis) and steady-state fluorescence spectroscopy. Using steady-state fluorescence spectroscopy, humic substances were found to be composed of fluorophores of humic and non-humic (protein) character. Organic fractions corresponding to the humic fluorophores were characterized by a higher content of oxygen substituents on the aromatic nukleus, a higher degree of aromaticity and also a higher molecular weight. The aim of this diploma thesis was to design and optimize the process of organic matter fractionation for the purpose of detailed understanding of the structure and properties of humic substances, which were isolated from the sedimentary rock alginite. Based on this, the practical applicability of the thin layer chromatography method to significantly reduce the molecular heterogenity of the studied humic substances was evaluated.

Sledování forem arsenu v potravinách / Monitoring of arsenic forms in foodstuffs

The diploma thesis is dealing with monitoring of arsenic in foodstuffs. The aim of this thesis is the determination of arsenic in samples of fish and rice and the study of forms, in which arsenic occurs, using speciation analysis. Arsenic is known as a toxic element, but its measure of toxicity depends on the chemical form it occurs in. Arsenic is present in the environment naturally or it gets in the environment by human activities. Complete characterization of arsenic compounds is necessary to understand intake, accumulation, transport, detoxification and activation of this element in the natural environment and living systems. The field of arsenic speciation analysis has grown rapidly in recent years, because determination of the total element content is not sufficient in the case of arsenic. Speciation method was done using separation by high performance liquid chromatography and detection by atomic fluorescence spectrometry with hydride generation. Extractable arsenic was present in the form of nontoxic arsenobetaine in all analysed samples of fish. In samples of rice there was confirmed the presence of toxic inorganic species of arsenic, esspecially As (III), but their concentration was at low level.

Hodnocení obsahu glutathionu v rostlinách jako markeru znečištění životního prostředí těžkými kovy / Evaluation of glutathione content in plants as a marker of heavy metals environmental contamination

Dependence of glutathione concentration on the amount of thallium in the plant was studied. Observed plant was maize (Zea mays) which was divided to two parts – root and overground. Two culture procedures were elaborated where seeds and young seedlings were cultivated in a solution of thallium of concentration 0, 1, 3, 5, 8, a 10 µmol/l. Extraction agents used during extraction were phosphate buffer and solution of ascorbic acid. Determination of glutathione was realized by capillary electrophoresis (CE) and high performance liquid chromatography (HPLC). Diode array detector (DAD) was used in both methods. Quantification of the thallium amount in the plant was done by method of inductively coupled plasma-atomic emission spectrometry (ICP-AES).

The aim of this diploma thesis is focused on determination of B vitamin in food especially in malting barley and malt. This diploma thesis is consists of theoretical and experimental parts including the conclusion and supplements. Theoretical part describes thiamine characteristic, chemical structure, chemical and physical properties, function of thiamin in organism, impact it´s insufficiency to the human health, occurrence in common food and methods for it´s determination. Further are describing theoretical principles of high-efficiency liquid chromatography, chromatography on reverse phase, derivatization and ways of validation. In the Experimental section is detailed description of used method for determination of thiamine in barley and malt, fresh mash and beer but also in common food supplements as are B-kompex, Pangamin and Thiamin. Determination was made by high-efficiency liquid chromatography on reverse phase with isocratic elution and fluorescent detection. Next part introduce the results of theses, discussion of experimental work, volume of thiamine in analyzed food and validation parameters of used method. In the conclusion are valorized results of experimental work. In the attachment are graphs of standard curves, chromatogram and calculations of validation parameters. Diploma thesis was realized in laboratory of Chemistry and Technology of Foodstuff, Faculty of chemistry, Brno University of Technology.
